        Study on chemotaxis and chemokinesis of bone marrow-derived mesenchymal stem cells in hydrogel-based 3D microfluidic devices

        Background: Controlling the fate of mesenchymal stems cells (MSCs) including proliferation, migration and differentiation has recently been studied by many researchers in the tissue engineering field. Especially, recruitment of stem cells to injury sites is the first and crucial step in tissue regeneration. Although significant progress has been made in the chemotactic migration of MSCs, MSC migration in three dimensional environments remains largely unknown. We developed a 3D hydrogel-based microfluidic-device to study the migration behavior of human MSCs in the presence of stromal-cell derived factor-1α (SDF-1α), interleukin 8 (IL-8) and Substance P (SP) which have been utilized as chemoattractant candidates of human mesenchymal stem cells (hMSCs). Results: We systematically investigated the chemotactic migration behaviors of hMSCs and their responses to SDF-1α, IL-8, and SP. SDF-1α was shown to be the most fascinating chemoattractant candidate among those factors at a certain time point. We also found that each chemokine showed different chemoattractant abilities according to their concentration. In the case of SP, this factor showed chemokinesis not chemotaxis. Especially at a 7–8 × 10−8 M concentration range, the chemokinesis ability driven by SP was further increased. The data suggest that some factors at the optimal concentration exhibit chemokinesis or chemotaxis in a 3D hydrogel-based microfluidic device. Conclusion: In this study on chemotaxis and chemokinesis of hMSCs, the system parameters such as chemokine concentration, system stability, and 2D or 3D microenvironment are critically important to obtain meaningful results.

        Prevalence and Clinical Characteristics of Metabolically Healthy Obesity in Korean Children and Adolescents: Data from the Korea National Health and Nutrition Examination Survey

        Metabolically healthy obesity (MHO) and metabolically unhealthy obesity (MUO) are differentiated by the presence of cardiometabolic risk factors (CMRFs) and insulin resistance (IR). This study aimed to evaluate the prevalence and clinical characteristics of MHO in Korean children and adolescents and to investigate the anthropometric, laboratory, and lifestyle predictors of MHO. This study included data from 530 obese subjects, aged 10–19 years, obtained from the Fourth Korea National Health and Nutrition Examination Survey. Subjects were classified into MHO and MUO groups according to the presence of CMRF (MHOCMRF/MUOCMRF) and degree of IR (MHOIR/MUOIR). Demographic, anthropometric, cardiometabolic, and lifestyle factors were compared between the groups. Logistic regression analysis and receiver operating characteristic curve analysis were performed to identify factors that predicted MHO. The prevalence of MHOCMRF and MHOIR in obese Korean youth was 36.8% (n = 197) and 68.8% (n = 356), respectively. CMRF profiles were significantly less favorable in MUO children. Longer and more vigorous physical activity and less protein intake were associated with MHOCMRF phenotype. The best predictors of MHOCMRF and MHOIR were waist circumference (odds ratio [OR], 0.82; 95% confidence interval [CI], 0.77–0.88; P < 0.001) and body mass index (BMI) standard deviation score (OR, 0.24; 95% CI, 0.15–0.39; P < 0.001), respectively. The prevalence of MHO differed depending on how it was defined. To adequately manage obesity in youth, the approach to individuals with MHO and MUO should be personalized due to variation in clinical characteristics. Longitudinal studies are needed to evaluate long-term consequences of MHO.

