        In Vitro Antiviral Activity of Ribavirin Against Severe Fever with Thrombocytopenia Syndrome Virus Myung Jin LEE1, Kye-Hyung KIM1, Jongyoun YI2, SuJin CHOI1, Chung-Jong KIM1, Nak- Hyun KIM1, Kyoung-Ho SONG1, Pyoeng Gyun CHOI1, Ji-Hwan BANG1, Wan Beom PARK1, Eu Suk KIM1, Sang-Won PARK1, Hong Bin KIM1, Nam Joong KIM1, Myoung- Don OH1 Seoul National University College of Medicine, Korea1, Pusan National University School of Medicine, Korea2 Background: Severe fever with thrombocytopenia syndrome (SFTS) is an emerging infectious disease caused by a novel Bunyavirus, severe fever with thrombocytopenia syndrome virus (SFTSV). No effective antiviral therapy is proven yet, but clinical use of ribavirin (RBV) has been tried. We investigated the antiviral effect of RBV against SFTSV in vitro. Methods: To test for cytotoxicity of RBV, Vero cells were treated with different concentrations of RBV (3.90 to 500 μg/mL, two-fold dilution) and analyzed by cell viability MTS assay 48h post-infection. To determine antiviral activity of RBV against SFTSV, Vero cells were infected with SFTSV strain Gangwon/Korea/2012 at 100 TCID50 (50% tissue culture infective dose) per well in a 96-well plate, and RBV was added at the concentrations showing no or minimal cytotoxicity. Viral RNAs were extracted from the culture supernatants and quantifi ed using one-step real-time reverse transcription- PCR to amplify the partial large segment of SFTSV. Statistical analysis was done by one-way ANOVA with Tukey`s post hoc test. Results: Cytotoxicity due to RBV was not observed at RBV concentration =31.3 μg/ mL. Viral RNAs at 24h post-RBV treatment were reduced with increasing RBV concentrations (1-32 μg/mL), compared with those of mock-treated cells (P <0.01, Figure). Half maximal inhibitory concentration (IC50) of RBV was 3.69 μg/mL at 24h post-RBV treatment. Conclusions: Our study shows that RBV has antiviral effect against SFTSV in a dose-dependent manner. Further studies are required to evaluate the effi cacy of RBV in SFTS.

      • 단면형상변화에 따른 점용접 부재의 에너지흡수 특성

        The front-end side members of vehicles absorb the most of the energy during the front-end collision. These side members are mostly consist of single hat shape. The side members absorb more energy under axial load if they have higher strength, and stable folding capacity (local buckling). When structural members are subjected to axial loadings, stress is concentrated on their edges. Thus, in this study, the energy absorption characteristics of the member were analyzed according to change of sectional shape for increase of strength. The side members consist of single hat, double hat, single cap and double cap section member. Based on the static test result of these side member, the impact simulation were carried out and compared with test result.

      • Bacillus sp. CS-17을 이용하여 제조한 청국장의 발효기간에 따른 향기성분 변호

        전통식으로 담근 청국장과 Bacillus sp. CS-17을 이용하여 제조한 청국장의 향기성분을 비교하였다. 전통식으로 제조한 청국장에서 동정된 향기성분은 hydrocarbone류가 5종, alcohol류가 5종, ester류가 4종, acid류가 3종, aldehyde류가 2종, ketone류가 2종, phenol류가 3종, furan류가 2종, pyrazine류가 4종 등 30종이 확인되었다. Bacillus sp. CS-17을 이용한 청국장에서 동정된 향기성분은 hydrocarbone류가 3종, alcohol류가 2종, aldehyde류가 2종, ketone류가 2종, phenol류가 3종, furan류가 3종, pyrazine류가 5종, pyridine류가 3종, 질소함유화합물로는 2종, thiazole류가 1종, 기타 화합물로 1종 등 모두 27종이 확인되었다. 동정된 성분 중에서 2-hydroxy-6-methylbenzaldehyde, 2-heptanone, 2-furancarboxaldehyde, 2.3-dimethyl-5-ethylpyrazine, 2-picoline, 3-picoline, 5-ethy1-2-picoline, trimethyloxazole, 1,2,6-trimethylpiperidine, 2,4,5-trimethylthiazole 및 chloroform 등의 성분들은 전통식으로 제조한 청국장에서는 확인이 되지 않았으나, Bacillus sp. CS-17을 이용한 청국장에서는 동정되었다. This study was conducted to compare flavor components of chunggugjang fermentated with Bacillus sp. CS-17 to those of traditional chunggugjang. 30 flavor components identified in traditional chunggugjang. 30 flavor components identified in traditional chunggugjang were five hydrocarbones, five alcohols, four esters, three acids, two aldehydes, two ketones, three phenols, two furans and four pyrazines. 27 flavor components identified chunggugjang fermentated with Bacillus sp. CS-17 were three hydrocarbones, two alcohols, two aldehydes, two ketones, three phenols, three furans, five pyrazines, three pyridines, two nitrogen containing compounds, one thiazole and one other compounds. Among the detected components, 2-hydroxy-6-methylbenzaldehyde, 2-heptanone, 2-furancarboxaldehyde, 2,3-dimethyl-5-ethylpyrazine, 2-picoline, 3-picoline, 5-ethy1-2-picoline, trimethyloxazole, 1,2,6-trimethylpiperidine, 2,4,5-trimethylthiazole and chloroform were identified in chunggugjang fermentared with Bacillus sp. CS-17, but not in traditional chunggugjang.

