        FK506이 T 림프구 사멸에서 활성산소 생성에 미치는 영향

        Purpose: Tacrolimus (FK506) has been widely used as an immunosuppressant in organ transplanted recipients to suppress organ rejection phenomenon. We investigated the role of oxidative stress and heme oxygense-1 by FK506 on human Jurkat T cells. Methods: The cells viability was examined by DAPI stain, enzyme activity of caspase family proteins, and western blotting for Baks, PUMA, iNOS, HO-1. Cells were cultured in the absence or presence of CoPPIX or ZnPPIX and the fluorescence intensity was analyzed using a flow cytometry. Results: Treatment with FK506 increased the generation of reactive oxygen species (ROS), including hydrogen peroxide and superoxide anion, and NO in Jurkat cells in a dose-dependent manner. Immunohistochemistry and Western blot analysis data revealed the hemoxygenase-1 (HO-1) was induced by the addition of FK506 in Jurkat cells. Induction of CoPP, HO-1 inducer, resulted in decreased intracellular H₂O₂ and NO concentrations. Instead ZnPP, an HO-1 competitive inhibitor did it reversely. In addition, ZnPP regulates iNOS protein synthesis by inhibition of HO-1. Conclusion: Increase of HO-1 expression would induce to decrease the intracellular H₂O₂ and NO concentrations. Also, HO-1 would regulate iNOS protein synthesis. Consequently, we can expect the regulation of HO-1 expression with concomitants use of FK506 to suppress organ rejection phenomenon by enhancing apoptosis.

        Omeprazole 투여시 위산분비 억제능에 대한 CYP2C19 의 영향

        Background/Aims: CYP2C19 has an important role in the catabolism of several proton pump inhibitors. However, the relative contribution of CYP2C19-mediated metabolism varies among the different proton pump inhibitors. We investigate to determine the effect of CYP2C19 genotype status on intragastric pH during dosing with omeprazole. Methods: The subjects were 16 healthy volunteers. Their CYP2C19 genotype status was determined by a polymerase chain reaction-restriction fragment length polymorphism method. Twenty-four-hour monitoring of intragastric acidity were performed on the day without medication and day 4 after omeprazole administration. Results: A single dose of omeprazole significantly decreased 24 hour intragastric acidity (1.7±0.3 vs. 5.1±0.4). Subjects were divided into three groups on the basis of their CYP2C19 genotype status: homozygous extensive metabolizers (Ho-EMs, n=5), heterozygous extensive metabolizers (Ht-EMs, n=6), and poor metabolizers (PMs, n=5). Median 24 hour intragastric pH in the Ho-EM group was 3.1 compared with 5.5 in Ht-EM group and 5.9 in PM group(P<0.05). The median pH during omeprazole administration was influenced by CYP2C19 genotype. On the other hand, the Helicobacter pylori infection did not influence the median intragastric pH during omeprazole administration (p= NS). Conclusions: The effects of omeprazole on intragastric acidity is influenced by the CYP2C19 polymorphism. (Korean J Gastroenterol 2001;38:399-404)

      • Captopril과 Nifedipine 병합요법의 강압효과에 관한 임상적 연구

        Background: The hypotensive effect of captopril(angiotensin converting enzyme inhibitor) and nifedipine(calcium channel blocker) was studied. Method: Captopril(75mg) and nifedipine(30mg) were administered in 30 hypertensive adults during twelve weeks after a week observation for washout with stepwise increments of the dose according to the patient's blood pressure in every two weeks. Result: The supine blood pressure were decreased from 169.5±22.3/104.0±20.3 mmHg to 130.4±21.3/82.4±19.5mmHg at the end of twelve weeks during therapy.(p<0.05) The standing blood pressure were also decreased conferrably and to the some lower level. Hematologic examination and blood chemistry revealed no discernible abnormal findings before and after the treatment. During the period of the study a few probably drug-related symptom such as cough and headache, gastrointestinal disturbance developed but not troublesome enough to administering. Conclusion: Captopril and nifedipine regimen is well tolerated and effective in the treatment of hypertensive patients.

        울금(鬱金)에서 분리된 demethoxycurcumin이 백서의 혈관재협착에 미치는 효과

        The pathobiologic process of arterial stenosis following balloon angioplasty continues to be an enigmatic problem in clinical settings. This study investigates the ability of demethoxycurcumin, a curcuminoid isolated from Radix Curcumae, to attenuate balloon injury-induced neointima(NI) formation in the rat carotid artery. It was found that demethoxycurcumin induced inducible heme oxygenase(HO-1) expression and inhibited dose-dependently cellular proliferation in rat vascular smooth muscle cells. Perivascular application of demethoxycurcumin immediately following injury significantly reduced NI area and NI thickness 2 weeks post-injury. Interestingly, treatment with tin-protoporphyrin IX, a HO inhibitor, reversed the effects of demethoxycurcumin on NI formation. These results implicate demethoxycurcumin as a potent new therapeutic agent that is capable of reducing post-angioplasty arterial stenosis through induction of the HO-1 expression.

