        여우주머니의 Phenol성 화합물

        The herbaceous species of phyllanthus in this genus are used as hyperglycemic, diuretic, and malaria agent in the world. For the phytochemical studies and the investigation of medicinal resources in the Phyllanthu species, phyllanthus species, (Euphorbiaceae) were used and the studies of constituents in this plant were carried out. From the aqueous fraction of methanolic extract, one flavonoid (quercetin-3-0-rutinoside), two gallotannins(gallic acid, methyl gallate), and two ellagitannins (corilagin, geraniin)were isolated through fractionation and repeated column chromatography using Amberlite XAD-4, ODS gel, and sephadex LH-20. The structures of these compounds were identified on the basis of spectroscopic evidences.

        다구찌 기법을 이용한 멸치 고압효소가수분해 공정의 최적화

        본 연구는 멸치 고압효소가수분해물의 산업화 생산 공정을 개발하기 위한 기초연구로서 다구찌 기법을 사용하여 고압과 효소를 이용하는 멸치 고압효소가수분해 공정의 운전 조건을 최적화하고, 최적화 공정 조건으로 가수분해물을 생산하여 품질특성 및 성능치의 개선 효과를 산출함으로써 신뢰성을 검증하였다. 다구찌 기법의 망대특성과 L<sub>9</sub>직교배열을 사용한 실험계획을 통하여 멸치 고압효소가수 분해물의 특성치를 최대화시키는 공정 조건으로, DH의 경우에는 압력 75-100 MPa, 반응온도 60℃, 반응시간 12 h, 복합효소농도 1%이고, NR의 경우에는 압력 100 MPa, 반응온도 50℃, 반응시간 12 h, 복합효소농도 1%이며, 수율의 경우에는 압력 100 MPa, 반응온도 60℃, 반응시간 12h, 복합효소농도 2%이었다. 최적 조건에 대한 확인 실험으로 압력을 75 MPa, 반응온도 50℃, 반응시간 12 h, 복합효소농도 1%로 설정하여 예측값과 실험값을 비교한 결과, 최적 조건의 신뢰성을 확보함을 확인하였다. 최적화 조건을 적용하여 생산한 멸치 고압효소가수분해물은 시판 멸치 어간장 및 액젓보다 더욱 풍부한 풍미증진 유리아미노산을 함유하고 있었으며, 필수아미노산 역시 풍부하였다. 뿐만 아니라 상압 조건에서의 가수분해물보다 가수분해도가 52.6% 이상 증진되었다. A Taguchi robust design method with an L<sub>9</sub> orthogonal array and larger-the-better characteristics was implemented to optimize experimental conditions for the hydrolysis of raw anchovy using a pressure-assisted enzymatic reaction method. The degree of hydrolysis (DH), nitrogen recovery (NR) and yield were considered as the response param-eters. Pressure, reaction temperature, reaction time, and mixed enzyme amount were chosen as control parameters. As a result of the Taguchi analysis in this study, the pressure was found to be the most influential parameter on DH and NR. The amount of mixed enzyme in the reaction also had a significant effect on DH and NR. Meanwhile, the optimum values were confirmed to be similar at 95% confidence and 5% significance level through analysis of variance (ANOVA). Furthermore, new hydrolysates at optimum conditions and control hydrolysates at atmospheric pressure were compared in terms of the DH, resulting in the improvement of DH by more than 52.6%.

