        금낭화(Dicentra spectabilis)의 배우체 형성 및 접합자배의 발달

        본 연구는 금낭화 배우체의 형성과 배 발달특성에 관한 기초 자료로 활용하기 위해 수행되었다. 시원세포로부터 발달된 소포자 모세포는 화뢰의 길이가 1 mm일 때 감수분열을 하여 4면체형(tetrahedral) 4분자가 형성되었다. 4개의 소포자는 분리되어 각각 웅성배우체로 발달하였다. 대포자 모세포는 화뢰의 길이 $4{\sim}5\;mm$ 수식 이미지에서 관찰되었다. 대포자의 발달유형은 정상형(polygonum)이었으며, 배낭의 형태는 굽어있는 곡생배주(amphitropous)였다. 3개의 뚜렷한 반족세포는 배낭이 성숙한 후에도 퇴화하지 않고 남아있었다. 개화 전 자웅배우체는 충분히 성숙하였다. 개화 시에 수술과 암술의 길이는 거의 비슷하거나 수술이 0.5 mm 짧아서 자화수정에 적당한 구조를 가지는 것으로 나타냈다. 수정 후 배는 구형, 심장형을 거쳐 자엽배까지 발달하였으며, 종자 산포시 초기 자엽배를 가지고 있음을 알 수 있었다. This study was carried out to obtain the basic informations on the characteristics of gametophytes formation and embryo developement in Dicentra spectabilis. Microspore mother cells developed from archesporial cells, start meiosis when flower bud length reaches around 1 mm, formed tetrahedral type tetrad. The 4 microspores were separated. They were developed to male gametophytes, respectively. Megaspore mother cells were observed when flower bud length was 4~5mm. The developemental type of megaspore was polygonum and embryo sac was amphitropous. Three large and distinctive antipodals did not degenerated and remained after embryo sac was developed. When the male and female gametophytes was fully developed, the length of stamen and style was very similar or stamen was shorter about 0.5 mm than that of style. This result indicates that self-fertilization can be occurred in this species. After fertilization, developing zygotic embryos showed various stages of development from globular to cotyledonary embryos, and zygotic embryo in seed scattering time seemed to have an early cotyledonary stage.

        장기간 심실세동상태의 돼지에서 고용량 epinephrine전처치의 효과

        There is a continual loss of high-energy phosphates in myocardium during prolonged ventricular fibrillation(VF), which cannot support myocardial contraction after defibrillation. Defibrillation should be attempted regardless of the duration of VF but the cumulative energy imparted to the fibrillating myocardium can induce structural myocardial injury. The myocardial blood flow requirements during VF are 50ml/min/100g, one third of blood flow, during normal sinus rhythm in swine. To attain this level of myocardial blood flow, epinephrine is required during CPR. The purpose of this study was to determine the effectiveness of epinephrine preceding defibrillation in a swine model of prolonged VF and comparing these results to intial defibrillation treatment only. Fourteen swine (20-25kg) were anesthezied and instrumented for hemodynamic monitoring. After eight minutes of VF, the control group(7) received defibrillation at 3J/kg. If unsuccessful, two additional defibrillation were attempted at 6J/kg. (AHA guideline) The other animals(7, experimental group) were given 0.025mg/kg epinephrine after eight minutes of VF. After two minutes of epinephrine injection, defibrillation was attempted in the same manner as the control group. If unsuccessful, the same dose of epinephrine was administered and external cardiac massage was begun. The mean weight of both group was 21±1kg. Systemic blood pressure was 110±15/68±12mmHg, 114±22/78±22mmHg in the control group, experimental group, respectively. There were no statistical differences in blood pressure and pulse rate between two groups. In the control group, 14% of the animals were resuscitated to a perfusing rhythm. In the experimental group, 71% of the animals were resuscitated to a perfusing rhythm. These results suggest that method of epinephrine pretreatment preceding defibrillation is more effective for restoration of spontaneous circulation than that of initial defibrillation in 8 minutes prolonged ventricular fibrillation in a swine model.

        Adrenaline-Cocaine gel의 국소마취 효과

        Lidocaine is commonly used in children as well as adults as a local anesthetics for dermal laceration repair. Local infiltration of wounds with lidocaine(local anesthetics) inflicts pain on pain, particularly distressing in children. To avoid this pain, the use of topical tetracaine-adrenaline-cocaine(TAC) solution and adrenalin-cocaine(AC) gel is proposed. We have evaluated the anesthetic effect of AC gel for minor dermal laceration repair. For three months(Jul to Oct, 1994), the residents of emergency medicine have sutured minor dermal laceration patients who were treated at to the Department of Emergency Medicine, Severance Hospital Yonsei University College of Medicine. The patients population ranged from 9 months to 46 years with an average of 10.2 years. Thirty of the patients were younger than 10 years of age. Lacerations were located on the face in 33 patients and on the scalp in 4 patients. Larger lacerations were encountered in 4 patients. The average dose of AC gel applied per laceration was 0.45 mL(containing 81.9mg cocaine). Two hundred thirty four sutures of total 240 (97.5%) were placed without eliciting any pain. The average anesthetic induction time was 123 seconds. There were no observed adverse reactions with AC gel application or reported complications of wound healing in any patient. Our results support the use of AC gel is an excellent anesthetic effect for dermal laceration.

