        Pretreatment Serum Amyloid A and C-reactive Protein Comparing with Epstein-Barr Virus DNA as Prognostic Indicators in Patients with Nasopharyngeal Carcinoma: A Prospective Study

        Purpose The measuring Epstein-Barr virus (EBV) DNA is an important predictor of nasopharyngeal carcinoma (NPC). This study evaluated the predictive value of pretreatment serum amyloid A (SAA) and C-reactive protein (CRP) comparing with EBV DNA in patients with NPC. Materials and Methods In an observational study of 419 non-metastatic NPC patients, we prospectively evaluated the prognostic effects of pretreatment SAA, CRP, and EBV DNA on survival. The primary endpoint was progress-free survival (PFS). Results The median level of SAA and CRP was 4.28 mg/L and 1.88 mg/L, respectively. For the high- SAA group (> 4.28 mg/L) versus the low-SAA ( 4.28 mg/L) group and the high-CRP group (> 1.88 mg/L) versus the low-CRP ( 1.88 mg/L) group, the 5-year PFS was 64.5% versus 73.1% (p=0.013) and 65.2% versus 73.3% (p=0.064), respectively. EBV DNA detection showed a superior predictive result, the 5-year PFS in the EBV DNA  1,500 copies/mL group was obviously different than the EBV DNA < 1,500 copies/mL group (62.2% versus 77.8%, p < 0.001). Multifactorial Cox regression analysis confirmed that in the PFS, the independent prognostic factors were including EBV DNA (hazard ratio [HR], 1.788; p=0.009), tumour stage (HR, 1.903; p=0.021), and node stage (HR, 1.498; p=0.049), but the SAA and CRP were not included in the independent prognostic factors. Conclusion The results of SAA and CRP had a certain relationship with the prognosis of NPC, and the prognosis of patients with high level of SAA and CRP were poor. However, the predictive ability of SAA and CRP was lower than that of EBV DNA.

        Enhanced delivery of liposomes to lung tumor through targeting interleukin-4 receptor on both tumor cells and tumor endothelial cells

        A growing body of evidence suggests that pathological lesions express tissue-specific molecular targets or biomarkers within the tissue. Interleukin-4 receptor (IL-4R) is overexpressed in many types of cancer cells, including lung cancer. Here we investigated the properties of IL-4R-binding peptide-1 (IL4RPep-1), a CRKRLDRNC peptide, and its ability to target the delivery of liposomes to lung tumor. IL4RPep-1 preferentially bound to H226 lung tumor cells which express higher levers of IL-4R compared to H460 lung tumor cells which express less IL-4R. Mutational analysis revealed that C1, R2, and R4 residues of IL4RPep-1 were the key binding determinants. IL4RPep-1-labeled liposomes containing doxorubicin were more efficiently internalized in H226 cells and effectively delivered doxorubicin into the cells compared to unlabeled liposomes. In vivo fluorescence imaging of nude mice subcutaneously xenotransplanted with H226 tumor cells indicated that IL4RPep-1-labeled liposomes accumulate more efficiently in the tumor and inhibit tumor growth more effectively compared to unlabeled liposomes. Interestingly, expression of IL-4R was high in vascular endothelial cells of tumor, while little was detected in vascular endothelial cells of control organs including the liver. IL-4R expression in cultured human vascular endothelial cells was also up-regulated when activated by a pro-inflammatory cytokine tumor necrosis factor-α. Moreover, the up-regulation of IL-4R expression was observed in primary human lung cancer tissues. These results indicate that IL-4R-targeting nanocarriers may be a useful strategy to enhance drug delivery through the recognition of IL-4R in both tumor cells and tumor endothelial cells.

        Linolenic Acid in Association with Malate or Fumarate Increased CLA Production and Reduced Methane Generation by Rumen Microbes