        고압 조건에서 멸치가수분해물 제조를 위한 산업용 단백질분해효소의 선별

        국내산 멸치의 효소가수분해를 개선하기 위하여 압력을 활용하고자, 중고압(200 MPa 이하) 하에서 비교적 효소활성이 안정하며 쓴맛이 제거된 가수분해물을 생산할 수 있는 상업용 단백질 분해효소를 선별하고자 하였다. 상업용 효소인 Alcalase® 2.4 L, FlavourzymeTM 500 MG, Neutrase®, ProtamexTM 및 AlcalaseTM 2.4 L와 FlavourzymeTM500 MG을 1:5의 비율로 혼합한 ME에 대한 안정성 평가로써 50oC에서 상압 및 100 MPa의 조건에서 처리시간에따른 잔존효소활성과 전기영동을 실시하여 고압 처리 시 효소 활성의 잔존율이 증가하고 효소의 형태학적 변화가 없이 안정함을 확인하였고, 고압에서 잔존 활성이 높은 FlavourzymeTM 500 MG과 ME를 1차적으로 선별하였다. 동일한 조건으로 멸치 효소가수분해물을 제조하여 고압의 효소반응 증진 효과, 짠맛증진물질 생산 가능성 및 쓴맛 감소 가능성을 검토하였다. 가수분해도를 분석하여 가수분해 반응 증진 효과는 고압에 의한 영향이 효소에 의한영 향보다 2배 이상 우월함을 확인하였고, GPC 크로마토그 램으로 짠맛증진물질의 생산 가능성을 확인하였으며 이 중에서 Alcalase® 2.4 L가 가장 우수하여 2차로 선별하였다. 1차 및 2차에서 선별된 효소로 100 MPa 및 50oC 조건에서 제조된 멸치 효소가수분해물의 쓴맛 강도와 지표성분으로 tryptophan 함량을 측정하여 mildly bitter taste와 tryptophan을 가장 적게 생산하는 ME를 최종적으로 선별하였다. Proteolytic enzymes were screened to obtain anchovy protein hydrolysates (APH) with low-bitterness under pressurization. First, the residual proteolytic activity (%) and SDS-PAGE of commercially available food-grade protease (Alcalase® 2.4 L, FlavourzymeTM 500 MG, Neutrase®, ProtamexTM) and mixed enzyme (ME, Alcalase® : FlavourzymeTM = 1 : 5) after treatments of ambient pressure and 100 MPa at 50oC were investigated. The proteolytic activity of enzymes was still largely retained after pressure treatment. The SDS-PAGE on tested enzymes showed that the electrophoretic mobility maps (EMMs) after pressure treatment were similar to those at ambient pressure. With these results, it seemed that tested enzymes were stable at 100 MPa and 50oC. Raw anchovy hydrolyzed with various protease at 50oC under ambient pressure and 100 MPa for 1, 2, 6, 12, and 24 h. The degree of hydrolysis (DH, %) of prepared APHs at 100 MPa was increased, which suggests that the medium pressure treatment accelerates protein hydrolysis. The APH by ME exhibited the best suitable characteristics, which had a relatively higher DH (36.2?}1.8- 57.5?}2.8), a high content of oligopeptides with a molecular mass lower than 1,100 Da, mildly bitter taste as the bitterness value was lower than that of 0.2 g/L tea alkaloid, and the lowest contents of tryptophan. Thus, ME was selected to obtain APH with low-bitterness under pressurization.

        Pilot Plant Scale 중고압장치를 이용한 멸치의 효소적 가수분해

        중고압효소가수분해 공정의 산업화를 위하여 설정한 생멸치고압효소가수분해 최적 조건(복합효소 1%, 압력 75MPa, 온도 59℃ 그리고 반응시간 12 시간)으로 pilot plant규모의 중고압 처리장치를 이용하여 중고압멸치효소가수분해물을 제조하고, 이를 pilot plant 규모의 연속식 원심분리기와 막여과 장치(UF와 NF)로 APH 및 분자량 분포가 다른 4가지 분획물을 제조하였다. 그리고 분획물을 분말 제형으로 제조하여 그 유용성을 확대하고자 분무건조 조건을 탐색한 결과, 가수분해는 본 조건에서 거의 달성되었고, APH의 단백질 용해도 및 수율은 각각 63.50%와 55.61%이었다. 분획물의 수율, 특성, 유리아미노산 조성 및 분자량 분포 확인한 결과, 몇몇 분자량 대의 피크들이 여러 분획물에서 발견되는 것을 확인할 수 있었다. 비록 여과막의 한계 분자량별로 peptides가 정확하게 분리되지는 않았지만, 여과막의 한계 분자량이 작아질수록 분획물에서 큰 분자량의 peptides가 감소하거나 제거된다는 사실을 확인할 수 있었다. 또한 짠맛증진물질로 알려진 arginine과 arginyl dipeptides는 10 kDa permeate에 다량 존재할 것으로 추측되었으며 분획 수율이 손실이 27% 이상 발생되어 가수분해물의 산업화를 위해서는 장비 사용법의 훈련 또는 분획 공정의 재검토가 필요하다고 판단하였다. 이에 APH 또는 분획물의 분말 제형화를 위해서는 70% 이상의 수율을 얻을 수 있는 최적화 연구와 좀 더 효율을 높힐 수 있는 공정개선 연구가 필요하다고 판단되었다. Medium pressure and mixed enzyme were used to hydrolyze raw anchovy under controlled conditions at a batch-pilot plant-scale process for the production of anchovy protein hydrolysates (APH). Mass balance calculations were carried out so that the degree of protein solubilization and yields could be estimated. Almost complete hydrolysis could be achieved in 12 h, at 50℃ and 75 MPa, with no pH adjustment, at 1% (10 g/kg) mixed enzyme using raw anchovy. This was achieved with the addition of water (1/2 raw anchovy/water). The degrees of protein solu-bilization and yield were 63.50% and 55.61%, respectively. Fractionation using UF/NF pilot scale systems was car-ried out for producing four different fractions on the APH. Successive fractionation on UF and NF membranes allowed the concentration of the peptides of selected sizes without, however, carrying out sharp separations, and with some MW classes being found in several fractions. Spray drying processes for 10 kDa permeate were described to increase their usability. The free amino acid profile of the fractions was identical to that of the APH.

