        The level of urinary aflatoxin M1 in Korean adults

        Competitive ELISA methods were used to measure the level of aflatoxin M1 (AFM1) from urine in 1008 Korean adults. Subjects were selected by random sampling in all areas of Korea, except Cheju-do. The recovery rate of AFM1 using this method was 105% (73-124%). The geometric mean of urinary AFM1 in all subjects was 3.43 pg/mL (3.67 ng/g creatinine). The level of AFM1 in males was statistically higher, compared with female subjects. However, the levels of AFM1 did not differ according to age. Subjects in Chungbuk-do showed the highest urinary AFM1 concentration, whereas subjects in Kyeongnam-do showed the lowest concentration. Assuming an excretion rate of 5%, this AFM1 excretion corresponds to approximately 0.1 microgram/day in Korean adults.

      • 동종 골수 이식한 백혈병에서 단반복 유전자로 살펴본 혼합 키메라 현상의 의의

        연구배경 : 동종 골수 이식한 백혈병에서 키메라 현상의 분석은 착상, 질병 재발과 이식 거부를 이해하는데 도움을 줄 수 있다. 최근 STR을 PCR로 증폭한 검사법에 의해 매우 민감하고 유용하게 혼합 키메라 현상을 검출할 수 있다고 보고되고 있으나, 이 방법의 민감도와 임상적 의의는 아직 완전히 정립되지는 않았다. 따라서 PCR-STR검사법이 혼합 키메라 현상을 민감하게 검출하는지 보고, 동종 골수 이식 후 혼합 키메라 현상의 변화 양상을 관찰하고, 키메라 현상의 임상적 의의를 보고자 하였다. 방법 : 형제로부터 동종 골수 이식한 급성 백혈병 6명과 만성 백혈병 2명의 모두 8명을 대상으로 하였다. 44개 골수 검체에서 phenol/chloroform방법에 의하여 단핵세포 DNA를 추출하였다. DNA는 CTT 삼중체, vWA와 amelogenin시발체를 이용하여 증폭하였다. 증폭 산물은 5% 폴리아크랄아미드 젤에서 전기영동 뒤 은 염색으로 확인하였다. 2명의 DNA를 여러 비율로 혼합하여 상기 검사를 하여 검출 가능 최소 농도를 결정하였다. 결과 : 상기 검사법의 검출 가능 최소농도는 0.25%였다. 혼합 키메라 현상은 8명중 4명에서 관찰되었다. 혼합 키메라 현상은 보인 1명에서 급성 및 만성 이식편대 숙주 질환이 나타났고, 완전 공여자 키메라 현상을 보인 1명에서 만성 이식편 대 숙주 질환이 나타났다. 점진적으로 증가하는 혼합 키메라 현상을 보인 2명은 이식 후 12개월에 질병 재발을 보였으나, 완전 공여자 키메라 현상을 보인 경우엔 재발이 관찰되지 않았다. 결론 : PCR-STR검사법은 동종 골수 이식 후 남아 있는 환자 세포를 검출하고 키메라 현상을 관찰하는데 유용한 방법이다. 또한 이 검사법은 동종 골수 이식 후 재발의 조기 진단, 이식편 대 숙주 질환의 평가와 면역 관용에 임상적으로 응용할 수 있으리라 생각한다. Background : Chimerism analysis after allogeneic bone marrow transplantation (BMT) for leukemia could be helpful understanding of the early marrow engraftment, disease relapse, and graft rejection. Recently, a PCR technique which amplifies short tandem repeats(STR) has been reported to be highly sensitive and reliable in detecting mixed chimerism. But its sensitivity and clinical significance has not been established. The purpose of this study was firstly, to confirm whether a PCR-STR in highly sensitive enough ti assess mixed chimerism, secondly to monitor the changing patterns of mixed chimerism after allogeneic BMT, and lastly to determine correlation between the chimeric status and the clinical outcome. Methods : A study was made eight patients (six with acute leukemia and two with chronic leukemia) who underwent unmanipulated allogeneic BMT along with matched donors. DNA was extracted from mononuclear cells of 44 bone marrow samples using a phenol/chloroform method. Amplification of DNA was done using CSFIPO-TPOX-TH01(CTT) triplex with or without vWA or amelogenin primer. The amplified product was separated on 5% polyacrylamide gel and was confirmed by silver staining. The sensitivity of CTT triplex method was determined by mixing DNA from two person in serial proportion using the same method. Results : The sensitivity for CTT triplex method was 0.25% Mixed chimerism was documented in 4 of 8 patients. ONe of four patients with mixed chimerism developed acute and chronic graft versus host disease (GVHD) and one of four patients with complete donor chimerism also developed chronic GVHD. Two patient with progressive mixed chimerism relapsed at 12 months post-BMT, whereas no patient with complete donor chimerism has relapsed. Conclusion : The study has found that PCR-STR was an effective method for the detecting residual host cell and monitoring the chimeric status after allogenic BMT. PCR-STR can also be clinically applicable in early prediction of relapse, appropriate assessment of GVHD, as well as tolerance after allogeneic BMT.

