The study of molecular markers to improve crops largely depends on the availability of rapid and of efficient DNA extraction methods. Here we developed a cheap and convenient method to isolate genomic DNA from rice grains suitable for large-scale microsatellite analysis. We confirmed that the isolated rice DNA is suitable for PCR analysis with STS marker and SNP marker, as well as microsatellite marker. Further, we established high-throughput DNA extraction system in a 96-well plate format which make it possible high-throughput analysis of microsatellite markers with rice grains. This implies that the new method could be a useful tool for other types of marker analysis in large scale.
단순반복염기서열의 변이 형태에 따른 위암 내시경 조직의 유전자형 분류

Purpose: Individual gastric cancers demonstrate complicated genetic alterations. The PCR-based analysis of polymorphic microsatellite sequences on cancer-related chromosomes has been used to detect chromosomal loss and microsatellite instability. For the purpose of preoperative usage, we analyzed the correspondance rate of the microsatellite genotype between endoscopic biopsy and surgical specimens. Materials and Methods: Seventy-three pairs of biopsy and surgical specimens were examined for loss of heterozygosity and microsatellite instability by using 40 microsatellite markers on eight chromosomes. Microsatellite alterations in tumor DNAs were classified into a high-risk group (baselinelevel loss of heterozygosity: 1 chromosomal loss in diffuse type and high-level loss of heterozygosity: 4 or more chromosomal losses) and a low-risk group (microsatellite instability and low-level loss of heterozygosity: 2 or 3 chromosomal losses in diffuse type or $1\∼3$ chromosomal losses in intestinal type) based on the extent of chromosomal loss and microsatellite instability. Results: The chromosomal losses of the biopsy and the surgical specimens were found to be different in 21 of the 73 cases, 19 cases of which were categorized into a genotype group of similar extent. In 100 surgical specimens, the high-risk genotype group showed a high incidence of nodal involvement (19 of 23 cases: $\leq$5 cm; 23 of 24 cases: >5 cm) irrespective of tumor size while the incidence of nodal involvement for the low-risk genotype group depended on tumor size (5 of 26 cases: $\leq$5 cm; 18 of 27 cases: >5 cm). Extraserosal invasion was more frequent in large-sized tumor in both the high-risk genotype group ($\leq$5 cm: 12 of 23 cases; >5 cm: 23 of 24 cases) and the low-risk genotype group ($\leq$5 cm: 7 of 26 cases; >5 cm: 16 of 27 cases). The preoperative prediction of tumor invasion and nodal involvement based on tumor size and genotype corresponded closely to the pathologic tumor stage (ROC area >0.7). Conclusion: An endoscopic biopsy specimen of gastric cancer can be used to make a preoperative genetic diagnosis that accurately reflect the genotype of the corresponding surgical specimen.
인체에 적용한 전기자극이 Serum myoglobin과 Aldolase에 미치는 영향에 대한 생리학적 효과

Prior studies have revealed that several stimulation to the muscle have released serum myoglobin into the blood vessel and increased aldolase activity. The present authors carried out a study which effect of electrical stimulation treatment (induced a isotonic wrist exerceise) on serum myoglobin(Mb) levels and aldolase(Al) activity were investigated in 6 healthy female. There were four groups of female: 1. no electrical stimulation control: 2. electrical stimulation 10min (EST10'); 3. electrical stimulation 20 min (EST20'); 4. electrical stimulation 30min (EST30'). Each groups is all the same one. Radioimmunoassay and Ultraviolet Spectrophotometry were performed to increased or decreased of serum myoglobin and aldolase. Serum myoglobin significantly increased in electrical stimulated groups[EST10' $(30.20{\pm}5.27ng/ml)$, EST20'$(31.65{\pm}3.96ng/ml)$, EST30'$(31.95{\pm}2.0ng/ml)$] to be compared with control group$(24.43{\pm}2.20ng/ml)$. Aldolase significantly increased in electrical stimulated groups [EST10' ($6.85{\pm}1.17$ Sigma U/mL), EST20'($6.70{\pm}1.46$ Sigma U/mL), EST30'($6.56{\pm}1.01$ Sigma U/mL)) to be compared with control group($5.03{\pm}1.86$ Sigma U/mL). The results of this study show that isotonic exercise result in electrical stimulation treatment increased serum myoglobin content and aldolase activity. In conclusion, our results support that stimulation release serum myoglobin and increase aldolase activity.