      • 고등학교 과학과 교육과정 개정에 대한 과학교사들의 인식과 준비도 연구

        여러차례 개정을 반복해온 고등학교 과학과 교육과정에 대한 교사들의 인식과 준비도에 대하여 전국적으로 설문조사를 하고 이에 응답한 77개교 495명의 응답을 분석하였다. 응답은 1. 현행(5차) 교육과정에 대한 인식, 2. 교육과정 개정에 대한 인식, 3. 공통과학에 대한 인식 4. 교육과정 개정에 대한 준비도의 4영역으로 구분하고 각 응답유형을 성별, 지역별, 설립자별, 담당과목별, 경력별, 연령별, 소지학위별로 나누어 분석하였다. 응답결과, 현행 교육과정은 문제점이 많아 개정의 필요성이 높으며 가장 큰 문제점은 과학Ⅰ, Ⅱ의 이름뿐인 통합에 있고 이는 과목별로 따로 분리하는 것이 타당하다고 하였다. 또한 이수단위에 대한 내용분량이 높고 단위수는 타과목에 비해 부족한 편이며 과학과 실생활과의 관련된 내용이 강조 보완되어야 한다고 지적하였다. 교육과정의 개정에 대해서는 대체로 관심이 높은 편이나 30%이상의 교사들이 관심이 없다고 응답했고 교육과정의 개정은 정권이 바뀜에 의해 이뤄진다는 응답이 높았다. 5-7년 주기의 교육과정 개정이 필요하나 기본틀은 유지하기를 바라고 있으며, 교육과정 개정에 대한 정보의 원천을 교육청 발행 교육과정 소개서를 이용한다는 응답이 높았다. 그러나 6차 교육과정에 대한 홍보는 보다 강화되어야 할 필요성이 제기 되었다. 공통과학에 대해 긍정적 견해를 나타낸 교사가 48%정도이고 응답자의 배경에 따라 다양한 견해차이를 나타내었으나 학생들에게는 과목부담만 늘게할 것이라는 등의 부정적 견해도 높았다. 공통과학수업은 교사들의 전공별로 나누어 맡거나, 공통과학을 위해 정규교육을 받은 사람이 맡아야 한다고 응답한 경우가 높았고, 공통과학담당을 의뢰할 경우 긍정적 수임의사를 나타낸 교사는 전체의 45%에 이르렀다. 외국 교육과정에 대한 인지정도는 약한 편이며 전공서적 독서량도 적은 편으로 교육과정 개정에 대한 준비도는 약한 것으로 나타났다. 교사들은 주로 참고서를 이용하여 교과내용 변화와 심화에 대처하며, 스스로 판단하여 탐구학습 지도능력이 부족하다고 생각하는 교사가 43%에 이르렀다. 그러나 교육과정 개정후 재교육이 실시된다면 참여하겠다는 외지를 나타낸 교사들이 83%에 이르러 적절한 교사재교육 프로그램을 운영해야할 필요성이 강력히 제기되고 있다. 본 연구 결과는 교육과정 개정작업에 있어서 교사들의 폭넓은 참여가 필요하고 이를 위한 연구가 심도깊게 이뤄져야함을 시사한다. The purpose of this study is to prove high school science teacher's appreciation and readiness about curriculum reform. For this study We developed a questionaire including four main themes - 1) The appreciation about the current(5th) curriculum, 2) The appreciation about the curriculum reform, 3) The appreciation about 'general science' 4) The readiness to curriculum reform. Some teachers have been sampled for the criterion of teacher's officially appointed region. The sample size is 495 person, 77 schools, and the rate of answer was 88.5% for school, 80.2% for person. SPSS/PC (Statistical Package for the Social Science) was used for the analysis of this data. 1. The result represents that current curriculum has many problems and so it must be reformed as soon as possible(85.1%). The main problem that teachers pointed was the formal integration of subject- science Ⅰ and Ⅱ(66.1%). Also, science teachers answered that the content to be stressed and fulfiled in High school science curriculum must be related to actual life(74.1%). It means that science teachers are much concerned about STS educational movement. 2. There were 66.8% of teachers interested some or much in curriculum reform but they have negative appreciation to the curriculum reform of past. The causes they pointed out mainly are that science curricula have been influenced severely by the policy change. 3. It was the main focus of this study that what teacher's appreciation is to the general science. About half of them answered general science is necessary subject, but 30% of them think it may be another load to students. To the question about who should be charged of general science, they answered that the teacher of Physics, Chemistry, Biology, Earth science should share each part(39.2%). And teachers who have the license of general science teaching should be charged it(31.7%). About 45% of science teachers expressed their opinion that they would take charge the subject willingly or without any complain. It means that general science have some foundations in high school. 4. Science teachers read not enough books relating manage and know little about curricula of foreign nations, but they have much concern about curriculum reform and have great eager to be reeducated after curriculum reform (83%). So Planning to reeducate teachers is as important as developing better science curriculum. The data from this study were analyzed in the factor or sex, region, charge of subject, career, foundation, age, and academic degree. The result of this study urges that science teacher's participation and concerns are necessary in the process of science curriculum reform, and that it must be performed more broad and systematic survey research before curriculum reform.

