      • Limulus Amoebocyte Lysate법에 의한 원유와 살균유의 세균수 측정

        LAL method for the determination of Gram-negative bacterial Lipopolysaccharides(LPS) in milk is proposed and its value for bacteriological quality control is investigated. The relationship between the LPS contents and total colony count(cfu/ml), which covers the Gram-negative bacteria, was determined in raw milk samples. The coefficient of correlation between Gram-negative bacterial counts(cfu/ml) and LPS contents was r^2 =0.969. And, the correlation coefficient between total bacterial counts(cfu/ml) and LPS contents was r^2 =0. 914. It was also investigated how much the LPS contents were in UHT milks from l0 different dairy industrial companies in Korea. It varied from 92 ng LPS/ml to 938 ng LPS /ml.

      • 공단지역 및 청정지역 식물 잎권의 잎표면세균 및 내산성세균의 분포

        산성강하물의 영향을 받는 대천공단지역과 영향을 받지 않는 청정지역인 대전 계족산 자연휴양림에서 자라는 밤나무(Castanea crenat)의 잎표면에서 서식하는 총세균수, 생존세균수, 종속영양세균수, 내산성세균수를 1996년 8월부터 1997년 8월까지 5회에 걸쳐 조사하였다. 공단지역 잎표면 평균 총세균수, 생존세균수 및 종속영양세균수는 각각 9.9×10^(5) cell/㎠, 1.6×10⁴cell/㎠, 7.1×10³cell/㎠,로서 청정지역에 비하여 각각 1.5배, 2배, 2.6배 정도로 관찰되었다. MPN법으로 측정한 pH 5.6애서의 잎표면 내산성세를수는 공단지역 3.3×10⁴, 청정지역 3.4×10⁴MON/㎠로 거의 같았고, pH 4.0에서의 내산성세균수는 공단지역애서 1.9×10^-(-1)MPN/㎠인 반면 청정지역에서는 전혀 검출되지 않았다. pH 3.0에서의 내산성 잎표면세균수는 공단지역과 청정지역의 잎권 어느 곳에서도 검출되지 않았다. 한편 계절별 잎표면세균수의 분포는 대체로 잎이 나기 시작하여 크기가 가장 작은 5월에 최대를, 그리고 낙엽이 지는 11월에 최소를 나타내었다. 이런 결과는 공단지역의 대기오염물질의 침적이 주변의 식물 잎표면 세균수를 감소시키지는 않으며, 특히 산성강하물의 영향으로 내산성세균수가 증가함을 보여주고 있다. Total, direct viable count, and acid-tolerant epiphytic bacterial population sizes were quantified on leaves of chestnut tree (Castanea crenata S. et Z.) near Taejon Industrial Estate affected by acid precipitation and deposition as well as in the clean natural forest area, Mt. Kyejok, in Taejon city from August 1996 to August 1997. Geometric mean numbers of total, direct viable count, and acid-tolerant epiphytic bacteria were 9.9×10^(5) cell/㎠, 1.6×10^(6)cell/㎠, and 7.1×10³cfu/㎠ respecfvely, being 1.5, 2, and 2.6 times those in the clean area. Acid-to-lerant epiphytic bfcterial numbers at pH 5.6 by MPN method were 3.3×10" in the industrial area, about the same as the number,3.4×104 MPNicni, of the clean area. Acid-tolerant bacterial number at pH 4.0 was 1.9×10^(-1)MPN/㎠ in the industrial area, whereas none was detected in the clean area. Acid-tolerant bacteria at pH 3.0 were not detected at all in the industrial area as well as in the clean area. Epiphytic bacterial population sizes were generally the greatest in May when leaves are emerged and grew but the lowest in November when defoliation occurs. These results showed that air pollutant deposition on leaves did not cause a deuease of epiphytic bacteria at least and acid deposition on leaves did cause an increase of acid-tolerant bacteria.

