        Evaluation of Porcine Pancreatic Islets Transplanted in the Kidney Capsules of Diabetic Mice Using a Clinically Approved Superparamagnetic Iron Oxide (SPIO) and a 1.5T MR Scanner

        Objective: To evaluate transplanted porcine pancreatic islets in the kidney capsules of diabetic mice using a clinically approved superparamagnetic iron oxide (SPIO) and a 1.5T MR scanner. Materials and Methods: Various numbers of porcine pancreatic islets labeled with Resovist, a carboxydextran-coated SPIO, were transplanted into the kidney capsules of normal mice and imaged with a 3D FIESTA sequence using a 1.5T clinical MR scanner. Labeled (n = 3) and unlabeled (n = 2) islets were transplanted into the kidney capsules of streptozotocin-induced diabetic mice. Blood glucose levels and MR signal intensities were monitored for 30 days post-transplantation. Results: There were no significant differences in viability or insulin secretion between labeled and unlabeled islets. A strong correlation (r 2 > 0.94) was evident between the number of transplanted islets and T2 relaxation times quantified by MRI. Transplantation with labeled or unlabeled islets helped restore normal sustained glucose levels in diabetic mice, and nephrectomies induced the recurrence of diabetes. The MR signal intensity of labeled pancreatic islets decreased by 80% over 30 days. Conclusion: The transplantation of SPIO-labeled porcine islets into the kidney capsule of diabetic mice allows to restore normal glucose levels, and these islets can be visualized and quantified using a 1.5T clinical MR scanner.       • 백서 대동맥 평활근의 Acetylcholine 이완반응에서 K^+ Channel의 역할

        To investigate whether the endothelium-dependent relaxation by acetylcholine (ACh) is due to the activation of K^+ channel, and what kind of K^+ channel plays an important role in vasorelaxation, the effects of K^+ channel blocker(glibenclamide, tetraethylammonium) were observed on the ACh-, sodium nitroprusside (SNP)- or 8-bromoguanosine 3' 5' -cyclic monophsphate (8-br-cGMP)-induced vasorelaxation in the rat thoracec aortic ring. In the aortic rings precontracted with 10^6 M norepinephrine (NE), ACh (10^9 ~ 10^5 M) and SNP (10^10 ~ 10^6 M) produced a dose-dependent relaxation. The relaxation by ACh was completely inhibited by 1065 M methylene blue (MB), an inhibitor of guanylate cyclase, as well as 10^4 M N^w -nitro-L-arginine methyl ester (L-NAME), a competitive inhibitor of nitric oxide synthase. However, the relaxant response by SNP was decreased only by MB. Glibenclamide (10^5 M) had no significant effect on relaxant response to ACh or SNP. However, TEA (5 mM) inhibited the ACh- or SNP-induced relaxation significantly. 8-br-cGMP-induced relaxation was also inhibited by TEA (5mM), not by glibenclamide. From the above results, it is suggested that the activation of K_Ca channel via cGMP-dependent pathway plays a significant role in the regulation of endothelium-dependent relaxation by ACh.

        Cyclo(his-pro) is a metabolite of thyrotrophin releasing hormone(TRH). It may be involved in the regulation of insulin metabolism and stimulates intestinal zinc absorption. In order to test the real function of cyclo(his-pro), a large amount of enantiomeric pure cyclo(his-pro) is needed. Cis-(L,L)-cyclo(his-pro) and trans-(L,D) -cyclo(his-pro) were successfully synthesized.

      • 한국산 시궁쥐(Rattus norvegicus)와 곰쥐(Rattus rattus)의 mtDNA RFLPs

        RFLPs(restriction fragment length polymorphisms) of mitochondrial DNA(mtDNA) of Rattus norvegicus and Rattus rattus in Korea were observed. All of the mtDNA of R. norvegicus individuals were shown no variatioin by the single digestion with EcoR Ⅰ, Hind Ⅲ, Ksp Ⅰ, Hind Ⅱ and Cfo Ⅰ as well as the double digestion with BamH Ⅰ/Sac Ⅰ or Hpa Ⅰ/Xba Ⅰ. Morph-1(18.75%) and morph-2(81.25%)) were observed by digestion with Hind Ⅱ in mtDNA of R. norvegicus. Morph-1(91.70%) and morph-2(8.30%) were observed by digestion with Cfo Ⅰ in mtDNA of R. norvegicus. In the mtDNA obtained from 3 R. rattus individuals, no variants were observed. The types of mtDNA obtained from 48 R. norvegicus individuals were four types(RN 1, RN 2, RN 3 and RN 4). The frequency of RN 1, RN 2, RN 3 and RN 4 was 66.67%, 14.58%, 6.25% and 12.50%, respectively. The value range of nucleotide diversity in mtDNA of R. norvegicus was between 0.0059 and 0.0153, and the value of nucleotide diversity of mtDNA between R. norvegicus and R. rattus was 0.13. The divergence time between the two species is believed to be 6.5 million years ago.

