        불가사리 부타놀 분획의 항알러지 활성

        We investigated the effects of butanol fraction of Asterias amurensis (BFA) on the anti-allergic activity. BFA inhibited both cyclooxygenase-2 dependent prostaglandin $D_2$ and 5-lipoxygenase dependent leukotriene $C_4$ generation in a concentration-dependent manner with $IC_{50}$ values of 174.6 and 22.2 ${\mu}g$/ml, respectively. In addition, BFA also inhibited the degranulation in a dose dependent manner. Furthermore, oral administration of BFA inhibited IgE-mediated passive cutaneous anaphylaxis in mice. These results suggested that BFA may be useful in regulating mast cell-mediated allergic diseases.

        Anti-allergic Activity of an Ethanol Extract from Salviae miltiorrhiza

        As part of an ongoing investigation aimed at the discovery of novel bioactive medicinal herbs with antiinflammatory properties, the effects of an ethanolic extract from the parts of Salviae miltiorrhiza Bunge (ESM) were evaluated using in vitro and in vivo animal model analysis. ESM inhibited cyclooxygenase- 2 (COX-2) and COX-1-dependent phases of prostaglandin D₂ (PGD₂) generation in bone marrowderived mast cells (BMMC) in a concentration-dependent manner with IC50 values of 3.96 μg/mL and 21.54 μg/mL, respectively. Furthermore, ESM inhibited leukotriene C₄ (LTC₄) production with an IC50 value of 2.6 μg/mL. These results clearly demonstrated the dual COX-2 selective/5-lipoxygenase inhibitory activity that ESM possessed. ESM strongly inhibited a degranulation reaction in a dose dependent manner within a BMMC system, with an IC50 value of 22.4 μg/mL. Additionally, ESM was tested in a rat passive cutaneous anaphylaxis (PCA) reaction assay by oral administration (25 to 100 mg/kg). ESM dose-dependently inhibited the PCA reaction, which was activated by anti-dinirophenyl (DNP) IgE. These results suggested that ESM might be beneficial in regulating various allergic reactions.

        새싹인삼 추출물의 항산화 활성 및 MMP-1 저해 활성

        Objectives : As a substitute for high-price ginseng, this study attempted to examine a possibility of the ferment extract of Panax ginseng sprout whether leaves and roots can be used together as a cosmetic ingredient with anti-oxidative and wrinkle-care effects. Methods : In terms of a test method, antioxidant activities were confirmed through total polyphenol contents, total flavonoid contents, DPPH radical scavenging activity and ABTS radical scavenging activity using the Panax ginseng sprout. In addition, to assess wrinkle-care effectiveness, the cytotoxicity of the extract was analyzed through MTT assay, and inhibition of collagenase activity in the cells was tested using the Panax ginseng sprout fermented by Saccharomyces cerevisiae. Resuits : The content of polyphenols and flavonoids in natural plants was highest in Panax Ginseng Sprout Extract at 100℃, which also demonstrated high DPPH, ABTS radical scavenging activity. MTT assay demonstrated that the Panax Ginseng Sprout Ferment Extract did not have a cytotoxic effect in CCD-986SK cell. Also, Panax Ginseng Sprout Ferment Extract was found to inhibit MMP-1 expression by 51.85±6.09% at a concentration of 10%. Conclusions : Therefore, this study has confirmed a possibility of Panax ginseng sprout ferment extract as a cosmetic ingredient with MMP-1-inhibitory effects.

        구속 스트레스 모델에서 석결명의 간손상 및 우울증 관련 인자에 미치는 영향

        Objectives : This study investigated anti-inflammatory effects of Nardotidis seu Sulculii Concha water extract (NSCE) against restraint-induced stress. Methods : In vivo, NSCE was orally administered to male white mice at concentrations of 250 mg/kg and 500 mg/kg for 3 days, and then restraint-induced stress was induced for 6 hours. The level of liver damage was measured by serum aspartate aminotransferase (AST), alanine aminotransferase (ALT) and lactate dehydrogenase (LDH). The stress-related hormones such as cortisol and corticosterone were measured by ELISA assay. Also, western blot analysis was performed to detect expression of mitogen-activated protein kinase (MAPK) and cyclooxygenase-2 (COX-2) proteins. Pathological changes were observed by hematoxylin and eosin (H&E) staining of the liver tissue, and Immunohistochemical (IHC) staining was performed to examine liver inflammation through macrophage infiltration. Results : The AST, ALT, LDH and the stress related hormones such as cortisol and corticosterone were significantly decreased in the NSCE treated group compared with stress group. In histological analysis, H&E staining of liver tissues did not detect the hepatic injury or damage in all groups. As a result of IHC staining, it was confirmed that infiltration of macrophages was increased in the stress-induced group, but decreased in the group treated with NSCE. The COX-2 and MAPK proteins expression was significantly increased by restraint-induced stress, but these proteins were decreased in the NSCE treated group. Conclusions : These results suggest that NSCE has the anti-inflammatory activity in restraint-induced stress model, and it is believed that NSCE can be used for the prevention of liver inflammation.

