Ultrasound elastography of the thyroid: principles and current status

Ultrasound (US) elastography has been introduced as a non-invasive technique for evaluating thyroid diseases. This paper presents a detailed description of the technical principles, peculiarities, and limitations of US elastography techniques, including strain elastography and shear-wave elastography. This review was conducted from a clinical perspective, and aimed to assess the usefulness of US elastography for thyroid diseases in specific clinical scenarios. Although its main focus is on thyroid nodules, the applications of US elastography for other thyroid diseases, such as diffuse thyroid diseases and thyroiditis, are also presented. Furthermore, unresolved questions and directions for future research are also discussed.

Vortex-Induced-Vibration (VIV) is one kind of the wind-induced vibrations, which may occur in the construction and operation period of bridges. This phenomenon can bring negative effects to the traffic safety or can cause bridge fatigue damage and should be eliminated or controlled within safe amplitudes. In the current VIV studies, one available mitigation countermeasure, the horizontal flow-isolating plate, shows satisfactory performance particularly in PI shaped bridge deck type. Details of the wind tunnel test are firstly presented to give an overall description of this appendage and its control effect. Then, the computational-fluid-dynamics (CFD) method is introduced to investigate the control mechanism, using two-dimensional Large-Eddy-Simulation to reproduce the VIV process. The Reynolds number of the cases involved in this paper ranges from 1×〖10〗^5 to 3×〖10〗^5, using the width of bridge deck as reference length. A field-filter technique and detailed analysis on wall pressure are used to give an intuitive demonstration of the changes brought by the horizontal flow-isolating plate. Results show that this aerodynamic appendage is equally effective in suppressing vertical and torsional VIV, indicating inspiring application prospect in similar PI shaped bridge decks.

Oxidation of cycloalkenes with O2 promoted by heterogeneous bis(acetylacetonato) cobalt (II) complex catalyst which can be recycled has been performed under mild conditions. It was found that β-ionone, cyclohexene,1-methylcyclohexene, and α-ionone were efficiently oxidized with O2 in the presence of Co (II) complex and butyraldehyde at 55 oC. A simple treatment of the resulting products led to epoxides as predominant products and a small amounts of allylic oxides, the chemoselectivity for the former being 82.1 - 90.8% with a 70.6 - 98.6% substrate conversion. On the other hand, oxidation of 1-phenylcyclohexene, 1-cyclohex-1-enylethan-1-one, α-pinene, and β-pinene gave allylic oxides as major products.

A real-time digital time-stamp sorting algorithm used in the In-Beam positron emission tomography (In-Beam PET) is presented. The algorithm is operated in the field programmable gate array (FPGA) and a small amount of registers, MUX and memory cells are used. It is developed for sorting the data of annihilation event from front-end circuits, so as to identify the coincidence events efficiently in a large amount of data. In the In-Beam PET, each annihilation event is detected by the detector array and digitized by the analog to digital converter (ADC) in Data Acquisition Unit (DAQU), with a resolution of 14 bits and sampling rate of 50 MS/s. Test and preliminary operation have been implemented, it can perform a sorting operation under the event count rate up to 1 MHz per channel, and support four channels in total, count rate up to 4 MHz. The performance of this algorithm has been verified by pulse generator and <sup>22</sup>Na radiation source, which can sort the events with chaotic order into chronological order completely. The application of this algorithm provides not only an efficient solution for selection of coincidence events, but also a design of electronic circuit with a small-scale structure.
Establishment of a novel myocarditis mouse model based on cyclosporine A

Background: Myocarditis is a myocardial injury that can easily cause adolescent death. Traditional research models of animal invasion with viral components, lipopolysaccharide (LPS) or porcine myocardial myosin, among others, have the shortcomings of potential biological safety hazards and high animal mortality. Objective: To explore the construction of a novel myocarditis model with cyclosporine A and the potential genes and pathways associated with it. Methods: BALB/c mice were used in this study, and cyclosporin A and LPS were injected into the peritoneal cavity of mice. The successful establishment of the model was assessed by detecting serum myocardial injury markers and inflammatory factors levels, HE, IHC staining, and RT-qPCR methods. Key genes were obtained using the GSE35182 dataset from the GEO database and validated with the RT-qPCR method. Results: We found that a large number of inflammatory cells infiltrated the myocardium of mice in each group of Cyclosporin A constructed model, while the expression of inflammatory factor indicators was increased, and this model has the characteristics of high degree of local inflammation in myocardial tissue, low mortality, and safe and non-toxic treatment. Using GSE35182 data, we selected 18 Hub genes and validated Hub genes in myocardial tissue with RT-qPCR and found that multiple signaling pathways such as Toll-likereceptor signaling pathway(TLRs), Rap1 signal pathway(Rap1), and Chemokine signaling pathway may be involved in the development of myocarditis. Conclusion: Cyclosporin A can construct a new myocarditis model, and TLRs, Chemokines and Rap1 signaling pathways may be the core pathways of myocarditis.
Influence of Device Parameters Spread on Current Distribution of Paralleled Silicon Carbide MOSFETs

