Isolation and Identification of Phytochemical Constituents from Taraxacum coreanum

Phytochemical constituents were isolated from the aerial parts of Taraxacum coreanum (Asteraceae)by repeated column chromatography and prep-HPLC. Their structures were identified as β-sitosterol (1), daucosterol (2), taraxasteryl acetate (3), chrysoeriol (4), diosmetin (5), luteolin (6),luteolin-7-O-glucoside (7), esculetin (8), and 5-hydroxypyrrolidin-2-one (9) by the interpretation of spectroscopic analyses including MS, 1H-, and 13C-NMR. This is the first report on the isolation of compounds 1-9 from T. coreanum. Among the compounds, 5 and 9 were isolated for the first time from Taraxacum.

A high performance liquid chromatography (HPLC) method was developed for simultaneous determination of two major flavonoid glycosides (poncirin and nanringin) in Poncirus trifoliata Raf. by different harvesting times and locations for two years. A SunFire C_18 column (4.6 mm×250 mm, 5 μm) was used at 40℃ for the determination of poncirin and naringin. The mobile phase using gradient flow consisted of two solvent systems. Solvent A was 1.0% (v/v) aqueous acetic acid and solvent B was acetonitrile with 1.0% (v/v) acetic acid. Flow rate was 1.0 mL/min and injection volume was 10 μL. The chromatogram was monitored by photodiode array (PDA) detection at 280 nm for the identification of two flavonoid glycosides in P. trifoliata. The contents of the two components in P. trifoliata ranged from 0.32~13.02%
Simultaneous Determination of Luteolin and Luteoloside in Dandelions Using HPLC

The content of the two flavonoids, luteolin and luteoloside, in dandelions was determined by high-performance liquid chromatography. A Nucleosil®100-5C18 column was used for the determination of luteolin and luteoloside concentrations. The mobile phase was a mixture of acetonitrile and water containing 0.1% formic acid (99.9:0.1, v/v). The gradient elution program was 10% to 23% acetonitrile within 60 min at a flow rate of 1.0 mL・min^(-1). The ultraviolet spectra were recorded at 350 nm for the identification of the two flavonoids. The contents of luteolin and luteoloside were highest in T. officinale flowers (5.817 ± 0.030 mg・g^(-1)) and in T. coreanum leaves (6.205 ± 0.087 mg・g^(-1)), respectively. The total content of luteolin and luteoloside in T. coreanum was about 1.3-2 times higher than that found in T. ohwianum and T. officinale.

This study was conducted to examine how the demographic-sociological characteristic, self-esteem, health state of elders in Daegu affects their satisfaction with life. The findings of this study were applied to seek a way to improve their satisfaction with life. First, age, religion, education, income, source of living expenses, subjective awareness of health, self-esteem, and health condition had a significant influence on the satisfaction with life at 0.05 or higher level of significance. Second, the satisfaction with life improved with higher subjective awareness of health, higher self-esteem, and better health condition.

The effects of luteolin (LT) and luteoloside (LS) from Taraxacum coreanum, using lipopolysaccharide (LPS)/interferon-gamma (IFN-γ)-induced RAW264.7 macrophage cells, on anti-inflammation were investigated. Our study was focused on the ethyl acetate fraction from T. coreanum (ETC) and its active compounds and their protective role against inflammation. The ETC and its active compounds, LT and LS, showed dose-dependent inhibitory activity against the production of nitric oxide (NO) and reactive oxygen species (ROS) in LPS/IFN-γ-stimulated RAW264.7 cells. In addition, ETC and its active compounds inactivated nuclear factor-kappa B and down-regulated inflammatory mediators. The results also showed that treatment with ETC, LT, and LS decrease pro-inflammatory cytokines, tumor necrosis factor-alpha, and interleukin-6. In conclusion, our studies indicated that ETC has anti-inflammatory activity owing to inhibition of NO/ROS generation and down-regulation of inflammatory mediators and cytokines. Moreover, LT and LS are bioactive compounds of ETC with protective effects against inflammation.
HPLC를 이용한 지치중의 부위별 Luteolin 정량

The content of luteolin in the different parts of Lithospermum erythrorhizon was determined by high performance liquid chromatography (HPLC). Luteolin was quantified by a reverse-phase column with gradient solvent program (water : ACN = 90 : 10 to 77 : 23 for 60 min). UV detection was conducted at 330 nm. The content of luteolin was measured in the seeds (0.28 mg/g), stems (0.30 mg/g), and roots (0.16 mg/g) of L. erythrorhizon. The presence of luteolin in Lithospermum species is important in agricultural crop production for increasing the amounts of clinically available medicine and health supplements.
흰민들레(Taraxacum coreanum) 활성획분인 Ethylacetate Fraction의 Peroxynitrite에 대한 산화적 스트레스 개선 효과

The EtOAc fraction from Taraxacum coreanum showed the strongest radical scavenging activity. The protective activity of active EtOAc fraction against the oxidative stress was investigated under cellular system using LLC-PK1 renal epithelial cell. The LLC-PK1 cells showed loss of the cell viability and elevation for lipid peroxidation by the treatment with generator of peroxynitrite (ONOO-), 3-mor- -pholinosydnonimine, and its precursors, nitric oxide (NO) and superoxide anion (O2 ), produced by sodium nitroprusside and pyrogallol, respectively. However, the treatment of the EtOAc fraction significantly, dose-dependently, recovered cell viability and inhibited lipid peroxidation against both ONOO- itself, and its precursors of NO and O2 . At 100μg/ml of the EtOAC fraction, the cell viability was significantly increased to 93.4% from 61.0%, and lipid peroxidation was inhibited to 0.104 nmol/mg protein from 0.494 nmol/mg protein. The present study suggests that T. coreanum would have the protective potential from ONOO--induced oxidative stress.
치자의 포장방법 및 보관기간에 따른 geniposide 함량 분석

Gardeniae Fructus (GF) is a traditional herbal medicine listed in “The Korean Pharmacopoeia” and has been used for jaundice, liver disease, and anti-inflammatory. The propose of this study is to establish basic data for setting the shelf life of GF. The content of geniposide, a major compound of GF, was investigated by the packaging method (general, general + silica gel, vacuum, vacuum + silica gel, nitrogen, and nitrogen + silica gel) and storage period (60 months) using high-performance liquid chromatography. As a result, the content of geniposide was initially detected as 4.40%, and then 3.85-5.86% in all packaging methods for 60 months. This result can be used as basic data for quality control and shelf life of GF.
