        Research on the Accessibility of Urban Green Space Based on Road Network- A Case Study of the Park Green Space in City Proper of Nanjing

        Accessibility of urban green space, as an important index of evaluating the service level of urban green space, not only shows the resident’s access to green space, but also reflects the environment quality of urban living. Since the quantity, area and space distribution of the patches in urban green space directly affect the function of urban green space and the quality of urban environment, the research on the accessibility of urban green space holds great significance to improve the service of urban green space and the living environment quality. Based on GIS software, this research uses the road network and the population distribution data of Nanjing’s city proper to conduct quantitative analysis of the accessibility of park green space and the convenience for residents to travel under different forms of transportation. After the case analysis, the author tests the application of road network in studying the accessibility of urban park green space and proposes a GIS-based method to study the accessibility and the rational layout of urban green space.

        Novel genetic variants of sine oculis homeobox homolog 3 gene are associated with body weight and average daily gain in Bos taurus

        Sine oculis homeobox homolog 3 (Six3) gene is responsible for normal mammalian pituitary development, and its genetic variations or deficiency will cause hypopituitarism, suggesting that this gene is a potential candidate gene for studying association with growth traits in animals. Herein, genetic variants within bovine Six3 gene was screened in 1031 individuals from four Chinese indigenous cattle breeds. Two novel polymorphisms (NC_007309:g.2515G>A and NC_007309:g.2607T>C) locating at positions nt1707 and nt1799 of intron 1 in bovine Six3gene, were found, and could be genotyped by TaqI ACRS PCR-RFLP and Alw26I PCR-RFLP, respectively. The frequencies of allele “A” of TaqI locus varied from 0.004 to 0.309,as well as the frequencies of allele “C” of Alw26I locus waved from 0.025 to 0.340. Association analysis revealed no significant association of TaqI locus with growth traits in Nanyang breed. However, significant relationships between Alw26I locus and body weight (BW) and average daily gain (ADG) in Nanyang breed was found (p<0.05). The individuals with genotype TC had greater body weight and average daily gain than those with genotype TT at 18 months old. Furthermore, based on combinated genotypes from these two loci, diplotypes was found to be associated with growth traits (p<0.05).The individuals with dihaplotype GG-TC had greater body weight and average daily gain at 18 month-old than those of other dihaplotypes. Therefore, the TaqI and Alw26I genetic variants of bovine Six3 gene were recommended as DNA markers related to growth traits through marker-assisted selection for genetics and breeding in cattle.

      • Prognostic Value of CD44 Variant exon 6 Expression in Non-Small Cell Lung Cancer: a Meta-analysis

        Background: CD44v6 (CD44 variant exon 6) is the chief CD44 variant isoform regulating tumor invasion, progression, and metastasis. The prognostic value of CD44v6 expression in non small cell lung cancer (NSCLC) has been evaluated in many studies, but the results have remained controversial. Thus, we performed a meta-analysis of currently available studies to investigate the prognostic value of CD44v6 expression in NSCLC patients and the relationship between the expression of CD44v6 and clinicopathological features. Materials and Methods: Two independent reviewers searched the relevant literature in Pubmed, Medline and Embase from 1946 to January 2014. Overall survival (OS) and various clinicopathological features were collected from included studies. This meta-analysis was accomplished using STATA 12.0 and Revman 5.2 software. Pooled hazard ratios (HRs) with 95% confidence intervals (95%CIs) were calculated to estimate the effects. Results: A total of 921 NSCLC patients from ten studies met the inclusion criteria. The results showed that CD44v6 high expression was a prognostic factor for poor survival (HR=1.91, 95%CI=1.12-3.26, p<0.05). With respect to clinicopathological features, CD44v6 high expression was related to histopathologic type (squamous cell carcinoma versus adenocarcinoma: OR=2.72, 95%CI=1.38-5.38, p=0.004), and lymph node metastasis (OR=3.02, 95%CI=1.93-4.72, p<0.00001). Conclusions: Our results suggested CD44v6 high expression as a poor prognostic factor for NSCLC, and CD44v6 expression is associated with lymph node metastasis and histopathologic type. Therefore, CD44v6 expression can be used as a novel prognostic marker in NSCLC cases.

        Epithelial-Mesenchymal Transition and Migration of Prostate Cancer Stem Cells Is Driven by Cancer-Associated Fibroblasts in an HIF-1alpha/beta-Catenin-Dependent Pathway

        Although cancer stem cells (CSCs) play a crucial role in seeding the initiation of tumor progression, they do not always possess the same potent ability as tumor metastasis. Thus, precisely how migrating CSCs occur, still remains unclear. In the present study, we first comparatively analyzed a series of prostate CSCs, which exhibited a dynamically increasing and disseminating ability in nude mice. We observed that the transcriptional activity of HIF-1 and -catenin became gradually elevated in these stem cells and their epithelial-mesenchymal transition (EMT) characteristic altered from an epithelial type to a mesenchymal type. Next, we further used cancer-associated fibroblasts (CAFs), which were cultured from surgically re-sected tissues of prostate cancer (PCa) to stimulate prostate CSCs. Similar results were reconfirmed and showed that the protein levels of both HIF-1 and -catenin were markedly improved. In addition, the EMT phenotype displayed a homogenous mesenchymal type, accompanied with increased aggressive potency in vitro. Most importantly, the aforementioned promoting effect of CAFs on prostate CSCs was completely repressed after “silencing” the activity of -catenin by transfection of stem cells with ShRNA. Taken together, our observations suggest that prostate migrating CSCs, with a mesenchymal phenotype, could be triggered by CAFs in a HIF-1alpha/beta-catenin-depen-dent signaling pathway.

