Background: Human skin pigmentation is regulated by various factors such as genetic, environmental, and endocrine factors. Among these factors, genetic factor is one of the most potent determinants in skin pigmentation phenotype and has polygenic quantitative traits. Objectives: We investigated the genetic factors influencing UV-induced skin pigmentation in Korean women. Methods: We carried out a multistage genome-wide association study after exposure to UV in Korean women and examined the effect of selected gene on melanogensis in normal human epidermal melanocytes. Results: In an initial discovery stage, meta-analysis of two GWA studies of 500K single nucleotide polymorphisms on 97 Korean women identified 9 SNPs as loci highly associated with tanning ability. We further genotyped these SNPs in an independent replication study. We found that a novel SNP in the intron of WWOX gene yielded highly significant replicated associations with skin tanning ability. To understand the functional consequences of this locus, we investigated the role of WWOX in human melanocytes and inhibition of WWOX expression significantly increased the expression and activity of tyrosinase in human melanocytes. Conclusion: Our results suggest that genetic variants in intronic region such as SNP in WWOX could be determinant of an individual’s UV-induced tanning ability. WWOX gene represents a new candidate gene to study the molecular basis of an individual’s UV-induced tanning ability.

사람의 피부색에 결정적인 영향을 미치는 멜라닌은 표피 기저층의 멜라닌형성세포(Melanocyte)에 의해 생성되어 피부 전반에 분포하고 있으며, 표피의 가장 외각인 각질층에서도 멜라닌은 존재하고 있다. 이러한 각질세포 내 멜라닌과 피부색과의 관계를 알아보고자 각질세포 내 멜라닌의 선택적 염색(Fontana-Masson method) 및 영상분석을 통한 정량적 평가법을 확립하였고, 각질세포 내 멜라닌 양과 색차계를 통한 피부색과의 상관관계를 조사하였다. 71명의 여성 피시험자를 대상으로 색차계를 이용한 피부색과 동일부위의 각질세포에 함유된 멜라닌을 정량하여 비교하였을 때, 각질세포 내 멜라닌이 차지하는 면적을 나타내는 MCA (Melanin covering area)값이 피부색의 L*값, ITAo (Individual Typology Angle)값과 비교적 높은 상관관계를 보였다(각각 r = 0.6049, r = 6651). 이러한 결과를 통해 각질세포 내 멜라닌 양이 피부색 분류의 새로운 기준이 될 수 있는 가능성을 확인했으며, MCA값에 따라 새로이 피부색을 분류하는 기준을 제시할 수 있었다. 본 실험을 통해, 각질세포 내 멜라닌 정량법의 개발과 멜라닌 양에 따른 새로운 피부색 분류기준을 세울 수 있었으며 개발된 각질세포 내 멜라닌 정량법은 향후 새로운 in vivo 미백 평가 및 피부 색상과 관련된 여러 분야에서 매우 유용하게 이용될 수 있을 것이다. Melanin is synthesized by the melanocytes in basal layer of epidermis and distributed over all the layers of skin including corneocytes in stratum corneum, outermost layer of the epidermis. Melanin of corneocytes was stained using Fontana-Masson method and quantified by image analysis. The correlation between melanin contents and skin color value was estimated in the skin of 71 Korean women. Melanin covering area (MCA) showed good correlation with L* value and ITAo (Individual Typology Angle) (r = 0.6049, 0.6651, respectively). MCA can be used as new parameter for skin color study and has potential application for evaluating the efficacy of the skin whitening cosmetics.
발아콩 및 목련박피 혼합추출물(SeleMix AN)에 의한 여드름 개선 효과

In vitro 및 in vivo에서 모두 효과를 나타내는 새로운 여드름 개선 성분을 개발하였다. 특히, in vitro에서 효과가 검증된 많은 원료들이 실제 여드름환자에게서 실질적인 효능을 나타내지 못하는 경우가 많아, 본 연구에서는 임상에서 실질적인 효과를 보일 수 있는 원료의 개발에 초점을 맞추었다. 우선적으로 여드름에 효과가 높은 것으로 알려져 있거나 여드름개선 효과가 기대되는 천연물질의 추출물을 대상으로 소규모의 예비임상시험을 통해 효능을 확인하였다. 여러 가지 후보물질 중에서 발아콩과 목련박피 2종의 추출물이 여드름 및 여드름에 의한 홍조현상과 흉터에 뚜렷한 효과를 나타내어 이 두 천연물의 혼합추출물을 SeleMix AN이라 명명하였다. 200여 명의 예비임상시험을 통해서 SeleMix AN의 임상적 효과를 확인한 후 상기 물질의 여드름 개선 작용기작을 규명하기 위한 in vitro 효능테스트를 실시하여 P. Acne 성장억제 효과(시료농도 0.0125%), 16.9%의 히스타민 분비저해효과와 함께, 인간유래 섬유아세포의 활성을 대조군 대비 57% 높여 주는 실험결과를 확인하였다. 최종적으로 분당 서울대병원과의 공동연구를 통해 23명의 여성여드름환자를 대상으로 한 임상평가를 실시하여 새로운 여드름 개선성분의 효능을 검증하였다. 여름에 실시된 최종임상에서 SeleMix AN이 함유된 시료는 피지분비량이 증가하고 여드름 발병율이 높아지는 계절적인 영향에도 불구하고 4주만에 특히, 염증성 병변을 대조군 대비 통계적으로 유의차 있게 감소시키는 뛰어난 여드름 개선 결과를 얻을 수 있었다. We investigated new ingredients with real efficacy in both in vitro and in vivo all together. Especially we focused on the real improving effort in the clinical experiments. Because most products containing effective materials evaluated in vitro failed to show a real improving effect in the human with acne. We evaluated the well-known ingredients in a small scale clinical experiment with half-finished goods containing each ingredient. Among these products, product formulating SeleMix AN composed with germinating soy bean and magnolias bark extract remarkably improved acne and acne scar. Moreover skin redness caused by severe acne was improved. There was statistical significance between placebo and sample. Two hundred volunteers participated in our pilot study with written informed consent. After then we performed in vitro efficacy test this ingredient. SeleMix AN inhibited the growth of propionibacterium acnes at a concentration of 0.0125% and suppressed histamine release by 16.9%. Moreover human fibroblast cell activity was increased by 57% compared to control. Lastly, we performed a clinical study. Consisting of groups of 23 volunteers. Although the period of the test was in summer accelerating sebum secretion and recurring a high tate of acne, inflammation lesions were especially improved after applying product containing SeleMix AN for 4 weeks.

