        A new method for immobilization of lipid-binding proteins to the glass slides

        <P><B>Abstract</B></P><P>Previously, we constructed nickel-coated glass slides to immobilize histidine-tagged proteins [Bull. Korean Chem. Soc. 23 (2002) 1724–1728]. Here, we further investigated whether the nickel-coated glass slide could be utilized for assaying molecular interaction between lipid and immobilized protein. Proteins interact with various lipid molecules in biological systems. More importantly, this interaction is responsible for a broad spectrum of physiological functions. Accordingly, to more efficiently analyze various types of interactions and understand the complicated biological systems, it is essential to develop a high-throughput analytical device. For the high-throughput analytical purpose, we immobilized an FITC (fluorescein isothiocyanate)-tagged lipid-binding protein to nickel-coated plates, incubated the plates with various lipid molecules and then measured fluorescence intensities to analyze its binding affinity. Our systems seemed to be reliable for the application to the high-throughput system consisted of lipid-binding proteins.</P>

      • A Study on the Discovery of IL-31 Receptor A in Astrocytes and Effect of Diospyros lotus Extract on IL-31-Mediated Pruritus in Microglia and Astrocytes

        Interleukin-31 is associated with hypersensitivity disorders such as atopic dermatitis, allergic rhinitis and asthma. IL-31 is also known to be one of the major "drivers" of pruritus. In the nervous system, pruritus may be enhanced through IL-31-related interactions. However, there were no reports of IL-31 receptor expression in astrocytes. In this study, the expression of IL-31 receptor in astrocytes and the effect of Diospyros lotus extract (DLE) on pruritus factors between microglia and astrocytes were investigated. As a result, the expression of IL-31 receptor A in astrocytes was discovered for the first time. Pretreatment with DLE suppressed the expression of IL-31 receptor A in IL-31-treated astrocytes. On the other hand, pretreatment of DLE in microglia inhibits lipopolysaccharide (LPS) plus interferon gamma (IFN-γ)-induced IL-31 production. DLE inhibited phosphorylation of MAPK, STAT1 and NF-κB in LPS plus IFN-γ stimulated microglia. Taken together, DLE down-regulated IL-31-mediated pruritus factors in both astrocytes and microglia. Therefore, the results of this study suggest the potential of DLE as a material that can relieve chronic itch conditions.

        CASE REPORT : Neuropathic Itch of the Back: A Case of Notalgia Paresthetica

        Notalgia paresthetica refers to an isolated mononeuropathy involving chronic localized itch or paresthesia most often at the skin of the scapula or surrounding regions. There are no specific skin manifestations except those arising from chronic scratching and rubbing. The specific etiology remains unknown; however, it has been theorized that the neuropathic itch is caused by sensory nerve entrapment involving the posterior rami of the T2 to T6 nerve root. The entrapment is due to degenerative changes in the vertebrae. We report here a particular case of notalgia paresthetica in a 55-year-old woman. The patient visited our hospital for tingling pain around the left inferior angle of the scapula. Pruritus was first reported seven years ago with tingling pain developing only four months ago. There were no specific skin lesions observed except for excoriation and vague hyperpigmentation. A skin biopsy revealed only epidermal thinning with pigmentary incontinence. The patient was treated with 600 mg of gabapentin daily as well as capsaicin cream. The response was deemed unsatisfactory. (Ann Dermatol 26(3) 392∼394, 2014)

        Feasibility of local interpretable model‑agnostic explanations (LIME) algorithm as an effective and interpretable feature selection method: comparative fNIRS study

        Many feature selection methods have been evaluated in functional near-infrared spectroscopy (fNIRS)-related studies. Thelocal interpretable model-agnostic explanation (LIME) algorithm is a feature selection method for fNIRS datasets that hasnot yet been validated; the demand for its validation is increasing. To this end, we assessed the feature selection performanceof LIME for fNIRS datasets in terms of classification accuracy. A comparative analysis was conducted for the benchmark(classification accuracy obtained without applying any feature selection method), LIME, two filter-based methods (minimumredundancymaximum-relevance and t-test), and one wrapper-based method (sequential forward selection). To ensure thefairness and reliability of the performance evaluation, several open-access fNIRS datasets were used. The analysis revealedthat LIME greatly outperformed the other feature selection methods in most cases and could achieve a statistically significantlybetter classification accuracy than that of the benchmark methods. These findings implied the effectiveness of LIMEas a feature selection approach for fNIRS datasets.