        Novel Turbo Receiver for MU-MIMO SC-FDMA System

        Single carrier‐frequency‐division multiple access (SC‐FDMA) has been adopted as the uplink transmission standard in fourth‐generation cellular networks to facilitate power efficiency transmission in mobile stations. Because multiuser multiple‐input multiple‐output (MU‐MIMO) is a promising technology employed to fully exploit the channel capacity in mobile radio networks, this study investigates the uplink transmission of MU‐MIMO SC‐FDMA systems with orthogonal space‐frequency block codes (SFBCs). It is preferable to minimize the length of the cyclic prefix (CP). In this study, the chained turbo equalization technique with chained turbo estimation is employed in the designed receiver. Chained turbo estimation employs a short training sequence to improve the spectrum efficiency without compromising the estimation accuracy. In this paper, we propose a novel and spectrally efficient iterative joint‐channel estimation, multiuser detection, and turbo equalization for an MU‐MIMO SC‐FDMA system without CP‐insertion and with short TR. Some simulation examples are presented for the uplink scenario to demonstrate the effectiveness of the proposed scheme.

        Hydroxydibenzoylmethane induces apoptosis through repressing ornithine decarboxylase in human promyelocytic leukemia HL-60 cells

        Ornithine decarboxylase (ODC) is the rate-limiting enzyme in polyamine biosynthesis and a target for chemoprevention. Hydroxydibenzoylmethane (HDB), a derivative of dibenzoylmethane of licorice, is a promising chemopreventive agent. In this paper, we investigated whether HDB would inhibit the ODC pathway to enhance apoptosis in human promyelocytic leukemia HL-60 cells. We found ODC enzyme activity was reduced during HDB treatment. Overexpression of ODC in HL-60 parental cells could reduce HDB-induced apoptosis, which leads to loss of mitochondrial membrane potential (${\Delta}{\psi}_m$), through lessening intracellular ROS. Furthermore, ODC overexpression protected cytochrome c release and the activation of caspase-3 following HDB treatment. The results demonstrated HDB-induced apoptosis was through a mechanism of down-regulation of ODC and occurred along a ROS-dependent mitochondria-mediated pathway.

        AuNP-Loaded Electrospinning Membrane Cooperated with CDs for Periodontal Tissue Engineering

        BACKGROUND: Guided bone regeneration (GBR) is commonly used to regenerate periodontal tissue. However, the bone inductivity and antibacterial properties of the GBR membranes currently in use are severely limited. This issue can be resolved by loading growth factors and antibiotics. Bioactive substitutes, such as Au nanoparticles (AuNPs) and carbon quantum dots (CDs), were proposed to prevent the denaturation of osteogenic growth factors and the induction of antibacterial drug resistance. METHODS: Ornidazole was initially used as the raw material to prepare the CDs, followed by the incorporation of an optimal ratio of nanoparticles to produce the electrospun membrane doped with AuNPs and novel traceable antibacterial CDs. The morphology of the membrane was characterized. The adhesion, proliferation, and osteogenic differentiation of cells on the membrane were evaluated in vitro. The antimicrobial characteristics of the membrane were also investigated. The electrospun membrane was implanted into a rat skull defect model in vivo to investigate its osteogenic potential. RESULTS: The blending of nanomaterials did not affect the micro morphology of the fiber, resulting in enhanced mechanical properties. Membranes doped with AuNPs and CDs exhibited excellent biocompatibility, increased ALP activity, improved calcified nodules, and increased expression of osteogenic-associated proteins, in addition to pronounced antibacterial effects. The membrane also demonstrated excellent osteogenic characteristics in rat models. CONCLUSION: The synergistic effect of loaded AuNPs electrospun fiber membrane with CDs can promote periodontal bone regeneration and exert antibacterial activity.

        An Autonomous Driving Approach Based on Trajectory Learning Using Deep Neural Networks

        Autonomous driving approaches today are mainly based on perception-planning-action modular pipelines and the End2End paradigm respectively. The End2End paradigm is a strategy that directly maps raw sensor data to vehicle control actions. This strategy is very promising and appealing because complex module design and cumbersome data labeling are avoided. Since this approach lacks a degree of interpretability, safety and practicability. we propose an autonomous driving approach based on trajectory learning using deep neural networks in this paper. In comparison to End2End algorithm, it is found that the trajectory learning algorithm performs better in autonomous driving. As for trajectory learning algorithm, the CNN_Raw-RNN network structure is established, which is verified to be more effective than the original CNN_LSTM network structure. Besides, we propose an autonomous driving architecture of a pilot and copilot combination. The pilot is responsible for trajectory prediction via imitation learning with labeled driving trajectories, while the copilot is a safety module that is employed to verify the effectiveness of the vehicle trajectory by the results of the semantic segmentation auxiliary task. The proposed autonomous driving architecture is verified with a real car on urban roads without manual intervention within 40 km.

