        Pseudomonas stutzeri IAM 12097 의 Exo - maltotetraohydrolase 에 관한 연구 : 제3보 . 각종기질(各種基質)에 대한 Exo - maltotetraohydrolase 의 분해산물 및 분해율 Part Ⅲ. Reaction products and hydrolysis rate on various carbohydrates of Exo - maltotetraohydrolase

        Exo-maltotetraohydrolase produced by Pseudomonas stutzeri IAM 12097 was characterized with respect to substrate specificity, the reaction products and hydolysis rate on various carbohydrates. Maltopentaose, maltohegtaose, soluble starch, amylose, amylopectin, oyster glycogen and gelatinized starch of corn, potato, glutinous rice, green banana and arrow root were hydolyzed by this enzyme, but α, β, γ-cyclodextin, sucrose, raffinose, lactose pullulan, maltose, maltotriose and maltotetraose were not hydolyzed. Among oligosaccharides, maltohexaose was favorably hydrolyzed by this enzyme and the main reaction product of oligosaccharides and polysaccharides was maltotetraose. Addition of pullulanase to this enzyme increased the hydolysis rate on gelatinized starches, but it did not on raw starches. Among various starches, corn starch was favorably hydrolyzed by this enzyme, whereas it acted on potato starch, arrow root starch and high amylose corn starch weakly.

        개별차원의 긍정적 행동지원이 지적장애 성인의 옷 찢는 행동에 미치는 효과

        The purpose of this study was to determine the effects ofindividualized positive behavior support on clothes ripping behavior ofadults with intellectual disabilities. The participant was 22 year-old malediagnosed with severe intellectual disability. The experiment took placeat the self-contained day care center for adults with intellectualdisabilities where the participant attended. A multiple baseline designacross settings was used with baseline, intervention, maintenance phases. The data for the maintenance phase was collected after two weeks ofintervention termination Multi-dimensional intervention strategies such asintervention with setting event, antecedent, teaching alternative behaviorand consequence were conducted after the functional behaviorassessment. Results of the study indicated that positive behavior supportintervention reduced the frequency of clothes ripping behavior and theeffectiveness remained after termination of intervention. Conclusionsuggests positive behavior support can be considered as effective strategyto decrease clothes ripping behavior of adults with intellectualdisabilities 이 연구에서는 개별차원의 긍정적 행동지원이 지적장애 성인의 옷 찢는 행동에 효과가 있는지를알아보자 하였다. 연구 참여자는 만 22세 지적장애 1급, 성인 남성이며, 실험은 연구 참여자가 생활하는 주간보호센터에서 실시하였다. 연구 설계는 상황간 중다기초선 설계를 사용하였고, 기초선, 중재, 유지의 세 단계로 실시하였다. 유지는 중재가 종료된 후 2주 후에 자료를 수집하였다. 연구 참여자의 옷 찢는 행동을 지도하기 위해 기능평가를 통해 문제행동의 기능을 파악한 후, 배경사건, 선행사건, 대체행동교수, 후속결과의 다차원적인 중재전략을 수립하여 적용하였다. 연구 결과,긍정적 행동지원이 지적장애 성인의 옷 찢는 행동을 감소시켰으며 중재 후 그 효과가 유지되었다. 이러한 결과는 긍정적 행동지원이 지적장애 성인의 옷 찢는 행동을 감소시키는 데 효과적인 전략으로 사용될 수 있음을 시사한다.

        귀리 첨가 호상 요구르트의 저장 중 항산화 활성 및 품질 특성

        Recently, consumers' awareness of the importance of the intestinal action of lactic acid bacteria and intestinal microbes is increasing, as well as interest in yogurt. In this study, yogurt was prepared with three mixed strains (lactic acid bacteria combination, Lactobacillus acidophilus, Lactobacillus delbrueckii subsp., and Bulgaricus, Streptococcus thermophilussei, 1:1:1) by adding oats flour, and the quality characteristics of yogurt were investigated, while stored at a storage temperature of 4℃ for 12 days. According to the storage period, the control as wel as the oat yogurt showed slight decrease in pH, and no significant change in acidity. Sugar content slightly increased. and brightness decreased, in the control and the oat yogurt. Visible cell numbers increased during storage, and decreased on the 12th day. Viscosity in the oat yogurt was 7,580 cP, which was approximately eight times higher than that of the control group, and decreased gradually according to the storage period. Antioxidant activity (DPPH) was approximately two times higher in the oat-added yogurt, and slightly increased with the storage period, decreased on the 12th day of storage, and β-glucan was detected only in oat-added yogurt.

