        Glucocorticoid-induced expansion of classical monocytes contributes to bone loss

        Classical monocytes are commonly involved in the innate inflammatory response and are the progenitors of osteoclasts. Excess endogenous glucocorticoids (GCs) can increase the levels of classical monocytes in blood and bone marrow. The role of this cell population in high-dose exogenous GC-induced osteoporosis (GIOP) remains to be elucidated. In this study, GIOP was established in rats and mice by daily methylprednisolone injection, and monocyte subsets were analyzed by flow cytometry. We demonstrated that classical monocytes accumulate in bone marrow during GIOP. Similarly, the monocyte proportion among bone marrow nucleated cells was also increased in patients with steroid treatment history. We sorted classical monocytes and analyzed their transcriptional profile in response to GCs by RNA sequencing. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis showed that classical monocytes isolated from GC-treated rats exhibited osteoclast differentiation potential. Deletion of classical monocytes by clodronate liposome treatment prevented GIOP via inhibition of osteoclastogenesis and restoration of CD31HiendomucinHi vessels. Regarding the molecular mechanism, classical monocytes express high levels of glucocorticoid receptors. In vitro treatment with GCs increased both the percentage and absolute number of monocytes and promoted their proliferation. In summary, classical monocytes mediated GC-induced bone loss and are a potential target for therapeutic intervention in GIOP treatment.

      • High Resolution Image Denoising Method Based on Vector Neighbor Domain

        It is assumed in the traditional total variation(neighbor domain, ND) algorithm that the pixel points are located at the edge and an edge-preserving model is set up. In the algorithm, pixels in flat regions of the image diffuse along the edge direction, leading to insufficient noise suppression and even presence of false edges. To carry on the edge-saving feature of neighbor domain algorithm and to make up its deficiency in omitting the image edge direction, this paper introduces direction neighborhood to the total variation algorithm so that edge points diffuse along the direction neighborhood. It changes the mode where edge points in the traditional ND algorithm diffuse along the multi-neighborhood, maximizing the smoothness along the edge direction and minimizing that at the vertical edge direction. The experimental results show that the image denoising method based on vector neighbor domain effectively addresses the drawbacks existing in the traditional ND algorithm and provides faster convergence efficiency, achieving both denoising and edge-preserving and improving PSNR and visual effects of the image.

      • Image Denoising Based on Surface Design Method and Grid Smoothing

        In order to solve the issues like the feature detection algorithm surface design too sensitive to the presence of noise, this thesis proposes the grid processing algorithm. Firstly, the constrained smoothing model is used to smooth the grid, in which the bound terms are the sparse constraint terms of the error term of l2 norm and l1 norm. In the process of smoothing, the points of the smooth location move less, while the there is a greater movement of feature points. And then through the anglicizing the movement distances the initial feature points are extracted; finally, after the generating of the initial feature points, the feature points are more complete. Experimental results show that: the proposed algorithm can handle the noise of too sensitive.

      • Quantitative Analysis for Risk Management in Flue Gas Desulfurization Engineering

        In China, the Flue Gas Desulfurization (FGD) system gradually becomes a necessary facility in coal-fired power plants for pollution reduction. The evaluation model of investment risk in FGD project was established in this paper. By researching on the probability distribution of uncertain risk variables impacting investment income, the distribution function as well as the eigenvalue of net present value was solved, and the investment risk in the Build-Operate-Transfer project of FGD engineering was analyzed quantificationally based on the method of Monte Carlo simulation. The result showed that there was relatively high risk in FGD project, and then some measures were proposed to reduce the investment risk.

      • Study of Hybrid DNA Physical Mapping Based on Approximation Algorithm with Errors

        A human chromosome is a DNA molecule with approximately 108 base pairs. The techniques developed to date for sequencing are restricted to pieces of DNA with up to tens of thousands of base pairs. This means that when a piece is sequenced, only an extremely small part of a chromosome can be seen. Molecular biologists use special techniques to deal with DNA molecules comparable in size to a chromosome. These techniques enable them to create maps of an entire chromosome or of significant fractions of chromosomes. Computational techniques were studied that could potentially aid biologists in the map-generation process. An algorithm that solves the consecutive 1s problem was studied. Such a problem is a good model of hybridization mapping when there are no errors and when probes are unique. If errors are present, another approach is needed, and the approximation algorithm is a prospective problem solver for hybridization physical mapping of DNA with errors.

        Investigation of synergistic adsorption between methyl orange and Cd(II) from binary mixtures on magnesium hydroxide modified clinoptilolite

        The simultaneous removal of Methyl orange (MO) and Cd2+ (mainly from organo-metallic dyes) onto magnesium hydroxide modified clinoptilolite (MHMC) was described and compared to a single adsorbate situation. The adsorption performance was studied by batch experiments. The adsorption mechanism of MO and Cd2+ on MHMC was investigated. Langmuir and Dubinin-Raduskevich (D-R) isotherm successfully predicted the adsorption of MO and Cd2+ in single and binary systems. Maximum adsorption capacity calculated from Langmuir isotherm equation in single solution for MO and Cd2+ was 0.305 and 0.282mmol/g, respectively. In a binary system of MO/Cd2+, the adsorption capacity for both MO and Cd2+ was higher than in single solutions. The results indicated that the adsorption system of MO/Cd2+ presented a synergistic effect, not competitive adsorption, which suggested that MHMC can be used as an adsorbent for removal of dyes and heavy metal in the multi-solute system.

