        H9c2 심근 세포주에서 외인성 nitric oxide가 허혈에 의한 세포 독성에 미치는 영향

        Background: Nitric oxide(NO) is known to have protective effects on an ischemic heart and to exert triggering effects on ischemic preconditioning. However, the effects of NO during the ischemic period have not been investigated. To investigate the role of exogenous nitric oxide in a model of ischemic heart cell death, we studied the effects of ischemic preconditioning and ischemia in a normal and an ischemic buffer. Methods: Rat cardiac myoblast cells(H9c2) were cultured in a normal and an ischemic buffered medium. For the ischemic culture of heart cells, the cells were cultured in a dessicator with GasPak for 5 hrs. In ischemic preconditioning, the cells were pretreated with ischemic buffer for 5 min and then perfused with normal medium for 30 min. For the measurement of the cytotoxicity, a MTT(3-4-Sdimethylthiazol-2-yl-2,5-diphenyl tetrazolium bromide) assay was performed. A DAPI(4',6-diamidino-2-phenylindole dihydrochloride) staining procedure and a flow cytometry analysis were performed to confirm apoptotic cell death by ischemia. Results: Cell viability, as determined by using a MTT assay, showed that the preconditioned group treated with NO showed more cell death than with the not-preconditioned groups in both normal and ischemic buffers. But, In normal medium and not-preconditioned groups, NO showed protective effect according to the concentrations(100,1000μM) . No treatment with NO produced the different results. In normal medium, the protective effect of ischemic preconditioning was demonstrated, but no protective effect of ischemic preconditioning could be seen in the case of the ischemic buffer. The DAPI staining and flow cytometry analysis of heart cells showed characteristic apoptotic features. Conclusion: NO added in the ischemic phase had deterious effects on heart cells. Ischemic preconditioning was more harmful than ischemia alone. The toxicity of the cells was characteristic apoptosis.

        간호대학생의 MBTI 성격유형과 학습태도, 학업성취도간의 상관성

        Purpose: This study was to explore the relationship between personality type and learning attitude and academic achievement of nursing students. Method: 91 students were sampled from freshman in nursing school. Data were collected from April 2 to October 5 2007. Analysis of the results were carried out using SPSS win 12.0 for frequency, mean and standard deviation, t-test, ANOVA, Pearson correlation. Result: The result of MBTI test was that nursing students in this study showed extraversion(52.7%), sensing(82.4%), thinking(56.0%), judging(57.1%). Academic achievement according to personality was that only feeling(F) type students had significantly higher score of English than thinking(T) type students(p=.013). Learning attitudes according to personality were that sensing(S) type students had significantly positive attitude toward their college than intuition(N) type students (p=.016). And think(T) type students had significantly higher confidence of learning than feeling(F) type students. There was significant positive correlations between learning method and score of English, Fundamental Nursing, Anatomy(p<05). Conclusions: Diagnosing personality can differentiate student's learning attitude. It is important to develop teaching-learning strategies according to their personality, which can help them to improve their academic achievement.

      • 활동혈압에 의한 Cicletanine의 강압효과 판정

        Prostaglandin 합성을 촉진하는 새로운 강압약인 cicletanine의 강압효과를 진료실 혈압과 24시간 활동혈압으로 평가했을 때 상당한 차이를 보여 그 이유를 검토해 보았다. To evaluate antihypertensive effect of cicletanine with both clinic and ambulatory blood pressure monitoring, 10 male hypertensive patients were administered cicletanine, a new antihypertensive stimulating prostaglandin synthesis, in 100∼200 mg daily for 12 weeks. The following were the results. 1.Mean of clinic BP decreased from 161/98 mmHg to 153/98 mmHg. 2.Mean of ambulatory BP changed from 140/91 mmHg to 153/99 mmHg. 3.Side reactions were mild, namely indigestion and constipation. 4.Discrepancy between clinic and ambulatory BP were discussed. In conclusion cicletanine revealed slight antihypertensive effect with clinic blood pressure, but failed to confirm it with ambulatory blood pressure.

      • 3-Cyano 및 3-Carbomethoxy Chromone과 Allenylstannane과의 반응

        t-Butyldimethylsilyl triflate 존재에서 3-Cyano 및 3-Carbomethoxy Chromone(1a,b)과 Allenylstannane(2) 과의 반응에서 Chromone의 C-2위치에 propargyl기가 도입된 유도체(3a,b)를 합성하였다. 합성한 유도체의 상대적 구조는 ¹H-NMR을 통해 분석하였으며, T.S구조의 모델을 통하여 가능한 메카니즘을 제시하였다. Reaction of 3-Cyano and 3-Carbomethoxy Chromone(1a,b) with Allenylstannane(2) in the presence of tert-butyldimethylsilyl triflate afforded the corresponding the propargylation product(3a,b) in the moderate yield. The relative stereochemistry of 3a,b are obtained on the basis of ¹H-NMR analysis. And the possible reaction mechanism are suggested by the molecular models in the transition state.

