        심근경색 모델 흰쥐에서 스트레스가 B-type Natriuretic Peptide 발현 및 심박변이율 변화에 미치는 영향

        심박변이율의 감소는 심근 경색 환자에서 예후의 악화와 관련되어 있다. 심박변이율 중 very low frequency가 심부전환자의 예후를 판단할 수 있는 독립적 인자로서 사용이 가능하다고 보고된 바 있으며, 심실의 압력 증가에 반응해서 생성되는 B-type natriuretic peptide가 심근 경색의 예후 인자로서 사용할 수 있음 역시 보고된 바 있다. 본 연구에서는 심근경색 모델을 제작하여 일정의 회복 기간을 거친 다음 다시 일정 기간의 스트레스를 겪게 한 후 심박변이율의 변화 및 심실 조직에서의 BNP 발현 정도를 비교하여 스트레스에 의한 영향을 관찰하고자 하였다. Sprague-Dawley계 수컷 흰쥐 15마리를 대상으로 하였다. 실험동물은 인위적 스트레스 및 수술을 받지 않은 대조군(CON, n=5), 심근경색 수술을 받은 후 restraint stress를 받지 않은 군(MI+No Stress, n=5), 심근경색 수술을 받은 후 1주일 동안 restraint stress를 받은 군(MI+Stress, n=5)으로 분류하였다. 심근경색 모델 제작 직후 15분 이상 심전도의 변화를 기록하였으며, 2개월 정도 후 심근경색 수술을 받은 동물을 두 군으로 나누어 그 중 한 군에는 1주일 동안 restraint stress를 가한 후 restraint stress를 가하지 않은 동물과 심박변이율을 비교 분석하였다. 심실 조직은 좌심실 전벽 부위의 위축을 관찰할 수 있었고, 대식세포에 의하여 응고, 괴사된 심근세포의 탐식과 혈관이 풍부한 육아조직 및 섬유 변화를 볼 수 있었다. 심박변이율은 심근경색 수술을 받은 직후 심박수는 유의한 증가를 보였고, standard deviation of the normal to normal intervals (SDNN), very low frequency (VLF) 및 low frequency (LF)의 유의한 감소를 보였다. 심근경색 수술을 받았던 동물에서 두 달 정도의 회복 기간을 거친 후 스트레스를 받지 않은 군은 심박수 및 기타 심박변이율 분석에서 SDNN 값을 제외하고는 정상군과 유의한 차이를 관찰할 수 없었던 반면 1주일 동안 스트레스를 받은 군에서는 심박수가 다시 유의하게 증가했을 뿐만 아니라 SDNN, VLF 및 LF 역시 정상군과 유의한 차이를 보였다. 이상의 결과를 종합하면 심근경색 동물에 대해 1주일 동안 스트레스를 가한 결과 심박변이율이 감소하고 심실에서의 BNP 발현은 더욱 증가하여 악화함을 보여주고 있다. Diminished heart rate variability (HRV) is associated with less favorable prognosis after myocardial infarction (MI). It has been reported that very low frequency (VLF) power in HRV analysis is an independent risk predictor in patients with congestive heart failure and B-type natriuretic peptide (BNP) can be used as a prognostic factor of MI. The purpose of this study was to investigate whether stress has an effect to the changes of BNP expression and/or heart rate variability in MI model in rats. Male Sprague-Dawley rats were divided into 3 groups: control group (CON), MI group (MI+No Stress), and MI followed by stress group (MI+Stress). MI+Stress group rats were raised for a two month recovery period after the operation, followed by being exposed to restraint stress for 2 hours per day for 1 week. Electrocardiogram was recorded after the operation and the last day after 1 week of stress. The frequency components of HRV were calculated in the frequency domain such as VLF, low frequency (LF), high frequency (HF) and so on. In HRV analysis, standard deviation of the normal to normal intervals (SDNN) was significantly reduced in both groups compared to the control group. VLF and LF also were significantly reduced in MI+Stress group compared to the control groups. In addition, BNP expression in western blotting was shown the strongest bands in MI+ Stress group among experimental groups. These results suggest that BNP and HRV were aggravated by stress in MI rat model. (Korean J Str Res 2010;18:275∼285)

        Crude saponin(100g) were obtained by extracting the radix(5kg) of Scrophalariae koraiensis Nakai. Crude saponin were positive in the Liebermann-Burchard test. We observed 5 sports by TLC using BuOH saturated with H_2O : ethyl acetate : water(4:1:5, upper phase) as solvent and 1% Ce(SO_4)_2 in 10% H_2SO_4 as a color former. We divided into 3 fractions by common column chromatography using BuOH saturated with H_2O : ethylacetate : water(4:1:5, upper phase) as solvent.

