        방사성 동위원소옥소(131I)에 의한 갑상선질환의 임상적 연구

        서울대학교 의과대학 내과학교실 및 방사성 동위원소 진료실에서 1960년 5월부터 1969년 10월까지 진료한 2,658명의 각종 갑상선 질환 환자에 대하여 131I에 의한 각종 갑상선 기능 검사 및 기능 항진증 환자에 대한 131I의 치료 성적을 종합 검토하여 아래와 같은 결론을 얻었다. 1) 2,658명의 갑상선 질환 환자중 독성 미만성 선종이 929명(34.9%)으로 가장 많고 비중독성 미만성 선종 및 비중독성 결절성 선종이 각각 762명(28.7%), 699명(26.3%)이며 기능저하가 210명(7.9%), 독성 결절성 선종이 58명(2.2%)였다. 2) 갑상선 질환의 성별 발생 빈도는 남자 300명(11.4%), 여자 2,358명(88.6%)로서 그 비는 1:8였다. 3) 연령별 발생 빈도는 20∼49세에서 전체의 79.1%인 2,102명이며 기능 항진증의 경우는 79.0%에 달하였다. 4) 각종 갑상선 기능 검사중 131I 섭취율, 131I 혈청내 방사능 BMR치등에 대한 고찰을 하는 한편 기능항진 및 저하증때 나타내는 각종 자학증세를 관찰하였다. 5) 갑상선 기능 항진증 환자 867명에 대하여 131I 치료를 하고 그 중 579명에서 47.8%의 초회 치료율을 확인하였다. 6) 131I 투여 후의 합병증인 기능 저하증의 발생 빈도는 초회 투여에서 6.75%였다. 7) 갑상선의 $quot; A summary of the clinical data of the (131)^I-thyroid function tests and the therapeutic results of 1(31I)^ among the 2,658 patients of various thyroid diseases treated over the past 10 years from May 1960 to Oct. 1969 at the Radioisotope Clinci and Laboratory, SNUH were presented and discussed. 1) The patients examined consisted of 929 cases (34.9%) of diffuse toxic goiter, 762 cases (28.7%) of diffuse nontoxic goiter, 699 cases (26.3%) of nodular nontoxic goiter, 58 cases (2.2%) of nodular toxic goiter and 210 cases (7.9%) of hypothyroidism. 2) There were 300 (11.4%) male and 2358 (88.6%) female, showing a ratio of 1:8. 3) The majority of patients (79.1%) were in the 3rd-5th decade of their lives. 4) The normal ranges, diagnostic values of (131)^I uptake test, 48 hrs serum activity, BMR and main subjective symptoms of various thyroid diseases were discussed. 5) In the 579 patients among 867 cases with hyperthyroidism treated with (131)^I, 47.8% were confirmed to be cured completely after single therapeutic doses. 6) The complications of 131I therapy were discussed and myxedema had developed in 6.75% of our patients. 7) The results of (131)^I thyroid function tests were analysed among the 160 cases of thyroid diseases which were confirmed the diagnosis with histopathological measures.

        운동후 측복통과 함께 발생한 급성 신부전 1 예

        We report a case of acute renal failure with severe loin pain and patchy renal vasoconstriction after exercise. An immediate postcontrast computed tomogram of this case shows multiple patchy areas of poor contrast enhancement in the cortex of the both kidneys. A 24-hours delayed scan without further injection of contrast dye shows multiple wedge-shaped contrast enhancement in the coincident areas of poor enhancement in the immediate scan. The 2 and 5 days delayed scans show that wedge-shaped contrast enhancement becomes disappeared gradually to the renal cortex, and the 8 days delayed scan shows complete disappearance of contrast enhancement. The renal scintigrams with technetium-99m-methylene diphosphonate and dimercaptosuccinic acid show diffuse increased uptakes and multiple patchy photon deficient areas in the both kidneys respectively. The follow-up renal scintigrams show nearly normal findings, This patient had an uneventful hospital course and recovered completely without dialysis.

        Syngas production by combined steam and CO<sub>2</sub> reforming of coke oven gas over highly sinter-stable La-promoted Ni/MgAl<sub>2</sub>O<sub>4</sub> catalyst

        Highly sinter-stable 10 wt% Ni-xLa/MgAl<SUB>2</SUB>O<SUB>4</SUB> (x = 0-5 wt%) catalysts were prepared by co-impregnation for the combined steam and CO<SUB>2</SUB> reforming (CSCR) of coke oven gas (COG). The physicochemical properties of the catalysts were analyzed by BET, XRD, H<SUB>2</SUB> chemisorption, and H<SUB>2</SUB>-TPR. To compare the sinterstabilities, aging treatment was performed at 900 <SUP>o</SUP>C under H<SUB>2</SUB>:H<SUB>2</SUB>O:N<SUB>2</SUB> = 1:10:1.25 for 50 h. Although the Ni dispersions decrease and Ni crystallite sizes increase for all of catalysts after the aging treatment, the La-promoted catalysts had higher Ni dispersion and smaller crystallite sizes than the Ni/MgAl<SUB>2</SUB>O<SUB>4</SUB> catalyst owing to the enhancement of strong metal-support interactions. The catalytic test was carried out under CH<SUB>4</SUB>:H<SUB>2</SUB>O:CO<SUB>2</SUB>:H<SUB>2</SUB>:CO:N<SUB>2</SUB> = 1:1.2:0.4:2:0.3:0.3 at 900 <SUP>o</SUP>C and 5 atm for 40 h. The Ni-2.5La/MgAl<SUB>2</SUB>O<SUB>4</SUB> (aged) catalyst exhibited the highest activity and sinterstability owing to its high surface area and Ni dispersion. It was also confirmed that La promotion prevents the agglomeration of Ni particles through TEM.

