Estimating the Wage elasticity of off-farm Labour supply of farmers in Kemubu, Kelantan

Off-farm labour supply is very important in the transformation of agricultural economies to industrial economies. Two econometric models were used to estimate the wage elasticity of off-farm labour supply in Kemubu, Kelantan, Malaysia. A wage elasticity of approximately 2.0 was obtained from both models. The study revealed that severe poverty exist in single-cropped areas outside Kemubu. Farm households are more pressed for time with off-farm employment in double-cropped areas inside Kemubu. The twin-strategies of intensifying paddy cultivation and creating off-farm employment opportunities can be carried out concurrently. To induce farmers to seek off-farm employment to augment farm incomes, off-farm employment opportunities should be created close to farm households and development policies should aim at improving the level of educational and vocational skills of farmers.

Validation of ALK/ROS1 Dual Break Apart FISH Probe probe in non-small-cell lung cancer

Background: ALK and ROS1 gene rearrangements are distinct molecular subsets of non-small-cell lung cancer (NSCLC), and they are strong predictive biomarkers of response to ALK/ROS1 inhibitors, such as crizotinib. Thus, it is clinically important to develop an effective screening strategy to detect patients who will benefit from such treatment. In this study, we aimed to validate analytical performance of Vysis ALK/ROS1 Dual Break Apart Probe Kit (RUO) in NSCLC. Methods: Study population composed of three patient cohorts with histologically confirmed lung adenocarcinoma (patients with ALK rearrangement, patients with ROS1 rearrangement and patients with wild-type ALK and ROS1). Specimens consisted of 12 ALK-positive, 8 ROS1-positive and 21 ALK/ROS1-wild type formalin-fixed paraffin-embedded samples obtained from surgical resection or excisional biopsy. ALK rearrangement was previously assessed by Vysis ALK Break Apart FISH Probe Kit (Abbott Molecular, Abbot Park, IL, USA) and ROS1 rearrangement was previously assessed by ZytoLight<SUP>®</SUP> SPEC ROS1 Break Apart Probe (ZytoVision, GmbH). All specimens were re-evaluated by Vysis ALK/ROS1 Dual Break Apart Probe Kit. FISH images were scanned on BioView AllegroPlus system and interpreted via BioView SoloWeb remotely. Results: For a total of 41 patient samples, the concordance of the results by Vysis ALK/ROS1 Dual Break Apart Probe Kit was evaluated and compared to the known ALK and ROS1 rearrangement status of the specimen. Of the 12 ALK-positive cases, hybridization with Vysis ALK/ROS1 Dual Break Apart Probe Kit was successful in 10 cases (success rate 10/12, 83%) and of these 10 cases, all showed ALK rearrangement (100% concordance with the results of Vysis ALK Break Apart FISH Probe Kit). Two of the ALK+ cases were excluded due to weak ROS1 signals that could not be enumerated. Of the 8 ROS1-positive cases, 6 cases were successfully evaluated using Vysis ALK/ROS1 Dual Break Apart Probe Kit. The success rate was 75% (6/8), and of these 6 cases, all showed ROS1 rearrangement, giving a 100% concordance with ZytoLight<SUP>®</SUP> SPEC ROS1 Break Apart Probe. Two of the cases were excluded due to weak ROS1 gold signal or high background. In the cohort of 21 wild-type cases, the success rate using Vysis ALK/ROS1 Dual Break Apart FISH Probe Kit was 85% (18/21) and the concordance with ALK and ROS1 probe kit was 100% (18/18). Conclusion: Vysis ALK/ROS1 Dual Break Apart Probe Kit (RUO) can detect ALK and ROS1 rearrangement simultaneously in NSCLC.

