      • Escherichia coli(M15)을 이용한 serotonin N-acetyltransferase 유전자의 발현

        AA-NAT cDNA was obtained by RT-PCR technique from total RNA of rat sacrified at night(02:00am). pCRNAT was cloned using the pCRII vector with insertion of AA-NAT cDNA(about 1.4 kb) at EcoRI site. For the expression of the gene, pQECNAT was subcloned, in which the coding region of AA-NAT was inserted into expression vector pQE30 at BamHI and HindIII sites. According to the experimental results, Escherichia coli strain M15, transformed by the expession vector pQECNAT, was selected as a host to express AA-NAT gene with the induction of isopropyl β-D-thiogalactoside(IPTG). Optimal condition for the expression of AA-NAT gene was achieve from the experimental results, showing that the expression of the protein was inducible much 19.800 ± 2,200 dpm per ml of culture volume in 4 hours at the concentration of 2 mM IPTG. Partial purification through the affinity column(Ni-NTA agarose) binding to the continuous 6 histidine residues of protein resulted in 5 times more increase in the specific activity of AA-NAT than that of the homogenate of bacterial pellet. These experimental results will provide basic data in further study for the enzymatic kinetics and antibody production of AA-NAT.

      • 임베디드 리눅스 기반의 영상처리기법에 관한 연구

        현재 embedded system에서 많은 기업체들이 리눅스를 채용하고 있고, 우리 생활에서 쓰이는 각종 전자기기, 정보가전, 제어장치 등에 적용되고 있다. 최근 멀티미디어 처리와 같은 점차 복잡한 기능이 요구되면서 임베디드 운영체제를 사용하기 시작하였고, 이러한 운영체제로 리눅스가 주목받게 되었다. 이는 가격 경쟁력뿐만 아니라 각 H/W에 따라 운영체제와 응용프로그램을 다양하게 변화시킬 수 있다. 본 논문에서는 CCD 카메라로부터 영상을 입력받아 압축하여 LCD 모니터에 출력하기 위한 임베디드 시스템 기반의 영상처리에 관해 기술한다. Recently, many industries adopts linux as an operating system for embedded system, which is commonly used in our life as the type of electronic machinery, electric home appliances and so on. The embedded operating system is getting more complex as lots of features such as multimedia are required. The operating system should be adjusted to the hard ware and software in order to satisfy a competitive price. In this paper, a operating system is discussed in order to compress the images form the CCD camera and then spread the LCD.

      • 근거리 수치사진측량의 정확도 향상에 관한 연구

        As state-of-the-art usually high-quality digital cameras are used for photogrammetric industrial metrology. Digital high-resolution consumer cameras are widely available. and are increasingly used in close-range photogrammetry. The mechanical construction of the cameras often do not meet photogrammetric requirements, thus they have to be modelled sufficiently. The close-range photogrammetry is a surveying technique the acquiring the 3D object from 2D geometric appearance shown in the image. Camera treatment is relatively so simple. Camera position refer to the object can be freely adjusted by locating the exposure station at any site. So it is used effectively a small area or object for survey. In this study, suing the digital camera which has capacity of keeping numerical value by itself and easy carrying, we analyze the positioning error according to various change of photographing condition. Also we try to find effective method of acquiring basis data for 3D monitoring of high-accuracy in sub-pixel degree through digital close-range photogrammetry with bundle adjustment for local terrain model generation.

      • 디지털 카메라의 자체검정을위한 도심측정방법과 타원 피팅 방법의 정확도 분석

        Digital high-resolution consumer cameras are widely available, and are increasingly used in close-range photogrammetry. The mechanical construction of the cameras often do not meet photogrammetric requirements, thus they have to be modelled sufficiently. An approach for camera modelling is discussed that introduces image-variant parameters of interior orientation. In addition a correction model based on finite elements is integrated that provides the correction of remaining errors in the sensor space. All functional parameters are estimated simultaneously in a bundle adjustment. Significant improvements of object accuracy have been achieved with respect to standard calibration techniques based on self-calibrating bundle adjustment. Rollei d7 metric 디지털 카메라를 이용하여 촬영조건의 다양한 변화에 따른 위치오차를 분석하며 대상물표면에 설치된 타켓의 3차원위치를 결정하기 위해 도심측정방법과 타원 피팅 방법으로 타켓중심을 구하였다. 각 타켓의 변위를 측정하고 3차원 정확도를 분석한 결과 다음과 같은 결론을 얻었다. 도심측정방법과 타원 피팅 방법을 여러 가지로 조합하여 적용한 결과 평균표준오차는 타원 피팅 방법과 도심측정방법을 동시에 적용되었을 경우 X좌표에서 ±0.478mm, Y좌표에서 ±0.478mm, Z좌표에서 ±0.478 mm로 가장 낮은 오차를 보였다. 도심측정방법은 타켓의 모양에 구애받지 않으나 적절한 임계값의 결정이 중요하며 경중률 을 고려함으로써 이를 보완할 수 있었다. 반면에 타원 피팅 방법은 타켓의 형상이 수학적으로 피팅 할 수 있는 형상이어야 한다는 점은 불편하나 임계값 영향을 받지 않았다. 자체 검정기법에 의한 촬영조건의 다양한 변화에 따른 위치오차를 분석하여 Rollei d7 metric 디지털 카메라의 기하학적 지식 및 수행성을 평가하기 위하여 내부표정요소 및 렌즈 왜곡 등에 관련된 기하학적 검정 매개변수들을 도출해 낼 수 있었으며 높은 정확 도 를 나타냄으로써 근거리 지상사진측량시스템 의 잠재성을 확인할 수 있었다. 자체검정 광속조정은 카메라의 계통적 오차들을 효과적으로 보정해줌으로써 정확도를 크게 향상시킬 수 있었다. 따라서 자체검정 기법을 적용하는 디지럴 카메라에 의한 광학적 근거리 지상사진측량시스템은 산업의 차원적 품질 검사,문화재의 3차원 모델링 및 형상재현 등을 효율적으로 구현하여 대상물의 시각적 분석에 활용할 수 있을 것이다.

