        Correlation between Meat Color and L-Carnitine Content in Livestock Meats

        In this study, the correlation between color of redness and L-carnitine content in meats was investigated using microplate enzymatic assays. The L-carnitine levels and its storage stabilities of domestic and imported livestock products in Korean markets were also studied. The results showed a high correlation (r=0.9764) between L-carnitine content and redness values of homogenized meat solution. Korean native cattle (‘Hanwoo’) meat showed the highest L-carnitine content (3.64±0.14 μ㏖/g) in meat samples analyzed in this study. The L-carnitine level of the meats decreases during periods of storage in cold and freezing conditions, and the level of decrease was more significant at 4℃ than at ?20℃, which suggests that the storage stability of L-carnitine is related to its storage temperature. This study gives reliable data about correlation between meat color of redness and L-carnitine content, and gives useful information to determine the characteristics of ‘Hanwoo’.

        Efficient hole injection in blue organic light-emitting devices by using a double hole injection layer to improve chromaticity and electrical characteristics

        A double hole injection layer consisting of Ag_2O//MoO_x was applied to a 4,40-bis (2,2-diphenylvinyl)-1,10-biphenyl-based blue organic light-emitting device (OLED). The hole current injection of the devicewas improved and the performance of the blue OLED was enhanced. We have observed that the insertion of such double layer leads to a striking improvement in chromaticity and electrical characteristics. This device showed much better chromaticity and less current-induced color shift as compared to the corresponding single hole injection layer counterparts. Using this strategy, a striking improvement in the electrical properties with lower driving voltage, higher power efficiency and a weak current-induced fluorescence quenching was achieved. This could be explained by the fact that an interlayer of Ag_2O//MoO_x provided a stepped energy level which greatly facilitated hole injection and hence enhanced injection current. The result is verified by using the J―V curves of ‘only’ devices and further explained with the help of Fowler-Nordheim (F-N) tunneling theory.

        A high performance of BPhen-based white organic light-emitting devices with a dual-emitting layer and its electroluminescent spectral property

        This work demonstrated the fabrication of white organic light-emitting devices (WOLEDs) using a dual emitting layer (d-EML) consisting of blue and ‘white’ emitters. In this d-EML system, the blue emitter not only emits but also assists the incomplete energy transfer. More importantly, it behaves as an effective trapping site for holes, which contributes to the efficient recombination of electron–hole pairs. The d-EML was constructed between the hole-transmitting layer (HTL) and the electron-transmitting layer (ETL) of Alq3 and BPhen. The thickness of the blue emitter used in the d-EML devices has an important effect on chromaticity and efficiency. Through the optimization of device structure, the reasonable white emission with Commission Internationale de L’Eclairage (CIE) color coordinates of (0.33, 0.33) and little color shift was obtained. The device showed an applicable luminance with its maximum luminance of 22,874 cd/m2 at a driving voltage of 16 V. The maximum luminance efficiency was achieved 8.10 cd/A,and the maximum power efficiency was reached 5.07 Lm/W. The result is explained with the help of the excitons generation and diffusion theory. According to the theory of excitons generation and diffusion,an equation has been set up which concerns electroluminescent spectra to the thickness of the two emitters and to the exciton diffusion length.

        물리적 처리에 따른 우육추출물중의 BSA와 BGG단백질의 SDS-PAGE 패턴 및 항원성의 변화

        In our previous report, we indicated that not only BSA but also BGG played an important role in the allergenicity of beef. In this study, the effect of heat or high pressure treatments to beef extract on the SDS PAGE patterns was examined. The antigenicity of each treated samples was also investigated by Western blots assay with the sera of BGG positive beef allergic patients. The BGG band and its antigenicity slightly disappeared but not generally in 100oC group, indicating 100oC treatment is not sufficient to totally eliminate the antigenicity of beef allergens. Compared with BGG band, BSA band significantly disappeared in SDS PAGE with 100oC treatment, indicating BSA is more heat sensitive than BGG. When the beef extract was heated at 120oC, not only BSA but also BGG bands was largely disappeared in both SDS PAGE and Western blots. High pressure (HP) treatment even at 600 MPa did not affect SDS PAGE and Western blots pattern of BSA. On the contrary, BGG treated with HP showed visible changes in SDS PAGE. 600 MPa treatment significantly reduced the antigencity. Interestingly, these behaviors of BGG were not found in the same experiments with pure BGG treated with HP. From these results, it was speculated that some kinds of proteolytic enzymes in beef extracts were involved in the BGG molecular degradation by HP treatment. The aging experiments of beef extracts treated with HP supported this hypothesis. Further studies are needed to clarify the function and working mechanism of enzymes associated with BGG degradation in beef extracts by HP treatment. 이전의 연구에서, 우육중의 주된 알레르겐은 우육추출물중의 BSA, BGG라고 보고하였다. 본 연구에서는 이들 단백질의 열처리와 초고압처리와 같은 물리적 처리에 따른 SDS-PAGE상의 변화와 우육알레르기 환자의 혈청을 이용한 알레르겐성의 변화를 Western blots로 관찰하였다. 우육추출물에 대한 열처리에 의한 알레르겐 단백질의 변화에서, 100oC 가열처리군에서 BSA band는 큰 감소를 보였으나, BGG band는 완전히 제거되지 못하고 남아 있었고, 항원성 역시 대조군에 비하여 감소는 하였으나 여전히 유지되고 있었다. 120oC이상의 열처리는 추출물중의 BSA와 BGG band의 소실과 이에 따른 항원성의 큰 감소를 가져왔다. 우육추출물중의 BSA단백질은 열처리에서와 달리 초고압처리에 의하여 전기영동상에 거의 변화를 보이지 않았다. 그러나 BGG 단백질은 400 MPa 가압처리군부터 전기영동상 band가 감소하였고, 항원성 역시 유의적으로 감소하였다. 이는 초고압처리 정도에 따른 우육추출물중의 BSA와 BGG를 포함한 여러 분자의 SDS-PAGE상의 특이적 변화가 최초로 시사된 결과이다. 이러한 BGG 단백질의 고압처리에 의한 변화는 추출물중의 어떤 효소에 의한 것으로 추측되었고, 고압처리 후 37oC숙성 실험으로 이러한 가설이 확인되었다. 우육추출물에 존재하고 초고압처리로 활성화되어 BGG분자에 특이적으로 관여하는 효소 및 이와 관련된 메커니즘에 대한 연구가 앞으로 필요할 것으로 보인다.

