Background: Type 2 diabetes mellitus (T2DM) is characterized by elevated fasting glucagon and impaired suppression of postprandial glucagon secretion, which may participate in diabetic complications. Therefore, we investigated the associations of plasma glucagon with estimated glomerular filtration rate (eGFR), albuminuria and diabetic kidney disease (DKD) in T2DM patients.Methods: Fasting glucagon and postchallenge glucagon (assessed by area under the glucagon curve [AUCgla]) levels were determined during oral glucose tolerance tests. Patients with an eGFR <60 mL/min/1.73 m2 and/or a urinary albumin-to-creatinine ratio (UACR) ≥30 mg/g who presented with diabetic retinopathy were identified as having DKD.Results: Of the 2,436 recruited patients, fasting glucagon was correlated with eGFR and UACR (r=–0.112 and r=0.157, respectively; P<0.001), and AUCgla was also correlated with eGFR and UACR (r=–0.267 and r=0.234, respectively; P<0.001). Moreover, 31.7% (n=771) presented with DKD; the prevalence of DKD was 27.3%, 27.6%, 32.5%, and 39.2% in the first (Q1), second (Q2), third (Q3), and fourth quartile (Q4) of fasting glucagon, respectively; and the corresponding prevalence for AUCgla was 25.9%, 22.7%, 33.7%, and 44.4%, respectively. Furthermore, after adjusting for other clinical covariates, the adjusted odds ratios (ORs; 95% confidence intervals) for DKD in Q2, Q3, and Q4 versus Q1 of fasting glucagon were 0.946 (0.697 to 1.284), 1.209 (0.895 to 1.634), and 1.521 (1.129 to 2.049), respectively; the corresponding ORs of AUCgla were 0.825 (0.611 to 1.114), 1.323 (0.989 to 1.769), and 2.066 (1.546 to 2.760), respectively. Additionally, when we restricted our analysis in patients with glycosylated hemoglobin <7.0% (n=471), we found fasting glucagon and AUCgla were still independently associated with DKD.Conclusion: Both increased fasting and postchallenge glucagon levels were independently associated with DKD in T2DM patients.

Purpose: To study enhancing effects of paclitaxel in the thermochemotherapy of osteosarcoma cell lines and related mechanisms. Materials and Methods: Paclitaxel and carboplatin were used alone or jointly on OS732 cell lines in the presence of hyperthermia. Inhibition of proliferation was measured by MTT assay and cellular changes were assessed with inverted phase contrast and fluorescence microscopy. Apoptosis was analyzed with flow cytometry (FCM) and Fas expression by immunocytochemistry. Results: At $43^{\circ}C$, one hour after the application of 10ug/ml paclitaxel and $5{\mu}g/ml$ carboplatin on OS732 cells jointly, the survival rate was 15.8% which was significantly lower than with $10{\mu}g/ml$ paclitaxel (45.8%) and $5{\mu}g/ml$ carboplatin (47.7%) respectively (P<0.01). Moreover, changes of morphology and apoptotic rates indicated that the apoptosis-inducing effect of combined application was also much enhanced, as evident also regarding Fas expression. Conclusion: Paclitaxel is conducive to thermochemotherapy of osteosarcoma cell lines, possibly accomplished by up-regulation of Fas expression with induction of apoptosis.

AMFR, autocrine motility factor receptor, also called gp78, is a cell surface cytokine receptor which has a dual role as an E3 ubiquitin ligase in endoplasmic reticulum-associated degradation. AMFR expression is associated with tumor malignancy. We here investigated the clinical significance of AMFR and its role in metastasis and prognosis in gastric cancer. Expression of AMFR, E-cadherin and N-cadherin in cancer tissues and matched adjacent normal tissues from 122 gastric cancer (GC) patients undergoing surgical resection was assessed by immunohistochemistry. Levels of these molecules in 17 cases selected randomly were also analysed by Western blotting. AMFR expression was significantly increased in gastric cancer tissues, and associated with invasion depth and lymph node metastasis. Kaplan-Meier analysis showed AMFR expression correlated with poor overall survival and an increased risk of recurrence in the GC cases. Cox regression analysis suggested AMFR to be an independent predictor for overall and recurrence-free survival. E-cadherin expression was decreased in gastric cancer tissues; conversely, N-cadherin was increased. Expression of AMFR negatively correlated with E-cadherin expression, whereas N-cadherin expression showed a significant positive correlation with AMFR expression. AMFR might be involved in the regulation of epithelial-mesenchymal transition, with aberrant expression correlating with a poor prognosis and promoting invasion and metastasis in GCs.
Selective miRNA Expression Profile in Chronic Myeloid Leukemia K562 Cell-derived Exosomes

