Background: Agranulocytosis is a rare but life-threatening side effect of antithyroid drugs. Several reports, including familial cases, suggested that there could be substantial genetic susceptibility determinants which could predict the risk of antithyroid drug induced agranulocytosis. However, no tests are currently available to predict the risk of antithyroid drug induced agranulocytosis. Methods: In this study, we performed whole-exome sequencing to comprehensively identify the genetic variations responsible for antithyroid drug induced agranulocytosis. For discovery stage 1, whole-exome sequencing was performed for seven antithyroid drug induced agranulocytosis cases. Variant based approach and gene based approach were performed to discover candidate variants or genes for antithyroid drug induced agranulocytosis. For HLA loci, HLA-sequence-based typing was performed. For validation stage 2, Sanger sequencing and HLA-sequence-based typing were performed for eight additional independent antithyroid drug induced agranulocytosis cases. Antithyroid drug users without agranulocytosis side effect, Korean local healthy controls and public HLA database were used as control group. Results: Discovery stage genome-wide association study results revealed strong signals in chromosome 6 (P=2.7×10-8) and chromosome 19 (P=1.7×10-14). HLA typing of seven discovery cases and eight validation cases revealed several HLA types, which showed strong association with antithyroid drug induced agranulocytosis (odds ratio=4.6-6.3, P=3×10-3-5×10-5). Gene based approach indicated several candidate genes for antithyroid drug induced agranulocytosis. In these genes, several loss of function variants were found, which were common among antithyroid drug induced agranulocytosis cases, but these variants were very rare among controls (P=2.0×10-3-1.8×10-4). Conclusions: In conclusion, several strong genetic predictors of antithyroid drug induced agranulocytosis were discovered. To avoid the risk of life-threatening agranulocytosis side effect, these pharmacogenomics markers could be tested in clinic before initiation of antithyroid drugs.
ORF Miner: a Web-based ORF Search Tool

The primary clue for locating protein-coding regions is the open reading frame and the determination of ORFs (Open Reading Frames) is the first step toward the gene prediction, especially for prokaryotes. In this respect, we have developed a web-based ORF search tool called ORF Miner. The ORF Miner is a graphical analysis utility which determines all possible open reading frames of a selectable minimum size in an input sequence. This tool identifies all open reading frames using alternative genetic codes as well as the standard one and reports a list of ORFs with corresponding deduced amino acid sequences. The ORF Miner can be employed for sequence annotation and give a crucial clue to determination of actual protein-coding regions.

Purpose: Nuclear Factor-κB p65 (NF-κB p65) and nuclear Factor-κB1 p50 (NF-κB p65) have been shown to play roles in cell proliferation, apoptosis, cytokine production and oncogenesis. This study was designed to investigate the expressions of NF-κB p65 and NF-κB p50 proteins in premalignant lesions and colorectal adenocarcinoma. Methods: Paraffin sections of 20 normal mucosa specimens, 20 low grade tubular adenoma specimens, 20 high grade tubular adenoma specimens and 64 adenocarcinoma specimens were analyzed immunohistochemically for the expressions of NF-κB p65 and NF-κB p50 proteins. Results: The expressions of NF-κB p65 and NF-κB p50 proteins were significantly higher in the adenocarcinoma tissue compared with that in the normal mucosa, the low grade tubular adenoma and the high grade tubular adenoma tissues. The frequency of a NF-κB p50 expression was higher in the poorly differentiated histologic grade specimens, in the presence of nodal metastasis and in the high stage specimens. There were significant correlations between the NF-κB p65 and NF-κB p50 proteins. Conclusion: The expressions of NF-κB p65 and NF-κB p50 proteins may play a role in the pathogenesis of colorectal carcinoma.
Microwave Sealing of Alumina Substrate with Sealing Glass

재래식과 마이크로파 열원으로 알루미나 기판을 PbO-ZnO-$B_2O_3$계 유리로 봉착하였을 때, 봉착용 유리의 결정화 거동 및 알루미나 기판과의 봉착상태를 조사하였다. 재래식 열처리된 시편에 비하여 마이크로파 열처리된 시편은 짧은 시간과 낮은 온도에서 열처리 되었음에도 불구하고 모유리 내에서 $\textrm{PbTiO}_3$ 결정이 잘 성장하였고, 높은 결정화를 보여주었다. 또한, 마이크로파 봉착 시편은 양호한 상태를 보여주었고, 봉착 곡강도는 거의 비슷하였다. Alumina substrate was sealed by PbO-ZnO-$B_2O_3$glass using microwave heating. The crystallization behaviour of sealing glass and the state of sealing joint were investigated and were compared with those of conventionally sealed one. Compared to the conventional one, the microwave heat-treated sample had well-grown $\textrm{PbTiO}_3$ crystals and high degree of crystallinity even though it had shorter heat-treatment time and lower heat-treatment temperature. Also, the microwave sealed sample exhibited relatively a good sealing state and almost same bending strength.
동종 신장이식후 Erythropoietin (EPO) 및 혈색소의 변화 양상

