        Effects of Phytosterol Ester on the Fatty Acid Profiles in Rats with Nonalcoholic Fatty Liver Disease

        Both serum and hepatic fatty acid (FA) compositions differ among nonalcoholic hepatic steatosis, nonalcoholic steatohepatitis, and healthy subjects. The severity of the above liver disease is closely associated with the concentration and composition of FAs. Our previous study found that phytosterol ester (PSE) could alleviate hepatic steatosis in nonalcoholic fatty liver disease rats. The aims of this work were to explore the effects of PSE (0.05/100 g·body weight) on FA profiles and the mRNA levels of FA metabolism-related genes. Compared with a high-fat diet alone group, PSE treatment significantly decreased hepatic saturated fatty acid levels (P < .05) and increased monounsaturated fatty acid (especially C16:1 n-7) levels in the liver, serum, and adipose tissue and polyunsaturated fatty acid levels in the serum and liver (P < .05) after 12 weeks of intervention. In particular, PSE treatment increased the level of C22:5 n-3, an FA that was negatively correlated with the degree of hepatic steatosis in the serum, liver, and adipose tissue. The increases in some unsaturated fatty acids are probably related to the upregulation of stearoyl-coenzyme A desaturase-1 and fatty acid desaturase-1.

        일반눈문 : 한족작가 이국문과 조선족작가 이홍규의 동명소설 「개선」 비교연구

        ‘생활에 참여하자’(干預生活)는 1950년대 중기, 중국문단 일각에서 제기한 창작주장으로 당시의 문학 창작에 커다란 영향을 주어, 중국문학사에서 ‘생활 참여’라 일컫는 일련의 문학 작품과 사조를 창출해냈다.‘생활에 참여하자’란 현실에 존재하는 모순이나 문제를 회피하지 말고대담하게 폭로, 비판하며 광범위한 노동대중들이 관심하는 문제를 관심하고 탐구해야 한다는 주장이다.따라서 당시 중국문단에서 가장 관심을 보였던 주제는 당정기관에 존재하는 관료주의에 대한 비판이었다.본고는 1950년대를 빛냈던 한족작가 이국문과 조선족작가 이홍규의 동명소설「개선」을 비교하면서 두 소설의 특징 및 문학사적 의미를 고찰하였다.무엇보다 두 소설은 주제에서 돌파를 보여주었다.두 소설은 진정으로 백성을 위하는 기층간부는 비판받고 강직되고 오히려 일은 하지 않고 위로는 아첨하고 아래로는 내리누르는 이기주의자, 관료주의자들이 득세하는 현실을 고발하면서 당시 당내에 존재하는 좌경사조, 관료주의, 기회주의, 교조주의 등 문제들을 폭로하였다.이국문의 「개선」에서 비극적인 결말은 “사회주의 사회에 비극이 있을 수 없다.”당시 문예이론에 대한 도전이고 돌파이며 이홍규의 「개선」에서 부업과 개인영농을 긍정한 것 등은 1950년대 당시의 시대적 조류나당의 정신과는 어긋나는, 상당히 파격적인 견해와 비판이다.사회를 꿰뚫어보는 예리한 관찰력과 그것을 대담하게 소설화한 두 작가의 용기가돋보인다.따라서 두 소설은 소설 기법에서도 돌파를 보여주었다.이 시기 대다수작품들이 무조건 신형의 노동자, 농민 등 긍정인물을 이상화하면서 평면화, 획일화에서 벗어나지 못하고 있을 때, 이국문의 「개선」과 이홍규의 「개선」에서는 긍정인물이라고 이상화하지 않고 학괴산과 박창길을 입체적으로 그려내었는데 학괴산의 몸에서 중국농민 고유의 인내와 나약성을 엿볼 수 있다면, 박창길의 몸에서는 옳다고 생각하면 물불을 가리지 않고 성급하고 직통배기인 한민족의 성격 특징이 그대로 들어나고 있다.인물 설정에서 이국문의 「개선」은 공회 위원 학괴산과 공회주석, 이홍규의 「개선」은 사 주임 박창길과 현에서 내려온 간부와의 갈등 속에서 상반되는 인간성과 운명을 대조적으로 그리어 두 성격을 부각시키고주제를 심화시키고 있는데 이국문의 「개선」에서는 대조의 수법, 이홍규의 「개선」에서는 오해, 반전의 수법이 더욱 뚜렷하다.이국문의 「개선」은 한차례 공회 위원 재선거, 이홍규의 「개선」은 사주임 재선거를 기본 플롯으로 하고 있는데 구조상 이국문의 「개선」은 이중플롯의 몽타주 기법, 이홍규의 「개선」은 액자 구조의 대화 형식을 취한다.