      • 벼葯培養에 있어서 低溫處理가 植物體分化에 미치는 影響

        This experiment was studied to determine the effects of low temperature treatment on culturability in anther culture of Tongil type(Indica/Japonica) rice varieties. Three Japonica varieties have better reaction to callus induction and plant regeneration than those of Indica and Indica/Japonica varieties. The mean frequency of plant regeneration of three Indica/Japonica varieties was less than 1%. But the frequency of callus induction and plant regeneration of Milyang 23(Indica/Japonica) was showed the best result in the anthers pretreated at 12℃ for 20 days, the frequency of callus induction and plant regeneration was 31.6% and 13.6%, respectively. Most of calli derived from pretreated-anthers, above 90%, were induced from 20 to 35 days after anther inoculation. Combination of NAA, Kinetin and ABA showed the best result on the growth of callus induced from pretreated-anthers.

      • 사람 혈청 알부민에 대한 단세포군 항체의 제조와 특성 연구

        Serum albumin은 혈청 단백질 중 50-60%를 차지한다. 알부민은 순환계 내에서 삼투압을 유지시켜 세포내외의 체액을 유지 및 지용성 물질의 이동에 관여한다. 신사구체와 기저막의 투과성이 증가하거나 세뇨관의 흡수감소 등으로 단백질의 요배설량이 하루에 150mmg 이상되는 단백뇨는 신장내의 병변이 있음을 나타내는 중요한 지표이다. 이러한 단백뇨는 크게 사구체성 단백뇨, 세뇨관성 단백뇨, 혼합성 단백뇨로 나눌수 있는데 이중 사구체 기저막의 손상으로 발생되는 사구체성 단백뇨는 알부민과 같은 고분자량의 단백질이 소변 내로 배설되는 현상이다. 일반적으로 알부민이 정상치 이상으로 소변을 통하여 배설되는 것을 'microalbuminuria'라고 하며 당뇨병으로 인한 신장병변을 예견하는데 중요한 지표로 알려져 있다. Microalbuminuria는 방사 면역확산법, 방사성 면역측정법, 면역혼탁측정법, 비탁측정법, 효소결합면역측정법(ELISA) 등으로 겸출할 수 있으나 최근에는 안전하면서도 민감도가 높고 측정이 용이한 방법인 ELISA를 이용한 면역분석방법이 많이 사용되고 있다. 면역특이성이 떨어지는 다세포군 항체를 사용한 방법의 문제점을 극복하기 위해서 세포융합기술을 이용한 단세포군항체를 생산하는 세포주를 개발하였고 그 특성을 규명하였다. 따라서 본 연구에서 세포융합기술을 도입하여 개발된 인간혈청 알부민에 대한 단세포군항체는 glycohemoglobin에 대한 단세포군항체와 더불어 당뇨병의 진행 정도를 진단할 수 있는 키트의 원료로 사용될 수 있다.

      • 담배거세미나방에서 분리된 항균펩티드 유전자의 대량 발현에 관한 연구

        The antibacterial gene isolated from the common cutworm. Spotoptera litura showed high degree of homologies in its nucleotide sequence to those of the cecropin genes from Hyalophora cecropia, implying that it may encode the antibacterial peptide. In this experiment, the antibacterial gene from S. litura was subcloned to the expression vector pGEX-1λT and overexpressed in BL21. The antibacterial gene is composed of total 114 nucleotides which encodes the relatively small peptide of 37 amino acid residues. This gene was designed to make a fusion protein with glutathione-S-transferase in response to the addition of IPTG. The total proteins from the cell extract were separated by SDS-PAGE and the overexpressed fusion protein was detected at the right positon in terms of its molecular weight. The fusion protein was also purified by use f the affinity column chromatography technique with the resin in which glutathione was cross-linked to 4% agarose bead.

      • 고교 씨름선수의 주특기에 따른 체력특성 연구

        Traditional Wrestling players from I High school and P High school in In-Chon District has been used in this research. Confronting list and recording papers from five recently participated match among the nationwide matches have been used as the basic resources. The main specialty has been chosen by the player himself, the specialist, and the coach I examined the physical strength differences among muscular factor, muscle endurance factor, the power of beginning, flexibility, and balance. By analyzing the correlation among these factors, I tried to provide the basic information for the choosing the player and coaching them. I got such a conclusion as a result of measuring and comparing the primary factors of physical strength. In these studies 10 players from each main specialty helped me. The total players were 40. 1) The power of the left hand's grip was higher for the group using the mixed skills than other players who were using other main specialty. The power of the right hand's grip was higher for the group using waist skills. The group using the waist skills as the main specialty has higher back muscular strength than other groups. (p<0.05) 2) The group using hand skills as the main specialty has higher power for sit-up among the muscle endurance factors. (p<0.05) 3) The group using mixed skills as the factor of power has higher power of beginning in the 50m run than other groups. (p<0.05) 4) For the trunk flexion among the flexibility factors, the group using hand skills as the main specialty had higher flexibility. As for the trunk extension, the group using mixed skills has higher flexibility than other groups. (p<0.05) 5) For the side step among the agility factors, the group using the hand skills as the main specialty has higher agility than other groups. (p<0.05) 6) Among the balance factors, standing in one foot with the eyes closed appeared to have no difference on each group using its own main specialty. (p<0.05)

