Selective COX (cyclooxygenase)-2 inhibitors including celecoxib have been shown to induce apoptosis and cell cycle changes in various tumor cells. New inhibitors are recently being developed as chemomodulating agents. We evaluated celecoxib and screened 150 synthetic compounds for anti-proliferative activities in vitro. Effects of celecoxib on COX activity, cell growth, cell cycle distribution, and apoptosis induction were determined in A549 COX-2 overexpressing human non-small cell lung cancer (NSCLC) cells. The COX inhibition of celecoxib increased with concentration up to 82% at 1μM after 24 hr exposure. Forty μM and 50μM of celecoxib induce G_1 arrest, and TUNEL-positive apoptotic cells, respectively. Among 150 compounds, several compounds were selected for having greater COX-2 inhibitory activity and higher selectivity than celecoxib with growth inhibitory activity. Celecoxib showed concentration-dependent COX inhibitory activity, and ability to induce cell cycle arrest and apoptosis in human NSCLC cells in vitro. Among synthetic analogues screened, several compounds showed promising in vitro activity as COX-2 inhibitory anticancer agents, which warrant further evaluation in vitro and in vivo.
Assessment of Soil Compaction Related to the Bulk Density with Land use Types on Arable Land

Soil compaction is affected by soil texture, organic matter (OM), strength (ST) and soil moisture, which is difficult to understand the degree and effects of related factors. The purpose of the study is to assess the impact of them on the compaction with bulk density (BD). The analysis was conducted with data collected from national-wide monitoring sites including 105 upland soils, 246 orchard soils, and 408 paddy soils between 2009 and 2012. The distributions of soil physical properties were measured. The correlation and multi linear regression analysis were performed between soil physical properties using SAS. The regression equation of BD(y) includes ST, gravitational water contents (GWC), and OM as variables commonly, having additional factors, clay content and sand content in paddy soil and upland soil for only subsoil (p<0.001). Our results show that the BD could be explained about 40~50% by various physical properties. The regression was mainly determined by ST in orchard and upland soil and by the GWC in paddy soil. To mitigate soil compaction, it is important to maintain the proper level of OM in upland soil and to consider the moisture condition with soil texture in paddy soil when making work plan. Furthermore, it would be recommended the management criteria classified by soil texture for the paddy soils.

Background: Hepatic venous pressure gradient (HVPG) is the gold standard for assessing portal pressure. In this study, we investigated which noninvasive fibrosis test most effectively reflects HVPG and predicts prognosis in patients with alcoholic liver disease (ALD). Methods: A total of 195 consecutive patients with ALD were included. Biochemical indices and liver stiffness assessed by transient elastography (TE) were compared with HVPG. Results: During a median follow-up period of 23.1 months, 51 patients died, including 21 liver-related deaths. The diagnostic values of liver stiffness in detecting clinically significant portal hypertension (CSPH; HVPG≥10 mmHg) was significantly higher (AUROC=0.87±0.03) than those of biochemical indices (i.e. APRI, FIB4, P2/MS and platelet count/spleen diameter ratio; all P<0.001). In multivariate analysis, liver stiffness was most significantly correlated to HVPG (P<0.001), whereas other biochemical indices were not. On the other hand, the prognostic values of liver stiffness for liver-related death (AUROC= 0.73±0.07) did not differ from those of FIB4 (0.78±0.04), HVPG (0.70±0.07) and APRI (0.69±0.04). In multivariate analysis, significant risk factors for liver-related death were Child score (hazard ratio [HR]=2.35, P<0.001), varices >F2 (HR=5.85; P=0.002) and FIB4 (HR=1.11, P=0.03), but not liver stiffness and HVPG. For all-cause death, age and FIB4 were independent predictors in compensated patients (P=0.02 and <0.001, respectively), whereas Child score was in decompensated patients (P<0.001). Conclusions: In patients with ALD, liver stiffness most accurately predicts CSPH, but did not improve the prognostic values of traditional risk factors for mortality, whereas FIB4 was independent predictor for liver disease-related death and allcause death.

Post-translational modifications of nucleosomal core histones play important roles in biological processes via altering chromatin structure and creating target sites for proteins acting on chromatin. Molecular genetic studies with Arabidopsis have verified several epigenetic factors that regulate flowering time. However, the roles of chromatin remodeling factors have not been well explored in rice. Here, we identified chromatin remodeling factors, OsVIL1, 2, and 4 (Oryza sativa VIN3-LIKE) genes, that regulate grain yield. OsVIL proteins contain a plant homeodomain (PHD) finger, which is a conserved motif of histone binding proteins. We showed that plant height and number of spikelets per panicle were increased in the OsVIL2-overexpression (OsVIL2-OX) and osvil4 plants, respectively. Each mutants (OsVIL2-OX and osvil4) exhibited longer internodes and thicker stems than wild type controls. Histochemical analysis revealed that cells are smaller in OsVIL2-OX and osvil4 plants. We performed an RNA-seq using 1st internodes of WT and OsVIL2-OX stems and got the suppressed target genes in the OsVIL2-OX. OsCKX2, which encodes cytokinin oxidase/dehydrogenase is one of the suppressed genes in the OX plants and we verified decrease of that gene using qRT-PCR and closed chromatins of OsCKX2 were enriched in the OX plants by using ChIP. As results of these, cytokinins were enriched in the OX plants. These demonstrate that OsVIL2 and OsVIL4 antagonistically regulate plant height and number of spikelets by controlling cytokinin contents. Like OsVIL2-OX and osvil4 plants, besides, OsVIL1-OX plants were also shown increased plant height and biomass. We propose that OsVILs may be used for improving grain yield by increasing biomass.

<P>To enhance high power characteristics for piezoelectric transformer, an alloy design approach in PZT base ceramic system was considered. Various Zr/Ti ratio compositions in 0.03Pb(Sb0.5Nb0.5)O3- 0.03Pb(Mn1/3Nb2/3)O3-(0.94-x)PbTiO3-xPbZrO3 (0.445x0.475 mol)[PSN - PMN - PZT] system were synthesized by the conventional bulk ceramic processing technique. To improve high power characteristics at their morphotropic phase boundary (MPB) composition of PSN-PMN-PZT system, various spectra of Zr/Ti ratio were systematically experimented, and their effects on the subsequent piezoelectric properties and dielectric properties for high power piezoelectric transformer application were investigated using an impedance analyzer and a laser vibrometer. Microstructure and phase information were characterized using X-ray diffractometer (XRD) and a scanning electron microscope (SEM). When the Zr/Ti ratios were 0.475/0.465, the mechanical quality factor and electromechanical coupling factor were shown to reach the maximum, indicating that this alloy design can be a feasible composition for high power transformer.</P>
