        MR-10 Enhances Men's Health by Improving Endogenous Male Sex Hormone Generation

        Although there is a clear need for improving men's health, treatment with suitable natural substances has not yet been well established. Previously, it was reported that MR-10, a novel complex of Korean dandelion and rooibos found by screening many natural products, improved sperm generation and activity. Here, the ability of MR-10 to increase testosterone levels and enhance men's health was tested. Treatment with MR-10 (400 mg/day) for a month significantly increased levels of free testosterone, total testosterone, and the testosterone precursor dehydroepiandrosterone by 22%, 14%, and 32%, respectively, in clinical studies. Also, men's health in terms of mental, physical, and sexual aspects, as determined by using the clinical questionnaires Androgen Deficiency of Aging Men and Aging Males' Symptoms, was improved. Furthermore, the safety of MR-10 was determined by testing levels of prostate-specific antigen, glutamic oxaloacetic transaminase, and glutamic pyruvate transaminase; and the lack of changes due to MR-10 treatment supports the safety of MR-10. In conclusion, this study suggests that MR-10 is a safe and effective natural product improving men's sexual health.

        Dynamics of a Bose-Einstein Condensate on Changing Speeds of an Atomchip Trap Potential

        We report experimental behaviors of condensed 87Rb atoms responding to changes in the trap potential of the atomchip. The two-types of adiabatic and non-adiabatic overall changes were implemented by changing the ramp-down speed of the chip-wire current, which can dominantly modify the one-axis magnetic field gradient. Under the adiabatic process, a pure condensate stayed in the initial spin state and collectively oscillated with both monopole and dipole modes, while an atomic cloud above the critical temperature exhibited sound waves in a dense ultracold gas. On the other hand, Bose-Einstein condensate atoms with non-adiabatic perturbation were split into spatially different positions by spin states through spin-flip. We investigated the split ratio among spin states depending on final evaporation frequency. Potential changes, of course, cause collective oscillations regardless of the changing process.

      • Improvement of Attention, Long-term and Short-term Memory by Brain Factor-7™ (BF-7™)

        Brain Factor-7™ (BF-7™) is a mixture of silk peptides obtained from hydrolyzed fibroin of the cocoon shell of Bombyx mori, which was developed by Famenity Co., LTD and approved as a health functional food by The Korea Ministry of Food and Drug safety. Several previous clinical studies showed that BF-7™ enhanced the learning and cognitive function in various age groups. In the present study, a clinical study was performed to assess whether BF-7™ enhances short-term, long-term memory and attention on 28 college students who were given 400 mg of BF-7™ orally once a day for 4 weeks. The memory was evaluated by Korean version of Memory Assessment Scales (K-MAS) and attention was by encephalogram of P300 wave. The results were as follows. Short-term verbal memory, short-term visual memory, long-term verbal memory and long-term word memory improved by 47.2, 42.2, 54.8 and 22.3%, respectively. The encephalogram of P300 wave showed that the attention level was enhanced significantly but with less stress. In the in vitro studies on SHSY-5Y cells (a neuronal cell line), BF-7™ were shown to prevent the toxic effects of Aβ1-42 on these cells such as the decrease of cell viability, apoptosis, decrease of mitochondrial membrane potential and generation of reactive oxygen species. These results suggest that BF-7™ enhances both short- and long-term memory as well as attention level and prevents harmful actions of Aβ that affects function and health of the brain.