        An in vitro study was conducted to investigate the effect of malate or fumarate on fermentation characteristics, and production of conjugated linoleic acid (CLA) and methane ($CH_4$) by rumen microbes when incubated with linolenic acid (${\alpha}-C_{18:3}$). Sixty milligrams of ${\alpha}-C_{18:3}$ alone (LNA), or ${\alpha}-C_{18:3}$ with 24 mM malic acid (M-LNA) or ${\alpha}-C_{18:3}$ with 24 mM fumaric acid (F-LNA) were added to the 150 ml culture solution consisting of 75 ml strained rumen fluid and 75ml McDougall's artificial saliva. Culture solution for incubation was also made without malate, fumarate and ${\alpha}-C_{18:3}$ (Control). Two grams of feed consisting of 70% concentrate and 30% ground alfalfa (DM basis) were also added to the culture solution of each treatment. In vitro incubation was made anaerobically in a shaking incubator up to 12 h at $39^{\circ}C$. Supplementation of malate (M-LNA) or fumarate (F-LNA) increased pH at 6 h (p<0.01) and 12 h (p<0.001) incubation times compared to control and linolenic acid (LNA) treatments. Both malate and fumarate did not influence the ammonia-N concentration. Concentration of total VFA in culture solution was higher for M-LNA and F-LNA supplementation than for control and LNA treatments from 6 h (p<0.040) to 12 h (p<0.027) incubation times, but was not different between malate and fumarate for all incubation times. Molar proportion of $C_3$ was increased by F-LNA and M-LNA supplementation from 6 h (p<0.0001) to 12 h (p<0.004) incubation times compared to control and LNA treatments. No differences in $C_{3}$ proportion, however, were observed between M-LNA and F-LNA treatments. Accumulated total gas production for 12h incubation was increased (p<0.0002) by M-LNA or F-LNA compared to control or LNA treatment. Accumulated $CH_4$ production for 12 h incubation, however, was greatly reduced (p<0.0002) by supplementing malate or fumarate compared to the control, and its production from M-LNA or F-LNA treatment was smaller than that from LNA treatment. Methane production from LNA, M-LNA or F-LNA treatment was steadily lower (p<0.01 - p<0.001) from 3 h incubation time than that from the control, and was also lower for M-LNA or F-LNA treatment at incubation times of 6 h (p<0.01) and 9 h (p<0.001) than for LNA treatment. Methane production from LNA, however, was reduced (p<0.01 - p<0.001) from 3 h to 9 h incubation times compared to the control. Both malate and fumarate increased concentration of trans11-$C_{18:1}$ from 3 h to 12 h incubation (p<0.01), cis9,trans11-CLA up to 6 h incubation (p<0.01 - p<0.01), trans10,cis12-CLA at 3 h (p<0.05) and 12 h (p<0.01), and total CLA for all incubation times (p<0.05) compared to corresponding values for the ${\alpha}-C_{18:3}$ supplemented treatment (LNA). In conclusion, malate and fumarate rechanneled the metabolic $H_2 pathway to production of propionate and CLA, and depressed the process of biohydrogenation and methane generation. Linolenic acid alone would also be one of the optimistic alternatives to suppress the $CH_4$ generation.

        Identification and evaluation of potent Middle East respiratory syndrome coronavirus (MERS-CoV) 3CL<sup>Pro</sup> inhibitors

        <P>Middle East respiratory syndrome coronavirus (MERS-CoV) causes severe acute respiratory illness with fever, cough and shortness of breath. Up to date, it has resulted in 1826 human infections, including 649 deaths. Analogous to picornavirus 3C protease (3C(PrO)), 3C-like protease (3CL(PrO)) is critical for initiation of the MERS-CoV replication cycle and is thus regarded as a validated drug target. As presented here, our peptidomimetic inhibitors of enterovirus 3C(PrO) (6b. 6c and 6d) inhibited 3CLPr0 of MERS-CoV and severe acute respiratory syndrome coronavirus (SARS-CoV) with IC50 values ranging from 1.7 to 4.7 M and from 0.2 to 0.7 M, respectively. In MERS-CoV-infected cells, the inhibitors showed antiviral activity with EC50 values ranging from 0.6 to 1.4 1.1M, by downregulating the viral protein production in cells as well as reducing secretion of infectious viral particles into culture supernatants. They also suppressed other cc and 8-CoVs from human and feline origin. These compounds exhibited good selectivity index (over 70 against MERS-CoV) and could lead to the development of broad-spectrum antiviral drugs against emerging CoVs and picornaviruses.         ANALYSIS OF BUS ROLLOVER PROTECTION UNDER LEGISLATED STANDARDS USING LS-DYNA SOFTWARE SIMULATION TECHNIQUES

