        Differential Expression of miR-34c and Its Predicted Target Genes in Testicular Tissue at Different Development Stages of Swine

        To verified the target genes of miR-34c, bioinformatics software was used to predict the targets of miR-34c. Three possible target genes of miR-34c related to spermatogenesis and male reproductive development: zinc finger protein 148 (ZNF148), kruppel-like factor 4 (KLF4), and platelet-derived growth factor receptor alpha (PDGFRA) were predicted. Then, the expression of miR-34c and its target genes were detected in swine testicular tissue at different developmental stages by quantitative polymerase chain reaction. The results suggested that the expression of PDGFRA has the highest negative correlation with miR-34c. Then immunohistochemical staining was done to observe the morphology of swine testicular tissue at 2-days and 3, 4, 5-months of age, which indicated that PDGFRA was mainly expressed in the support cells near the basement membrane during the early development stages of testicular tissue, but that the expression of PDGFRA was gradually reduced in later stages. Therefore, western blot analyzed that the highest expression of PDGFRA was generated in 2-days old testicular tissues and the expression levels reduced at 3 and 4-months old, which correlated with the results of immunohistochemical staining. In conclusion, PDGFRA is a target gene of miR-34c.

        Fuzzy identity-based signature scheme from lattice and its application in biometric authentication

        A fuzzy identity based signature (FIBS) scheme allows a signer with identity ω to generate a signature which could be verified under identity ω′ if and only if ω and ω′ are within a certain distance of each other as judged by some metric. In this paper, we propose an efficient FIBS scheme from lattice assumption, which can resist quantum-computer attacks. Without using the Bonsai Tree technique, we utilize the lattice basis delegation technique to generate the private key, which has the advantage of keeping the lattice dimension invariant. We also prove that our proposed scheme is existentially unforgeable under an adaptive chosen message and identity attack in the random oracle model. Compared with existing scheme, our proposed scheme is much more efficient, especially in terms of communication overhead. Since our FIBS scheme possesses similar error-tolerance property, it can be well applied in post-quantum communication biometric authentication environments, where biometric identifiers such as fingerprints, voice, iris and gait are used in human identification.

        An Efficient Anonymous Authentication Scheme with Secure Communication in Intelligent Vehicular Ad-hoc Networks

        Vehicular ad-hoc networks (VANETs) have become increasingly significant in intelligent transportation systems, they play a great role in improving traffic safety and efficiency. In the deployment of intelligent VANETs, intelligent vehicles can efficiently exchange important or urgent traffic information and make driving decisions. Meanwhile, secure data communication and vehicle’s identity privacy have been highlighted. To cope with these security issues, in this paper, we construct an efficient anonymous authentication scheme with secure communication in intelligent VANETs. Combing the ElGamal encryption technique with a modified Schnorr signature technique, the proposed scheme provides secure anonymous authentication process for encrypted message in the vehicle-to-infrastructure communication model, and achieves identity privacy, forward security, and reply attack resistance simultaneously. Moreover, except the trusted authority (TA), any outside entity cannot trace the real identity of an intelligent vehicle. The proposed scheme is designed on an identity-based system, which can remove the costs of establishing public key infrastructure (PKI) and certificates management. Compared with existing authentication schemes, the proposed scheme is much more practical in intelligent VANETs.

        Development of a simple nomogram to estimate risk for intraoperative complications before partial nephrectomy based on the Mayo Adhesive Probability score combined with the RENAL nephrometry score