        Capsaicin-Induced Apoptosis of FaDu Human Pharyngeal Squamous Carcinoma Cells

        Purpose: To investigate the anti-tumor effect of capsaicin on human pharyngeal squamous carcinoma cells (FaDu). Materials and Methods: The expression of apoptosis/cell cycle-related proteins (or genes) was examined by reverse transcriptase-polymerase chain reaction, western blotting and ELISA methods, while the apoptotic cell population, cell morphology and DNA fragmentation levels were assessed using flow cytometry, fluorescence microscopy and agarose gel electrophoresis. Results: Capsaicin was found to inhibit the growth and proliferation of FaDu cells in a dose- and time-dependent manner. Apoptotic cell death was confirmed by observing increases in nuclear condensation, nuclear DNA fragmentation and sub-G1 DNA content. The observed increase in cytosolic cytochrome c, activation of caspase 3 and PARP (p85) levels following capsaicin treatment indicated that the apoptotic response was mitochondrial pathway-dependent. Gene/protein expression analysis of Bcl-2, Bad and Bax further revealed decreased anti-apoptotic Bcl-2 protein and increased pro-apoptotic Bad/Bax expression. Furthermore, capsaicin suppressed the cell cycle progression at the G1/S phase in FaDu cells by decreasing the expression of the regulators of cyclin B1 and D1, as well as cyclin-dependent protein kinases cdk-1, cdk-2 and cdk-4. Conclusion: Our current data show that capsaicin induces apoptosis in FaDu cells and this response is associated with mitochondrial pathways, possibly by mediating cell cycle arrest at G1/S.

        Baculovirus titration method based on MOI values for optimizing recombinant protein expression of the anti‐cancer vaccine candidate GA733‐Fc using Sf9 insect cells

        The baculovirus expression system has been considered as a highly efficient tool for the production of recombinant biopharmaceutical proteins. The recombinant antigenic glycoprotein GA733 is a cell surface protein that is strongly expressed in human colorectal cancer. Efficient virus titration should be established to achieve optimal multiplicity of infection (MOI) conditions, which are in turn essential for strong expression of the recombinant GA733 fused to the human immunoglobulin IgG Fc fragment (GA733‐Fc) in the baculovirus‐insect system. In the present study, the Sf9 cell line was transfected with plasmid DNA containing the GA733‐Fc expression cassette under the control of the baculovirus polyhedron promoter. MOI values (0.05, 0.1, 0.5, 1, and 3) were calculated based on both microscope observations and results of titration assay and then used to determine the optimum recombinant expression and harvested sample [cell culture media (CM) or cell lysate (CL)]. The pFastBac dual vector carrying the GA733‐Fc gene was constructed to express GA733‐Fc and used to generate recombinant baculoviruses. Western blotting results showed that recombinant protein expression was dependent on the MOI. In addition, CM and CL showed significant differences in protein synthesis and protein secretion capacities. Our findings suggested that our proposed titration method can be used for reliable calculation of MOI values, which significantly influence recombinant GA733‐Fc protein expression in the baculovirus‐insect cell system.

        Epigenetic suppression of the anti-aging gene KLOTHO in human prostate cancer cell lines

        KLOTHO was originally identified as an aging-suppressor gene that causes a human aging-like phenotype when tested in KLOTHO-deficient-mice. Recent evidence suggests that KLOTHO functions as a tumor suppressor by inhibiting Wnt signaling. KLOTHO gene silencing, including DNA methylation, has been observed in some human cancers. Aberrant activation of Wnt signaling plays a significant role in aging, and its silencing may be related to prostate cancer and other types of cancers. Thus, we investigated whether the expression of the anti-aging gene KLOTHO was associated with epigenetic changes in prostate cancer cell lines. KLOTHO mRNA was detected in the 22Rv1 cell line while it was not detected in DU145 and PC-3 cell lines. The restoration of KLOTHO mRNA in the DU145 and PC-3 cell lines was induced with a DNA methyltransferase inhibitor. Methylation-specific PCR was performed to determine the specific CpG sites in the KLOTHO promoter responsible for expression. In addition, the level of methylation was assessed in each CpG by performing bisulfite sequencing and quantitative pyrosequencing analysis. The results suggested a remarkable inverse relationship between KLOTHO expression and promoter methylation in prostate cancer cell lines.

        본 논문은 hypercube의 재귀적으로 구성되는 성질을 이용하여 연구도니 subcube를 이용하는 고장감내 라우팅 알고리즘을 유사한 성질을 가지는 k-ary n-cube에 어떻게 적용할 수 있는가를 연구하였다. n-cube에 기초한 2n-2개의 고장을 허용하는 dimension cutting 알고리즘과 k-ary에 기초한 2 개의 고장을 허용하는 grouping 알고리즘을 보였다.