      • CWOS와 황토를 이용한 농축·소화슬러지의 탈수성에 관한 연구

        Every year, 190,000 tons of oystershell are wasted from many oyster farms. And the fact that the main components of oystershell are CaO and Al₂O₃ which are chemically similar to those of lime leaded to an assumption that oystershell can be used as raw materials for synthesizing dewatering conditioner. In this study, the Jar-Test and the Bu˝chner funnel test were proceeded for the assessment of dewaterability of a thickened sludge and digested sludge. And TTF(Time To Filter), SRF(Specific Resistence Filtration) and TS(Total Solids) were adopted as the valuation indices of sludge dewaterability. Dewatering conditioner which composed of both oystershell and loess is much dewaterable than the one composed of only oystershell. And in the course of combining of oystershell and loess, the following fact was found that the dewaterability of the combination which have the higher ratio of oystershelI than that of loess is superior. Eventually, the most suitable ratio of dewatering conditioner complex is 9 : 1 (oystershell : loess) in treating not only thickened sludge but also digested sludge. In conclusion, the conditioner synthesized from waste oystershell and loess seems to be used as a splendid alternative material for the dewatering conditioner treating the sludge of sewage.

      • Oyster-Zeolite 흡착탑을 이용한 하수의 유기물질과 질소제거

        The objective of this study is to investigated the removal possibility of organic matter and nitrogen contained final effluent of sewage treatment plant using oyster-zeolite adsorption column with waste oyster shell and natural zeolite. The raw water was collected the final effluent of Janglim sewage treatment plant in Busan city. The organic matter and T-N removal efficiency ranged from 2~60% and 38~97%, respectively. In conclusion, oyster-zeolite adsorption column could organic matter and nitrogen removal of final effluent of sewage treatment plant and its economical efficiency is enough because of using recycled waste oyster shell and a cheap natural zeolite.

      • Characterization of a Constitutive β-Lactamase from a Pseudomonas aeruginosa

        Objective : We analyzed some physicochermical chracteristics of a constitutive β-lactamase from a clinical isolate of a Pseudomonas aeruginosa which resistant to various β-lactams except imipenem and susceptible to aminoglycosides the proportion of its contribution to such resistance profile. Method : 1. Antimicrobial susceptibility test was done by the broth dilution method. 2. In hibition profile was done using EDTA, HgCl₂ and p-chloromercuribenzoate (pCMB). We also measured 50% inhibitory concentration(IC??) using β-lactamase inhibitors, such as clavulanic acid, sulbactam. 3. Kinetics study for substrate profile was performed by spectrophotometric assay using crude extracts and various antimicrobial agents. Relative hydropysis rates were determined. 4. The isoelectric point(pI) was determined by isoelectric focusing. The molecular weight was estimated by SDS-polycrylamide gel electrophoresis. Results : 1. The strain was resistant to ceftriaxone, sulperazon, piperacillin, ceftazidime, and aztreonam, while it was susceptible to gontamicin, tobramycin, amikacin and imipenem. 2. The β-lactamase was not inhibited by EDTA, and pCMB, whereas it was inhibited by HgCI₂. The IC?? of clavulanic acid was over 100μM and that of sulbactam was 22μM. 3. The relative maximal hydrolysis rate was calculated by setting V_(max) for cephaloridine at 100. The enzyme hydrolyzed cephalosporins more rapidly than penicillins. 4. The pI was 9.3 by isoelectric focusing. The molecular weight was estimated to be 40kDa. Conclusion : According to its substrate profile, pI and molecular weight, the β-lactamase from a clinical isolate of P. aeruginosa was a cephalosporinase belonging to group 1 in the Bush-Jacoby-Medeiros classification scheme.

        인터넷 쇼핑경험에서의 소비자만족 형성과정에 관한 연구

        This study aims to examine the forming procedure of consumer satisfaction from Internet shopping experience. Many studies on the satisfaction have primarily focused on the examination of the antecedents of satisfaction along with the expectation-disconfirmation paradigm dominating the former studies. This shady has built an Internet shopping expectation-disconfirmation model as to apply the expectation-disconfirmation model into the condition of the Internet shopping. The summary of study is represented in following. 1. The expectation of risk and performance of benefit had a significantly positive effect on the perceived disconfirmation. In addition, an expectation of benefit and performance of risk had a significantly negative effect on the perceived disconfirmation. 2. A performance of risk and performance of benefit not only had a significantly indirect effect, but also a significantly direct effect on the consumer satisfaction. But expectation of risk and expectation of benefit had no significant direct effect on satisfaction. 3. Not only the perceived disconfirmation had a significant positive effect on satisfaction, but also an effect of the perceived disconfirmation on satisfaction(0.472) is larger than that of the performance of risk or performance of benefit. Meaning that, it is desirable for one to apply the expectation-disconfirmation model into the Internet shopping circumstances as much as the off-line markets.

        열원이 존재하는 작업장내 기류 및 온도장 예측

        A CFD simulation of airflow and temperature filed in a heated room has been described in this paper. The thermal wall jet created by a radiator greatly influences the airflow pattern, temperature distribution. The area close to a heat source has a high risk of air-borne contamination and imposes a harmful effect on occupants in that area. The predicted flow field, temperature results show good agreement with the measured data. As the results were compared with experimental data. the applicability of CFD was satisfactorily verified. Also, the CFD simulation can capture the natural convective flow features. If a CFD simulation is applied ventilation design with a heat source, An effective design will be attained. Further study is required to improve the accuracy of CFD simulation.
