        Effect of aberrantly methylated androgen receptor target gene PCDH7 on the development of androgen-independent prostate cancer cells

        Background Androgen-independent prostate cancer (AIPC) is an extremely malignant tumor developed from the androgen dependent (ADPC). However, the mechanism of transition process from ADPC to AIPC remains unknown. Objective Here we aimed to identify the androgen receptor (AR) target gene and its roles in AIPC. Methods Target genes of AR were identified by ChIP-seq in AIPC cells. AR target gene PCDH7 was detected by real time PCR and western blot. Methylation of PCDH7 was measured by bisulfite sequencing and bisulfite amplicon sequencing. Cell growth, invasion and apoptosis were measured by CCK-8, transwell and flow cytometry, respectively. Results AR was significantly enriched in the upstream of PCDH7 gene. The expression of PCDH7 was significantly decreased, while the methylation of PCDH7 was increased in the AIPC cells compared to the ADPC cells. DNA methyltransferase inhibitor significantly suppressed the methylation and increased the mRNA and protein level of PCDH7. Moreover, overexpression of DNMT1 remarkably reduced the mRNA and protein level of PCDH7. DNA methyltransferase inhibitor decreased the cell growth and invasion while promote the cell apoptosis in the AIPC cells. AR significantly target PCDH7, whose hypermethylation may repress cell growth and invasion, and promote apoptosis in AIPC. Conclusions This study might provide a novel potential target for the treatment of AIPC.

        High expression of HOXC6 predicts a poor prognosis and induces proliferation and inflammation in multiple myeloma cells

        Background Multiple myeloma (MM) is a common blood system malignance accompanied by monoclonal plasma cell hyperplasia. Homeobox C6 (HOXC6) acts as an oncogene in various cancers, but its function on MM is elusive. Objective The role of HOXC6 on MM development was clarified in this study. Methods HOXC6 expression and its clinical significance were determined in the peripheral blood samples collected from forty MM patients and thirty healthy adult volunteers. The overall survival was evaluated by Kaplan-Meier analysis with the log-rank test. Cell viability, proliferation and apoptosis were measured by CCK-8, EdU assay and Flow cytometry in U266 and MM.1R cells. Tumor growth was estimated by a xenograft assay. The apoptosis of tumor tissues was evaluated using TUNEL staining. The protein level in tissues was tested by immunohistochemistry. Results The HOXC6 expression was elevated in MM and high HOXC6 level was associated with the poor overall survival of MM. Besides, the HOXC6 expression was associated with hemoglobin level and ISS stage. Furthermore, silencing HOXC6 suppressed cell proliferation, induced cell apoptosis, and restrained the secretion of inflammatory factors (TNF-α, IL-6, and IL-8) in MM cells through inactivating the NF-κB pathway. Moreover, silencing HOXC6 suppressed the tumor growth of MM, the inflammatory factors levels, and the activation of NF-κB pathway but enhanced apoptosis in vivo. Conclusion HOXC6 was elevated in MM and associated with poor survival. Knockdown of HOXC6 suppressed proliferation, inflammation and tumorigenicity of MM cells via inactivating the NF-κB pathway. HOXC6 may be a meaningful target for MM therapy.

        Comparison between laparoscopic and abdominal radical hysterectomy for stage IB1 and tumor size <2 cm cervical cancer with visible or invisible tumors: a multicentre retrospective study

