      • Generation of Transgenic Pigs Carrying a Construct for Endothelial Specific Expression of CD73

        Despite of the absence of hyperacute rejection and acute humoral xenograft rejection, the organ graft of the a1,3-galactosyltransferase (GalT) gene knockouted (KO) and complement regulatory protein (CRP) expressing pig into a nonhuman primate is rejected by development of a thrombotic microangiopathy and/or a consumptive coagulopathy. Thus further introduction of genes to overcome the coagulation incompatibilities between pig and primate under GalT KO/CRP genetic background has been strongly suggested. CD73 (ecto-5'-nucelotidase) is an enzyme attached via a glycosyl phosphoinositol anchor to the extracellular membrane of endothelial cells, which catalyses the hydrolysis of adenosine triphosphate to adenosine. Loss of activity of CD73 results in activation and aggregation of platelets by a reduced capacity to convert nucleotides to adenosine. In previous study, we reported generation of GalT KO fibroblasts concurrently expressing membrane cofactor protein and produced cloned pigs by nuclear transfer of the fibroblast cells (1). In this study, we constructed a vector for expression of human CD73 under control of promoter of pig Icam2 gene expressed specifically at endothelial cells. This vector was introduced into porcine fibroblasts using the nucleofection technology, by which we had forty three fibroblasts clones carrying pIcam2- CD73 vector. Somatic cell nuclear transfer resulted in generation of two transgenic piglets survived.

        University education is not enough with only lectures in order to achieve its educational goals. University is, by definition, a place where inividual academic researches and studies of professors and students are to take place. For this purpose, university students are required to do various kinds of home work, individual study, and report writing. This study tries to investigate the types and amount of individual learning tasks which is provided to the students of Inchon National Teachers University. There are various kinds of individual learning tasks : reading books, finding material, observation and experimentation, skill practice, and report writing. As the results of the study, we can find that : (1) Students are required to take more or less than 10 courses in a semester and every course require 2 or 3 reports. (2) Many students feel that the amount of individual tasks are too heavy to solve within the given time. Especially, the most heavy amount of tasks are given during the third year of university curriculum. (3) Most of the students think that individual study tasks are necessary for university education. But they hope the tasks should be inquiry-oriented rather than memory or mechanical work-oriented. (4) Students want to have a space like Curriculum Material Preparation Area(CMPA) in order to make individual learning tasks.

        In this paper, the use of versatile digital image storege and processing system for scanning acoustic microscope(SAM system) has been described. In addition to the data acquisition and storage functions, different types of image processing techniques of relevance to scanning acoustic microscope have been studied and analyzed. These include gray level-thresholding and edge enhancement, spatial filtering, and pseudo-color processing algorithms. In experiment, it has been shown that these techniques could enhance particular aspects of the acoustic image of filter out unwanted noise.

        In this paper, we proposed to use tone-burst signal instead of impulse signal for performance enhancement of ultrasonic concrete inspecting. In experimented results, it has been shown that input-to-output ratio of the tone-burst signal could obtain higher than that of impulse signal.

      • 加水分解 牛皮粉의 飼料的 價値에 관한 硏究

        Protein quality of hydrolyzed cattle skin meal was estimated by the pepsin-HCL digestion with various concentration and incubation time, and by chemical score to determine the limiting amino acids. Protein efficiency ratio(PER), net protein ratio(NPR) and net protein utilization(NPU) were also studied with chicks fed semipurified diet with and without supplementation of the limiting amino acids. Broiler chick growth trials were also conducted to determine the supplementation level of hydrolyzed cattle skin meal by replacing with soybean meal and feather meal. The results are summarized as follows: 1. Protein digestibility of hydrolyzed cattle skin meal was 66.31%, 80.69%, 83.72%, 84.65%, and 87.45%, respectively for 4, 8, 12, 16 and 20 hours incubation of 0.2% pepsin-HCl. Protein digestibility maintained above 80% after 8hours incubation and increased by increasing incubation time. 2. Protein digestibilities of hydrolyzed cattle skin meal incubated for 16 hours at 0.2%, 0.1%, 0.05%, 0.025%, and 0.0125% pepsin-HCl solution were 85.10%, 82.08%, 76.18%, 74.67% and 64.82%, respectively. 3. Chemical scores of methionine, isoleucine, threonine, leucine and histidine estimated based on NRC(1977) broiler(03 weeks) feeding standard were 27%, 38%, 44%, 48% and 51%, respectively 4. Growth rate, feed intake and feed conversion were improved by the addition of the limiting amino acids in the hydrolyzed cattle skin meal as compared with hydrolyzed cattle skin meal alone. 5. PER, NPR and NPU were also improved by the addition of the limiting amino acids. 6. Feed intake was not statistically significant upto 5% hydrolyzed cattle skin meal in the diet with was 100% replacement of hydrolyzed cattle skin meal with soybean meal and feather meal. Growth rate and feed conversion were not statistically significant at 3% but were significant 5% hydrolyzed cattle skin meal(p<0.05). These experiments were clearly indicated that hydrolyzed cattle skin meal could supplement in the diet upto 3% when mixed with 2 or 3 other protein sources.