        Purpose: This study aimed to develop a simple nomogram based on the Mayo Adhesive Probability (MAP) score combined with the RENAL nephrometry score (RNS) to predict intraoperative complications before partial nephrectomy (PN) in Asian populations. Materials and Methods: This study retrospectively collected patients undergoing PN at three medical centers. Each component of the MAP score and the RNS (6 variables) was evaluated to assess its association with intraoperative complications by multivariable logistic regression with backward elimination. Results: A total of 46 cases (7.2%) with intraoperative complications were identified among 637 patients. After backward elimination, three variables, including tumor diameter (4–7 cm vs. ≤4 cm: odds ratio [OR], 4.339; 95% confidence interval [CI], 1.943–9,692; ≥7 cm vs. ≤4 cm: OR, 8.434; 95% CI, 1.225–58.090), nearness to the collecting system (4–7 mm vs. ≥7 mm: OR, 2.988; 95% CI, 1.293–6.907; ≤4 mm vs. ≥7 mm: OR, 21.394; 95% CI, 6.122–74.756), and perirenal fat stranding type (type 1 vs. no stranding: OR, 3.119; 95% CI, 1.079–9.017; type 2 vs. no stranding: OR, 18.722; 95% CI, 6.757–51.868), were retained. The predictive power (measured by area under the curve [AUC]) of the nomogram was observed to be superior to the RNS or MAP score alone (RNS: 0.686, MAP score: 0.729, the nomogram: 0.837), but comparable to their combination (0.813). Conclusions: The simple nomogram contains fewer components than the combination of the RNS and MAP scores yet demonstrates equivalent predictive power for intraoperative complications.

        MALAT1 knockdown promoted cell viability and migration of LPS‑treated MG‑63 cells via sponging miR‑212

        Background Most fractures could heal after treatment, around 5–10% of patients still develop delayed union and nonunion. Evidence has increasingly shown that abnormal expression of long noncoding RNAs is closely related to the occurrence and development of various diseases including fracture healing. However, evidence regarding the efect of MALAT1 on fracture healing remains limited. Objectives In this study, we attempt to explore the role of MALAT1 during the process of femoral neck fracture healing and elucidate the underlying mechanism of this disease. Methods We frst detect the expression of lncRNAs in serums from 3 pairs of patients with delayed femoral neck fracture healing and healthy volunteers using lncRNA microarray. And the expression of long noncoding RNA MALAT1 in serums and LPS-treated MG-63 cells was measured using qRT-PCR. CCK-8 assay, cell migration and qRT-PCR were applied to the role of MALAT1 knockdown in LPS-treated MG-63 cells. ELISA was used for the measurement of infammatory cytokines in serums of patients and healthy volunteers. The bioinformatics analysis and the rescue experiment were devoted to the underlying mechanism. Results MALAT1 expression was up-regulated in serum of patients with delayed union of femoral neck fracture. MALAT1 knockdown promoted cell viability and migration, reduced infammation in LPS-treated MG-63 cells. The bioinformatics analysis showed MALAT1 acts as a molecular sponge for miR-212. And SOX6 was a target of miR-212. Besides, MALAT1 knockdown suppressed SOX6 expression via targeting miR-212 in LPS-treated MG-63 cells. Conclusions These data suggest MALAT1 knockdown promoted the biological behavior of LPS-treated MG-63 cells via sponging miR-212, which may provide a new therapeutic avenue for delayed union of femoral neck fracture.

        Pedestrian Choices of Vertical Walking Facilities inside Urban Rail Transit Stations

        This paper investigates pedestrians’ choices between escalator and stairway inside urban rail transit stations during peak hours. After the review of mode choice models, a logit model based on the utility maximization theory is selected. Binary logit models forthe passenger flow distribution are established with walking time, walking distance, gender and age of the passenger as the maininfluencing factors. The influences of these factors are quantitatively analyzed through data collected from three rail transit stationswith different arrangement forms of vertical walking facilities in Nanjing, China. The developed models are verified to produce ahigh accuracy in predicting passengers’ choices of vertical walking facilities. With developed models, sensitivity analyses forselecting escalator for upward and downward pedestrians in the three types of stations are also conducted and compared to furtherreveal the differences in pedestrians' choices of using vertical walking facilities. The modeling procedure and results can provide atheoretical reference for operation decisions, such as the planning of vertical walking facilities and passenger flow organization inbusy rail transit stations.

      • Investigation on the Data Processing in Automation Monitoring System Designed for Dam Deformation

        Deformation monitoring for dam is quite important and it can be directly used to reflect the running status to keep the operation safely. In order to meet the demand for monitoring the dam, global position system (GPS) is used to develop a monitoring system with high efficiency. In this paper, we developed a monitoring system for deformation monitoring for the dam with wireless data transmission. In order to reduce the energy consumption, the regulation of its working condition has been redesigned according to the monitoring object's own characteristics. At the same time, the data compression is also used in the GPS data processing to reduce the energy consumption. The compression method is divided into two steps. In the preprocessing, data amount can be removed about half of the amount, and Hoffman coding compression rate and the style conversion compression ratio are relatively small. The total compression rate is more than 60%. Compared with the existing compression algorithms, the new developed method has higher compression rate. When the data collection rate is higher, the compression will be higher, and the reason may be that the time information and position information are compressed when the data collection rate is higher. This method can be used to process the data effectively and improve the data transmitting efficiency.

