Molecular epidemiology of Korean porcine sapeloviruses

<P>To evaluate the prevalence and genetic diversity of porcine sapeloviruses (PSVs) in Korea, a total of 100 diarrhea fecal samples from pigs were analyzed by RT-PCR and nested PCR assays with primer pairs specific for the VP1 gene. Overall, 34 % of the diarrhea samples tested positive for PSV, and a high proportion of infections occurred along with a variety of other enteric viruses and bacteria. Genomic and phylogenetic analysis of the VP1 genes revealed pronounced genetic diversities between PSVs from Korean and elsewhere. Our results indicate that PSV infections are very common in Korean pigs with diarrhea. The infecting strains are genetically diverse.</P>
Micro-Forming Characteristics of a Zr-Based Amorphous Alloy with a Gear-Like Mold

The amorphous alloy Zr62Cu17Ni13Al8 exhibits a supercooled liquid state over a wide temperature range under high temperatures. In this present paper, the authors studied the micro-forming ability of the alloy by compressing specimens under a miniature pattern mold with a micro-gear shape. Micro-compressive formation is a formation method in which the surface of specimens of the Zr62Cu17Ni13Al8 amorphous alloy is compressed under conditions of dead loads and high temperatures and maintained for a given period for the fabrication of miniature patterns with a micro-gear shape. All the experiments were carried out under a vacuum environment to prevent the specimens from suffering deleterious effects, such as air traps in the miniature patterns and oxidation at the surface of the Zr62Cu17Ni13Al8 amorphous alloy. The characteristics of micro-compressive formation were investigated at temperatures higher than the glass transition temperature with different experimental parameters of loads and times. In addition, the micro-forming characteristics of the gear-like shape were investigated by means of scanning electron microscopes and a 3-D surface profiler system.

Micro forming is a suited technology to manufacture very small metallic parts(several ㎜~㎛). In this study, the micro forming property was studied, using Al5083 superplastic alloy with micro grain, suitable for the micro forming process and Zr-BMG amorphous with pseudo-superplastic phenomena in the supercooled liquid state. In a energy point of view, the vibrational load was useful for micro forming field. The vibrational load reduced friction and sticking slip between micro die and material and release the air static pressure bearing effect between micro die and material.

<P><B>Abstract</B></P> <P>Porcine epidemic diarrhea virus (PEDV) has emerged or re-emerged worldwide, posing a significant financial threat to major pig-producing countries. In the present study, a virulent Korean pandemic PEDV strain, KNU-141112, was serially propagated in Vero cells for up to 100 passages. Through cell culture adaptation, we obtained four distinct deletion (DEL) mutants by plaque purification followed by nucleotide sequencing of the spike (S)/ORF3 gene-coding region, which were designated KNU-141112-S DEL2, −S DEL5, −S DEL2/ORF3, and −S DEL5/ORF3. Further whole genome sequencing identified 12 or 14 amino acid changes in the cell-adapted DEL strains. Animal inoculation studies revealed that the virulence of both S DEL2/ORF3 and S DEL5/ORF3 viruses with a large 46-nt deletion in the intergenic portion of S and ORF3 was remarkably diminished, indicating viral attenuation in the natural host. Furthermore, these cell-adapted strains elicited potent neutralizing antibody responses in immunized pigs. Taken together, our data indicate that the cell-attenuated S DEL2/ORF3 and S DEL5/ORF3 strains are promising candidates for the development of a safe and effective live PEDV vaccine.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A virulent G2b PEDV strain KNU-141112 was subjected to Vero cell culture passage. </LI> <LI> Deletions (DEL) in S and ORF3 of KNU-141112 at high passage levels were identified. </LI> <LI> Cell-adapted KNU-141112 DEL strains were highly attenuated and immunogenic in pigs. </LI> <LI> Large deletions in ORF3 appear to be associated with PEDV attenuation in pigs. </LI> <LI> High-cell-passage KNU-141112 DEL strains can be live, attenuated vaccine candidates. </LI> </UL> </P>
