        A database of 5305 healthy Korean individuals reveals genetic and clinical implications for an East Asian population

        Despite substantial advances in disease genetics, studies to date have largely focused on individuals of European descent. This limits further discoveries of novel functional genetic variants in other ethnic groups. To alleviate the paucity of East Asian population genome resources, we established the Korean Variant Archive 2 (KOVA 2), which is composed of 1896 whole-genome sequences and 3409 whole-exome sequences from healthy individuals of Korean ethnicity. This is the largest genome database from the ethnic Korean population to date, surpassing the 1909 Korean individuals deposited in gnomAD. The variants in KOVA 2 displayed all the known genetic features of those from previous genome databases, and we compiled data from Korean-specific runs of homozygosity, positively selected intervals, and structural variants. In doing so, we found loci, such as the loci of ADH1A/1B and UHRF1BP1, that are strongly selected in the Korean population relative to other East Asian populations. Our analysis of allele ages revealed a correlation between variant functionality and evolutionary age. The data can be browsed and downloaded from a public website (https://www.kobic.re.kr/kova/). We anticipate that KOVA 2 will serve as a valuable resource for genetic studies involving East Asian populations.

      • Evaluation of the effects of food on levodropropizine controlled-release tablet and its pharmacokinetic profile in comparison to that of immediate-release tablet

        <P><B>Background</B></P><P>Levodropropizine is a non-opioid antitussive agent that inhibits cough reflex by reducing the release of sensory peptide in the peripheral region. To improve patients’ compliance, a controlled-release (CR) tablet is under development. The aim of this study was to compare the pharmacokinetic (PK) profiles of the CR and immediate-release (IR) tablets of levodropropizine. In addition, the effect of food on the PK properties of levodropropizine CR tablet in healthy subjects was evaluated.</P><P><B>Subjects and methods</B></P><P>A randomized, open-label, multiple-dose, three-treatment, three-period, six-sequence, crossover study was conducted on 47 healthy subjects. All subjects were randomly assigned to one of the six sequences, which involve combinations of the following three treatments: levodropropizine IR 60 mg three times in the fasted state (R), levodropropizine CR 90 mg two times in the fasted state (T), and levodropropizine CR 90 mg two times in the fed state (TF). Serial blood samples were collected up to 24 h after the first dose. Tolerability was assessed based on the vital signs, adverse events (AEs), and clinical laboratory tests.</P><P><B>Results</B></P><P>Levodropropizine CR showed lower maximum drug concentration (<I>C</I><SUB>max</SUB>) and similar total exposure compared to levodropropizine IR. The geometric mean ratios (GMRs) (90% confidence intervals [CIs]) of T to R for the <I>C</I><SUB>max</SUB> and area under the concentration–time curve from the 0 to 24 h time points (AUC<SUB>0–24h</SUB>) were 0.80 (0.75–0.85) and 0.89 (0.86–0.93), respectively. In the fed group, levodropropizine CR showed exposure similar to that in the fasted group. The GMRs (90% CIs) of TF to T for the <I>C</I><SUB>max</SUB> and AUC<SUB>0–24h</SUB> were 0.90 (0.85–0.97) and 1.10 (1.05–1.14), respectively. No serious AEs occurred with both levodropropizine CR and IR tablets.</P><P><B>Conclusion</B></P><P>Total systemic exposure for levodropropizine was similar in subjects receiving the CR and IR formulations in terms of the AUC. Although food delayed the absorption of levodropropizine CR, systemic exposure was not affected.</P>

        Sharpened VO<sub>2</sub> Phase Transition via Controlled Release of Epitaxial Strain

        <P>Phase transitions in correlated materials can be manipulated at the nanoscale to yield emergent functional properties, promising new paradigms for nanoelectronics and nanophotonics. Vanadium dioxide (VO2), an archetypal correlated material, exhibits a metal insulator transition (MIT) above room temperature. At the thicknesses required for heterostructure applications, such as an optical modulator discussed here, the strain state of VO2 largely determines the MIT dynamics critical to the device performance. We develop an approach to control the MIT dynamics in epitaxial VO2 films by employing an intermediate template layer with large lattice mismatch to relieve the interfacial lattice constraints, contrary to conventional thin film epitaxy that favors lattice match between the substrate and the growing film. A combination of phase-field simulation, in situ real-time nanoscale imaging, and electrical measurements reveals robust undisturbed MIT dynamics even at preexisting structural domain boundaries and significantly sharpened MIT in the templated VO2 films. Utilizing the sharp MIT, we demonstrate a fast, electrically switchable optical waveguide. This study offers unconventional design principles for heteroepitaxial correlated materials, as well as novel insight into their nanoscale phase transitions.</P>