        Antiviral effects of Bovine antimicrobial peptide against TGEV in vivo and in vitro

        Background: In suckling piglets, transmissible gastroenteritis virus (TGEV) causes lethal diarrhea accompanied by high infection and mortality rates, leading to considerable economic losses. This study explored methods of preventing or inhibiting their production. Bovine antimicrobial peptide-13 (APB-13) has antibacterial, antiviral, and immune functions. Objectives: This study analyzed the efficacy of APB-13 against TGEV through in vivo and in vitro experiments. Methods: The effects of APB-13 toxicity and virus inhibition rate on swine testicular (ST) cells were detected using 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT). The impact of APB-13 on virus replication was examined through the 50% tissue culture infective dose (TCID50). The mRNA and protein levels were investigated by real-time quantitative polymerase chain reaction and western blot (WB). Tissue sections were used to detect intestinal morphological development. Results: The safe and effective concentration range of APB-13 on ST cells ranged from 0 to 62.5 μg/mL, and the highest viral inhibitory rate of APB-13 was 74.1%. The log10TCID50 of 62.5 μg/mL APB-13 was 3.63 lower than that of the virus control. The mRNA and protein expression at 62.5 μg/mL APB-13 was significantly lower than that of the virus control at 24 hpi. Piglets in the APB-13 group showed significantly lower viral shedding than that in the virus control group, and the pathological tissue sections of the jejunum morphology revealed significant differences between the groups. Conclusions: APB-13 exhibited good antiviral effects on TGEV in vivo and in vitro.

        Study of upfront surgery versus neoadjuvant chemotherapy followed by interval debulking surgery for patients with stage IIIC and IV ovarian cancer, SGOG SUNNY (SOC-2) trial concept

        Background: Two randomized phase III trials (EORTC55971 and CHORUS) showed similarprogression-free and overall survival in primary or interval debulking surgery in ovariancancer, however both studies had limitations with lower rate of complete resection and lack ofsurgical qualifications for participating centers. There is no consensus on whether neoadjuvantchemotherapy followed by interval debulking surgery (NACT-IDS) could be a preferred approachin the management of advanced epithelial ovarian cancer (EOC) in the clinical practice. Methods: The Asian SUNNY study is an open-label, multicenter, randomized controlled,phase III trial to compare the effect of primary debulking surgery (PDS) to NACT-IDS instages IIIC and IV EOC, fallopian tube cancer (FTC) or primary peritoneal carcinoma (PPC). The hypothesis is that PDS enhances the survivorship when compared with NACT-IDS inadvanced ovarian cancer. The primary objective is to clarify the role of PDS and NACT-IDS inthe treatment of advanced ovarian cancer. Surgical quality assures include at least 50% of nogross residual (NGR) in PDS group in all centers and participating centers should be nationalcancer centers or designed ovarian cancer section or those with the experience participatingsurgical trials of ovarian cancer. Any participating center should be monitored evaluatingthe proportions of NGR by a training set. The aim of the surgery in both arms is maximalcytoreduction. Tumor burden of the disease is evaluated by diagnostic laparoscopy orpositron emission tomography/computed tomography scan. Patients assigned to PDS groupwill undergo upfront maximal cytoreductive surgery within 3 weeks after biopsy, followed by6 cycles of standard adjuvant chemotherapy. Patients assigned to NACT group will undergo 3cycles of NACT-IDS, and subsequently 3 cycles of adjuvant chemotherapy. The maximal timeinterval between IDS and the initiation of adjuvant chemotherapy is 8 weeks. Major inclusioncriteria are pathologic confirmed stage IIIC and IV EOC, FTC or PPC; ECOG performancestatus of 0 to 2; ASA score of 1 to 2. Major exclusion criteria are non-epithelial tumors as wellas borderline tumors; low-grade carcinoma; mucinous ovarian cancer. The sample size is 456subjects. Primary endpoint is overall survival.
