      • Expression of mosquitocidal crystal protein genes in non-insecticidal Bacillus thuringiensis subsp. israelensis

Bacillus thuringiensis NTB-1 isolated from soil samples in Korea produces ovoidal parasporal inclusions with proteins of approximately 24-40kDa in size. Although serological study indicated that the isolate has a flagella (H) antigen identical with subsp. israelensis, it seemed to be non-insecticidal against Lepidoptera and Coleoptera as well as Diptera. To investigate the activity of non-insecticidal B.thuringiensis transformed with insecticidal crystal protein genes, cryIVD and cytA genes of B. thuringiensis subsp. morrisoni PG-14, highly toxic to mosquito larvae, were introduced into the isolate NTB-1. The expression of mosquitocidal crystal protein genes in NTB-1 was characterized by SDS-PAGE analysis and electron microscopy. The results showed that crystalline inclusions of host, CryIVD and CytA were stably expressed in the transformant. However, the mosquitocidal activity of transformant was similar to that of B. thuringiensis subsp. kurstaki Cry-B harbouring cryIVD and cytA genes, demonstrating that a synergistic effect by an interaction of both introduced insecticidal and resident non-insecticidal crystal proteins was not observed.

        Universality of weak ferromagnetism in the magnetically frustrated system <i>R</i> <sub>1−<i>x</i> </sub>Y<sub> <i>x</i> </sub>B<sub>4</sub>

        <P>Emergence of anomalous weak ferromagnetism in geometrically frustrated magnetic system of R1-xYxB4 (R = Tb and Dy) is quite interesting [Phys. Rev. B 91, 024414 (2015)]. In order to study the weak ferromagnetism in other compounds, the magnetic properties of R1-xYxB4 (R = Sm, Gd, and Er) were investigated. The anomalous weak ferromagnetism was also found to occur in all compounds, R1-xYxB4 (R = Sm, Gd, and Er). The weak ferromagnetism is revealed in the magnetic easy plane, (001), for R1-xYxB4 (R = Sm and Gd) and along the magnetic easy axis, [001], for R1-xYxB4 (R = Er). The magnitude of saturated magnetic moment at T = 2 K shows a strong dependence on Y-concentration, i.e., the maximum magnitude occurs at x approximate to 0.3 for all compounds. The general magnetic properties of weak ferromagnetism is quite similar to the weak ferromagnetism in R1-xYxB4 (R = Tb, Dy). Therefore, it is experimentally demonstrated that the emergence of weak ferromagnetism, induced by non-magnetic substitution in a magnetically frustrated system, is the universal feature in RB4 compounds.       • Characterization of a wheat mutant missing low-molecular-weight glutenin subunits encoded by the B-genome

        <P>DH2O, a new wheat mutant missing low-molecular weight glutenin subunits encoded by the Glu-B3 locus, was discovered among double haploid lines obtained from a cross between the Korean wheat cultivars Keumkang and Olgeuru. Absence of the Glu-B3 LMW-GS proteins was determined by one-dimensional gel electrophoresis (SDS-PAGE) and confirmed by two-dimensional gel electrophoresis (2-DGE) and tandem mass spectrometry (MS/MS). The deletion of Glu-B3 genes was also demonstrated using allele specific DNA markers. Basic agronomic traits, protein content, dough mixing properties and bread loaf volume of DH2O and parental wheat cultivars were evaluated in field-grown wheat over a two year period. This mutant is a valuable resource for understanding the function of individual LMW-GS alleles or genes at the Glu-B3 locus using either breeding or biotechnology approaches. (C) 2017 Elsevier Ltd.         Development of Naturally Degradable “Rice Polymer" For Organic Weed Management of Red Pepper and Rice

        Among the developed bío-degradable polymer fílms as compared wíth transparent fílm(Whíte), black polymer fílm was sígnífícantly more effectíve for controllíng weeds ín red pepper. Also, we found that whíte and black polymer mulchíng had 81.8% and 97.9% of managing weed controllíng effects in rice, respectively. Compared to nonmulched rice paddy with water supply, the non-mulched rice paddy wíthout any water supply has stopped its growth at 41 days after transplantíng, whíle polymer -mulched rice paddy without water supply had about 60% of normally growing rice plants. This shows the polymer treatment has a remarkable effect on water and power saving, solution of herbicidal resistance, avoidance of herbícidal influence to eco-system etc. When the naturally decomposíng polymer was used, a temperature was elevated as high as 4.7 ℃ on maximum and 2.6 ℃ on average. Also the naturally decomposing polymer accelerated rooting by 7 days and lowered a stress level from transplanting. The weed control effect mulched by polymer was remarkable as 98. 7%. The p이ymer now, after 294 days treated on the rice paddy, has been completely decomposed.

      • Aberrant ventral striatal responses during incentive processing in unmedicated patients with obsessive–compulsive disorder

Aberrant ventral striatal responses during incentive processing in unmedicated patients with obsessive–compulsive disorder.</P><P><B>Objective: </B> Obsessive–compulsive disorder (OCD) is characterized by the dysfunction of control and reward mechanisms. However, only few neuroimaging studies of OCD have examined the reward processing. We examined the neural responses during incentive processing in OCD.</P><P><B>Method: </B> Twenty unmedicated patients with OCD and 20 age‐, sex‐, and IQ‐matched healthy controls underwent functional magnetic resonance imaging while performing a modified monetary incentive delay task.</P><P><B>Results: </B> Compared with controls, patients with OCD showed increased ventral striatal activation in the no‐loss minus loss outcome contrast and a significant positive correlation between the ventral striatal activation and compulsion symptom severity. In addition, patients with OCD showed increased activations in the frontostriatal regions in the gain minus no‐gain outcomes contrast. During loss anticipation, patients with OCD showed less activations in the lateral prefrontal and inferior parietal cortices. However, during gain anticipation, patients with OCD and healthy controls did not differ in the ventral striatal activation.</P><P><B>Conclusion: </B> These findings provide neural evidence for altered incentive processing in unmedicated patients with OCD, suggesting an elevated sensitivity to negatively affect stimuli as well as dysfunction of the ventral striatum.</P>

