        THE <i>SPITZER</i> INFRARED SPECTROGRAPH SURVEY OF PROTOPLANETARY DISKS IN ORION A. I. DISK PROPERTIES

        <P>We present our investigation of 319 Class II objects in Orion A observed by Spitzer/IRS. We also present the follow-up observations of 120 of these Class II objects in Orion A from the Infrared Telescope Facility/SpeX. We measure continuum spectral indices, equivalent widths, and integrated fluxes that pertain to disk structure and dust composition from IRS spectra of Class II objects in Orion A. We estimate mass accretion rates using hydrogen recombination lines in the SpeX spectra of our targets. Utilizing these properties, we compare the distributions of the disk and dust properties of Orion A disks with those of Taurus disks with respect to position within Orion A (Orion Nebular Cluster [ONC] and L1641) and with the subgroups by the inferred radial structures, such as transitional disks (TDs) versus radially continuous full disks (FDs). Our main findings are as follows. (1) Inner disks evolve faster than the outer disks. (2) The mass accretion rates of TDs and those of radially continuous FDs are statistically significantly displaced from each other. The median mass accretion rate of radially continuous disks in the ONC and L1641 is not very different from that in Taurus. (3) Less grain processing has occurred in the disks in the ONC compared to those in Taurus, based on analysis of the shape index of the 10 mu m silicate feature (F-11.3/F-9.8). (4) The 20-31 mu m continuum spectral index tracks the projected distance from the most luminous Trapezium star, theta(1) Ori C. A possible explanation is UV ablation of the outer parts of disks.</P>

        <P>A chiral foldamer that consists of three indolocarbazoles and chiral amide residues folds into a helical conformation with the orientation bias, thus displaying characteristic CD signals.         DED 공정을 이용한 부품 보수 시 기저부 경사각과 모서리 반경이 적층부 인근의 열-기계 특성에 미치는 영향 고찰

        에너지 제어형 용착 공정을 이용하여 기계 부품의 손상부에 대한 보수를 수행 시 적층량 및 잔류응력 최소화를 위하여 손상 제품에 생성되는 기저부의 적정 설계에 대한 연구가 필요하다. 이 연구에서는 3차원 유한요소해석을 이용하여 에너지 제어형 용착 공정으로 기계 부품 보수 시 기저부 경사각과 모서리 반경이 적층부 인근의 열-기계 특성에 미치는 영향을 고찰하였다. 해석 결과를 이용하여 기저부 경사각과 모서리 반경 조합에 따른 적층부 인근의 온도 및 잔류 응력 분포 변화를 예측하였다. 기저부 경사각과 모서리 반경과 최대 유효 응력 및 주응력 최대값과의 상관 관계를 고찰하였다. 최종적으로 적층부 인근의 잔류 응력과 적층 체적을 감소시킬 수 있는 적정 기저부 설계를 제안하였다. An appropriate substrate design is needed to reduce the deposited volume and the residual stress in the vicinity of the deposited region for the case of the repair of damaged regions of mechanical parts using a directed energy deposition (DED) process. The goal of the study is to investigate the effects of the inclined angle and the corner radius of the substrate on thermo-mechanical characteristics in the vicinity of the deposited region by the DED process to repair mechanical parts using finite element analyses. Temperature and residual stress distributions in the vicinity of the deposited region are estimated according to the combination of the inclined angle and the corner radius. The influence of the inclined angle and the corner radius on the maximum effective and 1<SUP>st</SUP> principal stresses in the designed model is examined. Finally, a suitable design of the substrate is proposed to simultaneously reduce the residual stress in the vicinity of the deposited region and the deposited volume.

        기저부 경사각과 깊이가 에너지 제어형 용착 공정으로 보수된 영역의 열 및 잔류응력 특성에 미치는 영향 고찰

        The design of the substrate significantly affects the thermal history and the residual stress formation in the vicinity of a repaired region by a directed energy deposition (DED) process. The occurrence of defects in the repaired region depends on the thermal history and residual stress formation. The objective of this study was to investigate the influence of the inclined angle and depth of the substrate on the thermal and residual stress characteristics in the vicinity of a repaired region by a DED process through two-dimensional finite element analyses (FEAs). The temperature and residual stress distributions in the vicinity of the repaired region were predicted according to the combination of the inclined angle and depth of the substrate. The effects of the inclined angle and depth on the depth of the heat affected zone and the maximum value of the residual stress were examined. A proper combination of the inclined angle and depth of the substrate was estimated to decrease the residual stress in the vicinity of the repaired region.