        HCBP6 upregulates human SREBP1c expression by binding to C/EBPβ-binding site in the SREBP1c promoter

        Sterol regulatory element-binding protein-1c (SREBP1c) plays an important role in triglyceride (TG) homeostasis. Although our previous study showed that hepatitis C virus core-binding protein 6 (HCBP6) regulates SREBP1c expression to maintain intracellular TG homeostasis, the mechanism underlying this regulation is unclear. In the present study, we found that HCBP6 increased intracellular TG levels by upregulating SREBP1c expression. HCBP6 increased SREBP1c transcription by directly binding to the SREBP1c promoter (at the -139- to +359-bp region). Moreover, we observed that HCBP6 interacted with C/EBPβ-binding site in the SREBP1c promoter both in vitro and in vivo. These results indicate that HCBP6 upregulates human SREBP1c expression by binding to the C/EBPβ-binding site in the SREBP1c promoter. [BMB Reports 2018; 51(1): 33-38]

        Prognostic Perspectives of STING and PD-L1 Expression and Correlation with the Prognosis of Epstein-Barr Virus-Associated Gastric Cancers

        Background/Aims: Epstein-Barr virus-associated gastric cancers (EBVaGCs) have unique molecular and clinicopathological characteristics. The cyclic GMP-AMP synthase-stimulator of interferon genes (STING) pathway is recently recognized as the critical innate immunity against pathogens and tumors. STING is also a master regulator in the cancer-immunity cycle and targeting STING could synergize with existing immune-checkpoint therapies. However, the role of STING in GC, especially in EBVaGC, and its correlation with programmed death-ligand 1 (PD-L1) remain largely unclear. Methods: We collected 78 cases of EBVaGCs and 210 cases of EBV-negative GC (EBVnGC) from a total of 1,443 cases of GC analyzed by EBV-encoded small RNA in situ hybridization. We investigated STING and PD-L1 expression and their concomitant prognostic value in EBVaGCs and EBVnGCs using tissue microarray and immunohistochemistry. The effects of STING and PD-L1 expression on the overall survival of patients with EBVaGC or EBVnGC were assessed by univariate and multivariate analysis. Results: We found that both STING and PD-L1 exhibited significantly higher expression in the EBVaGCs than that in the EBVnGCs. The expression of STING was positively correlated with that of PD-L1 in EBVaGCs. Simultaneous negative expression of STING and PD-L1, and positive expression of STING were independent prognostic risk factors for EBVaGC and EBVnGC, respectively. Conclusions: This is the first prognostic retrospective study of STING and PD-L1 expression and the prognosis among EBVaGC and EBVnGC. The expression and prognostic value of STING and PD-L1 are different in the two types of GCs. STING and PD-L1 are promising prognostic biomarkers and therapeutic targets for EBVaGC and EBVnGC.         Efficiently targeted therapy of glioblastoma xenograft via multifunctional biomimetic nanodrugs

        Background: Glioblastoma multiforme (GBM) is a fatal malignant primary brain tumor in adults. The therapeutic efficacy of chemotherapeutic drugs is limited due to the blood-brain barrier (BBB), poor drug targeting, and short biological half-lives. Multifunctional biomimetic nanodrugs have great potential to overcome these limitations of chemotherapeutic drugs. Methods: We synthesized and characterized a biomimetic nanodrug CMS/PEG-DOX-M. The CMS/PEG-DOX-M effectively and rapidly released DOX in U87 MG cells. Cell proliferation and apoptosis assays were examined by the MTT and TUNEL assays. The penetration of nanodrugs through the BBB and anti-tumor efficacy were investigated in the orthotopic glioblastoma xenograft models. Results: We showed that CMS/PEG-DOX-M inhibited cell proliferation of U87 MG cells and effectively induced cell apoptosis of U87 MG cells. Intracranial antitumor experiments showed that free DOX hardly penetrated the BBB, but CMS/PEG-DOX-M effectively reached the orthotopic ntracranial tumor through the BBB and significantly inhibited tumor growth. Immunofluorescence staining of orthotopic tumor tissue sections confirmed that nanodrugs promoted apoptosis of tumor cells. This study developed a multimodal nanodrug treatment system with the enhanced abilities of tumor-targeting, BBB penetration, and cancer-specific accumulation of chemotherapeutic drugs by combining chemotherapy and photothermal therapy. It can be used as a flexible and effective GBM treatment system and it may also be used for the treatment of other central nervous systems (CNS) tumors and extracranial tumors.
