      • Estrogen Receptor Alpha Gene Polymorphisms and Breast Cancer Risk: a Case-control Study with Meta-analysis Combined

        Molecular epidemiological studies have shown that gene polymorphisms of estrogen receptor alpha gene (ESR-${\alpha}$) are associated with breast cancer risk. However, previous results from many molecular studies have been inconsistent. In this study, we examined two polymorphisms (PvuII and XbaI RFLPs) of the ESR-${\alpha}$ gene in 542 breast cancer cases and 1,016 controls from China. Associations between the polymorphisms and breast cancer risk were calculated with an unconditional logistic regression model. Linkage disequilibrium and haplotypes were analyzed with the SHEsis software. In addition, we also performed a systematic meta-analysis of 24 published studies evaluating the association. No significant associations were found between the PvuII polymorphism and breast cancer risk. However, a significantly decreased risk of breast cancer was observed among carriers of the XbaI 'G' allele (age-adjusted OR = 0.80; 95% CI = 0.66- 0.97) compared with carriers of the 'A' allele. Haplotype analysis showed significantly decreased cancer risk for carriers of the 'CG' haplotype (OR = 0.79; 95% CI = 0.66- 0.96). In the systematic meta-analysis, the XbaI 'G' allele was associated with an overall significantly decreased risk of breast cancer (OR = 0.90, 95% CI = 0.82- 1.00). In addition, the PvuII 'C' allele showed a 0.96- fold decreased disease risk (95% CI = 0.92- 0.99). In subgroup analysis, an association between the PvuII 'C' and XbaI 'G' alleles and breast cancer risk was significant in Asians ('C' vs. 'T': OR = 0.93, 95% CI = 0.85- 1.00; 'G' vs. 'A': OR = 0.82, 95% CI = 0.68- 0.98), but not in Euro-Americans. Thus, our results provide evidence that ESR-${\alpha}$ polymorphisms are associated with susceptibility to breast cancer. These associations may largely depend on population characteristics and geographic location.

        Dynamic Characteristics of Saturated Soft Clays under Cyclic Loading in Drained Condition

        This study presents an experimental investigation on the effects of cyclic loading waveform and cyclic stress ratio (CSR) on dynamic behaviors through laboratory tests on a source of soft clay under cyclic loading in drained condition. Test results showed that under different cyclic loading waveforms at a given frequency (f = 1 Hz) and CSR = 0.6, the axial strain increases significantly at a relatively lower number of cycles (N) and then became stable, while the axial strain consistently decreases with the increasing N and tend to be stable. At a given N, specimen under sine wave cyclic loads shows a higher axial strain rate than that under triangle wave or square cyclic loads. Test results also indicated that at different CSRs, the axial strain increases rapidly at a relatively lower N and then gradually tends to be stable with the increasing N at lower CSR yet there is no stable trend for specimens at higher CSR, which indicates there is a critical value of CSR. It is recommended to apply the cyclic loading in sine waveform with CSR close to and less than the critical value to get a better treatment effect.

        Growth factors for silver nanoplates formed in a simple solvothermal process.

        <P>We synthesized both big and ultra-small silver nanoplates through a simple solvothermal process in a common autoclave. Using this approach, we achieved the reduction of silver nitrate in DMF or ethanol in the presence of PVP as a capping agent. The reduction capacity of PVP was also investigated. Growth factors such as the concentration of silver precursor, quantity of capping agent, reaction time, and temperature have been shown to play important roles in the formation of different sizes and shapes of silver nanoplates. The big plates, over 100 nm in edge length, were found mainly in the shape of a connected triangular. Single triangle plates, truncated triangular plates, and hexangular plates were also found under different reaction conditions. The ultra-small silver nanoplates, 20-100 nm in edge length, were successfully extracted from the solution under different centrifugation conditions. The UV-vis spectrum exhibited intense in-plane dipole absorption peaks varying from 470-630 nm, as reflected by the gradual color change of the solution from orange to red, and finally to blue.</P>

      • Development of a magnified flexure mechanism for fast tool servo system

        The flexure-hinge mechanisms are commonly used in the design of translational micro/nano-positioning stage. This paper presents the methodology for design and control of a lever-type magnified flexure mechanism used for the ultra-precision fast tool servo (FTS) system. A lever type hinge mechanism is designed and utilized to guide the tool stage and to preload the PZT actuator. A prototype of the FTS system was developed and tested. The influence of tile PZT actuator's capacitance on the output voltage of the given amplifier was investigated. The motion range of the FTS system can reach 11.3㎛ and a bandwidth of 600㎐, and the amplification of the lever flexure mechanism is approximately 5 as calculated from the experiment.

        An Improved CPTu-based Method to Estimate Jacked Pile Bearing Capacity and Its Reliability Assessment

        This paper presents an improved method for predicting the bearing capacity of single jacked pile based on piezocone penetration test (CPTu) data which contain tip resistance (qt), side friction ( fs) and excess dynamic pore water pressure (u2). Firstly, the average value of qt was determined to calculate the pile unit base resistance according to the influence zone of the logarithmic spiral concerning the effect of soil types. Secondly, fs and u2 were used to calculate the pile unit shaft resistance considering the effect of friction fatigue. The pile jacking force was obtained in terms of unit base resistance and unit shaft resistance. The ultimate bearing capacity was finally determined using pile jacking force multiplied by a force ratio denoted as Rf. The ultimate bearing capacities calculated for two case projects show a good agreement with the measured results, which indicates that the proposed method is feasible and effective. Reliability assessment was carried out on the proposed method and other five estimation methods with respect to the reliability index obtained by advanced first-order second-moment (AFOSM) method. Comparison result showed that the method proposed in this study has a higher reliability index over other five existing methods. Therefore, the proposed method can be used as a reliable alternative to predict the bearing capacity in the engineering practice of jacked pile foundation.

