        Radiation Hybrid Mapping and mRNA Expression of Chicken N- myc downregulated gene 4

        N-myc downregulated gene 4 (NDRG4) is a member of the N-myc downregulated gene family which belongs to the alpha/beta hydrolase superfamily. The protein encoded by this gene is a cytoplasmic protein that may be involved in the regulation of mitogenic signalling in vascular smooth muscle cells. To map NDRG4 gene in chicken chromosome, a 6,000 rads chicken- hamster radiation hybrid panel was used. Primers were designed according to the published human sequence for amplification of chicken NDRG4. We compared the corresponding human mRNA sequence with the predicted coding sequence of chicken NDRG4, and found that the assembled contig shared a high percentage of similarity with that of human gene. PCR of samples from ChickRH6 revealed the locations of NDRG4 to be linked to the maker PARD6G (5 cR away) with a LOD score 20.46. In addition, we detected the mRNA expression and distribution of chicken NDRG4 in various tissues by RT-PCR, and found that NDRG4 was highly expressed in chicken brain and heart, whereas lowly but detectable in thymus. The mRNA expression of this gene in chicken liver, spleen, lung and muscle was rarely detectable under present experimental conditions.

      • Variation of Urinary and Serum Trace Elements (Ca, Zn, Cu, Se) in Bladder Carcinoma in China

        Backgrounds: Deficiency or excess of trace elements can induce body metabolic disorders and cellular growth disturbance, even mutation and cancerization. Since there are few studies of the effect of trace elements in bladder carcinoma in China, the aim of this study was thus to assess variation using a case control approach. Methods: To determine this, 81 patients with bladder carcinoma chosen as a study group and 130 healthy persons chosen as a control group were all assayed for urinary and serum trace elements (calcium [Ca], zinc [Zn], copper [Cu], selenium [Se]) using an atomic absorption spectrophotometer, and the results were analyzed by independent sample t tests. The correlative factors on questionnaires answered by all persons were analyzed by logistic regression. Results: The results showed urinary Ca, Zn and serum Cu levels of the study group to be significantly higher (P<0.05) than those of he control group. Serum Ca and Se levels of study group were significantly lower (P<0.05) than those of control group. Conclusion: There were higher urinary Zn and serum Cu concentrations in bladder carcinoma cases. Bladder carcinoma may be associated with Ca metabolic disorder, leading to higher urinary Ca and lower serum Ca. Low serum Se and smoking appear to be other risk factors for bladder carcinoma in China.

        Effective mucosal live attenuated Salmonella vaccine by deleting phosphotransferase system component genes ptsI and crr

        Salmonella enterica is a major human pathogen that causes invasive non-typhoidal Salmonellosis (iNTS), resulting in significant morbidity and mortality. Although a number of pre-clinical and clinical studies have reported on the feasibility of developing a safe and effective vaccine against iNTS, there have been no licensed Salmonella vaccines available to protect against NTS strains. Vaccine formulations of highest priority for NTS are live attenuated vaccines, which can elicit effective induction of intestinal mucosal and intracellular bacteria-specific cell mediated immune responses. Since glucose is crucial for intracellular survival and replication in host cells, we constructed strains with mutations in components of the glucose uptake system, called the phosphotransferase system (PTS), and compared the relative virulence and immune responses in mice. In this study, we found that the strain with mutations in both ptsI and crr (KST0556) was the most attenuated strain among the tested strains, and proved to be highly effective in inducing a mucosal immune response that can protect against NTS infections in mice. Thus, we suggest here that KST0556 (ΔptsIΔcrr) is a potential live vaccine candidate for NTS, and may also be a candidate for a live delivery vector for heterologous antigens. Moreover, since PTS is a well-conserved glucose transporter system in both Gramnegative and Gram-positive bacteria, the ptsI and crr genes may be potential targets for creating live bacterial vectors or vaccine strains.

        Characteristics Comparison before and after Lyophilization of Transferrin Modified Procationic-Liposome-Protamine-DNA Complexes (Tf-PLPD)

        A novel non-viral gene delivery system, Procationic-Liposome-Protamine-DNA complexes (PLPD) which could further adsorb transferrin on the surface as a targeting ligand to form Tf- PLPD, was prepared and characterized before and after lyophilization. The size distribution of Tf-PLPD was in the range of 240 ± 12 nm and the zeta potential was -24.10 ± 2.5 mV. The transfection efficiencies of PLPD and Tf-PLPD were 12.18 ± 3.8 and 24.26 ± 2.6 mU β-galactosidase/ mg protein respectively. The lyophilization and the presence of serum didn’t affect the tansfectivities of PLPD or Tf-PLPD. Compared to LipofectamineTM 2000 (Invitrogen, U.S.A.), the procationic liposomes had less cytotoxicity to cells. In summary the procationic lipoplex described here, combining the condensing effect of protamine and the targeting capability of Tf, was a perspective non-viral vector for gene delivery system.

        Mining association rules is an important research direction in the field of data mining. Related studies have proposed many used to efficiently find large-scale database association rules algorithm, but the research on maintenance problem of association rules is less. Especially many transaction database is always in constant updates. Increase or decrease occurs when the database or dataset minimum support after the change, how to maintain the association rules have been, it got the attention of many researchers. Based on IFP-Growth increment of association rules mining model and to modify the FP-tree, put forward the suitable for transaction data and support the tree model of change, at the same time under different conditions is given incremental association rules mining algorithm, and reduce the frequency of the original dataset range query and query, and in a case of massive dataset multi-level tree structure decomposition, dynamic allocation rule tree branches, ensure load balancing, improve operation efficiency.