이국문의 「개선」은 비극과 풍자가 결합된 흑색유머, 이홍규의 「개선」은 오해, 해학과 유머가 기본 특징이다. “Interfering the life” was a kind of literature opinion claimed by some writers from the Chinese literary circle in the middle of 1950s, which has risen a huge influence on literary creation. Such kind of opinion has created a large amount of literary works and led a literary trend in the history of Chinese literature. “Interfering the life” means confronting with the realistic conflicts and problems, unafraid of revealing the truth and giving criticism, showing concern about people’s thoughts and opinions. At that time, the topic with the biggest concern was criticism on the bureaucracy existed in the Party and government’s organizations. This research is a comparative study of Han witer Li Guowen and Chinese-Korean writer Lee Hongkyu’s namesake novel Reelection, and the features and value of these two novels in the history of Chinese literature. First, both the two novels have made breakthroughs on topics. In both the novels, grass-root-unit officers who had truly served people has been criticized and demoted, otherwise, the egoists and bureaucrats who had done nothing for people but only knew flattering the superiors and oppressing the subordinates finally reached to power. The two novels also exposed the left-leaning trend, bureaucracy, opportunism and dogmatism in the Party. The tragical ending of Li Guowen’s Re-election was a challenge and breakthrough to the theory of literature and art which said “there should not be any tragedy in socialist society”. The positive opinion on avocation and personal agriculture in Lee Hongkyu’s Re-election, which was contradictory with the trend of 1950s and the policy of the Party, was a criticism breaking the rules. Second, the two novels have made breakthroughs on writing techniques. Most literary works at that time used socialist realistic techniques to create new workers and farmers with positive image, which made characters complanated and homogenized. However, the novels of Re-election of Li Guowen and Lee Hongkyu haven’t created idealized characters, but created protagonists with flesh and blood. Protagonist with strong personality was cubic and true. Readers can find the endurance and cowardice of traditional Chinese farmers on Hao Kuishan while they can also find Park Tsangkil an irritable and candid farmer, which had the typical Korean characteristics. Re-election of Li Guowen was written following the clue of a re-election of the labor union, while Re-election of Lee Hongkyu was describing a re-election of commune director. Structurally, Re-election of Li Guowen was written with montages following with two clues, while Re-election of Lee Hongkyu used dialogues with frame-work structure. When creating characters, Re-election of Li Guowen described the conflict between Hao Kuishan, a committee member of the labor union and the chairperson of the labor union, while Reelection of Lee Hongkyu described conflict between commune director, Park Tsangkil and officers from the county, which described opposite fate and humanity by comparison. The basic feature of Li Guowen’s Re-election was black humor combined by tragedy and irony, while the feature of Lee Hongkyu’s Re-election was misunderstanding, jocularity and humor.

