In the gingival tissues of patients with periodontitis, inflammatoryresponses are mediated by a wide variety of genes. In our previous screening study, plasminogen activator inhibitortype 1 (PAI-1) mRNA binding protein expression wasincreased in gingiva from periodontitis patients. In this study,we further investigated the signaling pathway involved inPAI-1 expression induced by Porphyromonas gingivalis LPS(Pg LPS) in human gingival fibroblasts (HGF). When HGFswere treated with Pg LPS, both PAI-1 mRNA expression andPAI-1 protein were induced in a dose-dependent manner. Pg LPS induced NF-κB activation and the expressions ofPAI-1 mRNA and protein were suppressed by pretreatingwith a NF-κB inhibitor. Pg LPS also induced ERK, p38, andJNK activation, and Pg LPS-induced PAI-1 expression wasinhibited by ERK/p38/JNK inhibitor pretreatment. In conclusion,Pg LPS induced PAI-1 expression through NF-κBand MAP kinases activation in HGF.
The Relationship between Mitochondria and NLRP3 Inflammasome

Mitochondria participate in various intracellular metabolic pathways such as generating intracellular ATP, synthesizing several essential molecules, regulating calcium homeostasis, and producing the cell’s reactive oxygen species (ROS). Emerging studies have demonstrated newly discovered roles of mitochondria, which participate in the regulation of innate immune responses by modulating NLRP3 inflammasomes. Here, we review the recently proposed pathways to be involved in mitochondria-mediated regulation of inflammasome activation and inflammation: 1) mitochondrial ROS, 2) calcium mobilization, 3) nicotinamide adenine dinucleotide (NAD+) reduction, 4) cardiolipin, 5) mitofusin, 6) mitochondrial DNA, 7) mitochondrial antiviral signaling protein. Furthermore, we highlight the significance of mitophagy as a negative regulator of mitochondrial damage and NLRP3 inflammasome activation, as potentially helpful therapeutic approaches which could potentially address uncontrolled inflammation.
Ginsan Enhances Humoral Antibody Response to Orally Delivered Antigen

There have been several reports describing the capability of ginseng extracts as an adjuvant. In this study,we tested if ginsan, a polysaccharide extracted from Panax ginseng, was effective in enhancing antibody response to orally delivered Salmonella antigen. Methods: Ginsan was treated before oral salmonella antigen administration. Salmonella specific antibody was determined by ELISA. mRNA expression was determined by RT-PCR. Cell migration was determined by confocal microscopy and flow cytometry. COX expression was detected by western blot. Results: Ginsan treatment before oral Salmonella antigen delivery significantly increased both secretory and serum antibody production. Ginsan increased the expression of COX in the Peyer’s patches. Various genes were screened and we found that CCL3 mRNA expression was increased in the Peyer’s patch. Ginsan increased dendritic cells in the Peyer’s patch and newly migrated dendritic cells were mostly found in the subepithelial dome region. When COX inhibitors were treated, the expression of CCL3 was reduced. COX inhibitor also antagonized both the migration of dendritic cells and the humoral immune response against oral Salmonella antigen. Conclusion: Ginsan effectively enhances the humoral immune response to orally delivered antigen, mediated by CCL3 via COX. Ginsan may serve as a potent vaccine suppliment for oral immunization.
장내세균의 발육억제 효과로 정한 Lactobacillus 균주 선정과 Probiotic으로서의 이용가능성

