      • AFB₁ 대사에서 phloretion의 이중 활성 효과

        Aflatoxm B₁(AFB₁) is a potent hepatocarcinogen in experimental animals and a hazard to human health in several parts of the world. AFB₁ is activated to its ultimate carcinogenic intermediate, AFB₁-8,9-epoxide, by cytochrome P450(CYP) 1A2 and CYP3A4 in human liver and the intermediate is decomposed by several glutathione S-transferase(GST) including GSTA2, GSTM1 and GSTP1. In this study, we investigated the effects of phloretin on the enzyme systems which are involved in the activation and detoxification of AFB₁. The metabolic intermediate of AFB₁ was measured with HPLC. We found that phloretin could strongly inhibit the activities of CYP 3A4 and CYP1A2 in a dose dependent manner. Phloretin induced the antioxidant-response element(ARE)-mediated gene expression, including GSTs. The expressions of GSTA2, T1, M1, and GSTP1 were induced by 10μM phloretin. The decomposition of AFB₁-8,9-epoxide was measured with GSH conjugating activity of the epoxide. The rate was increased to 1.5 fold when HepG2 cells were treated by 10μM phloretin for 12h. In the mean while, the total GST activitives toward CDNB in HepG2 cells were not changed by the treatment with phloretin. The results demonstrate that phloretin has strong chemopreventive effects against AFB₁ toxicity through the inhibition of AFB₁ activation and induction of GSTs.

        A new ferulic acid ester, 1'-methyl-2'-hydroxyethyl ferulate (1), together with methylcaffeate (2), 4-hydroxy cinnamic acid (3), ferulic acid (4), caffeic acid (5), diosmetin (6), luteolin (7), 5,3',4'-trihydroxy-3,7-dimethoxyflavone (8), eriodictyol (9), kaempferol (10), quercetin (11), acacetin-7-O-glcopyranoside (12), 4-($\beta$-glucopyranosyloxy) benzoic acid (13), luteolin-7-O-(6"-feruloyl) glucopyranoside (14), luteolin-7-O-glucopyranoside (15), kaempferide-3-O-rhamnopyranoside (16), quercitrin (17), kaempferol-3-O-glucopyranoside (18), prunasin (19), quercetin-7-O-glucopyranoside (20), quercetin-3-O-glucopyranoside (21), plantaginin (22), linarin (23), luteolin-7-O-rutinoside (24), and chlorogenic acid (25) were isolated from the aerial parts of Dacocephalum peregrinum. The structure of 1 was elucidated on the basis of spectroscopic and HR-ESI-MS analyses. In addition, compound 1 exhibited mild inhibitory effect on NO production in LPS-stimulated RAW264.7 cells.

      • Pre-treatment Metabolic Tumor Volume and Total Lesion Glycolysis are Useful Prognostic Factors for Esophageal Squamous Cell Cancer Patients

        Objectives: To study application of the maximum standardized uptake value (SUVmax), metabolic tumor volume (MTV) and total lesion glycolysis (TLG) with $^{18}F$-FDG PET/CT for predicting prognosis of esophageal squamous cell cancer (ESC) patients. Methods: Eighty-six patients with ESC staged from I to IV were prospectively enrolled. Cisplatin-based chemoradiotherapy (CCRT) or palliative chemoradiotherapy were the main treatment methods and none received surgery. $^{18}F$-FDG PET/CT scans were performed before the treatment. SUVmax, MTV, and TLG were measured for the primary esophageal lesion and regional lymph nodes. Receiver operating characteristic curves (ROCs) were generated to calculate the P value of the predictive ability and the optimal threshold. Results: MTV and TLG proved to be good indexes in the prediction of outcome for the ESC patients. An MTV value of 15.6 ml and a TLG value of 183.5 were optimal threshold to predict the overall survival (OS). The areas under the curve (AUC) for MTV and TLG were 0.74 and 0.70, respectively. Kaplan-Meier analysis showed an MTV less than 15.6 ml and a TLG less than 183.5 to indicate good media survival time (p value <0.05). In the stage III-IV patient group, MTV could better predict the OS (P < 0.001), with a sensitivity and specificity of 0.80 and 0.67, respectively. Conclusions: Pre-treatment MTV and TLG are useful prognostic factors in nonsurgical ESC.