        현탁배양을 통한 금낭화(Dicentra spectabilis L. Lemaire)의 대량증식

        Bleeding heart (Dicentra spectabilis L. Lemaire) is one of the most valuable wild flower in Korea. This work was conducted for the mass production of somatic embryos through suspension culture and more effective plant regeneration system in Dicentra spectabilis. High-frequency embryogenic callus proliferation was achieved in SH liquid medium supplemented with 1 mg/L 2,4-D. Half-strength SH medium was suitable concentration for somatic embryo induction and germination. About 5,000 embryos were produced per 250 ml flask after 4 weeks of culture. Germination rate of somatic embryos was decreased when GA3 was added in medium. The plantlets showed a 58% survival rate when transferred to pots after 1 month of culture.         항파상풍 면역 글로부린의 효과

        Despite the progress in treatment modality, tetanus still has a high mortality rate. However, the morbidity of tetanus can be decreased through proper prophylactic measures. Although Center for Disease Control(CDC) recommands Td or Td/TIG injection depending on prior immunization state and wound character, TIG alone is being injected to patient with open wound for tetanus prophylaxis in Korea. Therefore, we studied antitetanus antibody titers after injecting 250IU of TIG to evaluate TIG's duration of immunization effect, in vivo. TIG 250IU was injected to 42 healthy adult volunteers and their antitetanus antibody titers were measured at prior to TIG injection, 1 week and 4 weeks after infection. Antibody titers were measured by using ELISA assay with IMMUNOZYM-TETANUS and their results were verified by using the standard t-test. Mean antitetanus antibody titers were 0.167±0.031IU/mL, 0.211±0.031IU/mL and 0.186±0.031IU/mL at before injection, 1 week and 4 weeks after injection, respectively. Antibody titers showed significant difference between values at before injection and 1 week after injection. However, no difference was found between the values before injection and 4 weeks after injection. The practice of injecting TIG 250IU alone for prophylaxis has little effect on maintaining an increased antitetanus antibody titer until 4 weeks after injection.

        오가피(Acanthopanax sessiliflorus)의 체세포배로부터 식물체 재생에 미치는 GAa<sub>3</sub>와 Charcoal의 영향

        To establish the optimum condition for plant regeneration from somatic embryos of Acanthopanax sessiliflorus Rupr. et Maxim, a medicinal plant, somatic embryos were induced from zygotic embryo-derived embryogenic callus in hormoen-free MS medium. To induce plantlet conversion, cotyledonary somatic embryos were cultured on MS solid medium with GA$_3$at various concentrations (0~10 mg/L) for three weeks. Plantlets were transferred to 1/3 MS solid medium with 0.5% charcoal for 7 weeks. Stem length was increased proportionally to the concentration and treatment period of GA$_3$. Also, the highest leaf width (8.9 mm) and leaf number (2.84) of plantlet were obtained when plantlets were converted on 5,10 mg/L GA$_3$pretreatments, respectively. The highest plant conversion frequency (66.7%) was obtained when the somatic embryos were cultured on medium containing 5 mg/L GA$_3$ for 3 weeks and then were transferred to 1/3 MS medium with 0.5% charcoal. The highest survival rate of soil transfer was 90% when plantlets were regenerated on medium with 5 mg/L GA$_3$ for 3 weeks and then transferred to plastic pots containing vermiculite and sand mixture for 4 weeks.