        이용자 인식조사를 기반으로 한 공공도서관 비대면 서비스 운영 방향에 관한 연구

        Libraries are experiencing numerous modifications to multifunctional spaces in response to the declaration of WHO pandemic. Most libraries throughout the globe have reported their closures, including national libraries in every country, and most established facing services have been transformed to untact services. However, the breadth of the service varies by local autonomous entity and library, making it difficult for librarians to act according to their conviction, and also users are feeling inconvenienced. As a result, this study used a user perception survey to determine the extent of non-face-to-face services in the library and to suggest the direction of operation of non-face-to-face services in a pandemic condition. To that purpose, beginning February 12, 2020, an online survey of library customers in 37 public libraries was performed for about three weeks, and the 117 replies were examined using the SPSS statistics tool to perform frequency analysis, regression analysis, correlation analysis, and reliability analysis. By the survey and data survey results, it was recommended that library services be re-established in the direction of non-face-to-face library service operation, that library non-face-to-face services be expanded, that user education be conducted, that non-face-to-face services be promoted, and that user opinions be collected. WHO에서 팬데믹을 선포하면서 다중이용시설인 도서관 역시 여러 변화를 맞이하고 있다. 전 세계적으로 각국의 국립도서관을 비롯한 대다수의 도서관이 휴관을 발표하였으며, 기존에 대면하여 제공하던 서비스를 대부분 비대면 서비스로 전환하였다. 하지만 그 서비스의 범위가 지자체 및 도서관별로 상이하여 사서의 소신있는 대응이 어렵고, 이용자 역시 불편을 겪고 있다. 따라서 본 연구에서는 도서관 비대면 서비스의 범위를 파악하고, 이에 대한 이용자의 인식조사를 통해 팬데믹 상황에서 도서관 비대면 서비스 운영 방향을 제안하고자 하였다. 이를 위해 37개의 공공도서관에 도서관 이용자를 대상으로 2020년 2월 12일부터 약 3주간 온라인 설문조사를 진행하였으며, 회수된 117부의 응답지를 SPSS 통계 프로그램을 활용하여 빈도분석, 회귀분석, 상관분석, 신뢰도 분석 등을 실시하였다. 설문결과 및 자료조사 결과를 통해 도서관 비대면 서비스 운영 방향으로 도서관 서비스 재정립, 도서관 비대면 서비스 확대, 이용자 교육 진행, 비대면 서비스에 대한 홍보 진행, 이용자 의견 수렴을 제안하였다.

        Anion exchange membrane fuel cells (AEMFCs) have been considered as a promising energy device. To make highly efficient AEMs, high ion conductivity, mechanical stabilities and alkaline stabilities are required. High ion conductivity generally accompanies poor mechanical properties, and crosslinking has been widely applied to overcome these problems. Our groups introduced novel terminally-crosslinked piperidinium-functionalized poly(arylene ether sulfone)s as a anion exchange membrane. Unlike the typical AEMs, our terminally-crosslinked membranes are expected to have high conductivity, especially at low RH conditions due to enhanced water-holding capacity, while maintaining the high chemophyscial stabilities by crosslinking. The synthesis, and properties of the prepared membranes will be discussed in detail.

        Novel terminally-crosslinked ammonium-functionalized poly(arylene ether sulfone)s have been prepared as novel anion exchange membranes. The copper-mediated click chemistry for the propargyl-terminated PES induced the terminal-crosslinking of this polymer by triazole formation. Unlike the typical AEMs, our terminally-crosslinked membranes showed high conductivity, especially at low RH conditions due to the hydrogen bonding between triazole and water, while maintaining the high chemophyscial stabilities due to crosslinking. The synthesis, and properties of the prepared membranes will be discussed in detail.

        Click Chemistry-Induced Terminally Crosslinked Poly(ether sulfone) as a Highly Conductive Anion Exchange Membrane Under Humidity Condition

        New terminally crosslinked anion exchange membranes (AEMs) were developed by both thermal and click reaction of piperidinium-functionalized poly(ether sulfone) (PES) multiblock copolymer with both bis(aryl azide) and PEG-functionalized- azide (PEG-azide) as crosslinkers. The structure as well as physicochemical and electrical properties of the corresponding terminally-crosslinked membranes were analyzed and compared with their non-crosslinked counterparts. The hydroxide conductivity of our terminally-crosslinked membranes (especially the PEG-PI-xPES membrane) under room humidity and high-temperature conditions was found to be high because of their unique structures generated by terminal-crosslinking, and hence high water-holding capacity of these crosslinked systems. The amount of bound water was analyzed using both TGA and DSC technqiues to support the enhanced water-holding capacity for our terminally- crosslinked membranes.