        1. Purpose : The main purpose of this study is to examine the historical transition of the diagnosis of pulse and the pulse in the view point of Constitutional diseases. 2. Method : I collect and examine past literatures refered the diagnosis of pulse and the provisions of the pulse in the Dongyi-Suse-Bowon. 3. Results and Conclusion : ▶ Je-ma, Lee suggested that Taeumin's pulse is long and tensible and Soumin's is gentle and weak in the theory of differentiating syndromes of the Dongyi-Suse-Bowon on constitutionsl characters and regarded superficial, sunken, slow and rapid pulse as the principles of the diagnosis of pulse. ▶ According to the each constitutional diseases, the shapes of pulse are changed.

      • 신속한 간흡충증 진단과 혈청 및 혈구 변화양상 분석

        Diagnosis of Clonorchis sinensis requires specific proteins as antigens and it has been developed by using a rapid detection kit which identify circulating antibodies. In this study during Namseoul University Health/Medicine Care Service conducted in October 2010, with the residents in Wondeok-eup Samcheok Kangwon-do, the rapid diagnosis of C. sinensis infections was performed and patients' blood sera were analyzed. From 12 patients' sera of total 83, antibodies to C. sinensis were detected. According to the patients' sera analysis, about 73% of Total bilirubin, 40% of Cholesterol and 40% of Creatinine were abnormally exhibited. In the C. sinensis-positive sera detected by a rapid diagnostic kit, about 92% of Total bilirubin, 33% of Cholesterol and 50% of Creatinine were abnormally exhibited. According to analysis of blood cells, there was no abnormal result except for Hematocrit. In particular, high Eosinophil which means high incidence of C. sinensis infection was not observed. Taken together, it implied that the positive diagnosis of clonorchiasis would be previous infections of C. sinensis and its patients recovered from clonorchiasis have still antibodies in their sera. In addition, it also suggested that C. sinensis-positivity by the rapid diagnostic kit be not consistent with the present infections of C. sinensis.       • 고온 유리의 프레스 성형 공정 시뮬레이션

        This paper addresses a method for numerical simulation in the pressing process of hot glass. Updated Lagrangian finite element formulations are employed for the flow and energy equations to accommodate moving meshes. The model is assumed axi-symmetric and creep flow is assumed due to the high viscosity. Commercial software ANSYS is used to solve the coupled flow and energy equations. Moving contact points as well as free surface during the pressing are effectively calculated and updated by utilizing API functions of CAD software Unigraphics. The mesh distortion problem near the wall is overcome by automatic remeshing, and the temperatures of the new mesh are conveniently interpolated by using a unique function of ANSYS. The developed model is applied to the pressing process of TV glasses. In conclusion, the presented method shows that the pressing process accompanying moving boundary can be simulated by effectively combining general purpose softwares without resorting to special dedicated codes.

      • 回傳三重水槽에서 장애물에 의해 三重點의 변화

        實驗流體의 수평 온도차를 12C로 유지하고 상대높이가 0.3, 0.5, 0.7, 0.9인 장애물을 설치한다. 그리고 회전속도를 2.0rpm에서 0.2rpm까지 증가시켜 장애물의 높이에 따른 轉移點의 변화를 분석했다. 위의 실험으로 다음과 같은 결론을 얻었다. 중간 높이 이하의 장애물은 유체의 흐름을 불안정하게 하는 효과가 있고, 그 이상 높이의 경우는 흐름을 안정하게 하는 효과가 있다. It is mounting each obstacle's height, which is 0.3, 0.5, 0.7 and 0.9 with horizontal temperature difference of experimental fluid maintained 12 C in an annulus rotating. At velocity from 2.0rpm each more 0.2rpm, the effect of this obstacle on the change of transition point was analyzed. From about experiment, these conclusions were drawn: Obstacle in less than middle height of fluid had a destabilizing effect, while obstacle in more than middle height of fluid had a stabilizing effect.

      • 멀티미디어 저작도구와 DATABASE를 활용한 웹 기반 형성평가 방안 연구

        인터넷의 발전으로 우리는 빠른 시간에 많은 정보를 접할 수 있을 뿐 아니라 웹을 이용한 동적인 개인신상관리와 교육에서 교사와 학생간에 웹을 이용한 새로운 인터페이스를 열 수 있게 되었다.형성평가는 교사가 기계적으로 채점하고, 틀렸을 때 이를 교정해 주기보다는 스스로 학습결과의 정답을 확인하고 틀렸을 때 이에 대한 정답을 학생 자신이 찾아내도록 함으로써 형성평가의 과정 그 자체가 학습과정의 일부가 되도록 하여야 한다.본 연구에서는 멀티미디어저작도구로 형성평가 문항을 만들어, 교사와 학생 모두 빠르고 정확하게 채점하여 즉시 결과를 확인하고 feedback 효과를 얻고자 데이터베이스 연동을 이용한 웹기반 형성평가 방안을 제안한다.

      • 계측용 포터블 ESPI장치에 의한 강구조물의 결함 및 진동해석에 관한 연구

        평판의 진동모드와 진동수의 실험적 해석을 비접촉 광학적 방법인 ESPI법을 이용하여 상세히 기술하였다. ESPI를 이용하여 진동모드와 진동수를 측정하기 위해 두 개의 다른 재료 로 된 시험편을 준비하고 세장비의 변화에 따라 두 개의 다른 가진점을 택해 가진 하였다. 측정된 결과는 유한요소해석(FEA) 및 Warburton이 제안한 이론해석과 정성적으로 비교하였다. 실험적 결과는 이론해석 결과는 물론 FEA 결과와도 잘 일치함을 알수 있었다. 결과적으로 ESPI 법이 진동모드와 진동수의 고정도 해석에 이용할 수 있음을 확인하였다.