        Intravenous Tenecteplase for Acute Ischemic Stroke Within 4.5–24 Hours of Onset (ROSE-TNK): A Phase 2, Randomized, Multicenter Study

        Background and Purpose Intravenous tenecteplase (TNK) efficacy has not been well demonstrated in acute ischemic stroke (AIS) beyond 4.5 hours after onset. This study aimed to determine the effect of intravenous TNK for AIS within 4.5 to 24 hours of onset. Methods In this pilot trial, eligible AIS patients with diffusion-weighted imaging (DWI)-fluid attenuated inversion recovery (FLAIR) mismatch were randomly allocated to intravenous TNK (0.25 mg/kg) or standard care within 4.5–24 hours of onset. The primary endpoint was excellent functional outcome at 90 days (modified Rankin Scale [mRS] score of 0–1). The primary safety endpoint was symptomatic intracranial hemorrhage (sICH). Results Of the randomly assigned 80 patients, the primary endpoint occurred in 52.5% (21/40) of TNK group and 50.0% (20/40) of control group, with no significant difference (unadjusted odds ratio, 1.11; 95% confidence interval 0.46–2.66; <i>P</i>=0.82). More early neurological improvement occurred in TNK group than in control group (11 vs. 3, <i>P</i>=0.03), but no significant differences were found in other secondary endpoints, such as mRS 0–2 at 90 days, shift analysis of mRS at 90 days, and change in National Institutes of Health Stroke Scale score at 24 hours and 7 days. There were no cases of sICH in this trial; however, asymptomatic intracranial hemorrhage occurred in 3 of the 40 patients (7.5%) in the TNK group. Conclusion This phase 2, randomized, multicenter study suggests that intravenous TNK within 4.5–24 hours of onset may be safe and feasible in AIS patients with a DWI-FLAIR mismatch.

        Genome-Wide Transcriptomic Analysis of n-Caproic Acid Production in Ruminococcaceae Bacterium CPB6 with Lactate Supplementation

        n-Caproic acid (CA) is gaining increased attention due to its high value as a chemical feedstock. Ruminococcaceae bacterium strain CPB6 is an anaerobic mesophilic bacterium that is highly prolific in its ability to perform chain elongation of lactate to CA. However, little is known about the genome-wide transcriptional analysis of strain CPB6 for CA production triggered by the supplementation of exogenous lactate. In this study, cultivation of strain CPB6 was carried out in the absence and presence of lactate. Transcriptional profiles were analyzed using RNA-seq, and differentially expressed genes (DEGs) between the lactate-supplemented cells and control cells without lactate were analyzed. The results showed that lactate supplementation led to earlier CA p,roduction, and higher final CA titer and productivity. 295 genes were substrate and/or growth dependent, and these genes cover crucial functional categories. Specifically, 5 genes responsible for the reverse β-oxidation pathway, 11 genes encoding ATP-binding cassette (ABC) transporters, 6 genes encoding substrate-binding protein (SBP), and 4 genes encoding phosphotransferase system (PTS) transporters were strikingly upregulated in response to the addition of lactate. These genes would be candidates for future studies aiming at understanding the regulatory mechanism of lactate conversion into CA, as well as for the improvement of CA production in strain CPB6. The findings presented herein reveal unique insights into the biomolecular effect of lactate on CA production at the transcriptional level.

      • Diagnostic Significance of Combined Detection of Epstein-Barr Virus Antibodies, VCA/IgA, EA/IgA, Rta/IgG and EBNA1/IgA for Nasopharyngeal Carcinoma

        The objective of this study was to investigate the diagnostic significance of EBV antibody combined detection for nasopharyngeal carcinoma (NPC) in a high incidence region of southern China. Two hundred and eleven untreated NPC patients, 203 non-NPC ENT patients, and 210 healthy controls were recruited for the study. The titers of VCA/IgA and EA/IgA were assessed by immunoenzyme assay, and the levels of Rta/IgG and EBNA1/IgA were determined by enzyme-linked immunosorbent assay. The levels of VCA/IgA, EA/IgA, Rta/IgG and EBNA1/IgA demonstrated no association with gender or age (p>0.05). The receiver operating characteristic curve and the area under the curve were used to evaluate the diagnostic value. The sensitivity of VCA/IgA (98.1%) and the specificity of EA/IgA (98.5%) were the highest. When a logistic regression model was used to combine the results from multiple antibodies to increase the accuracy, the combination of VCA/IgA+Rta/IgG, whose area under the curve was 0.99, had the highest diagnostic efficiency, and its sensitivity, specificity and Youden index were 94.8%, 98.0% and 0.93 respectively. The data suggest that the combination of VCA/IgA+Rta/IgG may be most suitable for NPC serodiagnosis.