Background: Chronic myeloid leukemia (CML) is a myeloproliferative disorder of hematopoietic stem cell scarrying the Philadelphia (Ph) chromosome and an oncogenic BCR-ABL1 fusion gene. The tyrosine kinase inhibitor (TKI) of BCR-ABL1 kinase is a treatment of choice for control of CML. Objective: Recent studies have demonstrated that miRNAs within exosomes from cancer cells play crucial roles in initiation and progression. This study was performed to assess miRNAs within exosomes of K562 cells. Methods: miRNA microarray analysis of K562 cells and K562 cell-derived exosomes was conducted with the 6th generation miRCURYTM LNA Array (v.16.0). Gene ontology and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were also carried out. GO terms and signaling pathways were categorized into 66 classes (including homophilic cell adhesion, negative regulation of apoptotic process, cell adhesion) and 26 signaling pathways (such as Wnt). Results: In exosomes, 49 miRNAs were up regulated as compared to K562 cells, and two of them were further confirmed by quantitative real-time PCR. There are differentially expressed miRNAs between K562 cell derived-exosomes and K562 cells. Conclusion: Selectively expressed miRNAs in exosomes may promote the development of CML via effects on interactions (e.g. adhesion) of CML cells with their microenvironment.
Numerical Study on the Damage Characteristics of Layered Shale Using 3D DEM

Layered shale is a kind of rock with obvious anisotropy, which is significantly influenced by the inclination angle of the rock. In this study, the influence of bedding dip on the strength characteristics, failure mode, and internal fracture evolution of layered shale was investigated through laboratory triaxial compression tests and three-dimensional discrete element numerical simulations. Results show that the numerical model of layered shale constructed using the bonded block model can simulate the layered shale in terms of U-shaped strength variation and failure modes very well. Based on the distribution of cracks and failure characteristics of layered rocks, the failure modes of layered rocks are classified as follows: Shear failure across the bedding plane, slip failure along bedding planes, combined tensile splitting and shear failure. The development of microcracks damage in the slip failure model is controlled by interlayer joints, while in other failure modes, it is jointly controlled by the rock matrix and interlayer joints. The relationship between microscopic joint parameters and the variations of Young's modulus and peak strength has been proposed.
Recent advances in the food application of electrospun nanofibers

Electrospinning (e-spinning) has excellent advantages in producing functional and versatile nanofibersand electrospun (e-spun) nanofibers have a variety of potential applications, including in food industry. This paper reviews the recent progress of e-spinning and food-related e-spun nanofibers, covering ediblenanofibers (natural materials and food ingredients), active food packaging (antibacterial packaging,antioxidant packaging, high temperature and humidity resistant packaging), food filtration membranes,and food sensors. Additionally, we discussed the new trend of combining nanomaterials with food andthe challenges of applying e-spinning in food science. The results indicated that e-spinning is a promisingand efficient alternative for the production of daily and functional food.

Lonicera caerulea L. var. edulis Turcz et Herd. with great nutrient and commercial edible value, is a kind of wild berry resource in regions of Changbai Moutains, China. Systematic cultivation technology for its large-scale production is developed and formation of cultivated land with area of 150 hectares is established. With high-yield technology, the production yield can be increased by twice. Systematic cultivation technology and establishment of technology demonstration garden lay the foundation for construction of production base and large-scale production of Lonicera caerulea L. var. edulis Turcz et Herd.

High levels of genetic variation enable species to adapt to changing environments and provide plant breeders with the raw materials necessary for artificial selection. In the present study, six AFLP primer pairs were used to assess the genetic diversity of Desmodium triflorum (L.)DC. from 12 populations in South China. A high percentage of polymorphic loci (P=76.16%) and high total gene diversity (HT=0.310) were found, indicating that the genetic diversity of D. triflorum is high at the species level. Genetic diversity was also relatively high at the population level (P=55.85%, He=0.230). The coefficient of gene differentiation among populations (GST) was 0.255, indicating that while most genetic diversity resided within populations, there was also considerable differentiation among populations. AMOVA also indicated 24.29% of the total variation to be partitioned among populations (ΦST=0.243). UPGMA clustering analysis based on genetic distances showed that the 12 populations could be separated into three subgroups: an eastern, a western, and a central-southern subgroup. However, a Mantel test revealed no significant correlation (r=0.286, p=0.983)between the geographical distances and genetic distances separating these populations; mountain barriers to gene flow and human disturbance may have confounded these correlations. The present study has provided some fundamental genetic data that will be of use in the exploitation of D. triflorum.

In this study, conductive polymer composites and conductive functional fabrics are combined to serve aselectromagnetic shielding planks. Polypropylene (PP), carbon black (CB), and short carbon fibers (SCF) are blended atdifferent ratios to form conductive polymer composites (i.e. PCS series). The mechanical property, electrical property,morphology, and electromagnetic interference shielding effectiveness (EMI SE) of the PCS series are evaluated. The testresults show that with 20 wt% of conductive fillers (i.e. CB and SCF), PCS20 exhibits the optimal tensile strength, flexuralstrength, and electrical property that is 7 order of magnitude higher than that of pure PP plates. Moreover, the EMI SE of thisgroup also reaches -30 dB, which meets level one of civil EMI SE standard. Therefore, PCS20 is used to combine with fourconductive sandwiches. The resulting multilayered functional PCS-sandwich planks are tested in terms of mechanicalproperty, morphology, and EMI SE. The test results show that the planks composed of a pure conductive woven sandwichhave the maximum tensile property and significantly improved impact resistance. All of the multilayered functional plankshave EMI SE that is higher than -50 dB and are qualified for the protection level of standard EMI SE electronic devices.