        알코올 유발 간 손상 마우스 모델에서 복합 추출물 MJY2018의 간 보호 및 항산화 효과

        Objectives : This study investigated the liver-protective effects of MJY2018, a Herbal formula, against alcoholic fatty liver disease and anti-oxidative effects. Methods : Its effects were investigated in an alcoholic fatty liver disease model in male C57BL/6 mice, which were fed Lieber−DeCarli liquid diet containing ethanol. MJY2018 (100 and 200 mg/kg bw/day) or silymarin (50 mg/kg bw/day) were orally administered daily in the alcoholic fatty liver disease mice for 16 days. Results : The results indicate that MJY2018 promotes hepatoprotection by significantly reducing aspartate transaminase (AST) and alanine transaminase (ALT) levels as indicators of liver damage in the serum. Furthermore, MJY2018 reduced accumulation of triglyceride and total cholesterol, increased levels of superoxide dismutase (SOD) and glutathione (GSH) in the livers of the alcoholic fatty liver disease mice model. Additionally, it improved the serum alcohol dehydrogenase (ADH) activity. Conclusions : These results indicate that MJY2018 were effective in improving and protecting oxidative stress and alcoholic liver disease.

        황련해독탕의 위염 치료 효과에 대한 단일비교 임상연구

        Objectives : The purpose of this clinical study was to investigate the efficacy and safety of Hwangryunhaedok-tang on the gastritis treatment. Methods : A total of 20 subjects who visited Pusan National University Hospital from April 2nd, 2019 to September 2nd, 2019 were included. In this study, we evaluated the individual symptoms through questionnaires, laboratory test, BIA (Bioelectrical Impedance Analysis), and adverse reaction according to medication. Statistical analysis was performed with paired t-test and Wilcoxon Signed Rank test. Statistical significance was achieved if the probability was less than 5% (p < 0.05). Results : After medications, the total score of individual symptoms, primary outcome, was significantly decreased. In particular, individual symptoms such as upper abdominal pain, bloating, heartburn and trim showed significant improvement. However, no statistically significant differences were found in nausea/vomiting and anorexia. In additions, the laboratory test showed a statistically significant decrease in total cholesterol, but no statistically significant differences in results of BIA. In the safety evaluation, adverse reactions were not reported. Conclusions : Considering this results, we have confirmed the possibility of Hwangryunhaedok-tang to the gastritis treatment.

        Anti-inflammatory Activity of 6-Hydroxy-2,7-dimethoxy-1,4-henanthraquinone from Tuberous Roots of Yam (Dioscorea batatas) through Inhibition of Prostaglandin D_2 and Leukotriene C4 Production in Mouse Bone Marrow-derived Mast Cells

        6-Hydroxy-2,7-dimethoxy-1,4-phenanthraquinone (PAQ) isolated from the tuberous roots of Yam (Dioscorea batatas) inhibited cyclooxygenase-2 (COX-2) and cyclooxygenase-1 (COX-1) dependent prostaglandin D_2 (PGD_2) generation in mouse bone marrow-derived mast cells in a concentration-dependent manner with IC_50 values of 0.08 μM and 0.27 μM, respectively. In the Western blotting with specific anti-COX-2 antibodies, the decrease of the quantity of PGD2 was accompanied by a decrease in the COX-2 protein level. But PAQ did not affect COX-1 protein level. In addition, this compound inhibited 5-lipoxygenase (5-LOX) dependent production of leukotriene C_4 in a dose-dependent manner, with an IC_50 of 0.032 μM. These results demonstrate that PAQ has a dual COX-2/5-LOX inhibitory activity. This compound also inhibited the degranulation reaction in a dose-dependent manner with an IC_50 of 2.7 μM. Thus, these results suggest that PAQ may be useful in regulating mast cell-mediated inflammatory diseases.