        Study of compensatory growth based on different nutrition conditions of Bombyx mori

        Organisms achieve compensatory growth after a period of nutrient restriction followed by recovering the nutrient status. However, the underlying mechanisms associated with such growth acceleration remain unclear. The silkworm Bombyx mori is a lepidopteran model insect. This study aimed to investigate the physiological characteristics and the underlying mechanisms in B. mori fed on mulberry leaves (MG), artificial diet (AG), and artificial diet + mulberry leaves (AG-MG), respectively. Silkworms in AG-MG which fed on artificial diet from 1st to 3rd instars followed by feeding on mulberry leaves from 4th to 5th instars exhibited a higher weight gain rate than that in MG and AG, indicating that compensatory growth occurred as a result of the switch in the silkworm food regime. Trypsin and lipase activities of silkworms in AG-MG were shown to be up-regulated at 72 h after changing food. Digital gene expression profiling (DGE) analysis revealed that genes related to metabolism and development in silkworm midguts were differentially expressed. The quantitative real-time PCR (qRT-PCR) re sults showed that the expression levels of IIS/PI3K-AKT pathway genes including INR, IRS, AKT, PI3K60 and PI3K110 of silkworms in AG were down-regulated compared with that in MG at 0 h. Whereas AKT, PI3K and PI3K60 of silkworms were significantly increased by 1.68-, 1.49-, 1.67-fold, respectively, at 72 h after switched to mulberry leaves than the same instar fed on artificial diet. PDK’s expression of silkworms in AG was higher than that in other two groups at each timepoint. Compared with MG and AG, PTEN and IRS were down-regulated at 48 and 72 h in AG-MG. Collectively, these results indicate that compensatory growth in B. mori is regulated by IIS/ PI3K-AKT signaling pathway.

      • Knowledge Acquisition Approach Based on Rough Set in Online Aided Decision System for Food Processing Quality and Safety

        For the problem that the use effect of food processing information system is poor due to absence of knowledge acquisition measures and knowledge self-updating function, a knowledge acquisition approach based on rough set is put forward. First, the approach establishes a set of predicted samples for the relationship between food processing parameters and product quality; then uses the discretization of continuous attribute, attribute reduction and rule extraction algorithm of rough set to acquire automatically predicted knowledge from a large number of predicted sample sets, and then saves the predicted knowledge in the knowledge base of expert system; finally, realizes the extraction of knowledge of food processing process based on the inference engine, which greatly enhances the effectiveness and practicality of the acquired knowledge in online aided decision system of the food processing quality and safety.

        Isoindolin-1-ones from the stems of Nicotiana tabacum and their antiviral activities

        In previous studies, several isoindolin-1-oneanalogs that exhibited signifi cant anti-tobacco mosaic virus(anti-TMV) activities were isolated from Nicotiana tabacum . Since gene-editing mutants provide a new sample for thediscovery of active metabolites, we focused on the stems ofYN-18–23 (a mutant N. tabacum for gene editing with thealkaloid metabolic pathway cultivated by Yunnan TobaccoCompany), which led to the isolation of four new ( 1–4 )and four known ( 5–8 ) isoindolin-1-ones. To the best of ourknowledge, nicindole C ( 3 ) is the fi rst subclass of isoindolin-1-one bearing a pentacyclic ketone, while nicindole D ( 4 )is the fi rst example of isoindolin-1-one bearing a methylpyridin-2-(1 H )-one moiety. Compounds 1–4 were testedfor their anti-TMV activities, and the results revealed thatcompounds 1 , 3 , and 4 exhibited high anti-TMV activities atconcentrations of 20 μM with inhibition rates of 48.6, 42.8,and 71.5%, respectively. These rates are higher than the inhibitionrate of the positive control (33.2%). The mechanisticstudy of compound 4 , which had the highest anti-TMV activityrevealed that increased potentiation of defense-related enzyme activities and downregulation of expression of theNtHsp70 protein may induce resistance in tobacco againstthe viral pathogen TMV. Molecular docking studies alsorevealed that the isoindolin-1-one substructure is fundamentalfor anti-TMV activity. The methyl-pyridin-2-(1 H )-onemoiety in compound 4 and the 2-oxopropyl groups in compounds1 and 3 at the N -2 position may increase inhibitoryactivities. This study of the structure–activity relationshipis helpful for fi nding new anti-TMV activity inhibitors. Tostudy whether the isoindolin-1-ones have broader antiviralactivities, compounds 1–4 were also tested for their antirotavirusactivities. Compound 4 exhibited high anti-rotavirusactivity with a therapeutic index (TI) value of 20.7. This TI value is close to that of the positive control (20.2).