      • Synthesis of Nesseria meningitidis W135 capsular oligosaccharide with various lengths for immunogenicity comparison

        Nesseria meningitidis is a type of bacteria which causes meningococcal meningitis and meningococcal septicemia. It often infects children under two years old and causes infant dead. Meningococcal diseases are prevalent in developing countries, like China and Africa. Based on their surface capsular polysaccharides, they are classified into 13 serogroups, while group A, B, C, Y, W135 are the major epidemic strains in the world. Though, some efforts have devoted to develop polysaccharide vaccines against N. meningitidis, but these vaccines often elicit weak immune responses and short immune memories. Therefore, polysaccharide-protein conjugate vaccines were developed to tackle the problem and the immunogenicity improved greatly. However, the compositions of commercial available vaccines are very heterogeneous and the quality is hard to control. In this study, we synthesized the N. meningitidis W135 capsular oligosaccharide in different lengths and acquired the homogeneous form to detect the immunogenicity. Because the N. meningitidis W135 capsular oligosaccharide is silalic acid-galactose repeating structure, we used sialyl phosphate as a donor to conjugate thiol galactoside to form the basic repeat unit. By linking the repeat units, we have obtained various length of oligosaccharide from disaccharide to decasaccharide. We conjugated these different length of oligosaccharides to carrier protein CRM197 (a mutated diphtheria toxin) and injected these conjugates to mice to search for the best vaccine composition.

        Precoder Distribution and Adaptive Codebook in Wideband Precoding

        Based on wideband precoding (WBP) in the multiple-input multiple-output orthogonal frequency division multiplexing system, an adaptive nonuniform codebook is presented in this paper. The relationship between the precoder distribution and spatial correlation is analyzed at first. A closed-form expression based on overlapped isosceles triangles is proposed as an approximation of the precoder distribution. Then, the adaptive codebook design is derived with the approximate distribution to minimize quantization errors. The capacity and bit error rate performance demonstrate that the adaptive codebook with WBP outperforms the conventional fixed uniform codebook.

        Combined Assessment of Serum Alpha-Synuclein and Rab35 is a Better Biomarker for Parkinson’s Disease

        Background and Purpose It is essential to develop a reliable predictive serum biomarker for Parkinson’s disease (PD). Te accumulation of alpha-synuclein (αSyn) and up-regulated expression of Rab35 participate in the etiology of PD. Te purpose of this investigation was to determine whether the combined assessment of serum αSyn and Rab35 is a useful predictive biomarker for PD. Methods Serum levels of αSyn or Rab35 were determined in serum samples from 59 sporadic PD patients, 19 progressive supranuclear palsy (PSP) patients, 20 multiple system atrophy (MSA) patients, and 60 normal controls (NC). Receiver operating characteristics (ROC) curves were calculated to determine the diagnostic accuracy of αSyn or/and Rab35 in discriminating PD patients from NC or atypical parkinsonian patients. Results The levels of αSyn and Rab35 were increased in PD patients. The serum level of Rab35 was positively correlated with that of αSyn in PD patients. Compared to analyzing αSyn or Rab35 alone, the combined analysis of αSyn and Rab35 produced a larger area under the ROC curve and performed better in discriminating PD patients from NC, MSA patients, or PSP patients. When age was dichotomized at 55, 60, 65, or 70 years, the combined assessment of αSyn and Rab35 for classifying PD was better in the group below the cutof age than in the group above the cutof age. Conclusions Combined assessment of serum αSyn and Rab35 is a better biomarker for discriminating PD patients from NC or atypical parkinsonian patients, and is a useful predictive biomarker for younger sporadic PD patients.