      • 소아에서 발생한 골수이형성 증후군 2례

        Myelodysplastic syndrome(MDS) is a group of acquired conditions characterized by progressive bone marrow failure associated with normocellular or hypercellular bone marrow that cannot be attributed to nutritional deficiency, chronic infection, or other chronic systemic illness, and cannot be reversed by the successful treatment of those conditions. Whereas the cases of secondary MDS occur at all ages, primary MDS is essentially a disease of elderly, though no age group is exempt and predomonant in men. The MDS is fundamentally clonal disorders at the level of the haemopoietic totipotent stem cell. Clinically, they are characterized by varing degrees of peripheral cytopenia(s) with morphological and functional abnormalities of blood elements. We have experienced two unrelated pediatric cases, who showed pancytopenia on admission and the characteristic marrow findings of MDS. The first case, a 12 months-old girl, showed increased reticulin with abnormal localization of immature precursor cells(ALIP) in her bone marrow, in addition to dysplastic features such as bi-and multi-nucleation of late normoblasts and internuclear bridging. She also showed 22% of blasts in her bone marrow cells thus classified in the category of RAEB-t after FAB classification. Also found were a chromosomal abnormality namely 46 XX t(14q21q), which was an unusual one. Her family brought her home and, 4 month later, she deceased. The second case, a 12 year-old boy, showed pancytopenia with Pseudo-Pelger-Huet anomaly in neutrophils but no blasts in the peripheral blood. Bone marrow of this case also showed the picture of dyserythropoiesis with binuclearity, internuclear bridging, nuclear fragments, multinuclearity of late normoblasts, in addition to megaloblastoid change in the normoblasts and increased mitosis in granulocytic series. No chromosomal abnormality was found. Classified as RA after FAB classification of MDS. Showed good response to conservative management for anemia for more than one year.

        간질성 가성치매 1예

        Epileptic pseudodementia refers to a condition in which elderly patient with memory disturbance due to unrecognized complex partial seizures Memory disturbances in these patients can manifest in two ways discrete episodes of amnesia, or an insidious fluctuating course of memory dysfunction, simulating dementia Cases of epileptic pseudodementia have rarely been reported so far We herein report a patient who presented with progressive dementia and personality change associated with complex partial seizure A 68-year old previously healthy man with 16 years of school education visited our memory disorder clinic because of memory disturbance and changes in personality These symptoms developed insidiously and progressed quite rapidity over 6 months without fluctuation. Detailed history revealed that the occasionally mumbled or smacked his lips with facial flushing during watching TV or conversation Neurological examinations were within normal limits except for the decreased MMSE score (21/30) Neuropsychological tests were significant for profound memory loss accompaned by mild anomia and frontal executive dysfunction Brain MRI showed high signal intensities in bilateral (left worse than right) hippocampus and amygdala on T2W1 and FLAIR images Brain F18-FDG PET revealed focal hypermetabolism in the left mesial temporal area Intial surface EEG was normal but prolonged EEG monitoring showed intermittent epileptiform discharges originating from the left temporal lobe Despite seizure-free state after treatment with antiepileptic drug. his cognitive impairments persisted for about 13 months and then began to improve gradually Unlike the previous epileptic pseudodementia cases who had only memory disturbance. our case showed dementia associated with complex partial seizures We learn from this case that possibility of epileptic pseudodementia should be considered when elderly patient with memory disturbance or dementia of atypical course especially because complex partial seizure is often unrecognized without detailed history and epileptic pseudodementia is a reversible cause of memory dysfunction.