In this paper, we investigated the carbonization characteristics of lignin hydrochar prepared by hydrothermal carbonization and established a model for predicting the carbonization degree using near-infrared spectroscopy and partial least squares regression. The carbon content of the hydrothermally carbonized lignin at the temperature of 200 ℃ was higher by approximately 3 wt% than that of the untreated sample, and the carbon content tended to gradually increase as the heating time increased. Hydrothermal carbonization made lignin more carbon-intensive and more homogeneous by eliminating the microparticles. The discriminant and predictive models using near-infrared spectroscopy and partial least squares regression approppriately determined whether hydrothermal carbonization has been applied and predicted the carbon content of hydrothermal carbonized lignin with high accuracy. In this study, we confirmed that we can quickly and nondestructively predict the carbonization characteristics of lignin hydrochar manufactured by hydrothermal carbonization using a partial least squares regression model combined with near-infrared spectroscopy. 본 논문에서는 열수 탄화(hydrothermal carbonization)에 의해 제조된 리그닌 하이드로차의 탄화 특성을 조사하였고, 근적외선 분광법과 부분 최소 제곱(partial least squares) 회귀를 이용하여 탄화 거동을 예측하기 위한 모델을 수립하였다. 온도 200℃에서 열수 탄화된 리그닌의 탄소 함량은 무처리 시료 보다 약 3 wt% 높았으며 가열 시간이 증가할수록 탄소 함량도 서서히 증가하는 경향이 나타났다. 열수 탄화는 리그닌을 더욱 탄소 집약적으로 변화시키고 마이크로 파티클을 제거하여 더욱 균질한 특성을 부여하였다. 근적외선 분광법과 부분 최소 제곱 회귀를 이용한 판별 및 예측 모델은 수열 탄화의 적용 여부를 완벽히 구분했으며 높은 정확도로 열수 탄화 리그닌의 탄소 함량을 예측하였다. 본 연구로부터 근적외선 분광법과 결합된 부분 최소 제곱 회귀 모델을 이용하여 열수 탄화에 의해 제조된 리그닌 하이드로차의 탄화 특성을 빠르고 비파괴적으로 예측할 수 있다는 것이 확인되었다.

The GMR-SV (giant magnetoresistance-spin valve) device depending on the micro patterned features according to two easy directions of longitudinal and transversal axes has been studied. The GMR-SV multilayer structure was Ta(5 ㎚)/NiFe(8 ㎚)/Cu(2.3 ㎚)/NiFe(4 ㎚)/IrMn(8 ㎚)/Ta(2.5 ㎚). The applied anisotropy direction of the GMR-SV thin film was performed under the magnitude of 300 Oe using by permanent magnet during the deposition. The size of micro patterned device was a 1 × 18 ㎛² after the photo lithography process. In the aspects of the shape magnetic anisotropy effect, there are two conditions of fabrication for GMR-SV device. Firstly, the direction of sensing current was perpendicular to the magnetic easy axis of the pinned NiFe/IrMn bilayer with the transversal direction of device. Secondly, the direction of shape magnetic anisotropy was same to the magnetic easy axis of the free NiFe layer with the longitudinal direction of device.

Rheological properties and crystallization kinetics of the polypropylene (PP) block copolymer and recycled PP block copolymer were studied by advanced rheometric expansion system (ARES), differential scanning calorimetry (DSC), and optical microscopy. In the study of the dynamic rheology, it is observed that the storage modulus and loss modulus for the PP block copolymer and recycled PP block copolymer did not change with frequency. In the study of the effect of the repeated extrusion on the crystallization rate, half crystallization time of the PP samples was increased with the number of repeated extrusion in isothermal crystallization temperature ($T_c$). From the isothermal crystallization kinetics study, the crystallization rate was decreased with the increase of the number of repeated extrusion. Also, from the result of Avrami plot, the overall crystallization rate constant (K) was decreased with the increase of the number of the repeated extrusion. From the study of the optical microscopy, the size of the spherulite of the PP samples did not change significantly with the number of repeated extrusion. However, it was clearly observed that the number of the spherulite growth sites was decreased with the number of repeated extrusion. From the results of the crystallization rate, isothermal crystallization kinetics, Avrami plots, and optical microscopy, it is suggested that the crystallization rate of the PP block copolymer is decreased with the increase of the number of repeated extrusion.