      • 저염농도의 자리발효식품의 가공에 관한 연구

        This study was carried out to obtain the basic data on salted and fermented damsel fish(Jari-Jeot), one of the important traditional marine foods in Cheju-Do. and also to seek salt lowering method for the development of a new local tourist food. The results are as follows : The best organoleptic results were obtained after 60 days' fermentation for 25.0, 12.5 and 10% of salt-added group, and around 45 days for 7.5 and 5% of salt-added group. In raw fish ingredient IMP was abundant which marked 17.7 μmole/g while in fermented fish hypoxanthine was predominant but ATP and ADP were not detected after 30 days' fermentation. Sixteen kinds of free amino acids were detected and identified in Jari-Jeot instead of 17 kinds of those in raw sample. The abundant amino acids in Jari-Jeot after 60 days' fermentation were lysine. alanine, glutamic acid, glycine and leucine, and those were consisted of approximately 60% of the total free amino acids. During fermentation the content of TMAO was decreased gradually and no TMAO was detected after 75 days' fermentation while that of TMA was increased during fermentation up to 45 days' and decreased afterwards. The fatty acid of low-salt Jari-Jeot was composed of 31.9% of saturated acids, 13.2% of monoenoic acids and 54. 9% of of polyenoic acids. While the content of EPA in polyenoic acids was 1.9%, that of DHA was 23.0%. In order to lower salt concentaration of fermented damsel fish, additon of Pediococcus halophilus culture as a starter together with 10-12.596 of salt, 4-5% of KCI, and 2% of glucose gave as good quality as traditionally made Jari-Jeot.

      • 低鹽濃度의 자리醱酵食品의 加工에 關한 硏究

        제주도의 전통수산식품인 자리젓에 대한 식품학적 기초자료를 얻고 또한 저염화 방안을 찾아 향토관광식품으로 개발하고자 실험한 결과는 다음과 같다. 자리젓의 최적숙성기는 첨가식염농도가 25%, 12.5% 및 10%의 것은 각각 60일 전후였으며 첨가식염농도가 7.5% 및 5%인 것은 약 45일 경우에 가장 맛이 좋았다. 원료 자리돔에서는 핵산관련물질 중 IMP가 17.7 μmole/g으로 가장 많았으나 30일 이상 숙성시킨 경우 전시료에 걸쳐 ATP와 ADP는 검출되지 않았으며 hypoxanthine이 핵산관련물질의 대부분을 차지하였다. 원료 자리돔에서는 17종이, 자리젓에서는 16종의 유리아미노산이 검출, 동정되었다. 숙성60일째의 자리젓에서 함량이 많은 아미노산은 lysine, alanine, glutamic acid, glycine 및 leucine으로 이들이 총유리아미노산의 약 60%를 차지하였다. TMAO는 숙성기일의 지남에 따라 서서히 감소하여 75일째에는 거의 소실되었고 TMA는 45일 까지 급격히 증가하다가 이후 급격히 감소하였다. Pediococcus acidilactici를 starter 로 106cells/㎖접종한 시료에서는 숙성 15일만에 세균수는 최대치에 도달하였으며 45일 이후에는 급격히 감소하였다. 그러나 Pediococcus halophilus 를 ?? 접종한 경우 숙성기간중 유산균 수에는 큰 변화가 없었다. 저염자리젓의 지방산 조성은 포화산이 31.9%내외, 모노엔산이 13.2% 내외 그리고 폴리엔산이 54.9% 내외였다. 폴리엔산 중 EPA는 1.9%정도였으나 DHA가 23.0% 내외로 높은 분포를 보였다. 자리젓의 저염화를 위하여 Pediococcus halophilus를 starter로 첨가하고 식염 10-12.5% KCl 4-5%, 포도당을 2% 수준으로 첨가하면 재래식 젓갈에 손색이 없는 저염자리젓의 제조가 가능할 것으로 판단되었다.       • 항결핵 치료과정에서 호전되었다가 악화된 뇌 결핵종 1예 보고