        Multimodal Perturbation and Cluster Pruning Based Selective Ensemble Classifier and Its Iron Industrial Application

        The selective ensemble aims to search the optimal subset balanced accuracy and diversity from the original base classifier set to construct an ensemble classifier with strong generalization performance. A selective ensemble classifier named BRFS-APCSC is proposed in this paper, which realizes the generation and selection of a set of accurate and diverse base classifiers respectively. In the first step, a multimodal perturbation method is introduced to train distinct base classifiers. The method perturbs the sample space by Bootstrap and disturbs the feature space under a newly proposed semi-random feature selection, which is a combination of the core attribute theory and the improved maximum relevance minimum redundancy algorithm. Then, to search the optimal classifier subset, affinity propagation clustering is added to cluster base classifiers in the first step, then the base classifiers are regarded as features so that the improved maximum relevance minimum redundancy algorithm is applied to select parts of base classifiers from each cluster for integration. UCI datasets and an actual dataset of semi-decarbonization are employed to verify the performance of BRFS-APCSC. The experimental results demonstrate that BRFS-APCSC has significantly difference with other selective ensemble methods and improve the classification accuracy.

In this paper a brief review of the cut flower current situation of globe and current status of flower industry of China especially of cut flower in the world is presented. The acreage, yield, potential of cut flower in China along with distribution of major cut flower products and constraint of cut flower also was indicated in this paper was also presented.

        Intestinal segment and vitamin D3 concentration affect gene expression levels of calcium and phosphorus transporters in broiler chickens

        Two experiments were conducted in this research. Experiment 1 investigated the spatial expression characteristics of calcium (Ca) and phosphorus (P) transporters in the duodenum, jejunum, and ileum of 21-day-old broilers provided with adequate nutrient feed. Experiment 2 evaluated the effects of dietary vitamin D3 (VD3) concentration (0, 125, 250, 500, 1,000, and 2,000 IU/kg) on growth performance, bone development, and gene expression levels of intestinal Ca and P transporters in 1–21-day-old broilers provided with the negative control diet without supplemental VD3. Results in experiment 1 showed that the mRNA levels of calcium- binding protein 28-kDa (CaBP-D28k), sodium-calcium exchanger 1 (NCX1), plasma membrane calcium ATPase 1b (PMCA1b), and IIb sodium-phosphate cotransporter (NaPi-IIb) were the highest in the broiler duodenum. By contrast, the mRNA levels of inorganic phosphate transporter 1 (PiT-1) and 2 (PiT-2) were the highest in the ileum. Results in experiment 2 showed that adding 125 IU/kg VD3 increased body weight gain (BWG), feed intake (FI), bone weight, and percentage and weight of Ca and P in the tibia and femur of 1–21-day-old broilers compared with the negative control diet (p < 0.05). The rise in dietary VD3 levels from 125 to 1,000 IU/kg further increased the BWG, FI, and weights of the bone, ash, Ca, and P (p < 0.05). No difference in growth rate and leg bone quality was noted in the broilers provided with 1,000 and 2,000 IU/kg VD3 (p > 0.05). Supplementation with 125–2,000 IU/kg VD3 increased the mRNA abundances of intestinal Ca and P transporters to varying degrees. The mRNA level of CaBP-D28k increased by 536, 1,161, and 28 folds in the duodenum, jejunum, and ileum, respectively, after adding 1,000 IU/kg VD3. The mRNA levels of other Ca and P transporters (PMCA1b, NCX1, NaPi-IIb, PiT-1, and PiT-2) increased by 0.57–1.74 folds by adding 1,000–2,000 IU/kg VD3. These data suggest that intestinal Ca and P transporters are mainly expressed in the duodenum of broilers. Moreover, the addition of VD3 stimulates the two mineral transporter transcription in broiler intestines.

