        어성초 추출물의 세포독성과 향균효과 (IV)

        This study was carried out to evaluate cytotoxic effects of Houttuynia cordata Thunberg extracts on murine leukemia tumor cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazol iumbromide (MMT) assay. The comparison of $IC_{50}$ values of Houttuynia cordata Thunberg extracts on $L1210,\;P388D_1$ and Vero cell lines showed that the methanol extract of Houttuynia cordata Thunberg indicated the most antitumor activity in the MTT assay. In order to develop a antimicrobial agent, dried Houttuynia cordata Thunberg was extracted with several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of amocla and ketoconazole as references was compared to those of other solvent extracts such as $H_2O$, n-hexane, chloroform, ethyl acetate ethanol and methanol. The antimicrobial activity of all extracts from the sample had growth inhibition activity against gram-negative bacteria, yam-positive bacteria and fungi $(MIC,\;>\;200\;{\mu} g/ml)$. These results suggest that the methanol soluble extract of Houttuynia cordata Thunberg may be a valuable choice for the studies on the treaeent of murine leukemia tumor cell lines and antimicrobial agents.

        참쑥 정유의 항세균 및 항진균 효과

        참쑥의 정유성분을 추출하여 수종의 병원성 세균과 곰팡이를 대상으로 항세균 및 항진균 효과를 실험하여 다음과 같은 결과를 얻었다 그람 양성균인 S.epidermidis와 S. aureus는 200ppm의 농도에서, S. mutans는 1,000ppm의 농도에서 세균성장 억제효과가 나타났으며, 통계학적으로 유의성이 인정되었다.(P<0.05). 그람 음성균인 Z.mobilis와 En. faecalis는 200ppm의 농도에서. Ps. putida는 400ppm 농도에서 세포성장 억제효과가 나타났다(P<0.05) V. parahaemolyticus는 800ppm의 농도에서 Ps. aeruginosa는 1,000ppm의 농도에서 세포성장 억제효과가 나타났다. (P<0.05) 효모형 곰팡이인 C. albicans와 Cr. neoformans는 200ppm의 농도에서 세포성장 억제효과가 나타났다(P<0.05) 사상형 곰팡이인 Al. mali는 600ppm의 농도에서,A. nidulans는 400ppm의 농도에서, F. oxysporum은 100ppm의 농도에서 세포성장 억제 효과가 관찰되었으며. 이들 중 A nidulans에 대한 효과가 가장 높게 나타났다. Essential oil of Artemis a lavandulaefolia, the chrysanthemum family plant used in the chinese medicine. was extracted and antibacterial and antifungal activity with many kinds of the pathogenic bacterium and fungi was experimented by it. Staphylococcus epidermidis, and Streptococcus aureus, gram positive bacterium, at the concentration of 200ppm and Streptococcus mutans at the concentration of 1,000ppm showed the growth inhibition effect of the cell. These showed statistically significant difference(p<0.05). Zymomonas mobilis, Entrecoccus faecalis, gram negative bacterium, at the concentration of 200ppm and Pseudomonas putida at the concentration of 400ppm showed the growth inhibition effect of the cell (p<0,05), V. parahaemolyticus at the concentration of 800ppm and Pseudomonas aeruginosa at the concentration of 1,000ppm showed the growth inhibition effect of the cell(p<0,05). Candida albicans and Cryptococcus neoformans, yeast-type fungi, showed the gorwth inhibition effect of the cell at the concentration of 200ppm(p < 0.05). Altenaria mall, Aspergillus nidulans and Fusarium oxysporum, filamentous fungi, took the growth inhibition effect of the cell at the concentration of 600ppm, 400ppm and 100ppm respectively.

        고삼의 Streptococcus mutans에 대한 항세균 효과

        고삼에서 치아우식원인세균인 S. mutans에 대한 항세균효과와 GTase의 활성을 저해하는 물질을 탐색, 개발하여 식품 또는 치아우식예방제로 응용하기 위하여 연구를 수행하였다. 고삼 에틸 아세테이트추출물의 3차 분획의 소분획에서 S. mutans에 대한 최소억제농도가 3.13 ㎍/ml으로 나타나, 높은 항세균효과를 관찰할 수 있었다. GTase 활성은 고삼의 에틸 아세테이트 추출물에 의하여 glucan의 형성을 저해하였으며, 100 ㎍/ml의 농도에서 77%의 저해율이 나타났고, 통계학적으로 유의한 효과가 나타났다 (p<0.05). 이상의 결과로 고삼은 치아우식예방제로 개발 가능성이 있다고 기대된다. The purpose of this study is to investigate antibacterial activities of Sophora flavescens Ait. extracts for the prevention of dental caries caused by Streptococcus mutans. The fraction 5-4-3 of the ethyl acetate soluble extract of Sophora flavescens Ait. Showed strong growth inhibition activity against Streptococcus mutans (MIC, 3.13 ㎍/ml). The glucosyltransferase activity inhibited the formation of glucan by the fraction 5-4-3 and showed 77% of the antiproliferative effect at 100 ㎍/ml. This showed a significant activity (p<0.05). These results suggest that the fraction 5-4-3 of the ethyl acetate soluble extract of Sophora flavescens Ait. may be a valuable material as the prevention of dental caries.

        현호색 추출물질의 항진균효과

        Antifungal activities of the two antifungal substances, HWY-87 and HWY-89 extracted from Corydalis turtschaninovii were examined using several yeast types and filamentous fungi including C. albicans and A nidulans. The antifungal activities of the two substances were also compared with two comercial antifungal drugs, KCZ and MCZ and the result reads as follows. 1. The activity spectrum of the antifungal substances on C. albicons showed the concentration of 5ug in HWY-89, 25ug in HWY-87, 5ug in MCZ, and 50ug in KCZ and the inhibitory effect on cell growth showed statistically significant difference. (P<0.001) 2. Yeast type fungi, C. albicans, Cr. neoformans and S. sereviziae showed the inhibitory effect in the cell gorwth at the concentration of 25ug in HWY-89. 3. In filamentous fungi, Al, mali, the cell growth inhibitory zone was significantly formed at the concentration of 50ug in MCZ. (P<0.01) 4. In the index of the growth of C. albicans, the fungicidal effect showed at the concentration of 25ug in HWY-89 in four hours. 5. In the experiment of the survival of A. nidulans, fungicidal effect showed at the concentration of 50ug in HWY-89 and MCZ in eight hours. As the result, HWY-89 of antifungal substances extracted from Corydalis turtschaninovii showed the strong antifungal effect and showed higher antifungal effect in yeast type fungi than filamentous fungi.