        Edwardsiella tarda에 대한 계란난황항체의 분리와 정제

        Edwardsiella tarda로 면역한 닭은 난황항체(IgY) 정제 방법을 비교하였다. Anti-E.tarda IgY의 정제는 PEG법, chloroform-PEG법, ammonium sulfate법과 정제 kit를 사용한 4가지 다른 방법으로 실시하였다. 정제된 I&Y는 64kDa의 heavy chain과 27 kDa의 light chain을 나타내었다. E.tarda로 면역된 IgY는 면역되지 않은 대조 IgY보다 높은 ELISA가와 응집항체가를 나타내었으며, 정제된 IgY는 western blotting에서 anti-E.tarda 토끼혈청과 유사한 E.rarda 단백질을 인식하였다. PEG법과 ammonium sulfate법에 의해 정제된 IgY는 응집항체가가 1:512, chloroform-PEG법과 정제 kit에 의해 정제된 IgY를 정제하기 위한 가장 빠른 방법이었다. 이 연구의 결과로 PEG법이 IgY의 생물학적 활성을 유지함과 더불어 신속하고 효과적인 정제방법임을 알 수 있었다. The present study compared purification methods of hen egg yolk immunoglobulin (IgY) from the hen immunized with Edwardsiella tarda. The purification of anti-E. tarda IgY was performed by four different methods, polyethylene glycol (PEG), chloroform polyethylene glycol (Chloroform-PEG), ammonium sulfate and purification kit. Purified IgY had heavy chain of 64 kDa and light chain of 27 kDa size. IgY purified from the hen immunized with E. tarda showed higher ELISA values and agglutination titers than those with IgY purified from the non-immunized hen as a negative control. In addition, purified IgY recognized similar E. tarda proteins to those with anti-E. tarda rabbit serum by western blotting. Purified IgY had an agglutination titer of 1 : 5 12 by PEG method and ammonium sulfate method, and 1 : 128 by chloroform-PEG method and purification kit. Moreover, PEG method was the most rapid method among the four different IgY purification methods. These results indicate that PEG method is effective purification method maintaining biological activity of the IgY.

        Recently, Superconducting cable has attracted a great deal of interest from the viewpoint of energy saving. One of the most important and difficult problems to be solved to assure the high reliability and long life of cable is breakdown at cryogenic temperatures. In this paper, we will report on the breakdown characteristics of cryogenic liguid and gas for superconducting cable. In cryogenic gas N₂(300K-93K), the breakdown voltage depends on gas density and temperature. And it is obeyed by Paschen's law. But low breakdown voltage of cryogenic gas N₂requires caution in the design. in cryogenic liquid N₂(77K), the breakdown voltage depends on gas bubble.

        The purpose of this study was to suggest the ideal direction in job opportunities by identifying the relationships among the personal charateristics, type of sports and his/her expectations toward job. The subject of this study consists of 320 students who were enrolled as Martial Art major in Y university located Kyunggido area in Korea. The instrument used in this study was modified from the guestionnaires which was administered by Jung-Ha Kim(1995) and mainly concerned on the items of job expectations. The title of the survey was "Questionnaires of Job Expectation of the students with Martial Art Major." the reliability of this survey was examined by Cronbach's α(α = .89). The PC-SAS was used to analyze the achieved data for this study. In conclusion, the review and development of curriculum are suggested to enhance with martial art major. The results of this study were shown as follow: First, the curriculum which focused on the professional Knowledge beyond martial art area is needed. Second, the professional classes related to martial arts should be offered. Third, the curriculum for teachers' background such as teachers education and instructors education is recommended.

        Eu or Fe doped BGO scintillation crystals were grown by Czochralski method. In order to get information about traps in the grown BGO crystals, we measured trap parameters including activation energy, frequency factor and the kinetic order of thermoluminescence, and compared such parameters with thermoluminescent characteristics of pure BGO scintillation crystals. In addition, optical transmittance of the grown BGO crystals was measured.

      • 減少된 Na^+과 Cl^-濃度가 心房筋의 活動電壓과 收縮力에 미치는 影響

        To investigate the effects of decreased extracellular Na and Ci ion concentration on atrial muscle of frog, changing patterns of single cell action potential by means of glass ultramicroelectrode and tension curves of isometric contraction in atrial muscle strips were recorded simultaneously. The results were as follows: 1. Decreased Na^+ and Cl^- concentration had little effects on resting transmembrane potential but amplitude of action potential and size of the overshoot were decreased along with the decrease of the concentration. The peak of the action potential had not reached "O" potential difference after 25 minutes bathing 1/2, 1/4 of the normal Na^+ and Cl^- concentration. 2. -dV/dt in plateau phase were tended to increase according to the decrease of Na^+ and Cl^- concentration and significantly increased in less than 1/4 of the normal concentration. -dV/dt in repolarization phase were scarcely affected to Na^+ & Cl^-. 3. Duration of action potential were significantly decreased in low concentration such as 1/2 of the normal Na^+ and Cl^- concentration. 4. Atrial muscle contractility were increased in decreased Na^+ & Cl^- concentration but the degree of increment were relatively decreased with time. 5. Frequency of the contraction were dominantly affected by decreased Na^+ & Cl^- than any other parameters and decreased. Above results were considered as that Na^+ played a important role in maintaining upstroke phase and slow inward current of plateau phase in atrial muscle action potential and Na^+ also had influence on the flux of Ca^++ across the membrane or the control of available intracellular Ca^++ concentration.

      • 위 유문동근 K^+ - 경축에 미치는 Vanadate의 효과

        Effects of vanadate on the spontaneous contraction and the high K^+-induced contracture of guinea pig’s pyloric antral smooth muscles were observed in the various Na^+ and/or Ca^+ of bathing solutions. Following results were obtained; 1. Basal tension and developed tension of the spontaneous activity were increased by vanadate and the membrane potential tended to be depolarized. 2. Phasic contracture increased, tonic contracture decreased as the outside K^+ concentration increased and the both components were increased by vanadate. 3. In the case of absence of K^+ and/or Na^+, the contracture were hardly developed and the spontaneous, rhythmic contraction were sustained. 4. Effects of vanadate on the K^+-contracture and the Ca^2+-sensitivity of the Phasit and tonic components were prominent in the presence of Na^+. The above results suggested that vanadate would increase the voltage-dependent calcium current in smooth muscle cells of the pyloric antrum.