        Objective: To compare 5-year disease-free survival (DFS) and overall survival (OS) rates oflaparoscopic radical hysterectomy (LRH) and abdominal radical hysterectomy (ARH) forstage IB1 and tumor size <2 cm with visible or invisible tumors. Methods: We retrospectively compared the oncological outcomes of 1,484 cervical cancerpatients with IB1 and tumor size <2 cm on final pathology, who received ARH (n=899) or LRH(n=585) between January 2004 and December 2016. Patients were divided into visible tumorsubgroup (ARH: n=668, LRH: n=444) and invisible tumor subgroup (ARH: n=231, LRH:n=141) according to tumor type. Results: LRH and ARH showed similar 5-year DFS and OS rates (93.3% vs. 93.1%, p=0.997;96.2% vs. 97.5%, p=0.351) in total study population. LRH was not associated with worse5-year DFS rate (hazard ratio [HR]=0.96; 95% confidence interval [CI]=0.58–1.58; p=0.871)or OS rate (HR=1.37; 95% CI=0.65–2.89; p=0.409) by multivariable analysis. In the visibletumor subgroups, LRH and ARH showed similar 5-year DFS and OS rates (91.9% vs. 91.9%,p=0.933; 95.0% vs. 96.9%, p=0.276), and LRH was not associated with worse 5-year DFS orOS rate (p=0.804, p=0.324). In the invisible tumor subgroups, LRH and ARH also showedsimilar 5-year DFS and OS rates (97.3% vs. 97.1%, p=0.815; 100% vs. 99.5%, p=0.449), andLRH was not associated with worse 5-year DFS rate (p=0.723). Conclusions: Among patients with stage IB1 and tumor size <2 cm, whether the tumor isvisible or not, the oncological outcomes of LRH and ARH among cervical cancer patients arecomparable. This suggests that LRH may be suitable for stage IB1 and tumor size <2 cm withvisible or invisible tumors.Trial Registration: International Clinical Trials Registry Platform Identifier: CHiCTR180017778

      • Multi-focus Image Fusion with Cartoon-Texture Image Decomposition

        Multi-focus image fusion can fuse multiple source images with different focus settings into a single image that appears sharper. How to effectively and completely represent the source images is the key to multi-focus image fusion. A multi-component fusion method is proposed for multi-focus image fusion. The registered source images are decomposed into cartoon and texture components by cartoon-texture image decomposition. The salient features are selected from the cartoon and texture components respectively to form a composite feature space. The local features that represent the salient information of the source images are integrated to construct the fused image. According to the visual perception and objective evaluations on the fused images, the proposed method works better in extracting the focused regions and improving the fusion quality, compared with the other existing single-component fusion methods.

        Geochronology, geochemistry, Hf isotope, and electron probe analysis of the diorite in the Jianhei Mountain, East Kunlun Orogenic Belt, China, and their geological significance

        The spatial distribution of flow units and reductant within a sandstone uranium reservoir is very beneficial for exploring the uranium mineralization mechanism; however, such studies have been almost neglected. As such, this paper aims to provide the sedimentological interpretation and heterogeneity models of the sandstone of the Middle Jurassic Zhiluo Formation at Shiwan outcrop area, in the eastern Ordos Basin. The outcrops are sandstone-dominated subaqueous distributary channel successions, in which five distinct architectural elements are identified and interpreted: channel units (CU), downstream accretion elements (DA), lateral accretion elements (LA), sandy bedforms (SB) and no-channelized fine-grained sediments (NFS). Within the sandstone succession, six flow units are present in the middle and lower parts of CU, or the lower parts of DA and SB elements, with their geometry to be controlled by the fourth- or third-order bounding surfaces. Moreover, the carbonaceous debris-rich zones (CDZ) are places with the highest abundance of reductant and preserve exclusively upon the basal erosional surfaces of CU and DA elements. Based on those results, a prediction model is established and shows that the formation of interlayer oxidation zone is strongly selective and confined within the CU and DA elements. The favorable site of uranium mineralization is preferentially distributed in the transition zone between the front of flow unit and the CDZ.