        근로환경에서의 위험노출 정도에 관한 연구

        본 연구는 산업안전보건공단의 근로환경조사(Korean Working Conditions Survey) 4차(2014년) 자료를 이용하여 고용형태별(비정규직 세분류; 기간제, 비기간제, 시간제, 파견, 용역, 특수형태, 가정 내, 일일)로 근로환경에서의 위험노출 정도가 어떻게 차이가 나는지 ordered logit 모형을 이용하여 예측 확률(predicted probability)을 추정하였다. 분석 결과 첫째, 대부분의 위험요소영역에서 비정규직 근로자가 정규직 근로자에 비해 상대적으로 위험에 노출될 확률이 높은 것으로 나타났으며, 둘째, 비정규직 유형 가운데서도 비전형근로, 특히 용역, 일일(호출), 특수형태근로 종사자의 위험노출 확률이 상대적으로 높았다. 셋째, 각각의 위험요소별로 위험노출 확률에 대한 정도가 고용형태에 따라 차이가 있는 것으로 나타났으며, 근로환경 측면에서는 용역근로, 육체적 근로방식에 있어서는 일일(호출)근로, 정신적 근로방식에 있어서는 파견이나 기간제, 특수형태근로, 시간제 등의 고용형태에서 위험노출 확률이 큰 것으로 나타났다. 이와 같은 실증분석 결과는 비정규직이 정규직에 비해 일자리의 안정성뿐만 아니라 일자리의 안전성도 취약함을 의미하고, 비정규직에 있어서 일자리에서의 안전 강화를 위한 지원이 필요함을 시사한다. This study estimates the predicted probabilities of risk-exposure degree in working environment by employment type such as temporary workers, non-standard workers, non-regular workers, and atypical workers applying order logit analysis, using the 4th wave of Korean Working Conditions Survey (KWCS) which is conducted by Korea Occupational Safety and Health Agency (KOSHA) in South Korea. The empirical results are as follow: first, non-stand workers are likely to be relatively more exposed to risk than standard in most risk factors. Second, among non-standard workers, the atypical workers are likely to be relatively more exposed to risk. Third, subcontracting workers are the most likely to be exposed to risk in case of working conditions, one days workers are the most likely to be exposed to risk in case of physical working methods, and dispatch workers and part time workers are the most likely to be exposed to risk in case of psychological working methods. These results indicate that non-standard workers are more vulnerable than standard workers in job safety as well as job security. Also, in the case of non-standard workers, it is required to intensify the safety in working environment.

        Safety, Tolerability, and Pharmacokinetic Characteristics of a Novel Nonopioid Analgesic, VVZ-149 Injections in Healthy Volunteers: A First-in-Class, First-in-Human Study

        <P>VVZ-149, a dual antagonist of GlyT2 and 5HT(2)A receptors, is an investigational analgesic with a novel mechanism of action that is currently under early-stage clinical development as an injectable agent for the treatment of postoperative pain. Here, the safety, tolerability, and pharmacokinetics of VVZ-149 injections in healthy male volunteers were explored in a randomized, double-blind, single- and multiple-ascending-dose (SAD and MAD, respectively), placebo-controlled clinical study. Subjects randomly received a 4-hour intravenous infusion of 0.25-8 mg/kg VVZ-149 or placebo in the SAD study (n = 46) or a 4-hour intravenous infusion of 4-7 mg/kg VVZ-149 or placebo twice daily for 3 days in the MAD study (n = 20). Serial blood and urine samples were collected for the pharmacokinetic analysis of VVZ-149 and its active metabolite (VVZ-368). Noncompartmental and compartmental pharmacokinetic analyses were performed. Various dosing scenarios were simulated to identify the adequate dosing regimen for the subsequent trials. Plasma exposure to VVZ-149 and VVZ-368 showed a dose-proportional increase. VVZ-149 did not accumulate in the plasma, whereas the plasma concentration of VVZ-368 increased by 1.23- to 2.49-fold after the fifth and sixth doses, respectively, in the MAD trial. Among the simulated dosing regimens, a loading dose followed by a maintenance dose was found to be an optimal dosing regimen, yielding the effective concentration estimated from animal studies in rat models of neuropathic or inflammatory pain. Single- or multiple-dose administration of VVZ-149 was generally well tolerated. These results showed that 0.5-8 mg/kg VVZ-149 exhibited linear pharmacokinetic characteristics and can be safely administered in further clinical studies.</P>

