        Protective Effect of Survivin in Doxorubicin-Induced Cell Death in H9c2 Cardiac Myocytes

        Background and Objectives: Apoptosis has been known to be an important mechanism of doxorubicin-induced cardiotoxicity. Survivin,which belongs to the inhibitor of apoptosis protein family, is associated with apoptosis and alteration of the cardiac myocyte molecular pathways. Therefore, we investigated the anti-apoptotic effect and cellular mechanisms of survivin using a protein delivery system in a doxorubicin-induced cardiac myocyte injury model. Materials and Methods: We constructed a recombinant survivin which was fused to the protein transduction domain derived from HIV-TAT protein. In cultured H9c2 cardiac myocytes, TAT-survivin (1 µM) was added for 1 hour prior to doxorubicin (1 µM) treatment for 24hours. Cell viability and apoptosis were evaluated by 2-(4,5-dimethyltriazol-2-yl)-2,5-diphenyl tetrazolium bromide assay, caspase-3 ac-tivity, and terminal deoxynucleotidyltransferase-mediated dUTP nick end-labeling assay. We measured the expression levels of several apoptosis-related signal proteins. Results: The survivin level was significantly reduced in a dose dependent manner up to 1 µM of doxorubicin in concentration. Purified recombinant TAT-survivin protein was efficiently delivered to H9c2 cardiac myocytes, and its transduction showed an anti-apoptotic ef-fect, demonstrated by reduced caspase-3 activity and the apoptotic index, concomitantly with increased cell viability against doxorubicin injury. The phosphorylation of p38 mitogen-activated protein (MAP) kinase and the release of Smac from mitochondria were suppressed and the expression levels of Bcl-2 and cAMP response element-binding protein (CREB), the transcription factor of Bcl-2, were recovered following TAT-survivin transduction, indicating that survivin had an anti-apoptotic effect against doxorubicin injury. Conclusion: Our results suggest that survivin has a potentially cytoprotective effect against doxorubicin-induced cardiac myocyte apop-tosis through mechanisms that involve a decrease in the phosphorylation of p38 MAP kinase, mitochondrial Smac release, and increased expression of Bcl-2 and CREB.

        Prototype Development of a Three-wheel Riding Cultivator and Its Basic Performance

        Purpose: The aim of this study is to develop a three-wheel riding cultivator for improving the performance of the current four-wheel riding cultivators in the market. Methods: A prototype three-wheel riding cultivator with the rated power of 15.5-kW, a primary hydrostatic and a two-speed selective gear transmission shifts, front/rear three-wheel drive, a hydraulic wheel tread adjustment, and the mid-section attachment of the major implements was designed and constructed. Its specifications and basic performance are investigated. Results: The maximum speeds of the prototype at the low and high stages were measured to be approximately 7.31, and 11.29 km/h in forward travel, respectively, and approximately 3.60, and 6.37 km/h in rearward travel, respectively. The minimum ground clearance is shown to be 670 mm. The rotating speeds of the power takeoff (PTO) shaft at the low and high stages are shown to be approximately 795 and 1,140 rpm, respectively. The tread of the rear wheels, the minimum radius of turning, and the maximum lifting height of the parallel link device are measured to be within 1,320–1,720 mm, 2.80 m, and 390 mm, respectively. Approximately 25.3% and 74.7% of the total weight of the prototype are distributed in the front and rear wheels on flat ground, respectively. When the tread of rear wheels increased from 1,320 to 1,720 mm, the left and right static lateral overturning angles increased from 33.4° to 39.1° and from 29.0° to 36.1°, respectively. Conclusions: The prototype three-wheel riding cultivator showed a wide range of travel and PTO speeds, high minimum ground clearance, small minimum radius of turning, and easy control of the rear wheel tread. Further, the easy observation of cultivating operations by mid-mounting the implements can improve quality of work. Therefore, the prototype is expected to contribute to the riding mechanization of cultivating operations for various upland crops in Korea.

        도시침수 저감을 위한 by-pass 빗물침투성 우수관거

        본 연구는 기상이변과 도시열섬현상으로 국지성 호우가 잦아져 도시침수피해가 매년 반복되고 있으며, 건기시에는 비가 모자라 지하수 부족으로 이어져 하천 건천화 현상이 빈번히 발생하고 있어 이를 해결하고자 "도시침수 저감을 위한 by-pass 빗물침투성 우수관거"를 제안한다. 집중호우로 확률년수 이상의 호우발생시 관거를 확대 교체하지 않고 기존우수 관거를 그대로 사용하며 배제시스템과 연계하여 by-pass 빗물침투성 우수관거를 설치하여 본 연구지역 대한민국 서울특별시 강남구 강남역사거리 지역 3.07 ha에서 관거확대공사와 비교시 평균 27백만원(44%) 공사비절감 및 기존 우수관거 유지로 별도의 우수처리 대책이 불필요하다. "도시침수 저감을 위한 by-pass 빗물침투성 우수관거"는 여름철 확률년수 이상의 비가 오더라도 by-pass관로를 통하여 통수능력을 증가시키며, 봄, 가을에 내리는 소량의 비는 by-pass 침투공을 통하여 전량 지중 수자원 확보에 기여한다. The aim of this study is to determine and propose the by-pass rainwater sewer system in order to reduce the urban floodplain from the locality heavy rain every year during the dry season and the sinkholes in the city as well as the shortage of groundwaters due to extreme hot weather condition and urban heat island phenomenon. Heavy rain occurs more than the years of heavy rainfall probability, comparison between the place where uses the existing pipes and connect the sewer system with by-pass rain permeability and without expanding sewer pipe replacement at intersection of Gangnam station 3.07 ha at Gangnam-gu, Seoul Metropolitan area, it indicates that average of 27 million KRW (44%) maintenance cost savings and maintain existing sewer system without any other countermeasures. For the city flooded reduction, by-pass rainwater permeable rainwater pipe multiplying the probability the number of years during summer season and increase the water flow capacity during spring and fall when a small amount of rain that, it also contribute to the total amount of underground water secured through the by-pass penetration.