        Abstract- In a plasma source ion implantation, the target is successively biased by negative voltage pulses with an intrinsic finite rise time, resulting in the time-dependent sheath formation around the target. Measurement of time-dependent sheath during the pulse rise show the ion matrix sheath development which is in proportion to square root of time and pulse rise rate over plasma density but independent of the ion mass.

        W/O 마이크로에멀젼에 의한 나노크기의 TiO₂/SiO₂ 합성에서 물/계면활성제의 몰 비(W_(0)) 영향 및 그 광분해 특성

        나노크기의 TiO₂/SiO₂입자는 음이온 계면활성제 AOT(sodium bis(2-ethylhexyl) sulfosuccinate)를 사용하여 W/O 마이크로 에멀젼에서 TTIP(titanium isopropoxide)와 TEOS(tetraethylorthosilicate)의 가수분해 반응에 의해 제조하였다. 그때 TEOS의 몰분율은 0.1이였다. 나노입자 제조시 W_(0)(H₂O/AOT)비에 따른 열적안정성, 표면적, 결정성 및 결정크기 등과 같은 물리적 특성은 TEM, XRD, BET, FT-IR, TGA-DTA 등을 사용하여 분석하였다. 또한 광촉매적 특성을 알아보기 위해 회분식 반응장치를 이용하여 p-니트로페놀의 광분해 활성을 조사하였다. 제조된 Ti0₂/SiO₂ 나노입자는 열처리 온도 105℃, 300℃에서 비결정구조를 가졌으며, 소성온도 800℃에서는 Ti0₂ 결정입자 내에 무정형의 SiO₂로 인해 열적 안정성이 증가되었고, 여전히 anatase 결정을 유지하였다. 그리고 Ti0₂/SiO₂(90/10) 입자에서 SiO₂ 결정은 관찰되지 않았다. 제조된 촉매는 대체로 구형이며 아주 균일한 분포를 갖는 것으로 관찰되었다. W_(0) 비가 증가할수록 결정크기는 증가되었고, 표면적은 감소하는 경향을 나타내었다. 또한 p-니트로페놀에 대한 광분해 활성은 순수한 Ti0₂ 보다 Ti0₂/SiO₂(90/10) 촉매가 우수하였다. Nanosized TiO₂/SiO₂ particles were prepared by hydrolysis of TTIP (titanium isopropoxide) and TEOT (tetra-ethylorthosilicate) in sodium bis (₂-ethylhexyl)sulfosuccinate (AOT) reverse micelles. The mole fraction of TEOS was 0.1. The physical properties, such as surface area. thermal stability, crystallite size and crystallinity according to W_(0), ratio have been investigated by TEM, XRD, BET, FT-IR, TGA and DTA. In addition. the photocatalytic degradation of p-nitrophenol has been studied by using batch reactor in order to compare the photocatalytic activity of prepared nanosized TiO₂/SiO₂, particles. It is shown that the XRD pattern of the particle heat treated at 105℃ and 300℃ indicates amorphous and the major phase of all the prepared particles were anatase structure. No significant rutile phase was observed although the calcination temperature at 800℃ and no peaks SiO₂ crystal were also observed for TiO₂/SiO₂ (90/10). The presence of amorphous SiO₂ in TiO₂/SiO₂ particle enhanced the thermal stability of TiOz particle resulting in the suppression of the phase transformation from anatase to rutile phase. The crystallite size of prepared particles decreased with decreasing W_(0) ratio. The surface area increased with decreasing W_(0) ratio. In addition, TiO₂/SiO₂ (90/10) particles shows higher photoactivity than that of pure TiO₂ particles.

