김치에서 분리한 Lactobacillus sp. JC-7과 Lactobacillus acidophilus 88간의 Electrofusion 최적조건 설정

김치 발효 숙성기간을 연장하고 신선도를 오랫동안 유지 할 수 있는 김치발효 starter의 개발을 목적으로 최적숙성기에 있는 김치로부터 bacteriocin생성능이 없는 유산균을 분리하여 동정하였으며, bacteriocin생성능이 우수한 L.acidophilus 88을 융합시키기 위해 electrofusion에 대한 최적 조건을 검토하였다. 분리균주는 Lactobacillus속으로 동정되어 Lactobacillus sp. JC-7이라 명명하였다. Electrofusion에 의해 생성된 융합주를 식별하기 위해 streptomycin(2.5㎎/ml)에 내성을 나타내는 Lactobacillus sp. JC-7변이주와 kanamycin(600㎍/ml)에 내성을 나타내는 L.acidophilus 88변이주를 분리하였다. Electrofusion을 100V/㎝, 120msec(72ohms,1670 capacitance) 에서 수행했을 때 융합효율이 가장 양호하였으며 전기장의 세기와 시간이 중가할수록 융합 효율이 현저히 감소하였다. 2가 양이온은 농도가 중가할수록 대체적으로 융합효율을 감소시키는 경향을 나타내었으나 1mM MGCl_2에서는 대조군에 비해 융합효율이 약간 증가하였다. PEG매개에 의한 융합법의 융합효율을 비교한 결과, 융합효율은 chemical fusion<electrofusion<electrofusion+20% PEG순을 나타내었다. Evaluation of Optimum Conditions for the Electrofusion between Lactobacillus sp. JC-7 Isolated from Kimchi and Lactobacillus acidophilus 88. Young-Bae Jo, Hyun-Jung Choi, Hyung-Suk Baik and Hong-Ki Jun*. Department of Microbiology, Pusan National University, Pusan 609-735,Korea-A lactic acid bacterium was isolated from kimchi. The isolated strain was identified as the genus Lactobacillus thrugh its morphological characteristics and named as Lactobacillus sp. JC-7. The optimum conditions for the electrofusion between streptomycin(2.5㎎/ml)resistant mutant of Lactobacillus acidophilus 88 and kanamycin(600㎍/ml) resistant mutant Lactobacillus sp. JC-7 were evaluated. The highest number of fusants were obtained at a capacitance value of 120msec(1670㎌), a field strength of 100V/㎝,and a pulse controller setting of 72Ω. The potimum pH of elecroporation buffer was 7.5 and the concentration of divalent cation was 1mM MG^2+. Electrofusants were efficiently obtained by addition 20% polyethylene glycol to electroporation buffer. The yield of fusion was better than that of using polyethylene glycol mediated chemical induction.

Development of a Reduction Algorithm of GEO Satellite Optical Observation Data for Optical Wide Field Patrol (OWL)

An algorithm to automatically extract coordinate and time information from optical observation data of geostationary orbit satellites (GEO satellites) or geosynchronous orbit satellites (GOS satellites) is developed. The optical wide-field patrol system is capable of automatic observation using a pre-arranged schedule. Therefore, if this type of automatic analysis algorithm is available, daily unmanned monitoring of GEO satellites can be possible. For data acquisition for development, the COMS1 satellite was observed with 1-s exposure time and 1-m interval. The images were grouped and processed in terms of “action”, and each action was composed of six or nine successive images. First, a reference image with the best quality in one action was selected. Next, the rest of the images in the action were geometrically transformed to fit in the horizontal coordinate system (expressed in azimuthal angle and elevation) of the reference image. Then, these images were median-combined to retain only the possible non-moving GEO candidates. By reverting the coordinate transformation of the positions of these GEO satellite candidates, the final coordinates could be calculated.

新羅 哀莊王代의 政治變動과 金彦昇

King Aejang(哀莊王 : 800-809), the 40th king of the Silla dynasty(新羅), ascended the throne at the age of 13, was under the regency of Kim Eon-seung(金彦昇), his uncle, for some time, and was killed by rebels led by Kim after 10 years of his reign. Although it was relatively short, his period of reign witnessed important political upheavals including a series of political reforms. The intent of this study is to examine political upheavals and changes during the period of King Aejang's reign. Kim Eon - seung, who defeated King Aejang's mother to become the regent, was the strongest man in politics at that time. He had started his political activity from the period of King Wonseong(元聖王). It seems that his regency ended in 805, when King Aejang began to rule in his own right. Generally speaking, the regent meant a man or a woman who ruled in behalf of the legitimate but young king for some time, usually until the king came of age. In other words, the regent was a temporary, proxy ruler. King Aejang seemed to rule in his own right from 805, on the sixth year of his reign. It was usually at the beginning of king's accession to the throne that Chinese Tang(唐) emperors invested Silla kings with kings, who in turn invested their mothers and wives with great queens and queens. It was the sixth year of his reign that King Aejang was invested with king by the Tang emperor and he invested his mother and wife with great queen and queen. These investiture ceremonies were as good as the kings accession to the throne in political implications, suggesting that the king started ruling in his own right free from Kim Eon - seung's regency. King Aejang reviewed his troops on the fifth year of his reign. Because king's review of his troops symbolized his authority as commander - in chief of all troops, King Aejang's action was intended to strengthen his regal power. Since then, he newly built and repaired his crown princes palace, which revealed his intention that his young son should legitimately succeed him. Following these actions, he dispatched an envoy to Chinese Tang and asked the Tang emperor to invest him with Silla king. After he began to rule in his own right, King Aejang carried out a series of political refonns for new politics. He redressed the central administrative system, reorganized the local government, and tightened the control of Buddhism. The reformation by King Aejang to reorganize the ruling system and strengthen his regal power seemed to be led by kings own political forces. It was quite natural that regent Kim Eon - seung should resist his nephew's behavior and conflict with him. Finally, in the tenth year of King Aejang's rule, Kim Eon - seung killed the king and ascended the throne as King Heondeok(憲德王), the 41st king of Silla.

