Altered Sulfate Metabolism of Arabidopsis Caused by Beet Severe Curly Top Virus Infection

Sulfur, an important component of plants, is regulated by a variety of stresses in sulfate assimilation and metabolism. Increase has been observed in the expression of O-acetylserine(thiol)lyase (OASTL) through two-dimensional electrophoresis with the shoot tips of Arabidopsis infected by beet severe curly top geminivirus (BSCTV). With the three- to six-fold increases in the transcript expression of OASTL, serine acetyltransferase (SAT) and γ-glutmylcysteine synthetase (GSH) were induced over the mock-inoculated organization in each organization through real-time RT-PCR analysis.The expression of those genes might affect the accumulation of anthocyanin in symptomatic tissues and the induction of abnormal callus-like structures formed by additional cell divisions as typical disease symptoms of BSCTV-infected Arabidopsis. This is the first report to describe the collaborative induction of OASTL, SAT,and GSH in virus-infected plants. The changed expressions of OASTL, SAT, and GSH in Arabidopsis infected with BSCTV raises new aspects regarding the biological function of symptomatic tissues related to sulfate metabolism.
벼의 엽신 및 캘러스에서 Cytokinin 유도성 A-type 및 C-type Cyclin 유전자의 발현 분석

세포 주기의 조절에 있어 중요한 역할을 담당하고 있는 cyclin 유전자들을의 발현 양상을 벼에서 유도한 캘러스와 엽신을 이용하여 관찰하였다. 특히 캘러스를 이용한 분석결과 2,4-D와 kinetin이 조합된 배지 러에 7일 동안 암 배양한 후 이를 cytokinin만이 첨가된 배치로 옮겼을 때 A-, B-, C-type cyclin 유전자들의 발현이 더 증가되었다. 또한 벼실생의 각 기관에서 cyclin 유전자의 발현양을 조사한 결과 다른 기관들에 비해 잎에서 A-, B, C-type cyclin 유전자들이 더 많이 발현하였으나 잎에 cytokinin의 종류와 농도를 다르게 처리하여 24시간동안 배양한 결과에서는 zeatin을 제외하고는 cytokinin의 종류와 양이 cyclin 유전자들의 발현에 큰 영향을 미치지 못함을 알 수 있었다. 본 실험에서는 벼 cyclin유전자의 발현과 식물생장조절제 처리와의 상관 관계에 대해서 조사하였다. The expression patterns of cyclin genes, which play a crucial role on cell cycle control, were analyzed with rice calli and leaf blades from seedlings. When callus was transferred from media containing the combinations of 2,4-D and kinetin under the dark conditions to medium supplemented with cytokinin-only on 7 days after the cultures, the expression levels of A-, B- and C-type cyclins from callus were increased significantly. Despite the fact that cyclin genes were well expressed on leaf blades rather than other organs in rice seedlings, rice leaf blades grown on the medium containing various combinations and concentrations of cytokinin for 24 hours had no major effect on the expression patterns of cyclins except zeatin. The relation between cytokinin regulation and the expression of cyclins of rice is discussed.

Recently, demand the ultra precision product which is increasing rapidly is used extensively frontier industry field such as semi-conductor, computer, aerospace, precision machine. Ultra precision processing is the portion that is very needed to NT in the field of mechanical engineering. The latest date, together with radical advancement of electronic and photonics industry, necessity of ultra precision processing is on the increase for the manufacture of various kernel parts those are connected with these industrial fields. Specially, require motion accuracy of high resolution of nm order in stroke of hundreds millimeters according as diameter of processing object great and processing accuracy rises. In this case, the response speed absolute delay because inertial mass of moving part is very large. Therefore, real time motion error compensation becomes very hardly. In this paper, we used ultra precision cutting unit(UPCU) to cope such problem. a UPCU is designed and tested to obtain sub-micrometer from accuracy in diamond turning of flat surfaces. The thermal growth spindle error is compensated for real time using a UPCU driven by piezoelectric actuator along with a laser encoder displacement sensor.

Due to global warming issue, There has been discussion about lowering greenhouse gas Since 1980. UNFCCC (United Nations Framework Convention on Climate Change) tried to reduce greenhouse gas emissions with the concept of common but differentiated responsibilities in 1992. The Korean Government is pushing through a plan about technical development of green ships against emissions trading. And private firms are considering introducing green ships. Also, They are considering importing integrated control system and Floating LNG Bunkering Terminal (FLBT) for efficient supply of fuel. Therefore, this paper introduces ‘FLBT Multi Docking Aid System’ can monitor ships in real time for making effort to prevent large accident such as fires and explosions when bring the ship alongside the FLBT.

<P>More than 100 copper/zinc superoxide dismutase 1 (SOD1) genetic mutations have been characterized. These mutations lead to the death of motor neurons in ALS. In its native form, the SOD1 protein is expressed as a homodimer in the cytosol. <I>In vitro</I> studies have shown that SOD1 mutations impair the dimerization kinetics of the protein, and <I>in vivo</I> studies have shown that SOD1 forms aggregates in patients with familial forms of ALS. In this study, we analyzed WT SOD1 and 9 mutant (mt) forms of the protein by non-invasive fluorescence techniques. Using microscopic techniques such as fluorescence resonance energy transfer, fluorescence complementation, image-based quantification, and fluorescence correlation spectroscopy, we studied SOD1 dimerization, oligomerization, and aggregation. Our results indicate that SOD1 mutations lead to an impairment in SOD1 dimerization and, subsequently, affect protein aggregation. We also show that SOD1 WT and mt proteins can dimerize. However, aggregates are predominantly composed of SOD1 mt proteins.</P>

<P>Fluorescence resonance energy transfer (FRET) and bioluminescence resonance energy transfer (BRET) are extensively used to analyze protein interactions occurring in living cells. Although these two techniques are broadly applied in cellular biology, comparative analysis of their strengths and limitations is lacking. To this end, we analyzed a small network of proteins involved in the amyloidogenic processing of the Alzheimer β-amyloid precursor using FRET based cytometry, BRET, and fluorescence lifetime imaging microscopy (FLIM). Using all three methods, we were able to detect the interactions of the amyloid precursor protein with APBB1, APBB2, and APP itself. And we found an unreported interacting pair, APP–APH1A. In addition, we show that these four interacting pairs exhibit a strong FRET correlation with the acceptor/donor expression ratios. Overall the FRET based cytometry was the most sensitive and reliable approach to screen for new interacting proteins.