        영아자(Phyteuma japonicum Miq.)의 성분 조성

        산채식물인 영아자(Horned Rampion ; Phyteuma japonicum Miq.)의 영양학적인 가치를 평가코져 야생 및 평지재배 시료를 잎과 줄기로 구분하여 일반성분, 비타민 C, 유리당, 무기물, 핵산관련물질, 구성 아미노산 및 유리아미노산을 분석하였다. 야생과 재배 영아자의 회분은 1.2∼2.7%의 범위였고, 조지방과 조단백질은 재배시료가 조섬유는 야생시료에서 더 높은 함량으로 정량되었고, 전당은 두 시료간에 대차를 보이지 않았다. 비타민 C는 줄기보다는 잎에서 재배시료보다는 야생시료에서 더 높게 정량되었다. 유리당은 야생 및 재배시료 모두 glucose, frucose 및 sucrose가 잎보다 줄기에서 높게 정량되었다. 무기물은 총 9종이 분석되었는데 이중 칼슘의 함량이 가장 높아 재배시료의 경우 잎은 34374.0㎎/㎏, 즐기는 9584.1㎎/㎏였고, 그 다음으로 칼륨, 마그네슘의 순으로 많았으며, 야생시료도 비슷한 경향을 보였다. 핵산 관련물질은 CMP, UMP, IMP, AMP 및 hypoxanthine이 동정되었는데 잎과 줄기 모두 야생시료에서는 hypoxanthine이, 재배시료에서는 AMP가 월등히 높게 정량되었다. 구성아미노산은 총 17종으로 야생시료에서는 glutamic acid, 재배시료는 잎의 경우 aspartic acid, 줄기의 경우 glutamic acid의 함량이 가장 많았다. 유리아미노산은 총 29종이 동정되었고 야생시료의 잎에서는 glutamic acid, 줄기에서는 γ-aminoisobutyric acid, 재배시료의 줄기에서는 asparagine이 가장 높은 함량으로 정량되었다. This research is to establish the basic data of the nutritive value and improve our diet. In the part of th leaf and stem of the wild cultivated horned rampion(Phyteuma japonicum Mig), the components such as chemical composition, vitamin C, free sugar, mineral, nucleotide and its related compounds, composition and free amino acid were analyzed one after another. Content of the crude lipids and proteins was determined much higher in its wildness than in its cultivated horned rampion ; while, that of carbohydrates was higher in the former than in the latter. The content of vitamin C was retained higher in the leaf than in the stem horned rampion. And the content of calcium among the detected minerals was outstanding in all of the samples collected, and potassium and magnesium was the next ones in its order. The main components of free sugars in both the wild and cultivated horned rampion were glucose and fructose, and their content was higher in the stem than in the leaf. Nucleotide and its related compounds were identified with 5 kinds of nuciotides such as CMP, UMP, IMP, AMP and hypoxanthine(Hx), and the content of Hx and AMP was the highest in the wild and cultivated samples, respcetively. In the composition amino acid of the wild horned rampion, glutamic acid, aspartic acid and phenylalanine was outstandingly abundant; while, such amino acid as methionine and proline was small and besides cysteine couldn't be detected in the stem. Total amounts of composition amino acid in the leaf was 2118.0 and 1120.1㎎% in the wild and cultivated sample, respectively. In the free amino acid of horned rampion, the total amount ranged from 8.5 to 50.1㎎%, which were lower level than that of composition amino acid. But the number of free amino acid was 29 kinds, which was bigger in its number than of composition amino acid detected 17 kinds.