        Coenurosis of Yak, Bos grunniens, caused by Taenia multiceps: A Case Report with Molecular Identification in Qinghai Tibetan Plateau Area, China

        Coenurosis is an important zoonotic helminthic disease caused by the larval stage of the tapeworm Taenia multiceps. This parasite typically infects the brain of the intermediate hosts, including sheep, goat, cattle and even hu- mans. We report a case of T. multiceps infection in a yak confirmed by clinical symptoms, morphological characteristics, and molecular and phylogenetic analyses. The coenurus was thin-walled, whitish, and spherical in shape with a diameter of 10 cm. The parasite species was identified as T. multiceps by PCR amplification and sequencing of the 18S rRNA, cox1 and nad1 genes. Three gene sequences all showed high homology (all above 97%) with the reference sequences from different hosts. Moreover, phylogenetic reconstructions with the 3 published Taenia gene sequences confirmed that the Qinghai yak isolate was closely related to T. multiceps. Although there are advanced diagnosis and treatment meth- ods for coenurosis, early infection is difficult to diagnose. Importantly, the findings of yak infection case should not be ig- nored due to its zoonotic potential.

        Molecular and enzymatic characterization of acid phosphatase from venom of Scleroderma guani

        Acid phosphatase (ACPase) is a common component in venom of parasitoids. Although extensive researches regarding this enzyme have been conducted in many other organisms, its characteristics as a venomous enzyme are still sparsely known. In this study, we aimed to reveal the gene expression patterns, and structural and biochemical properties of an ACPase from the venom of Scleroderma guani. The cloned open reading frame of venomous ACPase gene of S. guani was 1218 bp encoding 406 deduced amino acids, shared 40% and 41% identities to ACPases from venoms of Apis mellifera and Pteromalus puparum, respectively. The structural analysis of this ACPase implied common functions and differences to the honeybee venom ACPase. qPCR analysis showed that this gene was abundantly expressed in the venom apparatus, and most highly expressed in the adult stage after one and three days emergence. Activity assay using p-nitrophenyl phosphate as the substrate revealed that the optimal pH and temperature for this venomous enzyme was 4.8 and 45 °C, respectively. NaF is an effective inhibitor for it. The results will enrich our knowledge for the ACPase as toxin, which may contribute to further uncovering its role involved in parasitism.

      • An Improved Plant Regeneration Protocol using Cotyledonary Explants from Inbred Lines of Chinese Cabbage (Brassica rapa ssp. pekinensis)

        We studied the effect of genotype, explant, age of explant, medium (plant growth regulators and gelling agents), and standardized an efficient regeneration protocol for inbred lines of Chinese cabbage (Brassica rapa ssp. pekinensis). Of the different concentrations of NAA and BA tested, the combination of $5\;\cal{mg/L}\;BA\;and\;0.5\;\cal{mg/L}$ NAA gave the highest frequency of shoot regeneration. The cotyledonary explants had more shoot regeneration frequency ($\ge40\%$) than the hypocotyl ex-plants. Besides, the cotyledonary explants, excised from the 4-day old seedlings, showed higher shoot regene-ration ($56.7\%$). Of the three gelling agents and their concentrations used, 16 g/L agar was found to be the best for shoot regeneration. Shoot regeneration frequency in-creased significantly by supplementing the medium with $4\;\cal{mg/L}\;of\;AgNO_3$ MS medium devoid of NAA showed higher frequency of rooting in the regenerated shoots than the ones supplemented with NAA. Our improved regeneration protocol will be especially useful for the genetic transformation of Chinese cabbage inbred lines to develop transgenic hybrids.

      • Autophagy Inhibition Promotes Gambogic Acid-induced Suppression of Growth and Apoptosis in Glioblastoma Cells

        Objective: To investigate the effects of gambogic acid (GA) on the growth of human malignant glioma cells. Methods: U251MG and U87MG human glioma cell lines were treated with GA and growth and proliferation were investigated by MTT and colony formation assays. Cell apoptosis was analyzed by annexin V FITC/PI flow cytometry, mitochondrial membrane potential assays and DAPI nuclear staining. Monodansylcadaverine (MDC) staining and GFP-LC3 localisation were used to detect autophagy. Western blotting was used to investigate the molecular changes that occurred in the course of GA treatment. Results: GA treatment significantly suppressed cell proliferation and colony formation, induced apoptosis in U251 and U87MG glioblastoma cells in a time- and dose-dependent manner. GA treatment also lead to the accumulation of monodansylcadaverine (MDC) in autophagic vacuoles, upregulated expressions of Atg5, Beclin 1 and LC3-II, and the increase of punctate fluorescent signals in glioblastoma cells pre-transfected with GFP-tagged LC3 plasmid. After the combination treatment of autophagy inhitors and GA, GA mediated growth inhibition and apoptotic cell death was further potentiated. Conclusion: Our results suggested that autophagic responses play roles as a self-protective mechanism in GA-treated glioblastoma cells, and autophagy inhibition could be a novel adjunctive strategy for enhancing chemotherapeutic effect of GA as an anti-malignant glioma agent.