        Clonging and Expression of Heparinase III Gene from Flavobacterium heparinum

        To enhance solubility of heparinase III when expressed in recombinant E. coli, the heparinase III gene (HepC) of 1980 bp from Flavobacterium heparinum was amplified by the PCR method, and then it was in inserted in the prokaryotic expression vector pMAL-c2x with the gene of maltose binding protein (MBP) which was identified by colony PCR, restriction enzyme digestion and gene sequencing. The results showed that the fusion strategy by using maltose binding protein (MBP) was effective to enhance solubility when induced at low temperatures, such as 15°C. By shake flask cultivation, the specific enzyme activity of MBP-hepC fusion protein could reach about 46.41 IU/mg which was higher than previous reported results. By one-­step affinity purification with amylose resin, the MBP-hepC fusion protein was easily purified to more than 95% purifty and the enzyme characteristics were studied. The optimum reaction pH value of MEP-hepC was 7.3. The optimum reaction temperature was 42-48°C, which was different from the native heparinase III from Flavobacterium heparinum. This enzymatic reaction to heparine followed Michaelis-Menten equation. The all above realize the process intergration of the enzyme production, separation, purification and applications in LMWH production for reducing the enzyme cost. The study laid the foundation for industrial production of low molecular weight heparin.

        Age quadratically affects intestinal calcium and phosphorus transporter gene expression in broiler chickens

        Objective: This research aimed to evaluate the effects of age on growth, tibia development, and intestinal calcium (Ca) and phosphorus (P) transporter gene expressions in broiler chickens. Methods: A total of 224 male Arbor Acres broilers were fed with nutrient-adequate diets and reared in eight cages (28 broilers per cage). Eight broilers (one broiler per cage) were selected and killed at 5, 10, 15, 20, 25, 30, 35, and 40 days of age, respectively. Results: Body weight continuously increased with age of broiler chickens from 5 to 40 days. The bone weight, ash weight, diameter, and length of the tibia also increased with broiler age. By contrast, the tibia ash, Ca, and P percentages quadratically changed with age (p<0.001), and the highest values of mineral contents were observed at 20, 25, and 25 days of age, respectively. The mRNA abundances of calcium-binding protein 28-kDa (CaBP-D28k), sodium-calcium exchanger 1 (NCX1), and plasma membrane ATPase 1b (PMCA1b) increased from 5 to 25 days and then decreased up to 40 days. Similar results were noted in the mRNA abundances of IIb sodium-phosphate cotransporter (NaPi-IIb), inorganic phosphate transporter 1 (PiT-1), inorganic phosphate transporter 2 (PiT-2), nuclear vitamin D receptor (nVDR), and membrane vitamin D receptor (mVDR). The mRNA abundances of Ca and P transporters and VDRs were the highest at 25 days of age. Conclusion: These data indicate that age quadratically affects intestinal Ca and P transporter gene expression and mineral absorption capacity in broiler chickens.

        Accurate delivery of pristimerin and paclitaxel by folic acid-linked nano-micelles for enhancing chemosensitivity in cancer therapy

        Chemoresistance remains a huge challenge for effective treatment of non-small cell lung cancer (NSCLC). Previous studies have shown Chinese herbal extracts possess great potential in ameliorating tumor chemoresistance, however, the efficacy is clinically limited mainly because of the poor tumor-targeting and in vivo stability. The construction of nano-delivery systems for herbal extracts has been shown to improve drug targeting, enhance therapeutic efficacy and reduce toxic and side effects. In this study, a folic acid (FA)-modified nano-herb micelle was developed for codelivery of pristimerin (PRI) and paclitaxel (PTX) to enhance chemosensitivity of NSCLC, in which PRI could synergistically enhance PTX-induced growth inhibition of A549 cancer cell. PTX was firstly grafted with the FA-linked polyethylene glycol (PEG) and then encapsulated with PRI to construct the PRI@FA-PEG-PTX (P@FPP) nano-micelles (NMs), which exhibited improved tumor-targeting and in vivo stability. This active-targeting P@FPP NMs displayed excellent tumor-targeting characteristics without obvious toxicity. Moreover, inhibition of tumor growth and metastasis induced by P@FPP NMs were significantly enhanced compared with the combined effects of the two drugs (PRI in combination of PTX), which associated with epithelial mesenchymal transition inhibition to some extent. Overall, this active-targeting NMs provides a versatile nano-herb strategy for improving tumor-targeting of Chinese herbal extracts, which may help in the promotion of enhancing chemosensitivity of NSCLC in clinical applications.