        황련 추출물의 LC-MS/MS 분석 및 항염증 효과

        Objectives : The main aim of this study was to examine the LC-MS/MS used to identify phenolic compounds of CRE(Coptidis Rhizoma 70% EtOH Extract). Also, we investigated antioxidative activities and Anti-inflammatory activities. Methods : LC-MS/MS Analysis HPLC and LC-MS/MS were performed on a 1260 series HPLC system (Agilent Technologies, Inc., California, USA) and 3200 QTrap tandem mass system (Sciex LLC) operated in positive ion mode (spray voltage set at −4.5 kV). The solvent used was DW and Acetonitrile containing 0.1% formic acid, a gradient system was used at a flow rate of 0.5 mL/min for analysis, and a Prontosil C18 column (length, 250 mm; inner diameter, 4.6 mm; particle size, 5 µm; Phenomenex Co., Ltd., California, USA, Biochoff Chromatography) was used. The solvent conditions used in the mobile phases were 0–10 min at 10–15% B, 10–20 min at 20% B, 20–30 min at 25%, 30–40 min at 40%, 40–50 min at 70%, 50–60 min at 95%, and 60–70 min at 95%. The analysis was performed at a wavelength of 284 nm and a temperature of 35℃. The cell viability was measured using a 3-(4,5-dimethyethiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. We examined the effects of CRE on the lipopolysaccharide (LPS)-induced production of nitric oxide (NO) in a RAW 264.7 cells Results : The chemical analysis CRE by Liquid chromatography-tandem mass spectrometry (LC-MS/MS) confirmed that Rosmarinic acid, Ferrulic acid, 3-O-feruloylquinic acid, and 5-O-feruloylquinic acid as phenolic components. DPPH radical scavenging activity was the inhibitory activity of CRE showed at 200 μg/mL a statistically significant level. MTT assay demonstrated that the CRE did not have a cytotoxic effect in RAW 264.7 and LPS-induced RAW264.7 cells. Also, CRE reduced NO production in RAW 264.7 cells stimulated with LPS. Conclusions : Based on these findings, The chemical analysis 4 major components CRE such as Rosmarinic acid, Ferrulic acid, 3-O-feruloylquinic acid, and 5-O-feruloylquinic acid. Moreover, we confirmed that CRE has effects antioxidant and anti-inflammatory. The results demonstrate that CRE can be used as an antioxidant and a powerful chemopreventive ingredient against inflammatory diseases.

        our continuing effort to investigate compounds having anti-inflammatory activity from natural products, Ail-anthus altissima was examined.         산화적 스트레스에 대한 석결명의 세포 보호 효과

        Objectives : This study investigated cellular-protective effects of Nardotidis seu Sulculii Concha water extract (NSCE) against oxidative stress induced by arachidonic acid (AA)+iron or tert-butylhydroperoxide (tBHP). Methods : In vitro, MTT assay was assessed for cell viability, and immunoblotting analysis was performed to detect expression of AMP-activated kinase (AMPK) signaling pathway and autophagy related proteins. In vivo, mice were orally administrated with the aqueous extract of NSCE of 500 mg/kg for 3 days, and then injected with CCl<sub>4</sub> 0.5 mg/kg body weight to induce acute damage. The level of liver damage was measured by serum aspartate aminotransferase (AST), alanine aminotransferase (ALT) and lactate dehydrogenase (LDH) analysis. Results : Treatment with NSCE inhibited cell death induced by AA+iron and tBHP. NSCE induced the phosphorylation of AMPK, and this compound also induced the phosphorylation of LKB1, an upstream kinase of AMPK, and Acetyl-CoA carboxylase (ACC), a primary downstream target of AMPK. NSCE increased the protein levels of autophagic markers (LC3II and beclin-1) and decreased the phosphorylation of mammalian target of rapamycin (mTOR) and simultaneously increased the phosphorylation of unc-51-like kinase-1 (ULK-1) in time-dependent manner. Conclusions : NSCE has the ability 1) to protect cells against oxidative stress induced by AA+iron or tBHP. NSCE 2) to activate AMP-activated protein kinase (AMPK), and 3) to regulate autophagy, an important regulator in cell survival.
