        Comparison of enzymatic activities between the recombinant CHT1 proteins from the hard tick Haemaphysalis longicornis expressed in E. coli and baculovirus-mediated Sf 9 cells

        A chitinase cDNA named CHT1 was cloned from the hard tick, Haemaphysalis longicornis, and the enzymatic properties of its recombinant proteins were characterized. The CHT1 cDNA encodes 930 amino-acid (aa) residues including a 22 aa putative signal peptide, with the calculated molecular mass of the putative mature protein 104 kDa. The E coli-expressed rCHT1 exhibited weak chitinolytic activity against $4MU-(GlcNAc)_3$. The rCHT1 protein with higher activity was obtained using recombinant Autographa californica multiple nuclear polyhedrosis virus (AcMNPV), which expresses rCHT1 under polyhedrin promoter. These findings suggest that the rCHT1 expressed in baculovirus-mediated Sf 9 cells has a high activity than E coli-expressed rCHT1.

        A simple and sensitive assay for chitinolytic activity of the recombinant CHT1 proteins from the hard tick H. longicornis using ethylene glycol chitin

        To determine effectively the chitinolytic activity of rCHT1 from the hard tick H. longicornis expressed in baculovirus-mediated Spodoptera frugtperda (Sf) 9 cells, a simple and sensitive assay system was established in solid phase using agarose gel containing ethylene glycol chitin as substrate. The various factors affecting the efficacy of the assay were also investigated. The effects of various temperature, dosages of proteins, pH of media and time courses of reaction were examined to verify the sensitivity of assay for chitinolytic activity of rCHT1 protein. It was found that the optimal reactive conditions were $37^{\circ}C$ of temperature, 12 to 15 hours of reactive times, $0.1{\mu}g$ of protein concentration and pH 5 to 7 of media. Using the assay system designed, the functional activities of H. longicornis rCHT1l protein could be evaluated simply and sensitively.

        The Effect of Seminal Plasma on Chilling and Freezing of Canine Spermatozoa

        Seminal plasma (SP) is usually removed from semen that is to be cryopreserved. However, some reports indicate that SP has beneficial effects on spermatozoa during chilling and freezing. The purpose of this study was to determine the effect of SP on sperm survival by adding SP to the extender before cooling and freezing canine spermatozoa. In replicate experiments, ejaculates obtained from four healthy dogs (1-4 years old) of various breeds were pooled, centrifuged at 300 × g for 10 min at 25oC, and the supernatant of seminal plasma was decanted. Spermatozoa were suspended in egg yolk-Tris (EYT) buffer. The study comprised two experiments: [Exp 1] Sperm were suspended in EYT extender containing either 0, 20, 40, 80 or 100% SP and were slowly cooled to 4oC for 2 h or held at 25 oC as controls. Sperm concentration was adjusted to 2 × 108/ml. [Exp II] Sperm samples, each of which contained 1 × 108/ml, were assigned to nine groups to be frozen. In the first four groups, sperm in EYT containing either 20, 40, 80 or 100% SP were cooled to 4oC, then diluted to contain final concentrations of EYT + 0.6 M glycerol and then were frozen. The final concentrations of SP were 10, 20, 40 or 50%. In the other four groups, sperm in EYT alone were first cooled slowly to 4oC, then diluted to contain final concentrations of EYT + 0.6 M glycerol plus 10, 20, 40 or 50% SP and then were frozen. Spermatozoa, which chilled in EYT alone and diluted to contain final concentrations of EYT + 0.6 M glycerol without seminal plasma, and then frozen, was regarded as control. Spermatozoa were frozen at 25oC/min of cooling rate in plastic straws that were suspended above liquid nitrogen and thawed in water at 38oC for 1 min. Sperm survival was assayed by determining progressive motility and integrity of plasma and acrosome membranes. Progressive motility was determined by microscopic examination at 200 × magnification. Membrane integrity was assessed by use of a double fluorescent dye, and acrosome integrity by staining sperm with Pisum sativum agglutinin. The results of the first experiment showed that adding SP did not improve motility of spermatozoa compared to those incubated without SP regardless of temperature. The results of the second experiment showed that spermatozoa suspended in EYT+0.6 M glycerol containing SP exhibited the higher progressive motility before being frozen (P < 0.05). However, frozen-thawed spermatozoa that had suspended in EYT + 0.6 M glycerol containing SP showed the similar or lower viability (P < 0.05). In summary, although seminal plasma did not affect spermatozoa that were chilled in EYT without cryoprotectant (CPA), addition of seminal plasma to EYT containing CPA did significantly improved progressive motility of canine spermatozoa that were chilled.

      • Synthesis of copper aluminate for laser direct structuring electronic device

        LDS(Laser Direct Structuring)는 사출 부품에 레이저를 활용하여 패턴을 형성하고 그 위에 무전해 도금을 통해 전도성 회로를 완성하는 기술이다. 본 연구에서는 Copper aluminate 소재를 합성하여 LDS 기술에의 적용 가능성을 고찰하였다. Copper aluminate 소재를 솔 젤법을 사용하여 약 30~40nm의 나노 크기로 합성하였으며, 열가소성 수지에 혼용하여 사출품 형태로 성형을 하였다. 사출된 시편에 UV 레이저를 활용하여 패터닝 후 무전해 도금 공정으로 전도성 패턴의 형성을 고찰하였다.