        Prototype Development of a Small Combine for Harvesting Miscellaneous Cereal Crops and its Basic Performance

        Purpose: The aim of this study is to develop a small combine for harvesting miscellaneous cereal crops. Methods: A prototype small combine was designed and constructed. Its specifications and basic performance were investigated. Results: The prototype small combine for harvesting miscellaneous cereal crops was designed and constructed to reflect similar specifications as those of the conventional combine. The prototype small combine comprises a diesel engine with the rated power/speed of 22.0 kW/2,600 rpm, three-stage primary and two-stage speed range transmission shifts, and a double acting threshing part. The maximum travel speeds of the prototype combine are approximately 0.72 m/s, 2.50 m/s, 0.30 m/s at the low, high speed range shifts in the forward direction, and while traversing in the reverse direction, respectively. The minimum radius of turning was approximately 1.50 m. In a static lateral overturning test, the prototype combine overturned neither to the right nor to left on a 30° slope. The results of an oilseed rape harvesting test included the maximum operating speed of 0.32 m/s, the grain loss ratio of approximately 9.0%, and the effective field capacity of approximately 10.3 a/h. Additionally, among the outputs in grain outlet, the whole grains, damage grains, and materials other than grain (MOG) ratios accounted for 97.4%, 0.0%, and 2.6%, respectively. Conclusions: The prototype small combine for harvesting miscellaneous cereal crops indicates good driving ability and stability. The results of the oilseed rape harvesting test reveal that the harvesting performance must be enhanced such that the separating and cleaning parts are more suitable for each type of crop, thus reducing grain loss and foreign substances among the outputs in grain outlet. An improved small prototype combine could be used effectively to mechanize the harvesting of miscellaneous cereal crops in small family farms or semi-mountainous areas.

      • 잡곡 수확용 25 kW급 소형 콤바인 시작기 설계 제작

        현재 국내에 공급되고 있는 잡곡류 수확기는 보행형 예취기, 탈곡기 위주의 저능률 기계화 수준으로 고능률의 콤바인 수확기 개발이 필요하며, 특히 잡곡류가 소규모 경작지의 영세농가 위주로 재배되어 저가격의 소형 콤바인 수확기 개발이 절실히 요구되고 있다. 따라서 본 연구는 소규모 밭의 두류 및 잡곡 수 확작업에 적응성이 뛰어나며, 농기계 임대사업소의 활용도를 높일 수 있고, 여성과 고령자도 쉽게 운전할 수 있어 수확작업의 노동력을 크게 절감할 수 있는 저가격의 25kW급 자주식 소형 콤바인을 개발하고자 시작기를 설계 · 제작하였다. 시작기의 주요부로 엔진은 25kW/2600rpm 3기통 디젤엔진을 탑재하였으며, 동력전달부는 주변속 3단, 부변속 2단의 선택맞물림 기어식의 변속장치를 이용하였다. 주행부는 궤도형으로 조향클러치와 습식 원판식 제동장치를 채용하였다. 전처리부는 선단거리 1700 mm의 디바이더와 상하좌우 수동 조절되는 회전속도 약 42 rpm의 정오각형 릴로 구성하였으며, 전처리부의 최대 승강높이는 740 mm이었다. 작물이송부는 돌기부착 오거와 체인컨베이어로 구성되어 있으며, 탈곡부는 단동형 축류식의 직경 440 mm, 길이 1180 mm의 급동과 높이 65 mm, 지름 10 mm의 46개 강봉형 급치, 격자형 수망으로 구성하였으며, 회전속도는 약 325 rpm으로 작동하도록 하였다. 선별 · 정선부는 요동 · 송풍선별식으로 곡립판, 볏짚체, 곡립체, 송풍팬으로 구성하였고 송풍팬의 회전속도는 약 850 rpm, 요동진동수는 약 5.8 Hz로 작동하도록 하였다. 곡물이송부와 재처리부는 수평이송 외경 103 mm, 수직이송 외경 110 mm의 피치가 모두 82 mm인 스크류컨베이어를 이용하였으며, 곡물탱크는 용량이 250 로 2개의 배출구로 곡물을 포대에 담도록 하였다. 그 외 시작기는 운전조작부, 유압장치부, 전기장치부 등을 갖도록 설계 · 제작하였다. 전체적인 기체의 크기는 길이x폭x높이 3935x1900x2440 mm이었으며, 기체 중량은 약 1753 kg이었다. 콩 대상 기초 성능시험결과 시작기의 작업속도는 약 0.5 m/s, 작업능률은 약 11 a/h로 나타났다.