        The effect of short-term creatine intake on blood lactic acid and muscle fatigue measured by accelerometer-based tremor response to acute resistance exercise

        [Purpose] The purpose of this study was to investigate the effects of short-term creatine intake on muscle fatigue induced by resistance exercise in healthy adolescent men, i.e., lactic acid concentration and wrist and head tremor measured by an accelerometer. [Methods] Twelve healthy adolescent men who had no experience with creatine intake were included. The subjects were randomly assigned to the creatine group and the placebo group, followed by 5 days of creatine and placebo intake, and 5 times of 5 sets of leg press, leg extension, bench press, and arm curl exercises at 70% repetition maximum (RM). The lactic acid concentration before and after exercising, rate of perceived exertion (RPE), and accelerometer-based wrist tremor and head tremor during exercise were measured. Subsequently, after 7 days to allow for creatine washout, the same exercise treatment and measurement were performed in each group after switching drug and placebo between the groups. [Results] The level of lactic acid before and after the acute resistance exercise trial was significantly lower in the creatine group than in the placebo group (P <0.05). The mean RPE during the resistance exercise was significantly lower in the creatine group than in the placebo group (P <0.05). There was no difference between the two groups in the mean wrist tremor during resistance exercise, but the mean head tremor values were significantly lower in the creatine group than in the placebo group in the arm curl, the last event of the exercise trials (P <0.05). [Conclusion] Short-term creatine intake reduces the blood fatigue factor increased by resistance exercise, and is thought to suppress fatigue, especially in the latter half of resistance exercise. Therefore, these findings indicate that short-term creatine intake can have an improved effect on anaerobic exercise performance.

        IP기업의 기술적 다각화 전략을 통한 비체계적 위험의 분산 효과에 관한 실증연구

        본 연구는 특허 출원을 통해 기업의 지식재산을 권리화하고 이를 통해 기업의 경쟁력을 제고시키는 IP기업에 대한 기술적 다각화 효과에 관해 연구한다. 기존의 선행연구들과 달리 본 연구에서는 기술적 다각화를 통한 기업성과로 재무적 성과, 기술적 성과, 사회적 성과 등을 사용하지 않고 비체계적 위험의 분산 효과로 설정했다는 차별점이 있다. 이러한 효과를 살펴보기 위해 본 연구에서는 엔트로피 기반의 방식으로 기술적 다각화 지수를 측정하고 이를 관련 다각화 지수와 비관련 다각화 지수로 구분했다. 그리고 이상의 다각화 지수에 대한 비체계적 위험의 분산 효과를 시간의 흐름에 따라 동태적으로 실증했다는 의의가 있다. 그 결과, 비관련 기술 다각화 전략만 IP 기업의 비체계적 위험의 분산 효과를 시간의 흐름에 따라 정(+)의 효과로 바꾸는 동적 효과가 있는 것으로 나타났다. 이는 국가과제발주 시에 입찰기업 평가지표로써 주로 요구하는 관련 기술개발 경험과 비교하면 그동안 상대적으로 외면해왔던 기업의 비관련 기술개발 경험의 가치를 정책 실무자들에게 새롭게 시사함으로써 잠재적 혁신기업의 육성에 이바지한다. This paper studies the effects of technological diversification on IP companies that make a company’s intellectual property rights through patent applications and thereby enhance the company’s competitiveness. Unlike previous prior studies, there is a difference in this study that the company’s performance through technological diversification was set as a decentralized effect of unsystematic risks without using financial performance, technical performance, and social performance. To examine these effects, in this work, the technological diversification index was measured in an entropy-based manner and it was divided into related and unrelated diversification indices. It is also meaningful to demonstrate dynamically the dispersion effect of unsystematic risks on the above diversification index over time. As a result, it is shown that only unrelated technology diversification strategies have a dynamic effect that transforms the dispersion effect of unsystematic risks of IP firms into the positive effect over time. This contributes to fostering potential innovators by suggesting to policy practitioners the value of unrelated technology development experiences that have been relatively neglected compared to the relevant technology development experience required by the state.