      • 망막아세포종 세포주(Y-79)의 Arylalkylamine N-acetyltransferase 활성에 미치는 Dopamine의 영향

        The arylalkylamine N-acetyltransferase(AA-NAT, EC 2.3.1.87) in vertebral pineal gland and retina is rate-limiting enzyme in the pathway of biosynthesis of melatonin. AA-NAT activities are remarkably increased during night with 10-150 folds. Neurohormone melatonin is thought to play roles in a broad range of physiological functions in human, primarily through effects on the biological clock in the suprachiasmatic nucleus of the hypothalamus, This rhythm reflects large changes in the activities of AA-NAT. So the important regulatory role of AA-NAT has made it central interest in research on melatonin biochemistry and neurochemical signal transduction. The neurotransmitter that regulates the AA-NAT activities is norepinephrine, which acts through a cyclic AMP mechanism. The effect of dopamine on AA-NAT in human retinoblastoma Y-79 cells was studied. the obtained results are as follows; AA-NAT activities in Y-79 cells were increased by cyclic AMP, and dopamine inhibited the AA-NAT activities stimulated by forskolin. The pattern of inhibition of dopamine on the AA-NAT activities in Y-79 cells was time and concentration dependent. The results suggest that dopamine participates in the regulation of the AA-NAT activities through the inhibitory mechanism in the human pineal and retinal tissues.

      • 박스-젠킨스법을 이용한 소양호 수질변화 시계열분석

        Statistical models were developed using the Box-Jenkins time series analysis to simulate changes of water qualities in Soyang Lake. The water qualities included in the models are total phosphorus(TP) and chlorophyll-a(CA). The model development was based on the data obtained from Feb. 1987 to Oct. 1992 for TP and from Mar. 1985 to Oct. 1992 for CA, and followed the typical procedures of the Box-Jenkins method including identification, estimation, and diagnostic check. From the computed autocorrelation function (ACF) and partial autocorrelation function(PACF), it was determined that the TP model should have two autoregression (AR) parameters and one moving average(MA) parameter, and two seasonal AR parameters and one seasonal MA parameter. The CA model was determined to have two AR parameters and two MA parameters, and two seasonal AR parameters and on seasonal MA parameter. Both models had seasonality of twelve months and were formulates as SARIMA(2,1,1)×(2,1,1)₁₂for TP and SARIMA(2,0,2)×(2,1,1)₁₂ for CA. The models were validated by testing normality and independency of the residuals. The model prediction ability was tested using the data collected from Nov. 1992 to July 1994 both for TP and CA. There were good agreements between the model predictions and the field measurements. Most of the predicted values were located within the 95% confidence limits except certain period of chlorophyll-a. It was concluded from this study that the Box-Jenkins time series analysis would be an effective tool to understand and simulate the long term variations of water qualities in Soyand Lake.

        Effect of Kettle Bell Swing Exercise on the Body Composition and Lower Limb Muscular Activity

        Purpose: This study examined the effect of Kettle bell swing exercise on the body composition and lower limb muscular activity. Methods: As subjects, 26 normal adults in their twenties were divided randomly into the experiment group (n=13), in which Kettle bell exercise was applied, and the control group (n=13), in which bar exercise was applied. Both groups participated in the interventions for eight weeks with three sessions per week. Each group was assessed before training and four and eight weeks into the training to determine the effects of training intervention within and between the groups. The body composition was measured with a body composition analyzer. The changes in muscle activity were measured with a surface electromyography. An independent t-test was conducted to test the significance between the groups according to the measurement points and Repeated measured ANOVA was performed to compare the groups according to the periods with the statistical significance level set to ${\alpha}=0.05$. Results: No significant changes to the main effects of time and the interactions of time according to the measurement points were observed in the Kettle bell and control groups in the body composition (p>0.05). The muscular activity of both muscles were significantly different between the Kettle bell and control groups according to the measurement points with the exception of the right lateral gastrocnemius muscle (p<0.05). Conclusion: These findings suggest that Kettle bell swing exercise can have positive effects on the development of lower limb muscles.