        Die Definition des Geschmacks, daß er das Vermögen der Beurteilung des Schönen sei. Das Geschmacksurteil ist ästheisch. Das Wohlgellen, welches das Geschmacksurteil bestimmt, ist ohne alles Interesse. Interesse wird das Wohlgefallen genannt, das wir mit der Vorstellung der Existenz eines Gegenstandes verbinden. Ein solches hat daher immer zugleich Beziehung auf das Begehrungsvermögen, entweder als Bestimmungsgrund desselben, oder doch als mit dem Bestimmungsgrunde desselben notwendig zusammenhängend. Das Wohlgefallen am Angenehmen ist mit Interesse verbunden. Angenehm ist das, was den Sinnen in der Empfindung gefällt. Das Wohlgefallen am Guten ist mit Interesse verbunden. Gut ist das, was vermittelst der Vernunft, durch den bloßen Begriff, gefällt. Das Schöne ist das, was ohne Begriffe als Objekt eines allgemeinen Wohlgefallens vorgestellt wird. Die Allgemeinheit des Wohlgefallens wird in einem Geschmacksurteile nur als subjektiv vorgestellt. Die subjektiv Notwendigkeit, die wir dem Geschmacksurteile beilegen, ist bedingt. Schön ist, was ohne Begriff als Gegenstand eines notwendigen Wohlgefallens erkannt wird. Das Schöne kommt darin mit dem Erhabenen überein, daß beides für sich selbst gefällt. Zum Schönen der Natur müssen wir einen Grund außer uns suchen, zum Erhabenen aber bloß in uns und der Denkungsart, die in die Vorstellung der ersteren Erhabenheit hineinbringt. Erhaben nennen wir das, was schlechthin groß ist. Die Größenschätzung durch Zahlbegriffe (oder deren Zeichen in der Algebra) ist mathematisch, die aber in der bloßen Anschauung (nach dem Augenmaße) ist ästhetisch. Macht ist ein Vermögen, welches großen Hindernissen überlegen ist. Ebendieslbe heißt eine Gewalt, wenn sie auch dem Widerstande dessen, was selbst Macht besitzt, überlegen ist. Die Natur im ästhetischen Urteile als Macht, die über uns keine Gewalt hat, betrachtet, ist dynamisch-erhaben. In Beziehung auf das Gefühl der Lust ist ein Gegenstand einweder zum Angenehmen oder Schönen oder Erhabenen oder Guten (schlechthin) zu zählen (incundum. pulchrum, sublime, honestum).

        The effect of chitosan in soybean koji preparation and characteristics of chitosan-koji were investigated. Aspergillus oryzae KCTC 6096 and Aspergillus kawachii KCCM 11458 was used for koji preparation. a-Amylase activity of soybean koji prepared with 0.25% 10cp chitosan was excellent relative to 30 cp chitosan and 1.6 times higher than non-added soybean koji. Acid and neutral protease activities of soybean koji prepared with 0.25%, 10 cp chitosan were showed simillary higher value than non-chitosna added koji. The period of koji preperation was reduced 30% by adding chitosan.

      • 세포질내 정자주입법(ICSI)에 있어서 정자흡입 및 난자내 주입방법에 관한 연구

        Study on Method of Sperm Aspiration and Injection into an Oocyte in Intracytoplasmic Sperm Injection(ICSI) Immobilization of spermatozoa prior to intracytoplasmic sperm iniection(ICSI) sometimes results in crooked tail and this makes it difficult to aspirate sperm into an injection pipette tail first. Head-first sperm aspiration into an injection pipette avoid this problem due to the bigger size of the sperm head. The effect of head or tail-first sperm injection into an oocyte on fertilization cleavage, percentage of grade I embryos and development to blastocyst stage in ICSI program has been studied. A single living immobilized spermatozoa from oligoasthenozoospermic patient was injected into an oocyte head-first or tail-first according to the treatment. Eighteen hours after microinjection, oocytes ware inspected for survival and fertilization Fertilized oocytes with two pronuclei were cultured in 30μl drop of mHTF supplemented with 10% heat-inactivated follicular fluid(FF) at 37℃. On day 2. embryo transfer was performed with cleaved embryos. The remaining 2-8 cell stage embryos were co-cultured with BRL cells in mHTF + 10% FF for 72 hours and the developmental stage was observed. The data were analyzed by Analysis of Variance. A total of 164 oocytes from 36 cycles were assigned to earth treatment and ICSI was performed(88 head-first, tail-first). The rates of normal fertilization were 81.8% and 76.3% for head-first and tail-first, respectively. Of the fertilized oocytes, the percentage of cleaved embryos and the percentage of grade 1 embryo among cleaved embryos were 88.9% and 68.8%, 93.1% and 74.1% for head-first and tail-first, respectively. Of the 2-8 cell embryos cultured, 44.4%(16/36) and 50.0%(10/20) for head first and tail first, respectively developed to blastocyst stage. There were no differences in fertilization, cleavage, rates of grade 1 embryos, and development to blastocyst stage. In conclusion, head-first or tail-first sperm injection into an oocyte in ICSI program does not affect fertilization and subsequent embryo development to blastocyst stage in vitro.