      • 야생너구리(Nyctereutes procyonoides)의 혈액 및 혈액화학치

        This study was performed to determine the blood and blood chemical values on captured 16 raccoon dogs (Nyctereutes procyonoides). Hematological examination revealed that RBC, Hb, PCV, MCV, MCH and MCHC values were 7.9±0.8x10^(6)/㎕, 14.0±2 g/dl, 44.1±5.5 %, 56.9±3.8 fl, 18.0±1.4 pg and 31.9±2.9 g/dl. respectively, and also revealed that WBC, neutrophil, lymphocyte. eosinophil and monocyte values were 13.6±2.4x10^(3)/㎕, 8.9±2.1x10^(3)/㎕, 2.7±0.8x10^(3)/㎕, 1.1±0.5x10^(3)/㎕ and 1.0±0.4x10^(3)/㎕, respectively. Blood chemical examination showed that total protein, albumin, globulin and A/G ratio values were 7.9±0.9 g/dl, 3.0±0.3 g/dl, 4.8±0.7 g/dl and 0.6±0.1, respectively, and also showed that ALT, AST, BUN. Creatinine and glucose values were 95.8±57.8 1U/ℓ, 64.4±30.1 1U/ℓ, 20.8±5.4 ㎎/dl, 0.7±0.1 ㎎/dl and 58.5±19.1 ㎎/dl, respectively.

        개의 전침마취에 관한 연구 2. 배부혈위의 효과에 대한 검토

        To elucidate the effect of dorsal acupoints for canine electroacupuncture anesthesia, mongrel dogs(6 heads: 2.6-10.5kg, 3 males and 3 females) were used. After needling perpendicularly to Tian-Ping(GV-5) and Bai-Hui(GV-20), the electrodes were connected with positive to Tian-Ping and negative to Bi-hui, respectively using with veterinary electroacupuncture apparatus(TEC AM-3000). The electric conditions were 2-4V and 30 Hz. The pain of body surface was checked by hemostatic forceps. The effect of electroacupuncture andesthesia was appreciated by application of laparotomy(2 heads), ear trimming and tail amputation(1 head), castration(2 heads) and enterotomy(1 head), respectively. Induction time, clinical findings and operation findings were investigated under the electroacupuncture anesthesia. The induction time of electroacupuncture anesthesia was very rapid, approximately 1 minute and the pain of body surface including the extremities was not detected. As for clinical findings during electroacupuncture anesthesia, consciousness was evident, pupil reflex was weak and blepharoreaction was existed. As for operation findings, pain was not detected during incision and suture, and bleeding was relatively small volume. The grade of electroacupuncture anesthesia was excellent in all cases applied with ear trimming and tail amputation, laparatomy, castration and enterotomy. Considering above all findings, it was thought that electroacupuncture stimulation at Tian-Ping and Bai-hui examined in the present study induced excellent electroacupuncture anesthesia for dogs.

      • Selective metallization of polyimide composite film for flexible printed circuit board via laser direct structuring

        Flexible Printed Circuit Board(FPCB)는 전자제품의 발전으로 소형화, 경량화, 집적화에 대한 요구들을 충족 시켜 줄 수 있는 폴리머 기반의 기판이다. 그 중에서도 Polyimide를 base로 한 FPCB는 유연성, 내화학성, 내열성이 뛰어난 소재이다. 본 연구에서는 Polyimde를 사용하여 Laser Direct Structuring(LDS) 공정을 통해 FPCB를 제작하였다. Copper aluminate를 Polyimide에 함량별로 첨가 하고 도포하여 Composite film 형태로 제작하였다. 1064nm laser를 사용하여 패터닝을 하고 무전해 동도금을 진행하였다. 함량별, Laser 공정조건, 무전해 도금의 조건 변수에 따른 변화를 Confocal microscope, SEM을 통해서 확인하였으며, 구현된 회로의 전도성과 비저항을 측정 하였다. 이번 실험을 통해 FPCB에 LDS 공정을 적용할 수 있었고, 회로 특성에 최적화된 laser 공정변수와 도금 조건을 찾을 수 있었다.

        Prevalence of Canine Giardiasis in the Daejeon and Chungnam Area

        A total number of 100 fecal samples were examined for the presence of Giardia intestinalis infection in dogs using a Giardia ELISA kit (SNAP® test, IDEXX Laboratories, Inc., Westbrook, ME). 49 males and 51 females were examined for a G. intestinalis infection in Daejeon and Chungnam area of South Korea. The overall positive rate of G. intestinalis infection was 12.00%. G. intestinalis infection resulted to be more prevalent in males (12.24%) than in females (11.76%), and in symptomatic dogs (18.18%) than in asymptomatic dogs (11.54%). There were no significant differences between the two groups. 19.60% being found in the < 2-year-old group, 4.08% in the over 2 year-old group. X2 analysis revealed a significantly higher prevalence (p < 0.05) in the < 2-year-old group than in the other, and a significantly higher prevalence in dogs kept in a shared kennel (36.00%, p < 0.001). This study is the first survey of G. intestinalis infection prevalence in South Korea according to life style (particularly between dogs kept in a shared kennel and that of dogs kept separately) using an ELISA kit, and this study is expected to provide a useful reference for clinicians and breeders.