꿩고기 가공제품의 저장중 미생물과 휘발성 염기태질소의 변화

To evaluate the storage characteristics of pheasant meat products, contents of aerobic, anaerobic and food poisoning bacteria as well as VBN were measured during storage at various temperatures. 1. During the storage Escherichia coli and food poisoning bacteria including Salmonella, Shigella and Staphylococcus were not detected from any of the pheasant meat products. 2. Total plate counts of aerobic and anaerobic bacteria increased with storage temperature, showing more than 10^6 CFU/g of most pheasant meat products within 5~10 days of storage at 20℃ and 30℃. However, frank sausage, loin ham, pressed ham and salad showed less than 10^5 CFU/g in 20~30 days of storage at 20℃. 3. When stored at 10℃, smoked product, electric roasted product and pressed ham showed the bacterial counts of more than 10^4 CFU/g within 10 days of storage. Frank sausage, loin ham and salad, however, showed less than 10^3 CFU/g in 10 days of storage at 10℃. 4. VBN contents of smoked product, electric roasted product and frank sausage exceeded edible limit of 20 mg%, showing more than 40 mg% and 8 mg% within 5 days and 10 days, respectively, of storage at 10℃. In contrast, loin ham, pressed ham and salad had the VBN of less than 20 mg% in 10 days of storage at 10℃. In summary, while pheasant meat products in general appear to be prone to microbial growth, loin ham and salad are thought to have a longer storage period than others, showing about 10 days of preservation at 10℃. Products other than loin ham and salad are suggested to be stored frozen or refrigerated at below 10℃.

간세포암종에 대한 간동맥 화학색전술 후 발생한 급성호흡곤란증후군

Transcatheter arterial chemoembolization (TACE) has become an effective alternative treatment strategy for patients with inoperable hepatocellular carcinoma (HCC). Although TACE is relatively safe, acute respiratory distress syndrome associated with pulmonary lipiodol embolism is a rare and potentially fatal complication. We report a rare case of acute respiratory distress syndrome after TACE for inoperable HCC. A 75-year-old man, with huge HCC in right lobe, was treated by TACE for the first time. Seven hours after uneventful TACE procedure, he felt dyspneic and his oxygen saturation recorded by pulse oximetry (SpO2) fell to 80% despite of applying non-rebreathing mask. He underwent mechanical ventilation with a protective ventilatory strategy. We experienced a case of acute respiratory distress syndrome after TACE for HCC.

인터넷 의료상담에 관한 분석 및 연구

Background and Objectives: Telemedicine uses computer and telecommunication technologies to provide medical information and services. Telemedicine can be useful in any situation in which there is a physical barrier between the doctor and the patient. We analyzed medical consultations from Chosun University Hospital is home page (http://hosp.chosun.ac.kr) to find problems on the home page and to offer high quality medical information services. Methods : We collected 544 queries of medical consultation on our home page from May 1998 to May 1999. From medical consultation data on the internet web site of Chosun University Hospital, consulted documents were analyzed with regard to medical department, patients’ age, consultation by attorney, and dermatologic consultations. Results : The telemedicine system is a relatively satisfactory method to both patients and physicians due to easier access and almost rea1-time medical consultation through the world wide web. But several problems should be solved to provide a more effective and lawful telemedicine environment. Conclusion : As a consequence to analyzed medical consultations on our home page, there were several problems in our store-and-forwarding system. To establish an effective telemedicine system, we have to prepare protocols to receive sufficient information and patients have to send digital images to make help an accurate diagnosis. Also problems of liability, reimbursement and coordinated national policy need to be solved.

Pityriasis rotunda is an uncommon cutaneous disorder consisting of asymptomatic, strikingly circular, scaly lesions on the trunk and extremities, with the histologic appearance of ichthyosis vulgaris. A case of pityriasis rotunda in a 35-year-old woman in whom no diagnosable disorder could be demonstrated is presented.

후두 및 하인두 편평세포암종에서 E-cadherin의 면역조직화학적 발현

Background and Objectives : The cell-cell adhesion molecule E-cadherin is necessary for the maintenance of the epithelial cellular structure. We were designed to confirm the significance of E-cadherin as a marker for differentiation and invasiveness of squamous cell carcinoma of the larynx and hypopharynx. Materials and Methods : Our study was investigated for the immunohistochemical expression of the epithelium-specific cell adhesion molecule E-cadherin in formalin fixed paraffin embedded tissue specimen of 32 cases of squamous cell carcinoma of larynx and hypopharynx. Results : The positive expression of E-cadherin was 56.3%(18 cases) in squamous cell carcinoma of the larynx and hypopharynx. Decreased E-cadherin expression was a stastistically significant correlation with a increased grade of lymph node stage and clinical stage. Reduced expression was seen in the large tumor size and poorly differentiated tumors, but these result was not statistically significant. Conclusion : The expression of E-cadherin may be related with progression of squamous cell carcinoma of the larynx and hypopharynx. But these correlation were not sufficient for the prognostic indicators in squamous cell carcinomas of the larynx and hypopharynx.