In this study, improvements on up-to-date actuator surface method are proposed to enhance the prediction capability and universality for rotor aerodynamic analysis. In order to improve accuracy for analysis of high-solidity rotor, the correction equation for reference line velocity was newly derived to take into account influences of neighboring blades. To describe more realistic flow field and wake around blades, source-doublet panel method was adopted to calculate the momentum source distribution along chord-wise direction which reflects the characteristics of sectional airfoil and flow condition. The validity of proposed methods is evaluated by carrying out the analyses for rectangular wing and open rotor, and comparing with available experimental and computational results. It is confirmed that suggested methods can provide improvement on prediction of velocity field and wake trajectory, as well as aerodynamic performance.

THE SUSCEPTIBILITY OF SCALELESS MUTANT CHICKENS TO VERY VIRULENT MAREK'S DISEASE VIRUS

This study evaluates the susceptibility of scaleless mutant chickens to very virulent Marek's disease virus (vvMDV) inoculation. One day old chickens were inoculated subcutaneously with Taiwanese isolates of LTB-1 and LTS-1 strains, and standard strain of Md/5. Compared with the non-inoculated group the vvMDV-inoculated chickens showed decreased body weights and atrophy of lymphoid organs before 35 days old. These results indicate that scaleless chickens show the same susceptibility as the wild type chickens to vvMDV infection. Furthermore, the protective effect of herpesvirus of turkey (HVT) vaccination at 1 day old against vvMDV challenge was evaluated. Scaleless mutant chickens of treated groups showed 20-30% early death, and 85.7-100% and 12.5-14.2% had lymphomatous lesions in visceral organs and peripheral nerves, respectively. No significant lesions were observed in non-challenged chickens of the control group. The HVT vaccination did not provide an effective protection against vvMDV infection. It is concluded that scaleless mutant chickens are susceptible to vvMDV infection.

Applications of Laser-Fabricated Plasma Structures in Laser-Wakefield Accelerators, X-ray Lasers and Plasma Nonlinear Optics

A general method for fabricating transient plasma structures with high-intensity laser pulses is developed to gain fine control over laser-plasma interactions. These structures have been used as programmable photonic devices in the development of laser-wakefield accelerators, soft X-ray lasers and plasma nonlinear optics driven by multi-terawatt laser pulses. Plasma ramps are used to control electron injection in laser-wakefield accelerators, plasma waveguides are used to enhance the efficiency of soft X-ray lasers by orders of magnitude and periodic plasma structures are used to achieve quasi-phase matching in relativistic harmonic generation. By scanning the interaction length with the same plasma-fabrication method, tomographic measurements are carried out to resolve the injection/acceleration process in laser-wakefield accelerators and amplification processes in X-ray lasers and relativistic harmonic generation. A theoretical analysis and a computer simulation are also carried out to provide insightful pictures of these processes. These research works show that by controlling plasma structures with optical fabrication methods, laser-plasma interaction can be engineered to expand and enrich the frontier of high-field physics.

Chlamydia trachomatis의 集團檢査 方法에 貫한 硏究 : 96Well microplate를 利用하여

Chlamydia trachomatis is the most common cause of nongonococcal urethritis in the world. This is detected by several methods: direct staining methods (Iodine, Giemsa, Direct immunofluorescence stain), serologic methods (ELISA, indirect immunofluorescence), Culture methods (Yolk sac, McCoy cell, HeLa cell, Lister cell culture). The standard method in isolation of Chlamydia trachomatis is the vial culture using monolayer of McCoy cells, but this method is not proper to mass screening for public health because this method was to time consuming and no economic. So we have tried to develop the massive sereening culture method using 96 well microplate in Korea. Clinical specimens were collected from urethral mucosa of 142 male health students who were volunteer in this study. This specimens were inoculated into cycloheximide-treated McCoy cell monolayer in 96 well microplate and incubated in the CO₂inoculator (37℃) for 48 and 72 hours. We used iodine stain for detection of Chlamydia trachomatis. The summaried results were following. 1. The positive rates of Chlamydia trachomatis in the 48 hour and 72 hours culture were 7.7% and 7.0% in respective and totally positive rate of 48 hours or 72 hours were 10.6% 2. The positive rates of Chlamydia trachomatis according to sample storage were decreased. Ⅰ: within 7 days (15.8%) Ⅱ: 9-14 days (9.9%) Ⅲ: 15-21 days (14.2%) Ⅳ: 22-28(6.6%) Ⅴ: more 29 days (6.3%) In summarized our study results, the reserve rate of Chlamydia trachomatis in the health male (students) were very high (10.6%) and the culture method using 96 well microplate was very easy and economic in Korea.