      • Heat-Shock Protein 70 as a Tumor Antigen for in vitro Dendritic Cell Pulsing in Renal Cell Carcinoma Cases

        Immunological functions of heat shock proteins (HSPs) have long been recognized. In this study we aimed to efficiently purify HSP70 from renal cell carcinoma and test it as a tumor antigen for pulsing dendritic cells in vitro. HSP70 was purified from renal cell carcinoma specimens by serial column chromatography on Con A-sepharose, PD-10, ADP-agarose and DEAE-cellulose, and finally subjected to fast protein liquid chromatography (FPLC). Dendritic cells derived from the adherent fraction of peripheral blood mononuclear cells were cultured in the presence of IL-4 and GM-CSF and exposed to tumor HSP70. After 24 hours, dendritic cells were phenotypically characterized by flow cytometry. T cells obtained from the non-adherent fraction of peripheral blood mononuclear cells were then co-cultured with HSP70-pulsed dendritic cells and after 3 days T cell cytotoxicity towards primary cultured renal cell carcinoma cells was examined by Cell Counting Kit-8 assay. Dendritic cells pulsed in vitro with tumor-derived HSP70 expressed higher levels of CD83, CD80, CD86 and HLA-DR maturation markers than those pulsed with tumor cell lysate and comparable to that of dendritic cells pulsed with tumor cell lysate plus TNF-${\alpha}$. Concomitantly, cytotoxic T-lymphocytes induced by HSP70-pulsed dendritic cells presented the highest cytotoxic activity. There were no significant differences when using homologous or autologous HSP70 as the tumor antigen. HSP70 can be efficiently purified by chromatography and induces in vitro dendritic cell maturation in the absence of TNF-${\alpha}$. Conspecific HSP70 may effectively be used as a tumor antigen to pulse dendritic cells in vitro.

      • SCYL1BP1 has Tumor-suppressive Functions in Human Lung Squamous Carcinoma Cells by Regulating Degradation of MDM2

        SCY1-like 1-binding protein 1 (SCYL1BP1) is a newly identified transcriptional activator domain containing protein with many unknown biological functions. Recently emerging evidence has revealed that it is a novel regulator of the p53 pathway, which is very important for the development of human cancer. However, the effects of SCYL1BP1 on human lung squamous carcinoma cell biological behavior remain poorly understood. In this study, we present evidence that SCYL1BP1 can promote the degradation of MDM2 protein and further inhibit the G1/S transition of lung squamous carcinoma cell lines. Functional assays found that reintroduction of SCYL1BP1 into lung squamous carcinoma cell lines significantly inhibited cell proliferation, migration, invasion and tumor formation in nude mice, suggesting strong tumor suppressive function of SCYL1BP1 in lung squamous carcinoma. Taken together, our data suggest that the interaction of SCYL1BP1/MDM2 could accelerate MDM2 degradation, and may function as an important tumor suppressor in lung squamous carcinomas.

        SiOx interlayer to enhance the performance of InGaZnO-TFT with AlOx gate insulator

        We have fabricated indium-gallium-zinc (IGZO) thin film transistor (TFT) using SiOx interlayer modified aluminum oxide (AlOx) film as the gate insulator and investigated their electrical characteristics and bias voltage stress. Compared with IGZO-TFT with AlOx insulator, IGZO-TFT with AlOx/SiOx insulator shows superior performance and better bias stability. The saturation mobility increases from 5.6 cm2/V s to 7.8 cm2/V s, the threshold voltage downshifts from 9.5 V to 3.3 V, and the contact resistance reduces from 132 Ucm to 91 Ucm. The performance improvement is attributed to the following reasons: (1) the introduction of SiOx interlayer improves the insulator surface properties and leads to the high quality IGZO film and low trap density of IGZO/insulator interface. (2) The better interface between the channel and S/D electrodes is favorable to reduce the contact resistance of IGZO-TFT.