      • Sampled-data H<sub>~</sub> fuzzy filtering for nonlinear systems with missing measurements

        <P>In this paper, a sampled-data H-infinity fuzzy filtering problem is considered for nonlinear systems with missing measurements. The nonlinear sampled-data system and missing measurements are assumed to be represented by a Takagi-Sugeno (T-S) fuzzy system and an independent, identically distributed Bernoulli random process, respectively. Based on the fuzzy system, the H-infinity fuzzy filtering problem is formulated to design the sampled-data fuzzy filter. By using the exponential mean-square stability definition, the stability condition with an H-infinity performance is guaranteed for the fuzzy system with the sampled-data fuzzy filter, and its sufficient condition is converted into the linear matrix inequality (LMI) format.         Prereforming of n-tetradecane over Ce-promoted 50 wt% Ni/MgO–Al2O3 catalyst with high coke resistance

        <P>The effect of Ce-promotion on 50 wt% Ni-based catalysts during the prereforming of n-tetradecane and its optimum content were investigated. The Ni catalyst was synthesized by deposition-precipitation method. Next, various amounts of Ce (0-13 wt%) were loaded on the Ni catalyst by impregnation. The characteristics of the prepared catalysts were analyzed by XRD, H-2-TPR, BET, BJH, and H-2-chemisorption analyses. The prepared catalysts were tested under the prereforming conditions (temperature = 400 A degrees C, GHSV = 3000 h(-1), and S/C = 3 and 4). The Ni catalyst was easily deactivated under the following conditions: temperature = 400 A degrees C, GHSV = 3000 h(-1), and S/C = 4. The stability of all Ce-promoted Ni catalysts was improved as compared to that of the Ni catalyst. Among the Ce-promoted catalysts, 5 wt% Ce/50 wt% Ni/MgO-Al2O3 catalyst showed excellent stability even under the severe condition of S/C = 3. SEM, TEM, and TG analyses were performed in order to identify the main factor responsible for the rapid deactivation of the Ni catalyst. In the case of 0Ce/50Ni, Ni particles were encapsulated by many folds of coke and it was related to the rapid catalyst deactivation. However, after Ce promoted on the Ni catalyst, the thickness of the coke layers and the number of encapsulated Ni particles decreased and the deposited amount of coke on the catalyst also decreased.</P>

        Pre-reforming of n-tetradecane over Ni/MgO–Al2O3 catalyst: effect of added potassium on the coke resistance

        <P>We investigated the pre-reforming of n-tetradecane (C14H30) over the K-promoted 50 wt% Ni/MgO-Al2O3 catalyst and also analysed the coke resistance. The 50 wt% Ni/MgO-Al2O3 catalysts were prepared by the deposition-precipitation method and 1-5 wt% of potassium was added to the catalyst using the impregnation method. The catalysts were characterized by ICP, XRD, H-2-chemisorption, H-2-TPR, and BET. The pre-reforming of n-tetradecane was performed under the reaction conditions of S/C = 4.0, GHSV = 3000 h(-1), and 400-450 A degrees C. The Ni crystallite size increased, whereas the Ni dispersion and surface area of catalysts decreased with increasing amounts of K. The 50 wt% Ni/MgO-Al2O3 showed large amounts of coke deposition compared to the K-promoted catalysts. It was found that the K addition over 50 wt% Ni/MgO-Al2O3 catalyst was beneficial for coke removal by promoting the coke gasification. A 1 wt%K/50 wt% Ni/MgO-Al2O3 catalyst showed stable activity during pre-reforming at S/C = 4.0, GHSV = 3000 h(-1), and 400 A degrees C. Furthermore, it had the lowest amount of surface coke, as observed by SEM, TEM, and TG analysis.</P>

        <P>Tulobuterol is a beta(2)-adrenergic agonist that was the first bronchodilator approved as a transdermal patch for humans. Previous studies have examined the pharmacokinetics of tulobuterol in humans but not in the veterinary species. In this study, the pharmacokinetics of tulobuterol was examined in healthy Beagle dogs after transdermal and intravenous administration. The C-max was 2.09 ng/mL at 16.0 h for a 0.2 mg/kg patch and 4.85 ng/mL at 13.6 h for a 0.4 mg/kg patch. The effective blood level in humans is 1-3 ng/mL, a concentration achieved using the 0.2 mg/kg patch in dogs. In conclusion, application of a 0.2 mg/kg tulobuterol patch to healthy dogs led to an apparently effective blood concentration for 24 h. (C) 2015 Elsevier Ltd. All rights reserved.</P>

      • Roles of AKT1 and AKT2 in non-small cell lung cancer cell survival, growth, and migration.