연구배경 : 저산소 자극에 따라 신장의 peritubular interstitial cell에서 만들어지는 당단백인 EPO로 인해 정상적으로 Hematocrit와 EPO는 negative feedback 관계에 따라 미세하게 조절된다. 그러나 만성 신부전환자에서는 이런 관계가 깨어지고 빈혈이 생기게 된다. 만성 신부전환자의 빈혈의 원인으로 요독성 조혈 억제인자, 용혈, 혈액손실 등을 꼽고 있으나, 그 중에서 빈혈에 비해 EPO의 상대적 생성부족이 가장 중요한 원인으로 알려져 있다. 지금까지 신장이식이후 EPO의 변화를 추적하는 여러 연구가 사체신 공여 신장이식자를 대상으로 있었지만 저자들은 cold ischemic time을 줄일 수 있는 생체신 공여 신장이식환자를 대상으로 신기능이 정상이 되면서 내인성 EPO의 수치와 혈색소의 변화를 알아보고자 본 연구를 시작하였다. 방법 : 1991년 12월 18일부터 1992년 3월 20일까지 연세대학교 의과대학부속 세브란스병원에서 생체신으로 동종 신장이식을 받은 말기 신부전환자 18명을 대상으로 수술전 7일부터 수술후 28일까지 BUN, creatinine, hemoglobin, hematocrit, reticulocyte count, serum EPO level, serum iron, TIBC 등을 전향적으로 일정한 간격으로 검사하여 분석하였다. 결과 : 1) 수술후 신장기능이 정상으로 돌아오면서 EPO수치는, 수술전 12.3±20.3U/L로 의의있게 증가하였다(p<0.05). 2) 수술후 EPO의 증가는 수술후 1일부터 3일까지에 peak을 보여주었고, 그 이후에는 약간 감소하는 경향을 보였지만 수술전보다는 의의있게 높은 수치로 계속 유지 되었다(p<0.05). 3) 수술후 EPO의 증가에 따른 망상구수는 수술전 0.5±0.3%에서 수술후 5일째 1.7±1.1%로 의의있게 증가하였으며(p<0.05), EPO의 peak 보다 4일후에 증가하여 계속 높게 유지되었다. 4) 혈소판, serum iron, TIBC, 그리고 혈청 ferritin 등은 수술전과 비교하여 수수후에도 의의있는 변화를 보이지 않았다. 결론 : EPO의 late peak을 보이지 않으면서 오히려 early peak 후에도 EPO가 지속적으로 의의있게 높이 유지되었는데, 생체신 동종이식을 통해 이식신 기능이 조기에 나타남으로 인하여 요독성 물질들이 조기에 제거 되어 late peak을 보이지 않더라도 골수에서 EPO의 자극에 의해 EPO의 peak에서 4일 후부터 망상구수의 증가가 보이는 것으로 생각된다. Background : In chronic renal failure, anemia develops due to uremic inhibitor, hemolysis and blood loss but mainly due to relative deficiency of Erythropoietin (EPO) compared to the level of anemia. Many studies about the changes of EPO after renal allograft have been done on the cadaveric-donor kidney recipient by other investigators but few studies were reported on living-donor renal allograft so far. We studied the changes of EPO and hemoglobin after living-donor kidney allograft with less ischemic time than cadaveric-donor renal allograft. Method : We studied prospectively the serial changes of BUN, serum creatinine, hemoglobin, hematocrit, reticulocyte count, serum EPO level, serum iron and TIBC with regular intervals from preop. 7th day to postop. 28th day on the 18 patients who had done living-donor renal allograft at Severance hospital from 18th Dec., 1991 to 20th Mar., 1992. Result : 1) As renal function became to normal after renal allogrft, serum EPO concentration increased significantly from 12.3±20.6 U/L on the preop. 1st day to 24.2±20.6 U/L on the postop 1st day and 25.2±16.6 U/L on the postop. 3rd day (p<0.05). 2) The peak serum EPO concentration was noted between postop. 1st day and 3rd day, and thereafter serum EPO concentration decreased slightly but maintained to be higher than preop. level significantly (p<0.05). 3) Reticulocyte count increased significantly from preop. 0.05±0.3% to postop. 5th day, 1.7±1.1% (p<0.05) which was 4 days later than the peak of EPO. 4) Platelet, serum iron, TIBC, and serum ferritin levels were not changed after renal transplantation. Conclusion : Although serum EPO concentration did not show the late peak which was usually reported in cadaveric-donor renal transplantation, we noted th serum EPO level maintained to be significantly high level with early peak after living-donor renal transplantation. But reticulocyte count increased significantly 4 days later than the peak of serum EPO level suggesting that the intial peak of EPO level came from renal ischemia during renal transplantation and the persisting high but slightly downed EPO level was from well-functioning renal allograft.
진공주조법에 의한 TiNi 형상기억합금 강화 6061Al 지적 복합재료의 계면 및 인장 특성

We investigated the change of mechanical properties for TiNi shape memory alloy by heat treatment. 6061Al matrix composites with TiNi shape memory alloy as reinforcement were fabricated by vacuum casting. TiNi alloy has the maximum tensile strength at 673K treated and there is no change of tensile strength and hardness at 448K treated. The composites, prepared by vacuum casting, showed good interface bonding by vacuum casting. It was about 3$\mu\textrm{m}$ of thickness of the diffusion layer. Tensile strength of the composite was in higher than that of 6061Al alloy as increased value of about 70MPa at room temperature and about 110MPa at 363K. We thought that the increase of the tensile strength at 363K was due to reverse transformation of the TiNi shape memory alloy.
금형주조법에 의한 TiNi/6061Al 복합재료의 미세조직에 미치는 냉간가공도의 영향

The 2.5 vol% TiNi/6061Al composites were fabricated by permanent mold casting. The microstructures and tensile test for the cold rolled composites with maximum 50% reduction ratio were investigated. In the case of TiNi fiber with 2mm interval in preform, the interface bonding of fabricated composites were good, interface diffusion layer of this composites was made by the mutual diffusion. Transverse section of TiNi fiber was decreased with increasing reduction ratio and longitudinal section of TiNi fiber showed multiple wave phenomenon. And the tensile strength of composites at 38% reduction ratio was the most high. In the case of over 38% reduction ratio, the decrease of the tensile strength was due to TiNi fiber rupture by excess working. The fracture mode was appeared brittle fracture with increasing reduction ratio.
