        Molecular Cloning and Functional Characterization of FOUR LIPS in Pomegranate, a Protein Involved in Regulating the Gravitropic Set-Point Angle of Adventitious Roots

        Self-rooted pomegranate seedlings are widely used in the horticultural industry to cut costs and time. However, these seedlings produce shallow roots that exhibit poor cold resistance. Thus, deeper adventitious roots generated through gravitropism are imperative for seedling survival, and understanding the molecular mechanisms of gravitropism can facilitate improved breeding techniques. We hypothesized that gravitropism in pomegranate is partially controlled by pomegranate FOUR LIPS (PgFLP), an R2R3-MYB protein that interacts with and controls the transcriptional expression of PgPIN10, which facilitates transmembrane auxin signaling. We studied subcellular localization of PgFLP, quantified auxin levels, and measured gravitropic set-point angle (GSA) to investigate the underlying mechanisms regulating PgPIN10 expression during the formation of GSA in pomegranate adventitious roots. We found that PgFLP was localized to the nucleus based on use of green florescent proteins, suggesting that this protein is a transcription factor. When using the tractable 35S::PgFLP, we observed stronger gravitational response in overexpression lines leading to a narrower GSA than in the wild-type Arabidopsis, and the expression of PgFLP and PgPIN10 in ‘Lanbaoshi’ (LBS; a deep-rooted cultivar) was higher than that in ‘Taishanhong’ (TSH; a shallow-rooted cultivar), which indicates that PgFLP may participate in regulating the GSA of adventitious roots via PgPIN10 in pomegranate. Taken together, our results indicate that the pomegranate R2R3-MYB transcription factor, PgFLP, plays a vital role in setting the GSA of adventitious roots in this crop species.

        A New Fine-grain SMS Corpus and Its Corresponding Classifier Using Probabilistic Topic Model

        Nowadays, SMS spam has been overflowing in many countries. In fact, the standards of filtering SMS spam are different from country to country. However, the current technologies and researches about SMS spam filtering all focus on dividing SMS message into two classes: legitimate and illegitimate. It does not conform to the actual situation and need. Furthermore, they are facing several difficulties, such as: (1) High quality and large-scale SMS spam corpus is very scarce, fine categorized SMS spam corpus is even none at all. This seriously handicaps the researchers’ studies. (2) The limited length of SMS messages lead to lack of enough features. These factors seriously degrade the performance of the traditional classifiers (such as SVM, K-NN, and Bayes). In this paper, we present a new fine categorized SMS spam corpus which is unique and the largest one as far as we know. In addition, we propose a classifier, which is based on the probability topic model. The classifier can alleviate feature sparse problem in the task of SMS spam filtering. Moreover, we compare the approach with three typical classifiers on the new SMS spam corpus. The experimental results show that the proposed approach is more effective for the task of SMS spam filtering.

        Parasite-insecticide interactions: The impact of Nosema ceranae and fenpyroximate on honeybees (Apis mellifera)

        Honeybees are susceptible to parasitic infections. As a consequence, beekeepers often apply pesticides in their hives. This approach will also affect the health of bees while eliminating parasites. We used Nosema ceranae (2 × 10 7 spores/mL), 0.002 % fenpyroximate, and the combination of N. ceranae-fenpyroximate to respectively treat newly emerged worker bees, to assess their survival curve, spore number and the expression levels of genes related to oxidative stress in the midguts. As a result, chronic exposure to N. ceranae and/or fenpyroximate significantly decreased honeybee survival. However, there were no different effects between the N. ceranae and fenpyroximate treatments (P > 0.05). Fenpyroximate inhibited the reproduction of N. ceranae in the early stage, but the number of N. ceranae increased gradually in the later stage (P < 0.01). Furthermore, the expression levels of genes related to oxidative stress were constantly changing with time under different treatments. In particular, the expression of the catalase gene was upregulated, and that of the ROS modulator 1-like gene and the superoxide dismutase gene was downregulated. In conclusion, this study indicated that the negative impact of N. ceranae and/or fenpyroximate on the health of honeybees caused the activation of their antioxidant system. Moreover, the redox status of the bees could increase their susceptibility and mortality.

        Beginning from the year 1990, various regions in South Korea paid more attention to make space for revitalisation. Murals were created in approximately 10 villages and because of the efforts made, the public art project made the old villages come back to life. The conclusion of this research was according to the advanced research result to conclusion about the space improvement at the village. The places of this study: AntVillage(Hongje-dong), Ewha Village(Ihwa-dong), Kang Full cartoon street(Gangdong-gu). First, the murals village used art works and plastic arts to ornamental space make it more vivid . Secondly, artists, students and local residents worked together to create themed murals with hopes and environmental protection. Thirdly, the mural village line is not clear. It has messy bare wires that cause visitors to cause disturbance to the lives of the residents during the tour.So there is an urgent need for post management.