        Optimal of black anti-static AZO/Al2O3-SiO2-VxOy coatings for spacecraft stray light on high silicon aluminum alloys

        It is still challenging to prepare an anti-static and anti-stray astigmatism coating on the high silicon aluminum alloy (>50 wt% Si, Al-50Si) for space material under extreme environments. To address these issues, the black anti-stray light coating with enhanced conductivity on the chemical etching surface of high-silicon Al-70Si alloy were successfully fabricated by the combination of plasma electrolytic oxidation (PEO) technology and deposited aluminum doped zinc oxide (AZO) transparent conductive film by magnetron sputtering technology. The microstructures, phase, elemental compositions, light absorption performance and electrical resistivity of coatings were tested and analyzed by SEM, EDS, XRD and XPS spectroscopy. The effects of process parameters including current density and AZO sputtering times on the structure, optical and electrical properties of the film layer were systematically studied. Convection through the fixation effect in the etched Al-70Si alloy porous layer promoted the internal heat and mass transfer during PEO process which is similar to previous reported mechanism of filling of cracks and pores by nanoparticles leading to the layer densification. The PEO coating had the highest absorptivity of 0.946 and it only decreased to 0.939 after AZO sputtering, at the same time the coating has the lowest resistivity of 1.04 × 107 Ω/sq, indicating excellent anti-stray light and antistatic ability. This two-step route pave the way to improve the conductivity without compromising optical properties of PEO coating on the high silicon aluminum alloy (>50 wt% Si, Al-50Si) for space material.

        Research on the characteristics of CO2 heat pump integrated with thermal energy storage for space heating

        The CO 2 air source heat pump (ASHP) has been widely used for space heating and domestic hot water because of its energy saving, high efficiency and environmental protection. However, when used for space heating, the high return water temperature causes higher throttle loss, which results in the energy efficiency of the system obviously decreasing. To solve this problem, a CO 2 ASHP integrated with thermal energy storage (TES) is proposed in this paper. A simulation model of the system was established by TRNSYS software, and the operating characters of the studied system were investigated under the typical daily operating conditions. Additionally, its performance was compared with the benchmark system. This research shows that the average unit COP of the studied system was increased by 14.1 % compared with the benchmark system. It indicates that TES is beneficial for promoting CO 2heat pump application in cold areas.

      • A Multi-keyword Ranked Search over Encrypted Cloud Data Supporting Semantic Extension

        With the emergence of cloud computing, many data owners outsource their local data to cloud server so as to enjoy high-quality data storage services. For the protection of data privacy, sensitive data has to be encrypted before outsourcing, which makes effective data utilization a challenging task. Although existing searchable encryption technologies enable data users to conduct secure search over encrypted data, the functionality of these schemes need to be further improved. In this paper, we construct a secure and efficient multi-keyword ranked search scheme which supports both the semantic extension search and the multi-keyword ranked search. The semantic extension is achieved through the mutual information statistical analysis of keywords. And the multi-keyword ranked search is achieved through a balanced binary tree whose nodes are the vectors of term frequency (TF) values. The splitting operation and secure transformation are utilized to encrypt the vectors of index and query. Note that, the encrypted vectors can be well used to calculate accurate relevance scores. Phantom terms are added to the index vector to blind the search results to resist statistical attacks. Due to the use of tree-based index structure, the proposed scheme can achieve the sub-linear search time. Finally, the experiments are conducted to demonstrate the efficiency of the proposed scheme.

        Exergy analysis of a frost-free air source heat pump system

        The main focus of this paper is to analyze the exergetic characteristics of a frost-free air source heat pump (ASHP) integrated with solid dehumidification and phase change heat storage. The exergy loss, exergy loss ratio and exergy efficiency of the system have been calculated to investigate the influence of the operation modes on the system characteristics. In addition, a comprehensive exergy loss ratio is defined to evaluate the system performance. It is concluded that the comprehensive highest exergy loss ratio is obtained as 51.4 % for compressor, followed by the energy storage device (ESD) with 15.8 %, the extra heat exchanger coated with a solid desiccant (EHECSD) with 10.7 %. Also the variation of the exergy loss, exergy loss ratio and exergy efficiency of the system and the key components with the ambient temperature changes from -20 to 5 °C are discussed. This study can provide guidance for optimizing the structure and improving the frost-free ASHP system in the engineering applications.
