      • 가토신장기능에 미치는 측뇌실내 Clonidine의 영향

        신장기능에 대한 중추신경계의 역할을 구명코자. presynaptic α-adrenoceptor의 선택적 작동약인 clonidine을 urethane마취 가토의 측뇌실내로 (icv)투여하여 신장기능의 변동을 관찰하였다. 5μg/kg icv이하의 양으로는 신장기능의 유의한 변동을 볼 수 없었으나, 15μg/kg icv 으로는 20분간에 걸쳐 현저한 Na 및 K배설 증가를 볼 수 있었다 이 때 신혈류 및 사구체 여과율은 유의한 변동을 나타내지 아니하였다. 또 이때 Na 재흡수율은 유의하게 감소하였으며, Na배설증가 작용이 세뇨관에서의 Na재흡수 억제에 기인함을 알 수 있었다. 전신혈압 변동은 이 작용에 기여하지 아니하였다. Presynaptic α-adrenoceptor에 대한 선택적 길항약인 yohimbine 100μg/kg을 clonidine투여 20분전에 측뇌실내로 투여하면 clonidine의 신장작용이 완전히 차단되었다. 이 량의 yohimbine은 측뇌실내 투여시 신장기능에 아무런 변동도 초래하지 아니하였다. 15μg/kg clonidine을 정맥내로 투여하면 투여 직후에 뇨량 감소와 신장기능 감퇴를 초래한 뒤 후기에 약간의 Na배설증가의 경향을 보였으나, clonidine을 icv로 투여하였을 때 볼 수 있던 만큼의 Na배설증가는 볼 수 없었다. 따라서 icv clonicine의 신장작용에는 신장에 대한 clonidine의 직접작용이 관여하지 않음을 알 수 있었다. 이 연구결과는 가토의 신장기능 조절에 있어서 중추의 교감신경 긴장도가 중요한 역할을 하고 있음을 시사하였다. To explore the regulatory roles of CNS on the renal function, clonidine, a specific presynaptic α-adrenoceptor agonist, was administered into a lateral ventricle of the brain (icv) and the changes of renal function were studies in urethane-anesthetized rabbits. 5μg/kg icv elicited no significant changes in renal function. However, 15μg/kg induced marked natriuresis and kaliuresis for 20 min. Neither RPF nor GFR changed significantly. The fractional sodium reabsorption was significantly reduced, indicating that the renal action was of the tubular origin. Changes of systemic blood pressure were not contributory to the renal action. Yohimbine, a specific antagonist for presynaptic α-adrenoceptor, when given icv in doses of 100μg/kg 20 min prior to clonidine, completely abolished the renal action of icv clonidine. Yohimbine icv did not produce any significant changes in renal function. Intravenous clonidine, 15μg/kg, elicited antidiuresis and decrement of renal function immediately after administration, followed by a slight tendency toward natriuresis, but no natriuresis corresponding to those seen after the icv clonidine were observed, indicating that in the renal action of icv clonidine no direct action is involved. These observations indicate that the central sympathetic tone plays a role in the regulation of renal function in the rabbit.

        윗눈꺼풀성형술 후 발생한 상윤부각결막염 양상을 보이는 결막 림프관종 1예

        Purpose: To report a case of conjunctival lymphangioma with clinical manifestations of superior limbic keratoconjunctivitis after upper lid blepharoplasty. Case summary: A 55-year-old woman who had upper lid blepharoplasty performed 1 year before complained of pain, injection, and blurred vision in her right eye, which lasted for 2 days. The slit lamp examination revealed a poorly demarcated hypertrophic lesion with central erosion on the superior bulbar conjunctiva and a diffuse corneal erosion and edema in the area of the superior limbus. Tenderness of the superior bulbar conjunctiva and corneal erosion were treated with artificial tears, antibiotic eye drops, and a therapeutic contact lens. Slight anterior chamber reaction was found and treated by steroid eye drops; however, there was no improvement. After eversion of the right upper eyelid with a Desmarres retractor, a hypertrophic lesion with central dimpling was found. However, no exposed sutures were observed. Resection and biopsy of the superior bulbar conjunctiva and a subtarsal triamcinolone injection were performed for diagnosis and treatment. A conjunctival biopsy specimen showed conjunctival lymphangioma. Conclusions: Conjunctival lymphangioma can occur due to pressure and friction of an eyelid lesion after upper lid blepharoplasty.         비유전독성 간발암물질인 Cloibrate 의 F344 랫드에 있어서 경구 아급성독성시험

        Clofibrate, a peroxisome proliferator, is hepatocarcinogenic in rats in a dose-dependent manner. A total of 70 male and female F344 rats, 5-week-old, were divided into three groups. Rats were fed clofibrate at 0, 0.25, or 0.5% in diet for 30 days. All rats were anesthetized with CO₂, blood samples were taken by cardiac puncture for hematology and clinical chemistry, and the rats were killed by exsanguination. Livers, kidenys, pancreas, adrenal glands, spleen, heart, lungs, thyroid gland, reproductive organs, and digestive organs were removed, weighed, later processed, and embedded with paraplast for histological examination. The relative liver and kidney weights with respect to final body weight in the clofibrate-treated group were significantly increased compared with those of control group at all dose levels (p<0.01). It has been suggested that clofibrate may influence on hepatotoxicity by increases in peroxisomal proliferation.

      • 개구리 皮膚의 Na 輸送에 對한 5-Hydroxytryptamine의 影響

        To ascertain the existence of tryptaminergic receptor involved in active transport of sodium in the frog skin and to delineate its physiological role, the effects of 5-hydroxytryptamine (5-HT)on the potential difference (PD), short·circuit current (SCC) and total skin-conductance (TSC) of the isolated frog skin, as well as influence of tryptaminergic antagonists and various agents upon the 5-HT effects were studied in Rana nigroma-culata. PD and SCC were determined by utilizing Ussing's technique. Both sides of the isolated frog skin were perfused with Ringer's solution for coldblooded animals. All the drugs, except Amiloride, were administered to the serosal side of the skin. Results were summarized as follows : 1. The responses to 5-HT were characterized by elevation of PD & SCC, roughly in a dose-related fashion in doses ranging from 1.7×10 exp (-5)M to 5.7×10 exp (-4)M. The more higher doses produced less effects. 5-HT induce greater increase in PD than SCC, indicating that TSC is decreased by 5-HT. 2. Similar responses were being observed in Na_2SO_4 -Ringer solution. But the magnitudes of the increase in both PD and SCC were approcimately equal, little changes in TSC were noted, seggesting that the changes of PD may be, at least in part, dependent upon the increased outflux of chloride ions. The stimulatory effects of 5-HT on PD & SCC were abolished by cAMP (5mM) and amiloride (Ⅸ 10^-5M). 3. The 5-HT induced increments of PD & SCC were significantly inhibited by methysergide (8.5×10 exp (-6)M), a specific 5-HT_1 receptor blocking agent, whereas ketanserine, a specific 5-HT_2 antagonist did not influence the effects of 5-HT. It is concluded from these results that the trypraminergic system might play role in the control of sodium transport in the frog skin. It is further suggested that the subtypes which indicate these effects might be of 5-HT_1 -type and cAMP might be involved in the effects as a second messenger.