      • Residual detection of buparvaquone, nystatin, and etomidate in animal-derived food products in a single chromatographic run using liquid chromatography-tandem mass spectrometry

        <P><B>Abstract</B></P> <P>A reliable and highly sensitive screening method based on liquid chromatography coupled withtriple-quadrupoleelectrospray tandemmass spectrometry (LC-MS/MS) analysis has been developed for the detection and quantification of three veterinary drugs, including buparvaquone, nystatin, and etomidate impurity B CRS. The tested drugs were extracted from samples of porcine muscle, pasteurized whole milk, and eggs using 10mM ammonium formate in acetonitrile followed by liquid-liquid purification with <I>n</I>-hexane. Chromatographic separation was achieved on a Phenomenex Luna C18 analytical column using 0.1% formic acid in ultrapure water (A) and acetonitrile (B) as mobile phases. All the matrix-matched calibration curves were linear (<I>R<SUP>2</SUP> </I> ≥0.9756) over the concentration levels of the drugs tested. Recovery at two spiking levels (equivalent to the limit of quantification (LOQ)=5ng/g and 2×LOQ) ranged from 72.88% to 92.59% with intra- and inter-day precisions <17%, except for porcine muscle spiked with 5ng/g nystatin (RSD=25.15%). Samples collected from markets located in Seoul, Republic of Korea, tested negative for all the drugs analyzed. In summary, this method is suitable for screening and quantifying the selected drugs in a single chromatographic run and with high selectivity in animal-derived food products meant for human consumption.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Buparvaquone, nystatin, and etomidate were analyzed in animal-derived food products using LC-MS/MS. </LI> <LI> A combination of 10mM ammonium formate in acetonitrile and <I>n</I>-hexane is sufficient to effectively extract the drugs without SPE. </LI> <LI> The performance parameters were satisfactory and acceptable. </LI> <LI> The target analytes were not detected in any of the market samples. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        간동맥색전술로 치료받은 간세포암종 환자의 예후예측인자로서 혈청 osteopontin의 의의