        Overexpressing <i>IbCBF3</i> increases low temperature and drought stress tolerance in transgenic sweetpotato

        <P><B>Abstract</B></P> <P>Dehydration-responsive element-binding/C-repeat-binding factor (DREB/CBF) proteins regulate the transcription of genes involved in cold acclimation in several species. However, little is known about the physiological functions of CBF proteins in the low temperature-sensitive crop sweetpotato. We previously reported that the <I>DREB1/CBF-like</I> sweetpotato gene <I>SwDREB1/IbCBF3</I> is involved in responses to diverse abiotic stresses. In this study, we confirmed that IbCBF3 is localized to the nucleus and binds to the C-repeat/dehydration-responsive elements (CRT/DRE) in the promoters of cold-regulated (<I>COR</I>) genes. We generated transgenic sweetpotato plants overexpressing <I>IbCBF3</I> under the control of the CaMV 35S promoter (referred to as SC plants) and evaluated their responses to various abiotic stresses. <I>IbCBF3</I> expression was dramatically induced by cold and drought but much less strongly induced by high salinity and ABA. We further characterized two SC lines (SC3 and SC6) with high levels of <I>IbCBF3</I> transcript. The SC plants displayed enhanced tolerance to cold, drought, and oxidative stress on the whole-plant level. Under cold stress treatment (4 °C for 48 h), severe wilting and chilling injury were observed in the leaves of wild-type (WT) plants, whereas SC plants were not affected by cold stress. In addition, the <I>COR</I> genes were significantly upregulated in SC plants compared with the WT. The SC plants also showed significantly higher tolerance to drought stress than the WT, which was associated with higher photosynthesis efficiency and lower hydrogen peroxide levels. These results indicate that IbCBF3 is a functional transcription factor involved in the responses to various abiotic stresses in sweetpotato.</P> <P><B>Highlights</B></P> <P> <UL> <LI> IbCBF3 binds to CRT/DRE in the promoters of <I>COR</I> genes. </LI> <LI> Overexpression of <I>IbCBF3</I> increases cold and drought stress tolerance in sweetpotato. </LI> <LI> <I>IbCOR</I> expression is higher in <I>IbCBF3</I>-overexpressing sweetpotato than in the WT. </LI> </UL> </P>

      • Protective Effects of Scutellaria barbata Against Rat Liver Tumorigenesis

        Scutellaria barbata D. Don (S. barbata), a traditional Chinese medicine, is used to treat cancers, inflammation, and urinary diseases. This study aimed to determine any protective effects of S. barbata crude extract (CE-SB) against rat liver tumorigenesis induced by diethylnitrosamine (DENA). Liver malfunction indices in serum were measured by biochemical examination. Hematoxylin and eosin staining was performed to examine liver pathology. Contents of malondialdehyde (MDA) and superoxide dismutase (SOD) were measured in liver homogenates to evaluate oxidative stress. The levels of liver malfunction indices in the CE-SB groups, especially in the CE-SB high dose group, were lower than that of the model group (P<0.05). The results from histological examination indicated that the number of liver nodules in the CE-SB groups decreased compared with the model group (P<0.05). Content of MDA determined in liver was significantly decreased, and level of SOD elevated by CE-SB. CE-SB can inhibit experimental liver tumorigenesis and relieve hepatic injury in rats.

        Fabrication and magnetic properties of hexagonal BaFe12O19 ferrite obtained by magnetic-field-assisted hydrothermal process

        High magnetic field effects on the microstructure and magnetic properties of BaFe12O19 hexaferrites synthesized hydrothermal method have been investigated. The obtained results indicate that the lattice constant decreases gradually as the magnetic field strength increases, which may be attributed to the lattice distortion resulted from the high magnetic field. Polycrystalline BaFe12O19 samples prepared under magnetic field strength at zero and 5 T are single phase. It is found that application of external magnetic field during synthesis can induce orientated growth of the hexaferrite crystals along the easy magnetic axis. The magnetic properties can be effectively regulated by an application of high magnetic fields. It is observed that the BaFe12O19 prepared under a 5 T magnetic field exhibits a higher room-temperature saturation magnetization (66.3 emu/g) than that of the sample (43.6 emu/g) obtained without magnetic field. The results can be explained as the enhanced crystalline, improvement of Fe3+ ions occupancy and the oriented growth induced by the external magnetic field. The growing orientation of particles gives rise to increased coercivity due to the enhancement in shape anisotropy. It is expected that an application of magnetic field during the formation of magnetic nanoparticles could be a promising technique to modify magnetic properties with excellent performance.
