        Reevaluation of protein standards which have been used in conventional gel filtration chromatography was conducted by high performance gel filtration column. Plotting the log molecular weight and its retention time, a linear calibration curve could be achieved using the well documented protein standards. However, an unexpected finding was the deviation of several protein standards approximately ranging from 13,000 to 15,000 from linearity. Although we did not clarify the question about the deviation of cytochrome C, lysozyme, ribonuclease A, and α-lactalbumin, we assume that the physico-chemical differences between two matrices, PROTEIN-PAK 125 and Sephadex G-75, may explain this behaviour.

      • 食生活 管理 實態에 關한 調査 硏究 : 全羅北道 主婦를 中心으로 Focused on the Housewives in Jeonbug Province

        The purpose of this study is to find out the problems on the meal management of housewives in Jeon Bug province. Specifically, the actual condition of the budget of dietary life, menu planning, food purchasing, food habits and meal improvement were analyzed by housewives age, educational levels and then economic status. The data in this study was obtained by the survey of 1, 093 wives who lived in Jeon Bug province by means of a questionnaire. Each question of the collected questionnaires was analyzed statistically with correlation coefficients of percentage(%) and x^2 (chi-square) inspection. The results are summarized as follows : 1. Most of the housewives (81. 0%) could understand the importance of the budget plan for meal, but they didn't make plans to carry out the meal which could take the food through everyday meal for good health. 2. For the reason that they didn't make a budget, the item, “understanding from experience”obtained 39.6%, and the item, “not forming a custom”obtained 27.2%. 3. Only 18% of them planned menu before the meal; also they didn't try to get several information for the improvement of the meal management through the books. 4. For the reason that they didn't prepare the menu, no formation of custom was 47.7% which was the highest. 5. Among the housewives, 83.3% of them weren't evaluating the menu after the meal. 6. As the basis of selecting a purchase place, the consideration of quality and price was high as 75.6% and for the method of purchase, the most common buying behavior occured while walking and looking around the market stores without the plan was 77.0%. 7. The kitchen structure as well as the sewage system has been unimproved. 8. The majority of the housewives understood the defectable nutrients through the meal but only the minority of them have been practising.

      • Inulin을 분해하는 한 박테리아의 특성과 그 효소에 관한 연구 : 제1보:균의 분리 및 그 효소의 성질 PartⅠ: Bacteria Isolation and the properties of Inulase

        A bacterium isolated from the laboratory was found to produce a strong inulin-hydrolyzing enzyme. Using this bacterium, the several characteristics of inulin-hydrolyzing enzyme in the culture filtrate were studied. The shape of becterium was rod type and the Gram straining was positive. Compared to the Kluyveromyces fragilis, a yeast which produces inulase in large amount, the isolated bacterium had a capacity of lower enzyme production. However, if the culture condition of the bacterium would be improved, inulase activity will be higher than that in the previous experiments. Surprisingly, the isolated bacterium produced inulase in large amount in other carbohydrate sources such as glucose and sucrose. The ratio of enzyme activity in two substrates, inulin and sucrose, was 3.7 to 1. These results show that the properties of inulase produced from this bacterium differ from those of previous studied microorganisms in enzyme action mode.

        中學校 家政科 學習指導의 形態에 따른 學習成果에 관한 연구 : 식생활 단원을 중심으로 Focused on the Dietary Life Unit

        The purpose of this study is to survey the learner's need achievement, interest and practical level of the learning objectives according to the teacher's teaching method in home economics of middle school(Dietary Life Unit), to examine the differences according to the local situation, and to get some information to improve the teaching method. This study surveyed the eight units of dietary life with 459 students in the rural community (224 students) and Seoul (235 students). The collected data were analyzed by T-test, F-test. To sum up this study are as follows: 1. When the need, achievement, interest and practical level of unit "food" are compared the students of seoul with those of rural community, the student of Seoul show high in "The use of Processed Foodstuffs" and low in "Cooking the Processed Foodstuffs" The student of rural community show in "A kind of Cooking Method" and low in "Environment and Food Life" 2. The need, achievement, interest, practical level of the whole unit in rural community is higher than those in Seoul. 3. According to teaching method, comparison Seoul with rural community is revealed as follows. Seoul region is revealed significance to discovery learning in "Cooking Foodstuffs(The achievement and interest level) and "Environment and Food Life"(interest level). Rural community is revealed significance to discussion learning in "The constituent and Food Life"(The need level, interest level). Rural community is revealed significance to explanation learning in "Environment and Food Life"(The achievement level).