        목적: 최근에 보고된 여러 연구들에 의하면, osteopontin의 과발현이 수술 후 재발하거나, 전이가 동반된 간세포암종에서 관찰되고 있다. 그러나 간세포암종 환자에서 간동맥색전술 후 예후평가인자로서 osteopontin의 의미는 아직까지 밝혀진 바가 없다. 따라서, 본 연구는 간세포암종 환자에서 간동맥색전술 전후 혈청 osteopontin 값의 변화를 중심으로 예후예측인자로서 혈청 osteopontin의 임상적 의의를 알아보고자 하였다. 대상과 방법: 조직검사로 진단된 총 46명의 간세포암종 환자가 본 연구에 포함되었으며, 간세포암종으로 진단된 후 간동맥색전술 전과 4주 후에 혈청 osteopontin을 ELISA법으로 측정하였다. 그 후 일반혈액검사, 생화학검사 및 전산화단층촬영검사를 정기적으로 시행하여 환자의 임상적 경과를 관찰하였으며, 이를 바탕으로 간동맥색전술 시행 전의 혈청 osteopontin 값 및 시술 후의 변화와 기저 임상양상, 치료반응, 누적생존율 간의 연관성을 분석하였다. 본 연구에서 혈청 osteopontin 치의 감소는 간동맥색전술 후 기저값에 비하여 10% 이상 감소한 것으로 정의하였고, 간동맥색전술에 대한 치료반응은 시술 후 4주 뒤에 시행한 전산화단층촬영에서 종양 괴사율이 50% 이상인 상태로 정의하였다. 결과: 대상 환자의 기저질환으로는 만성 B형간염이 우세하였고(33), 그 밖에 만성 C형간염(4), 비B형C형간염(9)으로 분류되었다. 기저 혈청 osteopontin 값이 높은 군은 상대적으로 큰 종양, 간문맥전이, 진행된 병기 등 종양 상태가 진행된 환자가 많았으며, 치료반응이 낮고(P=0.043), 누적생존율도 낮았다(P=0.036). 또한, 간동맥색전술 후 혈청 osteopontin이 감소한 군에서 그렇지 않은 군에 비해 치료반응을 보인 예가 많은 경향을 보였으며(P=0.055), 누적생존율이 유의하게 높았다(P<0.05). 그러나 다변량 분석에서는 혈청 osteopontin의 기저값과 시술 후의 변화는 생존율의 유의한 독립적 예측인자는 아니었다. 결론: 혈청 osteopontin 값은 간세포암종의 병기가 진행된 환자군에서 높으며, 간동맥색전술 시술 전 혈청 osteopontin 값이 낮거나 시술 후 감소하는 경우 치료반응이 좋고 생존율도 높다. Background/Aims: Osteopontin (OPN) is overexpressed in hepatocellular carcinoma (HCC) with postoperative recurrence or extrahepatic metastasis. However, its prognostic value in patients treated with transarterial chemoembolization (TACE) is unclear. We investigated the utility of serum OPN levels and changes therein as prognostic markers in HCC patients who have received TACE. Methods: Forty-six patients with HCC were enrolled. Serum OPN levels were measured before and 4 weeks after TACE. Serum biochemistry and computed tomography (CT) scans were analyzed. We evaluated baseline serum OPN levels and subsequent changes therein in relation to tumor responses and cumulative survival rates following TACE. A decreasing pattern was defined as a decrease after TACE of more than 10% relative to baseline levels. A "responder" was defined as a patient who exhibited a tumor necrosis rate of higher than 50% on the follow-up CT scan. Results: Higher initial serum OPN levels were associated with a large tumor, portal vein invasion, and an advanced tumor stage. Patients who had lower initial serum OPN levels and those who exhibited decreasing patterns after TACE tended to have more favorable tumor responses (P=0.043 and 0.055, respectively) and exhibited better cumulative survival rates (P=0.036 and 0.030, respectively). However, the initial serum OPN level and subsequent changes in serum OPN levels were not independent predictors for survival on multivariate analysis. Conclusions: Serum OPN levels were significantly higher in patients with advanced HCC. In addition, HCC patients with low pretreatment serum OPN levels and those for whom serum OPN declined following TACE exhibited better tumor responses and survived for longer.       • A clinico-epigenetic combination accurately defines the prognosis of cervical cancer

        Objective: To assess the genes with aberrant promoter methylation and their clinical role in cervical cancer. Methods: A genome-wide assessment of the methylation status of CpG sites using a microarray-based strategy of a HumanMethylation 27 BeadChip (Illumina, San Diego, CA, USA) was performed with genomic DNA from six normal and 17 carcinomas originated from human uterine cervix. For validation of array data, methylation specific real time PCR after bisulfate treatment was used. Then, quantitative methylation values were assessed with pyrosequencing in 79 cancer tissues from different cohort. Cox-regression analysis was used to assess the association between disease-free survival and clinicopathologic findings. Linear discriminant analysis was used to find a linear combination of disease-free survival. Results: Univariate analysis showed lymph node metastasis (LNM), parametrial invasion (PMI), and FIGO stage were significantly associated with disease-free survival (P <0.01, P =0.01, P=0.01, respectively). Multivariate analysis adjusted LNM, PMI, and FIGO stage showed that LNM was an independent prognosis factor (P=0.02). Linear discriminant analysis based on the clinical factors including LNM and PMI, and the quantitative methylation values on five loci of ATP8A2 gene and three loci of CALCA showed that this clinico-epigenetic combination classified more accurately the patients with respect to recurrence (hit ratio=97.6%) than LNM (hit ratio=78.5%). Conclusion: LNM is an independent clinical predictor of recurrence and the clinico-epigenetic combination accurately defines the prognosis of cervical cancer.