This paper systematically investigates the influence of device parameters spread on the current distribution of paralleled silicon carbide (SiC) MOSFETs. First, a variation coefficient is introduced and used as the evaluating norm for the parameters spread. Then a sample of 30 SiC MOSFET devices from the same batch of a well-known company is selected and tested under the same conditions as those on datasheet. It is found that there is big difference among parameters spread. Furthermore, comprehensive theoretical and simulation analyses are carried out to study the sensitivity of the current imbalance to variations of the device parameters. Based on the concept of the control variable method, the influence of each device parameter on the steady-state and transient current distributions of paralleled SiC MOSFETs are verified separately by experiments. Finally, some screening suggestions of devices or chips before parallel-connection are provided in terms of different applications and different driver configurations.

miR-214 plays a major role in the self-renewal of skin tissue. However, whether miR-214 regulates the proliferation and differentiation of human hair follicle stem cells (HFSCs) is unknown. Primary HFSCs were isolated from human scalp skin tissue, cultured, and identified using flow cytometry. An miR-214 mimic and inhibitor were constructed for transfection into HFSCs. The MTS and colony formation assays examined cell proliferation. Immunofluorescence detected the localization and expression levels of TCF4, b-catenin, and differentiation markers. Luciferase reporter and TOP/FOP Flash assays investigated whether miR-214 targeted EZH2 and regulated the Wnt/b-catenin signaling pathway. Western blot determined the expression levels of enhancer of zeste homolog 2 (EZH2), Wnt/b-catenin signaling-related proteins, and HFSC differentiation markers in cells subjected to miR-214 transfection. miR-214 expression was remarkably decreased during the proliferation and differentiation of HFSCs into transit-amplifying (TA) cells. Downregulation of miR- 214 promotes the proliferation and differentiation of HFSCs. Overexpression of miR-214 led to decreased expression of EZH2, b-catenin, and TCF-4, whereas downregulation of miR-214 resulted in increased expression of EZH2, b-catenin, and TCF-4 as well as TA differentiation markers. Immunofluorescence assay revealed that inhibiting miR-214 triggered the entry of b-catenin and TCF-4 into the nucleus. The luciferase reporter and TOP/FOP Flash assays demonstrated that miR- 214 directly targets EZH2 and affects Wnt/b-catenin signaling. The miR-214/EZH2/b-catenin axis could be considered a candidate target in tissue engineering and regenerative medicine for HFSCs.
Characterization of P450 monooxygenase gene family in the cotton aphid, Aphis gossypii Glover

Cytochrome P450 monooxygenases play an important role in the biosynthesis and detoxification of endogenous compounds. In this study, we analyzed a number of P450 genes through transcriptome-wide and genome analyses in Aphis gossypii Glover, the most notorious cotton pests. A total of 49 CYP-putative sequences were identified and classified into 13 families and 21 subfamilies. We identified a putative P450 motif (FXXGXXXCXG), and further analyzed other conserved sequences, which indicate that forty-seven of the fortynine genes seem to be functional, but one is likely non-functional and one is predicted pseudogenes. We found that P450-encoding genes are conserved in gene structure and organization (number, position, and phase of introns) in cotton aphids. The size, transcription orientation, and comparison with other insects of the cotton aphid P450 family suggested that it has evolutionary differentiation. The distribution across the genome and phylogenetic clustering analysis indicated past occurrence of gene duplication events. These results may provide valuable clues for understanding the evolution of the aphid genome, studying the functions of P450s in cotton aphid, and further selecting candidate genes for novel insect control methods.
