18S-srRNA 분석에 의한 Acantbamoeba 한국 분리주들의 분류 검토

Acanthamoeba spp. spread in soil, ponds, air and swimming pool, cause a granulomatous amoebic meningitis and amoebic keratitis in human and experimental animals. Their classification had depended upon morphological characteristics and physiological conditions such as thermophilic status and generation times. However, because amoeba species showed the morphological and physiological diversity in isolates, many attempts have been made on identification of Acanthamoeba spp. Restriction fragments length polymorphism (RFLP) analysis of IBS-small subunit ribosomal RNA (srRNA) was an useful tool for classification among morphologically and genetically closely-related species. In this study, 18S-srDNAs of amoebae were amplified by PCR with primer encoding 18S-srRNA gene and digested with restriction endonudeases, and RFLP analysis was applied on classification of Acanthamoeba Korean isolates (YM-4, YM-5 and YM-7), comparing with reference amoebae, A. culbertsoni, A. polyphaga and A. royreba. Trophozoites of six Acanthamoeba spp. had typical acanthopoda, but did not show any morphological differences. Cyst of Acanthamoeba sp. YM-7 was similar to that of A. polyphaga which was designated as group Ⅱ Acanthamoeba. Morphologically other amoebae belonged to group Ⅲ Acanthamoeba. Digestion of 18S-srDNA with six enzymes, Dde Ⅰ, Hae Ⅲ, Hind Ⅲ, EcoR Ⅰ, Rsa Ⅰ and Sph Ⅰ, resulted in various DNA fragments. Using the method of Nei and U (1979) for RFLP analysis, genetic divergence was not observed between A culbertsoni and Acanthamoeba sp. YM-4. Between A. culbertsoni and Acanthamoeba sp. YM-5, genetic distance was 0.070, 0.364 between Acanthamoeba sp. YM-4 and A. polyphaga, and 0.277 between Acanthamoeba sp. YM-7 and A. polyphaga. In comparison with A. culbertsoni and A. polyphaga, Acanthamoeba sp. YM-7 showed genetic distance of 0.330 and 0.154, respectively. Thus, Acanthamoeba sp. YM-7 is a similar species to A. polyphaga. Acanthamoeba sp. YM-5 is a different species from A. polyphaga, but closely-related to A. culbertsoni. Acanthamoeba sp. YM-4 can be classified as a subspecies or a strain of A. culbertsoni.

Laser surface hardening is an effective technique used to improve the tribological properties and also to increase the service life of automobile components such as camshafts, crankshatfs, lorry brake drums and gears. High power CO₂ lasers and Nd:YAG lasers are employed for localized hardening of materials and hence are of potential application in the automobile industries. The heat is conducted rapidly into the bulk of the specimen causing self quenching to occur and the formation of martensitic structure. In this investigation, the microstructure features occurring in Nd:YAG laser hardening SM45C steel are discussed with the use of optical microscopic and scanning electron microscopic analysis. Moreover, This paper describes the optimism of the processing parameters for maximum hardened depth of SM45C steel specimens of 3mm thickness by using CW Nd:YAG laser. Travel speed was varied from 0.6m/min to 1.0m/min. The maximum hardness and case depth fo SM45C steel are 780Hv and 0.4mm by laser hardening.
다구찌 방법을 이용한 고속/정밀 위치제어시스템의 강인한 제어게인 선정에 관한 연구

This paper presents a dynamic modeling and a robust PID controller design process for the wire bonder head assembly. For the modeling elements, the system is divided into electrical system, magnetic system, and mechanical system. Each system is modeled by using the bond graph method. The PID controller is used for high speed/high accuracy position control of the wire bonder assembly. The Taguchi method is used to evaluate the more robust PID gain combinations than conventional one. This study makes use of an L18 array with three parameters varied on three levels. Computer simulations and experimental results show that the designed PID controller provides more improved signal to noise ratio and reduced sensitivity than the conventional PID controller.

2008년 2월 10일 국보1호 숭례문의 방화사건 이후로 정부에서는 문화재 보호를 위한 다양한 사업을 실시 하고 있다. 이중 문화재를 화재 및 각종 재난으로부터 보호하기 위한 인적 자원 활용사업인 ‘문화재 안 전경비인력사업’의 지정현황 및 운용현황을 검토하고 향후 예방적이고 체계적인 재난관리 활동이 이루 어질 수 있도록 안전경비인력 운용에 관한 발전방안을 제시하였다. Since the case of setting Sungnyemun which was No. 1 national treasure on fire on February 10, 2008, the government has been carrying out various businesses for protecting cultural properties. The designation and operation status of 'Safety Security Personnel System for Cultural Properties' which was the human resource utilization business for protecting cultural properties from fire and various disasters among those businesses were reviewed and the plans for developing the operation of security guards were presented for preventive and systematic disaster management activities in future.

본 연구에서는 목조문화재 안전관리 표준화 방안을 위해 전국에 국가지정 중요목조문화재 159개소의 실태조사를 실시하였다. 문화재 안전관리를 인적측면, 안전관리적 측면, 시설적 측면, 주변환경적 측면으로 종합적인 실태조사를 실시하였으며, 현 문화재 안전관리 현황 및 정책을 알아보고 이에 대한 시사점을 제시하였다. 이를 통하여 중요목조문화재의 향후 안전관리 표준화 방안에 대한 기초 결과를 제시했다.
고속/정밀 위치제어시스템의 모델링 및 제어에 관한 연구

This paper presents a dynamic modeling and a sliding mode controller for the high-speed / high-accuracy position control system. Selected target system is the wire bonder head assembly which is used in semiconductor assembly process. This system is a reciprocating one around the pivot point that consists of VCM(voice coil motor) as a actuator and transducer horn as a bonding tool. For the modeling elements, the system is divided into electrical circuit, magnetic circuit and mechanical system. Each system is modeled by using the bond graph method and united into the full system. Two major aims are considered in the design of the controller. The first one is that the horn must track the given reference trajectory. The second one is that the controller must be realizable by using the DSP board. Computer simulation and experimental results show that the designed sliding mode controller provides better performance than the PID controller.

For many years and primarily for economical reasons, Dissimilar Metal Welds have been used as transition joints in a variety of equipment and applications. But Dissimilar Metal Welds have several fabrication and metallurgical drawbacks that can often lead to in-service failures. In this paper, the laser weldability of STS304 stainless steel and SM45C at dissimilar metal welds using a continuous wave Nd:YAG laser are experimentally investigated. An experimental study was conducted to determine effects of welding parameters, on eliminating or reducing the extent welding zone formation at dissimilar metal welds and to optimize those parameters that have the most influence parameters such as focus length, power, beam speed, shielding gas, and wave length of laser were tested.

Utilizing the electrospinning technique nanofiber-based biodegradable scaffolds made of PLGA was suggested. Under various conditions, their diameters and porosity as well as mechanical strength were evaluated. In addition to those, cell(chondrocyte) proliferation and formation of extracelluar matrices were also investigated along with the conventional membrane type PLGA scaffolds for the potential use in tissue engineering. As conclusions, this type of scaffold showed a potential of application to tissue engineering in view of mechanical stability as well as cellular responses.