        <P>Although AKT ? protein kinase B is constitutively active in nonsmall cell lung cancer (NSCLC) cells and is an attractive target for enhancing the cytotoxicity of therapeutic agents, the distinct roles of the AKT isoforms in NSCLC are largely unknown. In the present study, we investigated the roles of AKT1 and AKT2 in NSCLC cells using RNAi. The siRNA targeting of AKT1 or AKT2 effectively decreased protein levels of AKT1 and AKT2, respectively, in A549 and H460 cells. Cisplatin treatment of these cells increased apoptotic cell death compared with control. The siRNA-induced knockdown of AKT1 in H460 cells significantly decreased basal MEK? ERK1 ? 2 activity, resulting in nuclear factor-κB activation, whereas knockdown of AKT2 resulted in anti-apoptotic Bcl-2 family protein MCL-1 (MCL-1) cleavage, the collapse of mitochondrial membrane potential, cytochrome c release, and activation of the caspase cascade. Consequently, both siRNA treatments enhanced the chemosensitivity of H460 cells to cisplatin. However, neither AKT1 nor AKT2 siRNA treatment had any effect of p27 expression, and although both treatments tended to induced G2 ?M phase arrest, the effect was not statistically significant. Treatment with AKT1 siRNA markedly decreased colony formation growth and migration, but AKT2 siRNA had no significant effects on these parameters. These data suggest that AKT1 and AKT2 both contribute to cell survival, albeit via different mechanisms, and that the effects on cell growth and migration are predominantly regulated by AKT1. These findings may aid in refining targeted strategies for the inhibition of AKT isoforms towards the sensitization of NSCLC cells to therapeutic agents.</P>

        Mucosally administered Lactobacillus surface-displayed influenza antigens (sM2 and HA2) with cholera toxin subunit A1 (CTA1) Induce broadly protective immune responses against divergent influenza subtypes

        The development of a universal influenza vaccine that provides broad cross protection against existing and unforeseen influenza viruses is a critical challenge. In this study, we constructed and expressed conserved sM2 and HA2 influenza antigens with cholera toxin subunit A1 (CTA1) on the surface of Lactobacillus casei (pgsA-CTA1sM2HA2/L. casei). Oral and nasal administrations of recombinant L. casei into mice resulted in high levels of serum immunoglobulin G (IgG) and their isotypes (IgG1 & IgG2a) as well as mucosal IgA. The mucosal administration of pgsA-CTA1sM2HA2/L. casei may also significantly increase the levels of sM2- or HA2-specific cell-mediated immunity because increased release of both IFN-γ and IL-4 was observed. The recombinant pgsA-CTA1sM2HA2/L. casei provided better protection of BALB/c mice against 10 times the 50% mouse lethal doses (MLD<SUB>50</SUB>) of homologous A/EM/Korea/W149/06(H5N1) or A/Aquatic bird/Korea/W8½005 (H5N2) and heterologous A/Puerto Rico/8/34(H1N1), or A/Chicken/Korea/116/2004(H9N2) or A/Philippines/2/08(H3N2) viruses, compared with L. casei harboring sM2HA2 and also the protection was maintained up to seven months after administration. These results indicate that recombinant L. casei expressing the highly conserved sM2, HA2 of influenza and CTA1 as a mucosal adjuvant could be a potential mucosal vaccine candidate or tool to protect against divergent influenza viruses for human and animal.

        A biosynthetic pathway for hexanoic acid production in Kluyveromyces marxianus

        Hexanoic acid can be used for diverse industrial applications and is a precursor for fine chemistry. Although some natural microorganisms have been screened and evolved to produce hexanoic acid, the construction of an engineered biosynthetic pathway for producing hexanoic acid in yeast has not been reported. Here we constructed hexanoic acid pathways in Kluyveromyces marxianus by integrating 5 combinations of seven genes (AtoB, BktB, Crt, Hbd, MCT1, Ter, and TES1), by which random chromosomal sites of the strain are overwritten by the new genes from bacteria and yeast. One recombinant strain, H4A, which contained AtoB, BktB, Crt, Hbd, and Ter, produced 154mg/L of hexanoic acid from galactose as the sole substrate. However, the hexanoic acid produced by the H4A strain was re-assimilated during the fermentation due to the reverse activity of AtoB, which condenses two acetyl-CoAs into a single acetoacetyl-CoA. This product instability could be overcome by the replacement of AtoB with a malonyl CoA-acyl carrier protein transacylase (MCT1) from Saccharomyces cerevisiae. Our results suggest that Mct1 provides a slow but stable acetyl-CoA chain elongation pathway, whereas the AtoB-mediated route is fast but unstable. In conclusion, hexanoic acid was produced for the first time in yeast by the construction of chain elongation pathways comprising 5-7 genes in K. marxianus.