In this study, it was investigated by comparing the experimental hemming by the 3 steps and 2 steps in order to stabilize the quality of the hemming process. In the experimental results, the three-step hemming superior to the two-step one and the dimensional stability of part that was made by the three-step on was high. When the second stage Hemming has been found that the deflection caused by the force to the wear of the punch becomes larger plane can be folded by the hemming crimping and crimp uncertain.
불안정한 지지면 운동이 여성 노인의 균형과 하지 근활성에 미치는 영향

Purpose The purpose of this study was to determine the effect of unstable surface strength trainingon balance and lower extremity muscle activity in elderly women. Methods Our study included relatively elderly women age of 65 years or so, 14 elderly women who consented were randomized as two groups, the control group of 7 people, the experimental group of 7 people, unstable surface strength training was performed for 40 minutes a day, twice a week, for a total of 8 week. Results The experimental group had a significant difference in OLST_S and OLST_H(p<.05), the control group had a significant difference in OLST_H(p< .05). And the comparison between two groups had no significant difference (p >.05). Two groups had no significant difference in TUG (p>.05), the comparison between groups had no significant difference (p>.05). The experimental group had a significant difference in muscle activity of vastus medialis oblique and vastus lateralis oblique (p<.05), the control group had a significant difference in muscle activity of vastus lateralis oblique (p< .05), but the comparison between groups had no significant difference (p >.05). Conclusion Unstable surface strength training in elderly women were more effective than general strengthening exercises and it is an appropriate exercise to prevent from falling in the elderly.

Steel and PHC composite pile is improved in structural performance combining steel pipe pile with PHC pile by connecting coupler, which are resisted the shear force and bending moment by earthquake load. This paper presented the evaluation of structural performance and field applicability of steel and PHC composite pile combined connecting coupler. The connecting coupler in steel and PHC pile are well performed to distribute the applied loading and combine performance. Experimental and analytical work was planned to evaluate the load carrying capacity in composite pile. And also field test was performed with dynamic and lateral loading applied. Using the results obtain from dynamic loading test and lateral loading test on real structures, the behavior of composite pile is almost the same to the behavior of steel pile. According to considering the characteristics of structures and ground condition, it is judged that the presented composite pile can be one of the best solutions by introducing both steel pile which has a benefit of excellent flexural stiffness and construction efficiency in case of applying proper connection position.
노인의 장악력(grip strength)과 손 활동, 질환유무 및 식품섭취 빈도에 따른 상관관계 연구

The purpose of this study is to understand how the handgrip strength affects the total hand activities, presence of illness, and food intake frequency. We have chosen two hundreds and twenty-four elderly people in each welfare center(Seoul Shin-Jung welfare center, Welfare center for elders in two cities of Annyang & Kunpo, and the elders home in Ansan city) from July to September in the year of 2000. The results were as followed: 1. There were statistically significant correlation in the effects of between the hand grip strength, as we have studied, and the total hand activities such as writing, buttoning of clothes, holding a book while reading, opening of jars, doing housework, carrying of grocery bag, bathing and dressing using chopsticks. However, it showed no signs of correlation on gripping of a telephone handle. 2. The study of presence of illness showed lots of statistical signs of correlation in the effects of between handgrip strength and arthritis but significant differences in hypertension, diabetic, stroke, and depression. 3. The effects of handgrip strength showed lots of statistical signs of correlation with meat, dairy, and fruits intake but not with vegetable.