        Efficacy of HVT-IBD vector vaccine compared to attenuated live vaccine using in-ovo vaccination against a Korean very virulent IBDV in commercial broiler chickens

        <P>The production performance, efficacy, and safety of two types of vaccines for infectious bursal disease virus (IBDV) were compared with in-ovo vaccination of Cobb 500 broiler chickens for gross and microscopic examination of the bursa of Fabricius, bursa/body weight (b/B) ratio, flow cytometry, and serologic response to Newcastle disease virus (NDV) vaccination. One vaccine was a recombinant HVT-IBD vector vaccine (HVT as for herpesvirus of turkeys) and the other was an intermediate plus live IBDV vaccine. A significant difference was detected at 21 d. Eight of 10 chickens that received the IBDV live vaccine had severe bursal lesions and a relatively low b/B ratio of 0.95, and an inhibited NDV vaccine response. On the other hand, the HVT-IBD vector vaccine resulted in mild bursal lesions and a b/B ratio of 1.89. Therefore, the live vaccine had lower safety than that of the HVT-IBD vector vaccine. To determine the protective efficacy, chickens were intraocularly challenged at 24 d. Eight of 10 chickens in the IBDV live vaccination group showed gross and histological lesions characterized by hemorrhage, cyst formation, lymphocytic depletion, and a decreased b/B ratio. In contrast, the HVT-IBD vector vaccinated chickens showed mild gross and histological lesions in three of 10 chickens with a b/B ratio of 1.36, which was similar to that of the unchallenged controls. Vaccinated chickens showed a significant increase in IBDV antibody titers, regardless of the type of vaccine used. In addition, significantly better broiler flock performance was observed with the HVT-IBD vector vaccine compared to that of the live vaccine. Our results revealed that the HVT-IBD vector vaccine could be used as an alternative vaccine to increase efficacy, and to have an improved safety profile compared with the IBDV live vaccine using in-ovo vaccination against the Korean very virulent IBDV in commercial broiler chickens.</P>

      • 단감 '부유' 과실의 저장특성에 미치는 수확기 동상해 및 과수원 온도조건의 영향

        단감 '부유'의 가을 동상해 발생과 과실생장에 미치는 식재위치의 표고와 농장의 온도환경의 영향과, 서리가 발생한 과수원의 과실 저장 특성을 알아보고자 경남 창원(과원 A)과 창녕(과원 B)의 경사지 과원에서 표고별(상, 중, 하)로 과실특성을 조사하고 서리가 발생한 후 수확한 과실을 MA저장 후 저장특성(품질, 저온장해)을 조사하였다. 수확기 전후(10월25일, 11월5일, 11월20일)에 조사된 두 과원의 과실의 생장 특성(과중, 당도, 경도)은 표고와 농장 요인의 유의미한 영향을 받지 않았다. 과실 착색은 표고 요인의 영향을 받았으며, 과실의 착색(hunter 'a')은 B 과원보다 A 과원에서 더 빨리 진행되었다. 수확기에 발생한 저온으로 인하여 과원 B의 하부에서만 약 2%의 과실에서 동상해 피해가 관찰되었다. 두 과원(A, B)에서 11월 5일과 11월 20일에 수확한 과실을 저밀도 폴리에틸렌 필름을 사용하여 MA저장 한 결과, 저온지 과원(B)보다 고온지 과원(A)의 과실 경도가 높게 유지되었다. 그리고 11월 5일에 수확한 과실은 약 80일 저장 후에도 건전과율이 과원 A에서 약 73%, 과원 B에서 약 85%로 높게 유지되었으나, 11월 20일(서리 발생 후) 수확한 과실은 A 과원의 과실은 약 80일 저장 후에도 76%의 높은 건전과율을 유지하였으나 B 과원의 과실 건전과율은 약 14%에 불과했다. We analyzed the effect of planting site (elevation: high, middle, low) and temperature condition of orchard on the frost injury occurrence and storage characteristics of persimmon (Diospyros kaki cv. 'Fuyu) fruits under an early fall frost at two orchards, Changwon (A) and Changnyeong (B), Gyeongsangnam-do Korea. Also, the fruits picked at Nov. 5 and Nov. 20 were MA stored to investigate the quality and chilling injury of fruits. On 2-way factor analysis, both planting elevation and orchard factor had not a significant effect on fruit characteristics (weight, firmness, soluble solid), while planting elevation factor did on the skin color (hunter 'a')(p<0.05). A fruit skin coloration at orchard A was faster than that at orchard B. At low elevation of orchard B, 2% fruits were frost injured at harvest season. On LDPE film MA storage with the fruits harvested at Nov. 5 and Nov. 20, the fruits at orchard A had higher fruit firmness than those of orchard B during storage. In addition, the 73% (orchard A) and 85%(orchard B) fruits harvested at Nov. 5 showed no chilling injury 80 days after storage. However, At harvest of Nov, 20 (after incidence of frost), only the 14% fruits of orchard B had no the chilling disorder 80 days after storage, even though 76% fruits at orchard A did.