        운동 및 고지방식이가 흰쥐의 Carnitine농도와 Carnitine Palmitoyltransferase - I mRNA 수준에 미치는 영향

        본 연구는 고지방식이, 지구력 운동 및 이들의 복합 상태가 혈중 및 간장 중의 카르니틴 함량 및 carnitine palmitoyltransferase-I(CPT-I) mRNA수준에 미치는 영향을 조사하고자 하였다. SD계 흰쥐를 대상으로 정상식이군과 고지방식이군(corn oil 35%)으로 나누고, 다시 각 식이군을 운동군과 비운동군으로 나누었다. 31일동안의 실험기간 동안 각 식이군 중의 운동군은 하루 90분 동안 treadmill을 이용하여 10℃경사에서 0.9㎞/hr 조건에서 훈련시켜, 혈중 및 간조직 중의 카르니틴 농도와 간조직 중의 CPT-I mRNA수준을 비교분석하였다. 1) 정상식이군에서는 운동이 비운동군과 비교시 에너지 효율 및 체중증가에 유의성이 없었지만, 고지방식이군에서는 운동군이 체중증가율 및 에너지 효율이 유의적으로 낮았다. 2) 정상식이군에 있어 혈중 ASAC 및 AIAC 농도가 운동군이 비운동군과 비교시 유의하게 높았으나, 고지방식이군에서는 ASAC는 차이가 없었으며, AIAC는 운동군이 낮은 값을 보였다. 3) Northern blot을 통한 CPT-I mRNA수준은 정상식이군은 운동군이 비운동군에 비해 월등히 높게 나타났으나, 고지방식이군에서는 그 차이가 미미했다. 또한 정상식이군에 비해 고지방식이군이 높은 CPT-I mRNA수준을 보였다. 이상의 결과들은 운동 및 고지방식이는 미토콘드리아 내에서의 지방산의 산화를 증가시키며, 이는 체내의 카르니틴 대사와 CPT-I 효소의 transcription level에서의 조절에 의한 것임을 제안해준다. The effect of exercise and/or high-fat diet on carnitine status and carnitine palmitoyltransferase-I(CPT-I) level were investigated in Weanling Sprague-Dawley rats. The rats were fed an AIN-76 diet or a modified high-fat AIN diet, supplemented with 35% corn oil, for 31 days. During the 31-day period half of the animals in each dietary group were exercised on a treadmill for 90 minutes per day. Carnitine concentrations were determined in plasma and liver and CPT-I mRNA levels were measured by Northern blot analysis with CPT-I cDNA probe in livers of rats. Exercise rats gained less weight than non-exercised rats during the study for high fat diet group. Exercise rats had a higher plasma acid-soluble acylcarnitine and acid insoluble acylcarnitine concnetrations than non-exercised rats for normal diet group. Exercise or high fat diet increased liver carnitine concentration, but a mixed effect was not shown. In exercised rats, CPT-I mRNA levels increased significantly relative to those of nonexercised rats. CPT-I mRNA levels also increased when compared high-fat fed rats with those of normal diet fed rats. These data suggest that there is a correlation between carnitine concentrations and CPT-I mRNA levels and that CPT-I can be regulated at the transcriptional level by exercise and/or high fat diet.

      • 東醫寶鑑에 나타난 食餌療法의 考察(II)

        This study was carried out to investigate the difference between the prescription for dietery management of renal diseases written in Dong Eui Bo Gam and that using in the modern medicine sitologically, and the following results were obtained. 1. As for the salt intake, salt-containing food has been absolute contraindciation for the patient of renal disease in the oriental medicine, while a negligible amount of salt can be afforded to the patient of good status in the modern medicine. These facts imply that salt is very hazardous for the renal diseases. 2. As for the protein intake, protein below 100g per day has been recommended in the oriental medicine. This amount of protein is not different from the daily amount taken by the healthy adult. Moreover, intake of a lot of kidney meat has been recommended in the oriental medicine. This concept came from the idea "Likeness helps Likeness". On the other hand, protein intake is strictly restricted to the pateint of the renal disease in the modern medicine. Daily amount of protein allowable to the patient is 0.2g per kg of body weight according to the patient's status. 2. The recommended foods for the patient of the renal disease listed in Dong Eui Bo Gam and in the food exchange table of the modern medicine are all of good qualities. Those have been recommended with the results from the long clinical experiences in the oriental medicine, and with the data obtained by the modern scientific experimental methods in modern medicine: i.e.,those foods which increase waste products are prohibited for uremic patient in the modern medicine.