        Long noncoding RNA DANCR promotes colorectal cancer proliferation and metastasis via miR-577 sponging

        Long non-coding RNAs (lncRNAs) play key roles in various malignant tumors, including colorectal cancer (CRC). Long non-coding RNA differentiation antagonizing non-protein coding RNA (DANCR) is overexpressed in CRC patients, but whether it affects CRC proliferation and metastasis via regulation of heat shock protein 27 (HSP27) remains unclear. In the present study, we found that DANCR was highly expressed and correlated with proliferation and metastasis in CRC. In addition, we demonstrated that DANCR and HSP27 were both targets of microRNA-577 (miR-577) and shared the same binding site. Furthermore, we revealed that DANCR promoted HSP27 expression and its mediation of proliferation/metastasis via miR-577 sponging. Finally, using an in vivo study, we confirmed that overexpression of DANCR promoted CRC tumor growth and liver metastasis. The present study demonstrated the function of DANCR in CRC and might provide a new target in the treatment of CRC.

        Structural behavior of the stiffened double-skin profiled composite walls under compression

        Steel-concrete composite walls have been proposed and developed for applications in various types of structures. The double-skin profiled composite walls, as a natural development of composite flooring, provide structural and architectural merits. However, adequate intermediate fasteners between profiled steel plates and concrete core are required to fully mobilize the composite action and to improve the structural behavior of the wall. In this research, two new types of fasteners (i.e., threaded rods and vertical plates) were proposed and three specimens with different fastener types or fastener arrangements were tested under axial compression. The experimental results were evaluated in terms of failure modes, axial load versus axial displacement response, strength index, ductility index, and load-strain relationship. It was found that specimen with symmetrically arranged thread rods sustained more stable axial strain than that with staggered arranged threaded rods. Meanwhile, vertical plates are more suitable for practical use since they provide stronger confinement to profiled steel plate and effectively prevent the steel plate from early local buckling, which eventually enhance the composite action and increase the axial compressive capacity of the wall. The calculation methods were then proposed and good agreement was observed between the test results and the predicted results.

        Compressive behavior of profiled double skin composite wall

        Profiled composite slab has been widely used in civil engineering due to its structural merits. The extension of this concept to the bearing wall forms the profiled composite wall, which consists of two external profiled steel plates and infill concrete. This paper investigates the structural behavior of this type of wall under axial compression. A series of compression tests on profiled composite walls consisting of varied types of profiled steel plate and edge confinement have been carried out. The test results are evaluated in terms of failure modes, load-axial displacement curves, strength index, ductility ratio, and load-strain response. It is found that the type of profiled steel plate has influence on the axial capacity and strength index, while edge confinement affects the failure mode and ductility. The test data are compared with the predictions by modern codes such as AISC 360, BS EN 1994-1-1, and CECS 159. It shows that BS EN 1994-1-1 and CECS 159 significantly overestimate the actual compressive capacity of profiled composite walls, while AISC 360 offers reasonable predictions. A method is then proposed, which takes into account the local buckling of profiled steel plates and the reduction in the concrete resistance due to profiling. The predictions show good correlation with the test results.

        L-carnitine treatment attenuates renal tubulointerstitial fibrosis induced by unilateral ureteral obstruction

        Background/Aims: Accumulating evidence indicates that L-carnitine (LC) protects against multiorgan damage through its antioxidant properties and preservation of the mitochondria. Little information is available about the effects of LC on renal fibrosis. This study examined whether LC treatment would provide renoprotection in a rat model of unilateral ureteral obstruction (UUO) and in vitro. Methods: Sprague-Dawley rats that underwent UUO were treated daily with LC for 7 or 14 days. The influence of LC on renal injury caused by UUO was evaluated by histopathology, and analysis of gene expression, oxidative stress, mitochondrial function, programmed cell death, and phosphatidylinositol 3-kinase (PI3K)/ AKT/forkhead box protein O 1a (FoxO1a) signaling. In addition, H<sub>2</sub>O<sub>2</sub>-exposed human kidney cells (HK-2) were treated with LC. Results: LC treatment inhibited expression of proinflammatory and profibrotic cytokines, and was followed by a significant attenuation of tubulointerstitial inflammation and fibrosis. The increased oxidative stress caused by UUO was associated with mitochondrial dysfunction and excessive apoptosis and autophagy via PI3K/AKT/FoxO1a-dependent signaling, and this was abrogated by administration of LC. In H<sub>2</sub>O<sub>2</sub>-exposed HK-2 cells, LC decreased intracellular production of reactive oxygen species, and suppressed expression of profibrotic cytokines and reduced the number of apoptotic cells. Conclusions: LC protects against the progression of tubulointerstitial fibrosis in an obstructed kidney.

        African Maternal Origin and Genetic Diversity of Chinese Domestic Donkeys

        The origin of domestic donkeys in China has been controversial. To clarify the origin of Chinese domestic donkeys, we investigated the partial mitochondrial D-loop sequences of 126 samples from 12 native breeds. The results revealed two mitochondrial origins, lineage Somali and lineage Nubian of African wild ass detected in Chinese domestic donkeys. Lineage Somali was predominant in Chinese domestic donkey breeds. The pattern of genetic variation in ass mtDNA D-loop sequences indicated that the two lineages Somali and Nubian from China had undergone population expansion events. In a combined analysis of lineages Somali and Nubian between previously published sequences from other countries/regions and sequences of Chinese domestic donkeys, the results indicated that the two lineages of Chinese domestic donkeys were from Africa and supported the African maternal origins of Chinese domestic donkeys. There was no obvious geographical structure in Chinese domestic donkey breeds, but the population showed abundant mtDNA diversity. The spread routes of Chinese domestic donkeys were also discussed.