목적 : 본 연구에서는 한국인 소아의 침 및 성인의 질부로부터 분리한 1367개의 유산 생성 균주 중 4가지의 장내 병원성 세균 ; Methicillin-resistant Staphylococcus aureus (MRSA), Listeria monocytogenes, Salmonella typhimurium, Escherichia coli O157에 대해 발육억제 효과를 나타내는 유산 균주를 분리하고 분리된 균주의 Probiotic으로서의 가능성을 조사하고자 하였다. 방법 : 네 가지 병원성 세균을 대상으로 agar spot test와 Catalse test를 시행하여 1367개 균주 중 최종적인 실험대상 균주를 선정하였다. 최종 선정된 유산균과 네 가지 병원성 세균을 혼합 배양하여 병원성 세균증식 억제 효과를 보았다. 또한 최종균주의 probiotic으로서의 가능성을 확인하기 위해 균을 동정하고 내산성 등 몇 가지 특성을 관찰하였다. 결과: Agar spot test 결과 전체의 15.7%에 해당하는 215 균주가 MRSA에 대해 억제 효과를 나타내었으며 215 균주 중 9.8%에 해당하는 21 균주가 L. monocytogenes에도 동시에 억제효과를 나타냈다. 이들 균주 중 가장 탁월한 효과를 보인 Lb 1250을 실험 대상 균주로 선정하였다. Lb (Lactobacillus) 1250과 상술한 병원성 세균을 각각 혼합 배양하면 Lb 1250 존재 하에서 MRSA의 증식은 10⁴배 억제되었으며, L. monocytogenes는 10³배의 억제 효과를 보였다. 그러나 S. typhimurium, E. coli O157에 대해서는 아주 미약한 효과를 나타냈다. Lb 1250의 몇 가지 특성을 조사한 결과 Lactobacillus delbrueckii subsp. delbrueckii로 동정되었으며, 이는 H₂O₂를 잘 생산해 내는 균주이었다. 또한 이 균주는 우유에서 curd를 약하게 생성하며 이 curd의 향은 달콤하고 최종 pH가 4.6까지 감소하였으며, pH 2와 같은 강산의 조건하에서 배양한 결과 2시간 후에도 상당한 생존도를 유지하였다. 결론 : 이상의 결과에서 Lb 1250은 병원성 세균의 성장을 억제함을 관찰하였고, probiotic으로서의 이용가능성을 확인하였다.
Synthesis and Immunosuppressive Activity of Novel Succinylacetone Analogues

This study describes the synthesis of novel enol esters (3) and triketones (4) as analogues of succinylacetone (SA) (Ed- this abbreviation is introduced here based on your use of it in the body of the paper) and the evaluation on the mouse allogeneic mixed lymphocyte reaction (MLR) and the murine model of antigen-induced paw edema formation for immunosuppressive activity. Enol esters (3a-f) were about 2-4 fold more potent than SA in in vitro activity.
Molecular Methods for Studying the Human Microbiota

Vast array of microbes colonize to each anatomical environment of human body. Culture based methods are importantin investigating the microbial structure, but they are extremely biased in their evaluation of microbial diversity byselecting particular population of microbiota. Recent advance in molecular technology has allowed sophisticated analysisof complex human microbiota by culture-independent methods. Here, we will discuss features of tools for humanmicrobiota studies including Roche-454 and Illumina platform. We will also briefly discuss features of some strategiesthat are commonly applied to these platforms including 16S rRNA targeting and shotgun sequencing. New platformssuch as PacBio and Oxford Nanopore are also introduced.
Caffeic acid-conjugated chitosan derivatives and their anti-tumor activity

In this study, we synthesized caffeic acid (CFA)-conjugated chitosan (ChitoCFA) as an anti-cancer compound. CFA was conjugated to the amine groups of chitosan(ChitoCFA) and its chemical composition was confirmedusing 1H nuclear magnetic resonance spectra, which indicatesthat specific peaks of CFA was observed at ChitoCFAconjugates. The anti-cancer effects of CFA and ChitoCFAwere studied using CT26 colorectal carcinoma cells. In thiscytotoxicity study, CFA and ChitoCFA revealed a dosedependentdecrease of cell viability while chitosan hadlower cytotoxicity against tumor cells. CFA and ChitoCFAalso proved an anti-proliferative effect against tumor cells. In comparison with CFA, ChitoCFA may accelerate anapoptosis of tumor cells. Furthermore, ChitoCFA demonstratedgood anti-invasive efficacy at Matrigel invasionassay against tumor cells. We suggest that ChitoCFA is apromising candidate as an anti-cancer compound.