      • 체육교육에서 피그말리온 효과

        A 1948 paper by Robert Merton established that certain social events were, to a large extent, influenced by the expectancy effects in different social settings. It wasn’t until 20 years later that Merton’s concept of the self-fulfilling prophecy came to the attention of educational researchers (Clark, 1963; Goldberg,1963). Since the Oak Elementary School experiment, researchers have conducted well over 100 studies relative to teacher expectations. It is also important to note that impressions about a certain student may not be the same for two or more teachers. Futhermore, after an extensive review of the literature, Brophy and Good(1974) concluded that teachers, irrespective of gender, are more negative toward young males, particularly regarding their potential as behavior problems. In a follow-up study(Martinek & Karper, 1984a), it was discovered that the attractiveness cue was more influential in expectancy formation for older age groups than for younger ones. These data suggested that as children get older, attractiveness becomes increasingly related to performance expectations.

      • 맥주맥 F_1세대의 조합능력 검정

        맥주맥의 이면교잡에 의한 F_1세대에 대한 조합능력을 검정한 결과를 요약하면 다음과 같다. 8개 형질에 대한 GCA, SCA, RCA효과를 검정한 바 조합능력은 전 형질에서 GCA, SCA, RCA가 모두 유의하였는데 간장, 수장, 망장, 간직경, 곡핍폭, 1수립수 1000립중은 GCA가 SCA보다 크고, 품종간 GCA효과는 진양보리가 간장에서 부(-)이고, 삼도보리가 수장·망장·간직경·1000립중에서, 사천6호가 망장과 간직경에서, 진광보리가 곡립장에서, 두산29호가 곡립폭·1수립수·1000립중에서 각각 정(+)으로 높았다. SCA효과에서 수장은 두산29호×두산8호 조합이, 망장은 진양보리×두산29호 조합이, 간직경은 사천6호×진광보리 조합이, 곡립장은 진양보리×두산8호 조합이, 곡립폭은 진광보리×두산8호 조합이, 1수립수는 진양보리×삼도보리 조합이, 1000립중은 사천6호×두산8호 조합 및 두산29호×두산8호 조합이 각각 정으로 높았고, 간장은 삼도보리×진광보리 조합이 부로서 높았다. RCA효과에서 수장은 두산8호×남향보리 조합이, 망장은 진양보리×삼도보리 조합이, 간직겨은 사천6호×진양보리 조합이, 곡립장은 사천6호×진광보리 조합이, 곡립폭은 진양보리×삼도보리 조합이, 1수립수는 사천6호×두산29호 조합이, 1000립중은 진양보리×두산29호 조합이 각각 정으로 높았고, 간장은 사천6호×두산29호 조합이 부로서 높았다. Seven barley varieties in F_1 generation of the possible crosses among them were used to estimate the combining ability for eight characters, i.e., culm length, spike length, awn length, culm diameter, grain length, grain width, number of grains per spike and 1000 grains weight. Data for F_1 hybrid and parents were subjected to the analysis method proposed by Griffing. The results obtained were summarized as follows. mean squares of general combining ability(GCA), specific combining ability(SCA) and reciprocal combining ability(RCA) were significant for all characters observed. Mean squares of GCA were higher than those of SCA and RCA in culm length, spike length, awn length, culm diameter, grain width, number of grains per spike and 1000 grains weight. Variety Dusan#29 showed the highest GCA effect for number of grains per spike and positively high effects of GCA were observed from variety Samdo bori in spike length, awn length, culm diameter, 1000 grains weight, and from variety Jinkwang bori I grain length, from Dusan#29 in grain width, umber of grains per spike, 1000 grains weight. Also Jinyang bori showed negatively high GCA effect in culm length. In SCA effects, hybrids in Jinyang bori × Samdo bori were exhibited positively high for number of grains per spike and high SCA effects for 1000 grins weight were found positively in Sacheun#6×Dusan#8 and Dusan#29×Dusan#. In RCA effects, hybrids in Sacheun#6×Dusan#29 were exhibited positively high for number of grains per spike and high RCA effects for 1000 grains weight were found positively in Jinyang bori×Dusan#29.

      • 갑상샘 유두암과 혼재하여 발견된 편평세포암 1예

        The occurrence of a mixed tumor containing papillary thyroid carcinoma (PTC) and primary squamous-cell carcinoma (SCC) is rare because there is no squamous epithelium in the thyroid gland. Reported herein is a 30-year-old female with mixed PTC and primary SCC of the thyroid presented as thyroid incidentaloma. Fine-needle aspiration biopsy of the thyroid nodule revealed the presence of malignant thyroid cells. The histopathological examination following total thyroidectomy yielded two mixed, morphologically distinct histotypes that included PTC and SCC. After total thyroidectomy, the patient underwent radioactive iodine therapy. No recurrence or metastasis occurred during the 20-month follow-up period after the operation.