        블랙베리(Rubus fruticosus L.)의 맹아절편체로부터 다경유도를 통한 식물체 재분화

        블랙베리의 다경유도를 통한 식물체 재생을 위하여, 노지에서 생육되고 있는 맹아를 채취하여 1.2% NaOCl 용액에 침지하여 표면 살균한 후, kinetin과 BA가 각각 포함된 MS 배지에서 4주간 배양하여 절편당 형성된 신초의 수를 조사하여 BA가 효과적임을 밝혔다. 동일방식으로 맹아를 IBA와 BA를 첨가한 MS 고체배지에서 4주간 배양하여 신초를 유도하였는데 1.0 mg/L BA가 첨가된 배지에서 신초유도는 100% 이루어졌다. 기내배양한 신초의 절편체를 1.0 mg/L BA가 첨가된 MS 배지에 치상하여 형성된 신초의 수는 절편체당 5.3개로 최고를 나타내었다. 약 10주 정도 배양을 더 오래하였을 때, 다경 형성율이 더 높아질 뿐 아니라 절편체당 신초 수도 많아졌다. 형성된 신초로부터 뿌리 발생을 위하여 glycine의 농도를 달리한 (0~2.0mg/L) MS 기본배지에서 4주간 배양하였을 때, 0.5 mg/L glycine을 첨가한 배지에서 고빈도 (85%의 뿌리발생을 관찰 할 수 있었다. 재생된 식물체는 모래 : 토양 : 버뮤큘라이트 (1:1:1, vol.) 혼합토양에서 순화시켜, 토양에 이식하였을 때 95%의 식물체 생존율을 나타내었다. This study was carried out to induce plant regeneration via shoot formation from sucker explants of Rubus fruticosus L. To induce adventitious shoots, sucker explants were sterilized in 1.2% NaOCl solution, and cultured on the MS solid medium supplemented with kinetin (0.5, 1.0, 3.0 mg/L) and BA (0.5, 1.0, 3.0 mg/L), respectively. As above, to induce adventitious shoots, sucker explants were cultured on the MS solid medium supplemented with IBA (0, 0.1, 1.0 mg/L) and BA (0, 0.1, 1.0, 2.0 mg/L). After 4 weeks of culture, the highest frquency (100% of shoot formation from sucker explants was obtained from the medium with 1.0 mg/L BA. The highest shoot number per explant from in vitro shoot explants was 5.3. After 10 weeks of culture, the number of shoot per explant was increased. The highest frequency (85% of root formation was obtained at 0.5 mg/L glycine medium, when the explant with shoot were cultured on the MS medium containing glycine at various concentrations from 0 to 2.0 mg/L. The survival rate of the plantlets after transfer to plastic pots containing sand, soil, and vermiculite (1:1:1, vol.) was 95%. The results indicate that micropropagation procedure can be applied for an efficient mass propagation of Rubus fruticosus.

        한국판 다차원 신체자각척도(K-MAIA)

        본 연구는 Mehling et al.(2012)의 다차원 신체자각척도(MAIA: Multidimensional Assessment of Interoceptive Awareness; 이하MAIA)를 한국판으로 개발하고 타당화하기 위한 것이었다. 이를 위해 예비 연구와 두 개의 연구를 실시하였다. 예비연구에서는 MAIA의 32문항을 번안한 척도를 심신통합적 수련을 하는 253명의 성인에게 실시하여 탐색적 요인분석, 내적합치도, 문항의 변별력을 분석하였으며, 이를 바탕으로 척도문항을 수정하였다. 연구 1에서는 심신통합적 수련을 할 것으로 여겨지는 또 다른 295명의 성인 자료를 수집하여, 수정한 척도의 탐색적 요인분석과 하위 요인의 신뢰도를 다시 분석하였다. 또한 이론적으로 관련이 있는 다른 척도들과의 상관계수를 검토하였고, 수련경험 기간과 만성질환 여부 및 성별과 나이에 따른 차이를 검증하였다. 연구 2에서는 일반성인 223명의 자료를 수집하여 확인적 요인분석을 통해 척도의 요인구조를 재검증하였다. 연구결과를 토대로 6개의 하위 요인으로 구성된 32문항짜리 한국판 다차원 신체자각척도(K-MAIA)를 제시하였다. 이 최종척도의 심리측정학적 특성은 K-MAIA가 관련분야의 연구에 사용될 수 있음을 보여주었다. In this study, based on a critical review of the 32-item Multidimensional Assessment of Interoceptive Awareness (Mehling, Price, Daubenmier, Acree, Bartmess & Stewart, 2012; MAIA), a 32-item Korean edition of MAIA was developed in a pilot study and validated in two studies. In a pilot study, the 32 items of MAIA were adapted to Korean language, then administered to 253 adults for item analysis. Based on the results on item discrimination, internal consistency, and exploratory factor analysis, the scale was revised and reconstructed. In the study 1, the scale was administered to 295 adults; reliability analysis and exploratory factor analysis were performed, and correlation coefficients with other related scales were examined. In addition, interoceptive awareness was compared among groups with varying experiences in training related to interoceptive awareness. In the study 2, the scale was administered to 223 adults; confirmatory factor analysis was performed. Results showed that the Korean edition of MAIA with a total of 32 items and six factors had sufficient reliability and validity, demonstrating a high reliability based on internal consistency of .94, theoretically consistent levels of correlation coefficients with other scales, and satisfactory construct validity. The study concludes with discussions on implications and limitations of the study.