        취반 후 백도가 좋고 베타글루칸 함량이 높은 찰성 쌀보리 "진주찰"

        신품종 "진주찰" 쌀보리는 취반 후 백도가 좋고, 베타글루칸 함량이 높은 소립, 내재해, 양질, 다수성 쌀보리 신품종이다. 1996년에 호남농업연구소에서 도복과 내한성이 강하고 찰성으로 립백도와 정맥율이 높으며 취반특성이 우수한 특성을 지닌 "진미찹쌀보리"와 "수원333호"를 인공교배한 후 계통육종법으로 육성하였다. 2003년부터 2년간 생산력검정시험을 실시한 결과, 수량성이 높고 도복이 강한 우수 계통으로 판명되어 "익산79호"로 계통명을 부여, 20 A new six-rowed naked barley cultivar "Jinjuchal" with high whiteness after cooking and high beta-glucan content was developed from the cross between 'Jinmichapssalbori' with high winter hardiness, lodging tolerance, grain whiteness and pearling yield, and 'Suwon 333' with waxy endosperm by the Honam Agricultural Research Institute (HARI), NICS, RDA in 2007. An elite line, SB962002G-B-B-B-84-4 was selected in 2002 and designated as 'Iksan 79' It showed good agronomic performance in the regional yield trials (RYT) from 2005 to 2007 and was released with the name of "Jinjuchal" possessing high whiteness and low proanthocyanidin waxy endosperm. The average heading and maturing dates of "Jinjuchal" were April 27 and June 1 in paddy field, which were same and one day later than those of the check cultivar 'Saechalssalbori' respectively. The new cultivar, "Jinjuchal" had 81 cm of culm length that was 1cm longer than that of 'Saechalssalbori' and 4.9 cm of spike length. It showed 673 spikes per m^2, 56 grains per spike, 27.0 g of 1,000-grain weight, and 752 g of test weight. "Jinjuchal" showed stronger hardiness and better resistance to powdery mildew and BaYMV (Barley yellow mosaic virus) than those of the check cultivar, 'Saechalssalbori' It showed higher beta-glucan content(8.4%) and water absorption rate than those of the check cultivar, 'Saechalssalbori' Its average yield of the pearled grain in the regional yield trial was 3.79 MT/ha in upland, and 3.73 MT/ha in paddy field, which were 1% lower and 3% higher than those of the check cultivar, respectively. Total phenol and proanthocyanidin contents were 4.2 and 0.2 mg/g, respectively. This cultivar is suitable for the area of the daily minimum temperature above -6^℃ in January in Korean peninsula.

        교사와 학생이 지각하는 디지털교과서의 교수학습 적합성 평가 및 개선방안

        본 연구의 목적은 우리나라 정부 주도로 지난 15년간 개발 및 적용연구를 하고 있는 디지털교과서를 교수·학습 활용 측면에서 교사와 학생이 직접 적용 및 평가하고 문제점 및 개선 방안을 도출해 내는 것이었다. 연구대상은 광주·전남지역 2010, 2011년 연속 2년 교과부지정 디지털교과서 초등연구학교 2개교 교사(15명)와 학생(21명) 총 36명을 선정하였다. 평가대상 디지털교과서는 초등 4,5,6학년 사회, 과학이었다. 교사와 학생들이 직접 수업에 적용해 본 경험을 기초로 평가설문지와 인터뷰를 통해 평가하였다. 자료 수집은 2011년 10-12월에 이루어졌다. 본 연구 결과, 디지털교과서의 여러 교육적 가능성에 대한 예상들과는 다르게 실제 평가 결과는 대체로 만족하지 못하고 문제점 지적이 많은 편이었다. 교수학습 측면 적합성에 대한 평가 전체평균은 2.8/5(교사 2.71, 학생 2.96)로 백분율 59%정도로 낮았다. 또한, 각 평가영역별 개선이 필요한 문제점과 대안들도 조사하였다. 조사결과 교사와 학생이 대체로 같은 문제점 지적을 하였다. 평가영역을 초월하여 가장 자주 반복 지적되는 문제점들은, 수준별 수업 지원 미흡, 고등사고능력 촉진 부족, 서책형교과서와의 차별성 낮음 등이었다. 지적내용을 바탕으로 이 문제점들과 구체적인 대안들을 제시하였다.