        This study was conducted to investigated the major characteristics of genetically modified rice of "Milyang 204" originated from Dongjinbyeo compared to a non-transgenic rice varieties Dongjinbyeo and Junambyeo. Basta resistant transgenic rice lines carrying bar gene produced by the Yeongnam Agricultural Research Institute were evaluated for their agronomic characters. The transgenic Japonica rice of "Milyang 204" showed inferior phenotypic traits compared to a non-transgenic rice variety Dongjinbyeo and Junambyeo. On the basis of UPOV (Union Internationale Pour la Protaection des Obtentions Vegetables) and NSMO(National Seed Management Office) the transgenic "Milyang 204" showed difference in some traits out of some agronomic traits, such as leaf color, angle of flag leaf, number of spikelets, culm length, white core and white belly compared to the nontransgenic varieties rice.

      • 웹기반의 가축전염병 발생자료 관리시스템(AIMS) 개발

        The AIMS(Anima1 Infectious Disease Data Management System) was developed to collect and analyze data on animal infectious disease outbreak (62 diseases). When an animal infectious disease is confirmed at a diagnostic laboratory, the disease outbreak is then reported to the regional government through AIMS. After the regional government confirms the number of animals infected or dead, outbreak date and etc, this information is reported to the Ministry of Agriculture & Forestry and the National Veterinary Research & Quarantine Service through AIMS. The AIMS analyzes the outbreak data and reports the outbreak statistics by region, month and year to them. In addition, the AIMS can identify the farm where there was an outbreak of the animal infectious disease. There are four user roles : 1) Administrator, responsible for the overall operation of the system. 2) Inspector, responsible for confirmation of the animal infectious disease outbreak. 3) Pathologist, responsible for input to an animal infectious disease at first. 4) Gallery, only able to read the outbreak statistics. The AIMS has been implemented using the ASP technology and is operated on the web to allow easy access. The data, middle and presentation tier are based on MS-SQL 2000 server, Microsoft transaction server and Internet Explorer, respectively. The database's stored procedure was used to improve the performance. In addition to above software, the dextupload software is used to upload the files. By using the AIMS, a more accurate statistics for the animal infectious disease outbreak is expected.

        Antimicrobial resistance and resistance patterns of S. aureus, coagulase-negative Staphylococcus spp., and E. coli isolated from bovine mastitis in Korea during 2010~2011

        To determine current rate of antimicrobial resistance, a total of 236 isolates from milk samples of dairy cattle with mastitis in Korea during 2010-2011 were examined against 12 antimicrobials using disc diffusion method: 67 Staphylococcus aureus, 74 coagulase-negative Staphylococcus spp. (CNS), and 95 Escherichia coli isolates. The isolates examined in this study were submitted by Local Veterinary Service Laboratories located in 13 provinces and metropolitan cities nationwide. The highest rates of resistance among S. aureus isolates were against ampicillin (56.7%) and penicillin (56.7%), followed by kanamycin (11.9%). All S. aureus isolates were sensitive to lincomycin, amikacin, and cephalothin. Only one isolate showed resistance to tetracycline and oxacillin, respectively. Less than 10% of the S. aureus isolates presented resistance to erythromycin, neomycin, and gentamicin. Among CNS isolates, the most frequently observed resistance was to lincomycin (44.5%), followed by penicillin (28.3%), ampicillin (18.9%), tetracycline (17.5%), kanamycin (13.5%), and erythromycin (9.4%). All or most of the CNS isolates were sensitive to cephalothin, amikacin, neomycin, and gentamicin. The highest rate of resistance among E. coli isolates was against tetracycline (26.3%), followed by streptomycin (21%), neomycin (15%), kanamycin (12.6%), and gentamicin (10.5%). Amikacin was the only antimicrobial to which no E. coli isolates showed resistance. Around 10% of the S. aureus isolates and 15% of the CNS isolates showed resistance against three or more antimicrobials simultaneously, while more than 30% of the E. coli isolates did.