Relationship between the abdominal sagging index and the reproductive performance of the Roman goose

Objective: This research aimed to explore the changes in the observed abdominal sagging index (ASI) and reproductive performance of Roman male and female geese during the breeding period. Methods: The 339 six-month-old breeding geese (109 male; 230 female) were used in this study, in which five male and five female geese were slaughtered on a monthly basis to record the ASI. Results: The short diameter of the testes of the male goose when the female goose lays eggs and in the second, third, and fourth months was significantly wider than in the fifth months (19.0, 20.8, 21.4, and 19.6 vs 12.7 and 14.0 mm/bird; p = 0.0105). On the other hand, the testicular weight of the male goose in the second and third months after the female goose lays eggs was significantly higher than that in the second and fifth months after laying (0.33% and 0.37% vs 0.11% and 0.19%; p = 0.0212). During the exploring period, the length and weight of the fallopian tube, the weight of the ovary, the number of follicles in 2 to 3 cm, the number of follicles in 3 to 4 cm, the fallopian tube weight in the carcass weight percentage, and the ovary weight in the carcass weight percentage all demonstrated a significant curve response. Further, female ASI was positively correlated with reproductive tract length (r = 0.815; p<0.05) and egg production per female (r = 0.790; p<0.05). Conclusion: The ASI classification method is more objective and easy to distinguish. This scoring method has a high correlation with the number of eggs laid by each goose and the length of the reproductive tract, inferring that the goose observation could take advantage of ASI during egg-laying and can predict the reproductive system development during the laying period and determine when the breeding goose begins to lay eggs.

The Release of Hepatic Triglyceride Lipase from Rat Monolayered Hepatocytes in Primary Culture

쥐간세포 배양시 간트리글리세리드 lipase의 유리 및 호르몬 조절에 관하여 연구하였다. 배양 2일째 헤파린 첨가구 배양액에 유리된 lipase 활성은 24시간 동안 계속 증가하였다. 반면에 헤파린 무첨가의 lipase활성은 헤파린 첨가가우에 비하여 10%에 지나지 않았다. 간세포를 anti-hepatic triglyceride lipase IgG와 배양시 lipase 활성이 92%까지 저해되었다. Monensin 첨가시 lipase활성 저해는 61%였다. 인슐린은 lipase활성을 20% 상승시켰으며 dexamethasone은 44% 저해시켰다. 이상의 결과로 미루어 보아 간트리글리세리드 lipase는 헤파린 존재하에 분비 및 유리되며 그 분비는 호르몬에 의해 조절됨을 시사한다. The release of hepatic triglyceride lipase from cultured rat hepatocytes and its hormonal regulation were studied. The activity of lipase released into the medium in the presence of heparin was increasing during 24 hours on the 2nd day of culture, while this was 10% in the absence of heparin as compared with the lipase activity in the presense of heparin. When hepatocytes were cultured with anti-hepatic triglyceride lipase IgG, the lipase activity was suppressed by 92%. The results suggest that the enzyme released into culture medium is identical to hepatic triglyceride lipase which can be released only in the presence of heparin, the model of release being similar to that of lipoprotein lipase from adipocytes. The addition of monensin to the medium resulted in the inhibition of lipase secretion by 61%. Insulin enhanced lipase activity only 20%, whereas dexamethasone suppressed the activity by 44%. These data indicated that hepatic triglyceride lipase is secreted and released from hepatocytes in the presence of heparin and its secretion is regulated by hormones.