        토목 및 건설, 건축 등의 현장에서 많이 사용되는 가설기기들은 기계의 자체적인 기계고장 뿐만 아니라 야외 현장의 환경에 따른 기후의 변화에도 고장이 발생할 수 있다. 이러한 고장들을 사후약방문의 형식으로 고장이 발생하는 경우에만 수리 후 사용한다면 시간적/경제적으로 많은 손실이 있을 것이다. 그러나 가설기기들의 종류별 기기적 특징을 미리 시스템화하여 발생할 수 있는 고장을 사전에 방지하고 예방한다면 불필요한 손실을 미연에 막을 수 있다. 따라서 본 논문에서는 가설기기들과 관련된 각종 빅 데이터를 이용하여 피로도를 예측하여 고장이 발생하기 전에 사전에 예방할 수 있는 시스템을 제안한다.

      • 푸새직물의 효소세척 시 계면활성제 적용 연구

        푸새는 직물에 풀을 먹이는 행위로 전통 의류 관리 방법 중 하나이다. 전분으로 이루어진 풀은 푸새직물 문화재에 생물학적 손상을 가속할 수 있어 고착된 풀을 제거하는 과정이 요구된다. 본 연구에서는 고착된 풀 제거 시 세척성 향상을 위해 효소(α-amylase)세척에 세척제의 침투력을 높이는 계면활성제(SDS, Triton X-100)의 적용 가능성을 확인해보고자 하였다. 계면활성제의 적용 평가는 직물 내 잔여 전분 검증, 세척성 및 재질 안정성 평가를 통해 진행하였다. 잔여 전분 검증에서는 효소세척에 계면활성제를 적용했을 때, 푸새하지 않은 원포와 유사한 경향을 띠어 직물 내에 풀이 남아있지 않음을 확인하였다. 세척성 평가에서는 계면활성제 중 SDS로 세척 후 효소 세척을 하였을 때 세척성이 우수한 것을 확인할 수 있었다. 세척성 평가에서 SDS는 세척으로 정돈된 표면 특성을 관찰할 수 있었으며, 세척률 평가에서도 면과 견 각각 66.3%, 29.8%로 가장 높게 나타났다. 또한 안정성 평가에서도 효소세척보다 SDS를 적용한 세척 방법이 pH가 중성에 가깝게 나타났고, 미생물 오염도가 가장 낮아 안정성을 확인하였다. 따라서 열화된 푸새직물의 효소세척 시 계면활성제의 적용이 세척 방법으로 활용될 수 있을 것으로 기대된다. Sizing(Pusae) is the act of starching fabrics and is one of the traditional ways to take care of clothes. Rice powder based starch can accelerate biological damage to sized historic textile, so removing the rice powder is needed. The objective of this study is to investigate applicability of surfactants(SDS, Triton X-100), which increase permeability of the cleaning agent in enzyme cleaning, in order to improve detergency when cleaning the sized fabrics. Applicability evaluation was performed by comparison of residual starch, and evaluation of detergency, and material stability. In the comparison of residual starch, rice starch in the fabrics of enzyme cleaning after surfactants did not remain as they were similar to the fabrics without sizing. In the evaluation of detergency, enzyme cleaning after SDS had excellent detergency. As enzyme cleaning after SDS had the neat and tidy surface, and in the evaluation of soil removal, those of cotton and silk were very high at 66.3% and 29.8%, respectively. As a result of material stability, when using enzyme cleaning after SDS. pH was nearly neutral compared to enzyme cleaning, also the result of microbial contamination was the lowest. As a result, it is likely that the surfactant will be used when cleaning the degraded sized fabric with amylase.