        A bus rollover is one of the worst vehicle accidents that can occur. Because of the large numbers of passengers, the casualties in a bus rollover are often high and severe. The compliance with rollover safety standards for buses and coaches is mandated by law. This paper presents a comparative analysis of the physical meanings of regulation number 66 of the Economic Commission for Europe (ECE R66) and standard number 220 of the American Federal Motor Vehicle Safety Standards (FMVSS 220). This comparison was carried out using a LS-DYNA finite-element analysis. After performing a comparative analysis following ECE R66 and FMVSS 220 assessments, the investigation further demonstrated the distortion configuration of the vehicle superstructure through the absorbed energy and its distribution over the vehicle and in sections of vehicle superstructure as well as the violation of the passenger compartment under the rollover testing conditions of both ECE R66 and FMVSS 220. Great differences were found between ECE R66 and FMVSS 220 in distortion configuration, reflecting differences in capability and rollover testing conditions. These findings provide a means of evaluating bus superstructure strength and provide guidelines useful in the assessment of regulations applied to the evaluation of bus rollover strength.

      • Observation of the topological surface state in the nonsymmorphic topological insulator KHgSb

        <P>Topological insulators represent unusual topological quantum states, typically with gapped bulk band structure but gapless surface Dirac fermions protected by time-reversal symmetry. Recently, a distinct kind of topological insulator resulting from nonsymmorphic crystalline symmetry was proposed in the KHgX (X = As, Sb, Bi) compounds. Unlike regular topological crystalline insulators, the nonsymmorphic glide-reflection symmetry in KHgX guarantees the appearance of an exotic surface fermion with hourglass shape dispersion (where two pairs of branches switch their partners) residing on its (010) side surface, contrasting to the usual two-dimensional Dirac fermion form. Here, by using high-resolution angle-resolved photoemission spectroscopy, we systematically investigate the electronic structures of KHgSb on both (001) and (010) surfaces and reveal the unique in-gap surface states on the (010) surface with delicate dispersion consistent with the 'hourglass Fermion' recently proposed. Our experiment strongly supports that KHgSb is a nonsymmorphic topological crystalline insulator with hourglass fermions, which serves as an important step to the discovery of unique topological quantum materials and exotic fermions protected by nonsymmorphic crystalline symmetry.</P>

        OPTIMIZATION OF BUS ROLLOVER STRENGTH BY CONSIDERATION OF THE ENERGY ABSORPTION ABILITY

        Buses are an integral part of the national transportation system of each country. A rollover event is one of the most important hazards that concerns the safety of the passengers and the crew in a bus. In the past, it was observed after the accident that the deforming superstructure seriously threatens the lives of the passengers. Thus, the stiffness of the bus frame is the first thing that needs to be considered. The unfortunate side of strengthening the bus superstructure is that it usually causes the bus weight to increase. This paper presents an efficient and robust analysis method with which to design the bus superstructure for a reduction in occupant injuries from rollover accidents while the weight of the strengthened bus is maintained at the same level. First, the absorbed energy of the bus frame and its components during a rollover were investigated by using a LS-DYNA numerical study. The highest energy absorption region, which is the side section of the bus frame, was found and focused on for the investigation of a means to re-distribute the energy-absorption ability of the side frame component. Then the thickness parameters that were obtained from the re-distribution of the energy-absorption ability were used in the analysis to optimize the design. Finally, a prototype of the bus with a reasonable thickness for the window pillars and the side wall bars, which was based on the optimized parameters, was verified to ensure it satisfied ECE R66. In this paper, an effective usage of materials and an efficient and robust analysis method were presented to design the bus superstructure. Although the optimization process for increasing the stiffness is simple, this study improves the upper displacement by 39.9% and the lower displacement by 49.3% (versus the bus survivor space) while maintaining the bus weight at the existing level.

        Measurements of the Quantum Lifetime in Semiconductor Heterostructures by Using Microwave-Detected SdH Oscillations

        Low-temperature magnetotransport measurements were performed on Al0:25Ga0:75As/In0:20 Ga0:80As/Al0:25Ga0:75As and Al0:25Ga0:75N/GaN two-dimensional electron systems. By studying the fast Fourier transform spectrum of both conventional Shubnikov-de Haas (SdH) oscillations and microwave-modulated SdH data, we were able to measure both the temperature (T)-independent and the T-dependent parts of the quantum lifetime. Our experimental results indicate that at low T, a T-independent background, such as alloy disorder and interface roughness scattering in nitride-based semiconductors, are more dominant than those in GaAs-like heterostructures.