        Antimicrobial resistance of streptococci isolated from mastitic bovine milk samples in Korea.

        <P>The aim of this study was to determine the antimicrobial resistance of streptococci isolated from mastitic bovine milk samples. A total of 178 isolates belonging to 6 different Streptococcus species were examined: S. uberis (n = 99), S. bovis (n = 30), S. oralis (n = 24), S. salivarius (n = 13), S. intermedius (n = 7), and S. agalactiae (n = 5). Only 8.9% (16/178) of the isolates were susceptible to all antimicrobial agents tested in this study, and S. agalactiae and S. intermedius isolates were all resistant to at least 1 antimicrobial agent tested. Overall, the most frequently observed resistance was to tetracycline (61.2%), followed by lincomycin (43.2%), gentamycin (35.3%), oxacillin (34.3%), and erythromycin (28.6%). Cephalothin and penicillin were the only antimicrobial agents to which most of the streptococci (>or=92%) were susceptible. Wide differences in the prevalence of resistance are apparent among the individual species: S. salivarius displayed exceptionally high resistance to cephalothin (23.0%) and oxacillin (76.9%) and S. agalactiae (20%) and S. intermedius (14.2%) to penicillin. Streptococcus salivarius and S. agalactiae were all susceptible to erythromycin, but others showed various rates of resistance ranging from 12.5% to 42.8%. Resistance to 3 or more of 7 antimicrobial agents was observed in all species (37.6%, 67/178).</P>

        Culture results from quarter milk samples submitted to veterinary diagnostic laboratories during January∼November 2012 in Korea

        During January-November 2012, a total of 2,041 quarter milk samples were collected from dairy cattle of 82 dairy farms nationwide. About 42% (870/2,041) of the samples that had somatic cell counts (SCC) of ≥ 200,000 cells/ml were subjected to microbiological examination. No bacteria was isolated from 95 of 870 (10.9%) samples. Among 1,237 bacteria isolated from the rest 775 samples, 1,085 were identified with VITEK: more than half (52.1%, 645/1,237) of the isolates were gram negative bacillus. Gram positive cocci including Staphylococcus accounted for 35% of the isolates and almost none of gram positive bacilli isolated. Excluding Coagulase-negative Staphylococcus (CNS), the most frequently isolated bacterial species was Escherichia coli (11.2%, 138/1, 237), followed by Klebsiella pneumoniae (8.1%,100/1,237), Staphylococcus aureus (7.1%, 88/1, 237), Enterobacter cloacae (6.0%, 74/1, 237) and Serratia marcescens (3.5%, 43/1, 237). The most common resistance of S. aureus was to penicillin (77.4%) and ampicillin (73.0%), while no resistance was observed against gentamicin and cephalothin. Although CNS presented resistance to all antimicrobials tested but the most prevalent resistance was to penicillin (35.6%) and ampicillin (37.0%). The pattern of antimicrobial resistance observed in CNS was similar to that of S. aureus, but the rates were much lower than those of S. aureus. E. coli also showed resistance to all the antimicrobials tested, although the rates were not very high. The highest resistance of E. coli was to cephalothin (39.4%) and ampicillin (36.2%), while most of the strains (98.0%) showed sensitivity to amikacin. The results of this study provide information on current situation of bovine mastitis in Korea.

        Epidemiological characteristics of bovine tuberculosis in the Republic of Korea, 2000~2004

        This paper describes the epidemiological characteristics of bovine tuberculosis in Korea during January 2000 to September 2004, when the incidence of bovine tuberculosis increased markedly: a total of 1,054 herds (4,197 cattle) were confirmed to be infected with Mycobacterium bovis during this period. Based on the record of epidemiological investigation, introduction of purchased cattle (22.9%, 125/545) into a farm was the most frequent transmission route of M. bovis infection. On 31.7% (335/1,054) of the infected farms, recurrent infection occurred more than once before the disease has been eradicated completely. The highest rate of recurrence was detected around 70 days after the initial test of the infected herd, which seems to be related to current regulation on the test of animals that cohabited with those previously diagnosed with infection in farms, rather than to the characteristic of the disease. Although the current eradication program has been effective in controlling the disease in dairy cattle in Korea, control measures more specific to beef cattle may be needed because infection rate in beef cattle continues to increase in recent years.