        Investigation of the Lower Bound of Outage Threshold in Indoor Power-Line Networks

        <P>We theoretically investigate lower bound of the outage threshold based on channel capacity degradation in indoor power-line networks and its practical applicability to a realistic indoor power-line network existing in practice. We estimate the channel capacity degradations of arbitrary channels in networks with uniformly distributed branches with regular length. The channel capacity degradations are estimated under various network configurations, such as network size, branch characteristics (including the number and length), and loading conditions (including the number, type, impedance, and location). Based on the channel capacity degradation, the lower bounds of the outage threshold of arbitrary channels, which can evaluate the quality of the channels, are given. To evaluate the practical applicability of the simulated results, we compare the lower bounds of the outage threshold between a realistic network and simple networks. It is shown that the Pearson's correlation coefficients between the realistic network and the simple networks are predominantly greater than 0.85 (-value 0.01) in almost all cases, which shows a strong correlation in terms of the outage behavior. Therefore, our investigation enables estimating the outage behavior of a realistic network, without sufficient prior information of the network configuration using a simple network with a similar network configuration.</P>

      • A Ternary Hybrid EEG-NIRS Brain-Computer Interface for the Classification of Brain Activation Patterns during Mental Arithmetic, Motor Imagery, and Idle State

        <P>The performance of a brain-computer interface (BCI) can be enhanced by simultaneously using two or more modalities to record brain activity, which is generally referred to as a hybrid BCI. To date, many BCI researchers have tried to implement a hybrid BCI system by combining electroencephalography (EEG) and functional near-infrared spectroscopy (NIRS) to improve the overall accuracy of binary classification. However, since hybrid EEG-NIRS BCI, which will be denoted by hBCI in this paper, has not been applied to ternary classification problems, paradigms and classification strategies appropriate for ternary classification using hBCI are not well investigated. Here we propose the use of an hBCI for the classification of three brain activation patterns elicited by mental arithmetic, motor imagery, and idle state, with the aim to elevate the information transfer rate (ITR) of hBCI by increasing the number of classes while minimizing the loss of accuracy. EEG electrodes were placed over the prefrontal cortex and the central cortex, and NIRS optodes were placed only on the forehead. The ternary classification problem was decomposed into three binary classification problems using the “one-versus-one” (OVO) classification strategy to apply the filter-bank common spatial patterns filter to EEG data. A 10 × 10-fold cross validation was performed using shrinkage linear discriminant analysis (sLDA) to evaluate the average classification accuracies for EEG-BCI, NIRS-BCI, and hBCI when the meta-classification method was adopted to enhance classification accuracy. The ternary classification accuracies for EEG-BCI, NIRS-BCI, and hBCI were 76.1 ± 12.8, 64.1 ± 9.7, and 82.2 ± 10.2%, respectively. The classification accuracy of the proposed hBCI was thus significantly higher than those of the other BCIs (<I>p</I> < 0.005). The average ITR for the proposed hBCI was calculated to be 4.70 ± 1.92 bits/minute, which was 34.3% higher than that reported for a previous binary hBCI study.</P>

      • Evaluation of a Compact Hybrid Brain-Computer Interface System

        <P>We realized a compact hybrid brain-computer interface (BCI) system by integrating a portable near-infrared spectroscopy (NIRS) device with an economical electroencephalography (EEG) system. The NIRS array was located on the subjects' forehead, covering the prefrontal area. The EEG electrodes were distributed over the frontal, motor/temporal, and parietal areas. The experimental paradigm involved a Stroop word-picture matching test in combination with mental arithmetic (MA) and baseline (BL) tasks, in which the subjects were asked to perform either MA or BL in response to congruent or incongruent conditions, respectively. We compared the classification accuracies of each of the modalities (NIRS or EEG) with that of the hybrid system. We showed that the hybrid system outperforms the unimodal EEG and NIRS systems by 6.2% and 2.5%, respectively. Since the proposed hybrid system is based on portable platforms, it is not confined to a laboratory environment and has the potential to be used in real-life situations, such as in neurorehabilitation.</P>

        Association between Ultrasonography Findings and Abnormal Karyotypes in Early Pregnancy Loss

        Objective: Chromosomal abnormality in the fetus is a major cause of early pregnancy loss (EPL). It is considered that maternal age is a risk factor of chromosomal abnormality in the fetus. The objective of this study was to evaluate the association between ultrasonography findings and abnormal karyotypes in EPL. Methods: This retrospective analysis assessed 217 cases of EPL occurring between 2009 and 2018, which have the results of cytogenetic analysis following miscarriage, as well as the ultrasonography finding. The correlations between the ultrasonography findings and the karyotypes were evaluated. Results: Of the 217 cases, after excluding cases with no mitotic cells, karyotypes analysis was performed in 190 cases. The overall rate of abnormal karyotypes was 32.1% (61/190). Mean maternal age was significantly higher in the abnormal karyotype group (35.66±4.22 vs. 33.79±4.02 years, respectively, P=0.006). The embryo/gestational sac ratios was significantly smaller in the abnormal karyotype group (0.3±0.18 vs. 0.43±0.3, P=0.022). In the multivariate logistic regression analysis, smaller embryo/gestational sac ratios (≤0.4) was a significant risk factor of abnormal karyotype (adjusted odds ratio [OR] 2.43, 95% confidence interval [CI]: 1.086-5.437, P=0.031), after the adjustment with the number of previous abortion. The chromosomal abnormality rate was significantly higher in male miscarriage (adjusted OR 2.36, 95% CI: 1.003-5.443, P=0.049). Conclusion: By identifying a predictive factor of chromosomal abnormalities in embryonic abortion, this study presented ultrasonography findings to consider cytogenetic analysis and an easy-to-use cut off value, small embryo/gestational sac ratio (≤0.4) at a patient’s request of chromosomal study in the fetus.