        Effect of serotonin on the cell viability of the bovine mammary alveolar cell-T (MAC-T) cell line

        5-Hydroxytryptamine (5-HT), a monoamine, as a local regulator in the mammary gland is achemical signal produced by the mammary epithelium cell. In cows, studies have shown that5-HT is associated with epithelial cell apoptosis during the degenerative phase of the mammarygland. However, studies in other tissues have shown that 5-HT can effectively promotecell viability. Whether 5-HT could have an effect on mammary cell viability in dairy cows isstill unknown. The purpose of this study was to determine: (1) effect of 5-HT on the viability ofbovine mammary epithelial cells and its related signaling pathways, (2) interaction betweenprolactin (PRL) and 5-HT on the cell viability. The bovine mammary alveolar cell-T (MAC-T)were cultured with different concentrations of 5-HT for 12, 24, 48 or 72 hours, and then wereassayed using cell counting kit-8, polymerase chain reaction (PCR) and immunobloting. Theresults suggested that 20 μM 5-HT treatment for 12 or 24 h promote cell viability, which wasmainly induced by the activation of 5-HT receptor (5-HTR) 1B and 4, because the increasecaused by 5-HT vanished when 5-HTR 1B and 4 was blocked by SB224289 and SB204070. And protein expression of mammalian target of rapamycin (mTOR), eukaryotic translationelongation factor 2 (eEF2), janus kinase 2 (JAK2) and signal transducer and activator of transcription5 (STAT5) were decreased after blocking 5-HT 1B and 4 receptors. When MAC-Tcells were treated with 5-HT and PRL simultaneously for 24 h, both the cell viability and thelevel of mTOR protein were significantly higher than that cultured with 5-HT or PRL alone. Inconclusion, our study suggested that 5-HT promotes the viability of MAC-T cells by 5-HTR1B and/or 4. Furthermore, there is a reciprocal relationship between PRL and 5-HT.

        Transcriptome analysis of response strategy in Hemerocallis fulva under drought stress

        Background Hemerocallis fulva is an important ground cover plant widely used in urban greening. The analysis of the molecular mechanism underlying the drought response of H. fulva can lay a foundation for improving its adaptability and expanding its planting area. Objective To reveal the drought response mechanisms of H. fulva, identify candidate unigenes associated with drought response, and lay a foundation for further unigenes functional study and drought resistance improvement of H. fulva via genetic engineering. Methods RNA was isolated from H. fulva under different experimental conditions. De novo transcriptomic analysis of the samples was performed to screen drought response unigenes. The transcriptional changes of candidate drought response unigenes were verified by quantitative real-time PCR. Results The differentially expressed unigenes and their functions were analyzed after H. fulva treated by PEG-simulated drought stress and rewatering. The candidate unigenes, associated with H. fulva drought response, were identified after transcriptome analysis. Then, the transcription level of drought response unigenes of H. fulva under different conditions was further verified. Abscisic acid, protein phosphorylation, sterol biosynthesis and ion transport were involved in drought response with quick restore in H. fulva. The response unigenes, involved in hormone (ABA, JA, CK and GA) signaling pathways, defense response, high light response, karrikin response and leaf shaping, can maintain at changed expression levels even after stress withdraw. Conclusion Hemerocallis fulva has unique drought response mechanism. Negative regulation mechanism may play more important roles in drought response of H. fulva. The analysis of candidate unigenes, associated with drought response, lays a foundation for further drought resistance improvement of H. fulva.

        Functional significance of cholesterol metabolism in cancer: from threat to treatment

        Cholesterol is an essential structural component of membranes that contributes to membrane integrity and fluidity. Cholesterol homeostasis plays a critical role in the maintenance of cellular activities. Recently, increasing evidence has indicated that cholesterol is a major determinant by modulating cell signaling events governing the hallmarks of cancer. Numerous studies have shown the functional significance of cholesterol metabolism in tumorigenesis, cancer progression and metastasis through its regulatory effects on the immune response, ferroptosis, autophagy, cell stemness, and the DNA damage response. Here, we summarize recent literature describing cholesterol metabolism in cancer cells, including the cholesterol metabolism pathways and the mutual regulatory mechanisms involved in cancer progression and cholesterol metabolism. We also discuss various drugs targeting cholesterol metabolism to suggest new strategies for cancer treatment.

        A fluorescent probe for colorimetric detection of bisulfite and application in sugar and red wine

        A new fluorescent probe made from (E)-2-(benzo[d]thiazol-2-yl)-3-(6-hydroxynaphthalen-2-yl) acrylonitrile(Probe 1) was synthesized for the determination ofbisulfite concentrations in real food samples (red wine andsugar). Adding bisulfite to a Probe 1 solution caused amarked decrease in fluorescence intensity and a visualcolor change from yellow to light yellow. This distinctcolor response indicates that Probe 1 could be used as avisual sensor for bisulfite. Probe 1 can detect bisulfitequantitatively in the range 0–400 lM with a detection limitof 0.10 lM. This makes Probe 1 a convenient signalinginstrument for determining bisulfite levels in sugar and redwine samples.