        질소시비수준이 콩의 생육 및 isoflavone 함량에 미치는 영향

        The nitrogen fertilization effect on growth characteristics and isoflavone content was investigated in this study, and isoflavone analyzed by HPLC with photodiode array (PDA) detector and reverse-phase C18 column. Fertilization levels were no-fertilization, no nitrogen, 50% decreased in nitrogen, standard and 50% increased in nitrogen fertilization. The 50% increased nitrogen fertilization showed the highest growth characteristics then other fertilization level and the number of pod and seed showed maximum value 31.9 and 72.3, respectively. and seed yield was 2,460 kg/ha. During growth stages, isoflavone content in leaf, stem and root of soybean plants decreased to R5 stage then increased to R7 stage. Isoflavone content according to various nitrogen fertilization condition, in case of none fertilization, no nitrogen, 50% decreased nitrogen fertilization showed higher value than those of standard and 50% increased nitrogen fertilization levels. Aglycon content among the isoflavone isomers showed much higher in plant than in seed. The highest isoflavone content was found in the root of soybean plant parts. Isoflavone content of seed was higher in none, no nitrogen, 50% decreased nitrogen than those of standard and 50% increased nitrogen fertilization. Isoflavone은 다양한 식물군에 광범위하게 발견되고 있으나, 콩과 식물에 상대적으로 많이 존재하는 phytochemical로서 암, 심장질환, 골다공증과 여성의 폐경기 증후군과 같은 질병을 예방하는데 효과가 있는 물질로 알려져 있다(Arther & William, 1983). 특히, 콩에 포함되어 있는 isoflavone은 antiestrogen, antioxidant와 tyrosine protein kinase inhibitor 기능을 가지고 있어 콩 소비가 많은 아시아인들의 전립선암, 유방암, 직장암 등에 대한 발병률을 낮추는 것과 관계가 있다는 연구결과가 보고되면서(Record et al., 1995) 양질의 단백질과 지방 및 생리활성 물질을 함유한 기능성 식품으로서 콩의 식품영양학적 가치가 재인식되고 있다. 콩은 곡류 위주의 식습관을 지닌 우리 나라 사람들에게 두부, 장류, 콩나물 등 다양한 식품소재로 이용되고 있는데, 콩과 콩 가공식품 중에 주로 존재하는 isoflavone은 genistein과 daidzein 및 glycitein의 배당체인 genistin, daidzin, glycitin 형태로 존재한다(Murphy, 1982; Walter et al., 1941; Kodou et al., 1991). 이 중에서 genistein과 daidzein은 생리기능이 가장 우수한 대표적인 isoflavone으로서 암세포의 성장과 분화에 관련된 protein tyrosin kinase의 활성(Levitzki et al., 1995) 및 다른 항암제들처럼 DNA 복제와 관련된 topoisomerase II를 억제하는 물질로도 알려져 있으며(Yamashita et al., 1991), 독성이 거의 없다고 한다(Barnes et al., 1996). 일반적으로 콩의 isoflavone은 100~300 mg %정도 함유되어있으며(Anlin et al., 1995), 국내산 콩 품종의 isoflavone 함량은 46~232 mg % 범위였다(Choi et al., 1996; Kim & Kim, 1996; Choi & Sohn, 1998; 김 등, 1999). Isoflavone 함량과 조성은 품종 및 재배환경에 따라 큰 차이가 있다고 보고(소 등, 2001; Tony et al., 2002)되고 있으나 생육시기나 시비수준에 따른 함량변화에 대한 연구결과는 비교적 많지 않은 실정이다. 본 연구에서는 생육시기별 콩과 콩 식물체의 isoflavone 함량변화를 알아보고, 질소시비 수준이 콩의 생육, 수량 및 isoflavone 함량에 미치는 영향을 검토하여 특수목적 콩 및 고품질 콩 품종육성을 위한 기초 자료로 활용하고자 수행하였다.

      • 비만아동의 신체상 만족도와 자아존중감 및 학교생활적응관계

        The purpose of this study is to provide the basic data for understanding of and psychological and emotional support for obese children by measuring the level of their physical satisfaction, self-esteem and adaptability at school to identify the differences according to the level of obesity and by finding out correlation among the variables. Out of 22 elementary schools in Cheongwon-gun, Chungbuk province 16 elementary schools where health teachers are were selected for the study out of convenience and among students in 5th and 6th grades of the 16 schools, 305 students who agreed to participate in the study with awareness of its objective and also were classified as obese as a result of physical tests done in 2003 participated in the study as subjects. But ultimately the data of 260 students were used for the study excluding 45 who gave us insufficient responses. Data was collected from February 2-20, 2004. SPSS Win 10.0 was used for the analysis of data and average, standard deviation, cross analysis, ANOVA and Pearson's Correlation Coefficient were used for verification while Duncan test was done as post hoc test. The findings of the study were as follows : 1) The value of concerning for one's body by fatness were showed that 'a little' was 59.7% and 54.5% in each slight obesity group and moderate obesity group and severe was 81.3% in normal(p<.001), and the value of satisfaction for one's body were that slight obesity group was showed 54.5% in normal, moderate was 45.5% in normal, and severe was each 35.5 % in very not satisfaction and not satisfaction(p<.05). 2) The satisfactory values for one's body by obesity value(slight, moderate, severe) were not showed meaningful differences, and average value of each groups were 76.8 in slight obesity group, 73.2 in moderate, and 74.5 in severe(F=2.358, p>.05). 3) The self-esteem by obesity value(slight, moderate, severe) were not showed meaningful differences, and average value of each groups were 66.9 in slight obesity group, 66.4 in moderate, and 60.6 in severe(F=1.900, p>.05). 4) The adaptability at school by obesity value (slight, moderate, severe) were not showed meaningful differences, and average value of each groups were 132.8 in slight obesity group, 131.8 in moderate, and 130.5 in severe(F=.120, p>.05). 5) There is clear correlation among the level of physical satisfaction, self-esteem and adaptability at school according to the level of obesity (slight, moderate, severe). In other words, the level of physical satisfaction and adaptability at school (r=.390, p<.001). the level of physical satisfaction and self-esteem (r=.525, p<.001) and self-esteem and adaptability at school (r=.646, p<.001).