        『성학십도』에 내재된 인성교육 연구

        내면적 자기수양을 추구하면서 참된 자아를 실현하려는 퇴계의 사상은 오늘날 이 시대에 필요한 인성교육의 내용이자 한국적 인성교육의 표본이 될 수 있다. 본고는 우리나라를 대표하는 학자퇴계의 주저(主著)인 『성학십도』의 체제와 내용을 살펴서 『성학십도』에 내재된 인성교육을 제시하는데 목적이 있다. 『성학십도』의 체제와 내용을 기반으로 `『성학십도』에 내재된 인성교육`을 세 가지 소주제로 구성하였다. 첫 번째 주제는 인성교육의 근본과 교육의 내용으로, 『성학십도』의 제1도부터 제5도까지의 내용이 여기에 해당된다. 제1도와 제2도는 `나는 누구인가『`에 대한 인간의 근원과 표준을 설명하였고 제3도에서 제5도까지는 교육과 연관 있는 『소학』, 『대학』 그리고 `백록동 서원의 규칙`을 통해 `어떻게 살아야 하는가?`에 대한 답으로 학문의 내용을 살펴보았다. 두 번째 주제는 심성교육으로 제6도에서 제8도까지의 내용이 해당된다. 인성교육에 있어 가장 중요한 심성교육은 마음공부로 귀결된다. 사사로운 욕심을 절제하고 공동체의식을 고양하여 사랑의 마음을 확충하고 보존하는 마음의 수양이 필요하다. 세 번째 주제는 실천교육으로 제9도와 제10의 내용이 여기에 해당된다. 『성학십도』 전체를 관통하는 `경(敬)`은 실천덕목이다. `경`은 관념적이고 추상적인 정신 훈련이 아니라 일상생활에서 몸과 마음을 바르게 하는 훈련이며 이 공부는 마음의 안정과 자기주체를 이루게 되어 자신이 주인이 되는 주체적인 삶을 살게 된다. 전체를 종합하면 『성학십도』에 내재된 인성교육은 나에 대한 존재론적 인지와, 삶에 대한 의미와 가치를 인식하여 그 앎을 마음공부와 실천교육인 행(行)으로 연결할 수 있도록 하는 것이다. In the flow of Korean history, begun humanitarianism as founding principle, the Humanistic Education of T`oegye, to seek internal self-discipline and realized true self, can be content of Humanistic Education necessary for this age and specimen of Korean Humanistic Education. This article is to suggest the Humanistic Education inherent in 『The Ten Diagrams of Sage Learning』, by examining the system and content of 『The Ten Diagrams of Sage Learning』, book of T`oegye, scholar representing Korea. Suppose it examines the system of 『The Ten Diagrams of Sage Learning』 and content in structure, it structuralized the sub-themes into three, at `Humanistic Education inherent in 『The Ten Diagrams of Sage Learning』.` The first theme is about the source of humanistic education and content of study, and it applies the contents from 1 diagram to 5 diagram. In diagram 1, 2, it explains the source and standard of humans for `who am I『`, and it examines the 『Sohak』, 『Daehak』, and `Rule of Baekrokdong auditorium` relevant to education about `how do we live』` from diagram 3 to 5. The second theme is the Mind Education, contents from diagram 6 to 8. Mind Education, the most important in Humanistic Education, concluded to mind study. Mind encompasses nature and emotion, and nature is revealed as emotion. By preventing selfish desire, it requires cultivating heart to expand and preserve the mind of `perfect virtue` learned by sense of community. The third theme is the Practice Education, applying the contents from diagram 9 to 10. `Concentrate` is the Practice Education going through the whole 『The Ten Diagrams of Sage Learning』. `Concentrate` is not the ideal and abstract mental training but training to straighten out the body and mind in everyday life. This study makes the stability of mind and self subject, so one can live an independent life which self is the owner. To integrate the whole, Humanistic Education inherent in 『The Ten Diagrams of Sage Learning』 makes realize the realistic realization for myself, meaning and value for life, and it connects the knowing to Practice, learning of mind and practice education.

        단일 혈액검사센터의 Rh 및 기타 항원 음성률 도출 경험

        Background: The demand for Rh-related and other specific antigen negative donations has increased recently, and in response, we need to improve work efficiency while decreasing reagent consumption. Thus, we desire to create a ‘table of Nambu Blood Laboratory Center’s own Rh-related and other antigen negative rate’. Methods: We analyzed the results of Rh-related specific antigen test for 2,806,330 donors using ‘PK-7300’ and the results of manual test for 10,024 other blood type antigen-related specific and compound antigens. We made a table summarizing the results using two cases. Results: The negative rate of Rh-related specific antigens was approximately 12.9% for C, 41.7% for c, 49.5% for E, and 9.2% for e antigens. The negative rate of compound antigens was increased when compared with the number of inspections. The negative rate for C and e antigens was increased from 90.1% to 97.3%, and that for E, c, and JKa was increased from 7.6% to 31.6%. Conclusion: The negative rate table made in July 2016 has been determined to be very effective in screening for specific and compound antigen negative blood. Also it was very efficient in terms of examining and supplying the compound antigen negative blood. It would be helpful to improve the supply of specific antigen negative blood to better meet the unique needs of each medical institution.

      • 고혈압 및 정상혈압백서의 대동맥 평활근 세포에서 ATP-sensitive K^+ Channel에 대한 Cromakalim의 영향

        To determine wherther the properties of ATP-sensitive K^+ channel (K_ATP channel) in aortic smooth muscle cells from spontaneously hypertensive rats are altered of not, the effect of a K^+ channel opener, cromakalim, on membrane currents in aortic smooth muscle cells from spontaneously hypertensive rate (SHR) and Wista-Kyoto rats (WKY) were compared, using a patch-clamp technique. 1. The internal and external K^+ were 140 and 60 mM, respectively. The currents were recorded at -70mV. The inward current was evoked by cromakalim and this current was inhibited by glibenclamide. 2. The cromakalim-evoked current was time-independent and voltage-insensitive. 3. The reversal potential of cromakalim-evoked current depended on external K^+ concentration, and this current was reversed at near K^+ equilibrium potential. 4. The cromakalim-evoked current was reduced when the intracellular ATP concentration was raised from 0.1mM to 3 mM. 5. In SHR, crmakalim-evoked current, normalized by cell capacitance, was not changed when compared to WKY. From the above results, it is suggested that cromakalim-evoked current was carried throuhg K_ATP channel and the function of cromakalim on K_ATP channel in aortic smooth muscle cells of SHR is not impaired when compared to WKY cells