        In spite of nationmide tuberculosis control program, tuberculosis is still prevalent in Korea. Accordingly, tuberculous meningitis also poses one of the main trouble some problems. Recently, We experienced a perplexing case of tuberculous menin-gitis and tubercdoma in a 24-year old female patient. She was admitted to our hospital owing to meningeal irritation sign, aphasia, VI nerve palsy, and ,fever. CSF analysis showed a finding consistent with that of presumptive tuberculous meningitis.. Initial MRI of the brain revealed a large mass(tuhercu1oma) on left parieto-temporal area. Antitubeculous regimen(1soniazid (INH), rifampin(RMP), ethambutol(EMB), pyrazinamide(PZA)} of meningeal dose was given immediately, and her symptoms and signs were dramatically disappeared with a few days after treatment. She was discharged with improved condition and under maintenance treatment of tuberculosis. Follow-up one year after treatment. revealed that the previous lesion was nearly diminished. She showed no symptom at all until she recognized a total blindness of sudden onset on her left eye a t the 18th month of treatment. Brain MRI checked at that time showed multiple masses of tuberculoma scattered over the brain base and one of them invaded left optic nerve. In our opinion, the cause of relapse in this patient might he explained by the selection and ultimate replication of a minor population of resistant strain. Although such relapse is an extremely rare event, we think that we should always pay attention to this possible resistance problem every time we meet patients with tuberculous meningitis.

        고등학생 스트레스의 횡문화적 연구 : 연변의 한족, 조선족 및 서울의 한국인 비교

        In this transcultural comparison, we examined areas of stress, coping strategies and psychosomatic symptoms resulting form such strategies in Chinese, Korean-Chinese and Korean high school students. A total of 1,042 second-year high school students participated in this study : 213 Chinese from Yanbien, China ; 368 Korean-Chinese from Yanbien, China ; and 466 Koreans from Seoul, Korea. We administered the Stress Questionnaire Form for High school Students(Won & Lee 1995), the Ways of Coping-Revised(Lazarus & Folkman 1984) and the Physical Symptom Scale of Stress(Allen & Hyde 1980) to investigate areas of stress, coping stategies and psychosomatic symptoms, respectively. The findings were understood and interpreted from focus group discussions. Although there were no differences in total stress scores among the three groups, there were significant differences in areas of stress. Chinese students manifested the highest stress scores in peer relation, family problem, heterosexual problem, the future-employment, and religious problem. Koreans scored the highest in academic performance, school life, extracurricular activity, psychological-personality problem, value system, and the future-employment. The Korean-Chinese students scored the highest in extracurricular activity, family-economic problem. In terms of collectivism versus individualism, Chinese students' stress was related more toward the collective situation, whereas the Korean students faced increased stress regarding the individual situation. In coping strategies, the Chinese were characterized by active coping, Koreans by passive coping and Korean-Chinese by a mixture of the two. Koreans had the highest scores in psychosomatic symptoms, suggesting unfavorable coping strategies. The Korean-Chinese students seemed to lie intermediate between Koreans and Chinese in all three study areas, implying their chltural pluralism.

        치과용 도재분말의 제조 및 물리적 성질

        In this study, the prepared glasses were heat-treated to precipitate the leucite crystal in order to control the thermal expansion coefficient which is an important property of dental porcelain powder. The effect of addition of Li₂O and Na₂O on the amount of the precipitated leucite crystal were also examined. The mixing ratio of the leucite-containing glass must be controlled to match the thermal expansion coefficient of dental porcelain to that of metal. The physical properties of the dental porcelain powder prepared in this study were characterized. The results of this study were obtained as follows : 1. Conditions for the preparation of the maximum amount of leuctie crystal were compostion L-2(2.0 wt% Li₂O added) heat-treated at 950℃. 2. The phase of the precipitated crystals was turned out to be the tetragonal leucite crystal. 3. Maximum relative crystallinity was 64.2% for composition L-2, heat-treated at 950℃. Its linear thermal expansion coefficient was 16.1×10-6/℃ in the range of room temperature to 500℃. 4. The optimum mixing ratio of the leucite-containing glass was 45% in order to match the thermal expansion coefficient of dental porcelain to that of metal. In this case, the thermal expansion coefficient of the prepared dental porcelain was 10.6×10-6/℃ (∼500℃). 5. The physical and chemical properties of the prepared dental porcelain were suitable for the requirements of ISO 6872 and 9693.
